Resistance of the New Varieties Milyang $\#21\;and\;\#23$ to Plant-and Leaf-hoppers (멸구$\cdot$매미충에 대한 수도 신품종 밀양 21 및 23호의 저항성)

  • Laboratory experiments were conducted to evaluate the resistance of the new rice varieties Milyang $\#$21 and $\#$23 at the seedling stage to the brown planthopper(Nilaparvata lugens), small brown planthopper(Laodelphax striatellus), white-back planthopper(Sogatella furcifera), green rorice leafhopper(Nephotettix cincticeps) and zigzag-striped leafhopper (Recilia dorsalis) The varieties Yushin and T(N)-1 were used as susceptible check and the resistant check varieties were Mudgo to brown planthopper, ASD-7 :o small brown planthopper, Colombo to white-back planthopper, IR2061 (46763) to green rice leafhopper and Vellailanalgayan to zigzag-striped leafhopper. The varieties Milyang $\#$21 and $\#$23 were moderate in plant reaction only to the green rice leafhopper and $\#$23 was moderately resistant in plant rection only to the small brown planthopper. The nymphs of plant-and leaf-hoppers were more prefered Milyang $\#$21 and $\#$23 and susceptible check-varieties were more preferred for feeding than the resistant check-varieties. The green rice leafhopper and aigzag·striped leafhopper much more prefered Milyang $\#$21 and $\#$23 for oviposition, while brown planthopper and small brown planthopper more prefered the resistant cheeks than test varieties and susceptible checks. However, there was no any steady relationship in ovipositional preference between resistant and susceptible to the hoppers. Shorter nymphal-periods and higher rate of adult emergence were observed in the test varieties and the susceptible checks compared with the resistant checks. In conclusion, the varieties Milyang $\#$21 and $\#$23 seemed to be lack of true resistance to the plant-and leaf-hoppers from the viewpoints of antibiosis and feeding preference.

A Co-inhibitory Molecule, B7-H4, Synergistically Potentiates Oral Tolerance by Inducing CD4+CD25+FoxP3+ T Cells

  • Background: A co-inhibitory molecule, B7-H4, is believed to negatively regulate T cell immunity by suppressing T cell proliferation and inhibiting cytokine production. However, the mechanism behind B7-H4-mediated tolerance remains unclear. Methods: Balb/c $(H-2^d)$ mice were fed with dendritic cell line, DC2.4 $(H-2^d)$ every day for 10 days. Meantime, mice were hydrodynamically injected with recombinant plasmid expressing B7-H4 fusion protein (B7-H4.hFc) or hFc via tail vein. One day after last feeding, mice were immunized with allogeneic B6 spleen cells. 14 days following immunization, mice were challenged with B6 spleen cells to ear back and the ear swelling was determined the next day. Subsequently, a mixed lymphocyte reaction (MLR) was also performed and cytokines profiles from the reaction were examined by sandwich ELISA. Frequency of immunosuppressive cell population was assayed with flow cytometry and mRNA for FoxP3 was determined by RT-PCR. Results: Tolerant mice given plasmid expressing B7-H4.hFc showed a significant reduction in ear swelling compared to control mice. In addition, T cells from mice given B7-H4.hFc plasmid revealed a significant hyporesponsiveness of T cells against allogeneic spleen cells and showed a significant decrease in Th1 and Th2 cytokines such as IFN-${\gamma}$, IL-5, and TNF-${\alpha}$. Interestingly, flow cytometric analysis showed that the frequency of CD4+CD25+FoxP3+ Tregs in spleen was increased in tolerant mice given recombinant B7-H4.hFc plasmid compared to control group. Conclusion: Our results demonstrate that B7-H4 synergistically potentiates oral tolerance induced by allogeneic cells by increasing the frequency of FoxP3+ CD4+CD25+ Treg and reducing Th1 and Th2 cytokine production.

EVALUATION OF CONDYLAR DISPLACEMENT USING COMPUTER TOMOGRAPHY AFTER THE SURGICAL CORRECTION OF MANDIBULAR PROGNATHISM (전산화단층촬영법을 이용한 하악전돌증 환자의 외과적 악교정술후 하악과두 위치 변화 검토)

  • This study was intended to perform the influence of condyle positional change after surgical correction of skeletal Class III malocclusion after orthognathic surgery in 37 patients(male 13, female 24) using computed tomogram that were taken in centric occlusion before, immediate after, and long term after surgery and lateral cephalogram that were taken in centric occlusion before, 7 days within the period of intermaxillary fixation, at the 24 hours later removing intermaxillary fixation and long term after surgery. 1. Mean intercondylar distance was $84.42{\pm}5.30mm$ and horizontal long axis of condylar angle was $12.79{\pm}4.92^{\circ}$ on the right, $13.53{\pm}5.56^{\circ}$ on the left side. Condylar lateral poles were located about 12mm and medial poles about 7mm away from the reference line(AA') on the axial tomogram. Mean intercondylar distance was $83.15{\pm}4.62mm$ and vertical axis angle of condylar angle was $76.28{\pm}428^{\circ}$ on the right, $78.30{\pm}3.79^{\circ}$ on the left. 2. In amount of set back, We found the condylar change(T2C-T1C) which had increasing tendency in group III (amount of setback : 10-15mm). but there was no statistical significance(p>0.05). 3. There was some correlation between condylar change(T2C-T1C) and TMJ dysfunction. It seemed that postoperative condylar change had influenced postoperative TMJ dysfunction, through there was no statistical significance (p>0.05). As we have observed the change of condylar axis in the group that complained of TMJ dysfunction in cases of large amount of mandibular setback. So we consider that the more trying to conserve condylar position will decrease occurrence rate of post operational TMJ dysfunction.

  • Many myocutaneous flaps have been used for the reconstruction of intraoral defects caused by the excision of oral cancer. Among these myocutaneous flaps, cervical island flap has been introduced by Farr et al. Although different in detail, this flap was designed as the platysma myocutaneous flap by Futrell et al in the supraclavicular site. Since many authors applied this flap to cover intraoral defect, they discussed deeply the blood supply of this flap. To improve further flap survival, it was modified by Tashiro et al. This flap makes its vascularity highly reliable. The amount of tissue needed for reconstruction can be accurately planned. The surgical and reconstruction procedure can be performed simply, rapidly, and effectively. Oral functions including deglutition, speech, and denture fitting are not compromised. With it's minimal deformity, new donor fields is not necessory. Of course, we keep in mind that this flap has limitations in patients where much bulk of tissue defects is needed and more than 3000 rad radiation due to the metastasis of neck lymph node is exposed. In three patients with intraoral squamous cell carcinoma($T_{1-3}N_0M_0$), we performed induction chemotherapy with FP regimen including pepleomycin. Thereafter, we ablated oral cancer and peformed reconstruction of intraoral defects with cervical island flap designed by Tashiro et al. Due to these significant benefits and minimal limitations, we have found that this flap is adequate for reconstruction of most intraoral defects following cancer ablation.

Effects of Thermal and Mechanical Fatigue Stress on Bond Strength in Bracket Base Configurations (열적, 기계적 피로응력이 교정용 브라켓의 결합강도에 미치는 영향)

  • The purpose of this study is to evaluate the effects of mechanical and thermal fatigue stress on the shear, tensile and shear-tensile combined bond strengths(SBS, TBS, CBS) in various orthodontic brackets bonded to human premolars with chemically cured adhesive(Ortho-one, Bisco, USA). Five types of commercially available metal brackets with various bracket base configurations of Photoetched base(Tomy, Japan), Non-Etched Foil Mesh base(Dentaurum, Germany), Micro-Etched Foil Mesh base(Ortho Organizers, USA), Chessboard base(Daesung, Korea), and Integral base(3M Unitek, USA) were used. Samples were divided into 3 groups, the first group was acted with shear-tensile combined loads($45^{\circ}$) of 200g for 4 weeks(mechanical fatigue stress), the second group was subjected to the 5,000 thermocycles of 15 second dwell time each in $5^{\circ}C\;and\;55^{\circ}C$ baths(thermal fatigue stress), and the third group was the control. Bond strengths were measured at the crosshead speed of 0.5mm/min. The cross-section of bracket base/adhesive interface and the fracture surface were examined with the stereoscope and the scanning electron microscope. The resin remnant on bracket base surface was assessed by ART(Adhesive Remnant Index). The obtained results were summarized as follows, 1. In static bond strength, Photoetched base bracket showed the maximum bond strength and Integral base bracket showed the minimum bond strength(p<0.05). In all brackets, shear bond strength(SBS) was in the greatest value and shear-tensile combined strength(CBS) was in the least value(p<0.05). 2. After mechanical fatigue test, Photoetched base bracket showed the maximum bond strength and Integral base bracket showed the minimum bond strength(p<0.05).   • Vitamin D is known to exert its action by activating DNA and RBA within target cells to produce proteins and enzymes that can be used in bone resorption process. Particularly, the active form of vitmain D, 1,25-dihydroxycholecalciferol $[1,25-(OH)_2D_3]$, is considered to be one of the most potent stimulators of osteoclatic acitivity in vitro. The purpose of this study was to evaluate the effect of 1,25-Dihydroxyvitamin $D_3$ on the avtivity of periodotal ligament cells and, the experimental tooth movement. Human periodontal ligament cells were collected from the first premolar tooth extracted for the orthodontic treatment, and were incubated in the environment of $37^{\circ}C$, 5% $CO_2$ and 95% humidity. Microtitration(MIT) assay was done at 10, 25, 50 and 100ng/ml of 1,25-Dihydroxyvitamin $D_3$. 21 Sprague-Daft rats were divided into a control gmup(3), and experimental groups(18) where 100g of force from helical spring was applied across the maxillary incisors 1,25-Dihydroxyvitamin $D_3$ was injected into periodontal ligament at the mesial or distal surface of maxillary incisors so that we can compare the control side and the experimental side. Expreimental groups were sac rifled at 12, 24, 36, 48, 72hours and 7 days after force application, respectively. And the obtained tissues were evaluated histologically. The observed results were as follows. 1. The activity of periodontal ligament cells in l0ng/ml or 25ng/ml of 1,25-Dihydroxyvitamin $D_3$ 1,25-Dihydroxyvitamin $D_3$ was not significantly different to the control at the cultivation of 1, 2 and 3 days. 2. The activity of periodontal ligament cells was significantly increased at 3 days in 50 ng/ml of 1,25-Dihydroxyvitamin $D_3$ and 2, 3 days in 100g/ml of 1,25-Dihydroxyvitamin $D_3$. 3. Up to 7 days after force application, there was no difference in osteoblastic activity, tearing of periodontal ligament and proliferation of capillary at tension side between 1,25-Dihydroxyvitamin $D_3$ injection side and the control side. 4. The osteoclastic activity and the resorption of alveolar bone was greater in 1,25-Dihydroxyvitamin $D_3$ injection side than the control side at 36 hours after force application.

Decomposition of Leaf Litter Containing Heavy Metals in the Andong Serpentine Area, Korea (안동 사문암지대의 중금속 함유 낙엽의 분해)

  • The present study attempts to compare the soil chemical characteristics and biological activities (i.e. microbial biomass and soil enzyme activities), and litter decomposition rate of Arundinella hirta and Miscanthus sinensis var. purpurascens) collected from serpentine and non-serpentine sites by litter bag techniques at serpentine and non-serpentine field experiment sites over a 9-month period. The serpentine soil showed higher pH and soil alkaliphosphatase activity, and lower soil dehydrogenase and urease activities than the non-serpentine soil. Microbial biomass-N at the serpentine soil was larger than the non-serpentine soil, although the microbial biomass-C and microbial biomass-N represented no significant difference between serpentine and non-serpentine soil. These results suggest that the larger microbial biomass-N caused the lower C/N in serpentine soil. At the end of the experiment, the litter samples of A. hirta and M. sinensis collected from serpentine soil revealed a 39.8% and 38.5% mass loss, and the litter sample from non-serpentine soil also showed a 41.1% and 41.7% mass loss at the serpentine site. On the other hand, at the non-serpentine site, 42.2%, 37.4%, and 46.8%, 44.8% were respectively shown. These results demonstrate that the litter decomposition rate is more intensely affected by the heavy metal content of leaf litter than soil contamination. Moreover, the litter collected from the serpentine soil had a lower C/N, whereas the litter decomposition rate was slower than the litter from the non-serpentine soil, because the heavy metal inhibition activities on the litter decomposition process were more conspicuous than the effect of litter qualities such as C/N ratio or lignin/N. The nutrient element content in the decomposing litter was gradually leached out, but heavy metals and Mg were accumulated in the decaying litter. This phenomenon was conspicuous at the serpentine site during the process of decomposition.

Physicochemical Quality Characteristics of Traditional Kamju Using Extruded Rice Collet Powder (압출 쌀 콜렛 분말을 사용한 전통 감주의 제조 및 이화학적 품질 특성)

  • This study was conducted to investigate the physicochemical quality properties and provide basic data for the activation of traditional Kamju of juice type product prepared by mixing malt and extruded rice collet powder. Malt extracts were prepared by extracting the mixture of malt and water at a weight ratio of 25:75 after soaking for 2 h at $45^{\circ}C$. Rice collet powder was prepared by adjusting the barrel temperature to $95^{\circ}C$, screw speed to $3.07{\times}g$, discharge port diameter to 7 mm and a raw material input to 50 kg/h, the powder was then ground to a particle size of 80 mesh. The physicochemical characteristics (pH, color, viscosity, reducing sugars, number of viable cells, free amino acids) and sensory evaluations were conducted at various time points during the saccharification and at different mixing ratios of the extruded rice collet powder to malt extract (5:95, 15:85, 25:75, 35:65, each at $55^{\circ}C$ for 9 h). As a result, with an increase in the proportion of the extruded rice collet powder and saccharification time, the physicochemical properties of traditional Kamju significantly improved (p<0.05). A mixing ratio of 35:65 rice collet powder to malt extract and a saccharification time of 9 h were found to be the most desirable conditions. However, based on the sensory evaluation, a mixing ratio of rice collet powder and malt extract of 25:75 and a saccharification time of 5 h resulted in the most preferable palatability of traditional Kamju (p<0.05). Therefore, the mixing ratio and saccharification time should be determined to provide a better choice with respect to the taste and economic aspects of traditional Kamju.

Suggestions for improving data quality assurance and spatial representativeness of Cheorwon AAOS data (철원 자동농업기상관측자료의 품질보증 및 대표성 향상을 위한 제언)

  • Providing high-quality meteorological observation data at sites that represent actual farming environments is essential for useful agrometeorological services. The Automated Agricultural Observing System (AAOS) of the Korean Meteorological Administration, however, has been deployed on lawns rather than actual farm land. In this study, we show the inaccuracies that arise in AAOS data by analyzing temporal and vertical variation and by comparing them with data recorded by the National Center for AgroMeteorology (NCAM) tower that is located at an actual farming site near the AAOS tower. The analyzed data were gathered in August and October (before and after harvest time, respectively). Observed air temperature and water vapor pressure were lower at AAOS than at NCAM tower before and after harvest time. Observed reflected shortwave radiation tended to be higher at AAOS than at NCAM tower. Soil variables showed bigger differences than meteorological observation variables. In August, observed soil temperature was lower at NCAM tower than at AAOS with smaller diurnal changes due to irrigation. The soil moisture observed at NCAM tower continuously maintained its saturation state, while the one at AAOS showed a decreasing trend, following an increase after rainfall. The trend changed in October. Observed soil temperature at NCAM showed similar daily means with higher diurnal changes than at AAOS. The soil moisture observed at NCAM was continuously higher, but both AAOS and NCAM showed similar trends. The above results indicate that the data gathered at the AAOS are inaccurate, and that ground surface cover and farming activities evoke considerable differences within the respective meteorological and soil environments. We propose to shift the equipment from lawn areas to actual farming sites such as rice paddies, farms and orchards, so that the gathered data are representative of the actual agrometeorological observations.

N- and P-doping of Transition Metal Dichalcogenide (TMD) using Artificially Designed DNA with Lanthanide and Metal Ions

  • Transition metal dichalcogenides (TMDs) with a two-dimensional layered structure have been considered highly promising materials for next-generation flexible, wearable, stretchable and transparent devices due to their unique physical, electrical and optical properties. Recent studies on TMD devices have focused on developing a suitable doping technique because precise control of the threshold voltage ($V_{TH}$) and the number of tightly-bound trions are required to achieve high performance electronic and optoelectronic devices, respectively. In particular, it is critical to develop an ultra-low level doping technique for the proper design and optimization of TMD-based devices because high level doping (about $10^{12}cm^{-2}$) causes TMD to act as a near-metallic layer. However, it is difficult to apply an ion implantation technique to TMD materials due to crystal damage that occurs during the implantation process. Although safe doping techniques have recently been developed, most of the previous TMD doping techniques presented very high doping levels of ${\sim}10^{12}cm^{-2}$. Recently, low-level n- and p-doping of TMD materials was achieved using cesium carbonate ($Cs_2CO_3$), octadecyltrichlorosilane (OTS), and M-DNA, but further studies are needed to reduce the doping level down to an intrinsic level. Here, we propose a novel DNA-based doping method on $MoS_2$ and $WSe_2$ films, which enables ultra-low n- and p-doping control and allows for proper adjustments in device performance. This is achieved by selecting and/or combining different types of divalent metal and trivalent lanthanide (Ln) ions on DNA nanostructures. The available n-doping range (${\Delta}n$) on the $MoS_2$ by Ln-DNA (DNA functionalized by trivalent Ln ions) is between $6{\times}10^9cm^{-2}$ and $2.6{\times}10^{10}cm^{-2}$, which is even lower than that provided by pristine DNA (${\sim}6.4{\times}10^{10}cm^{-2}$). The p-doping change (${\Delta}p$) on $WSe_2$ by Ln-DNA is adjusted between $-1.0{\times}10^{10}cm^{-2}$ and $-2.4{\times}10^{10}cm^{-2}$. In the case of Co-DNA (DNA functionalized by both divalent metal and trivalent Ln ions) doping where $Eu^{3+}$ or $Gd^{3+}$ ions were incorporated, a light p-doping phenomenon is observed on $MoS_2$ and $WSe_2$ (respectively, negative ${\Delta}n$ below $-9{\times}10^9cm^{-2}$ and positive ${\Delta}p$ above $1.4{\times}10^{10}cm^{-2}$) because the added $Cu^{2+}$ ions probably reduce the strength of negative charges in Ln-DNA. However, a light n-doping phenomenon (positive ${\Delta}n$ above $10^{10}cm^{-2}$ and negative ${\Delta}p$ below $-1.1{\times}10^{10}cm^{-2}$) occurs in the TMD devices doped by Co-DNA with $Tb^{3+}$ or $Er^{3+}$ ions. A significant (factor of ~5) increase in field-effect mobility is also observed on the $MoS_2$ and $WSe_2$ devices, which are, respectively, doped by $Tb^{3+}$-based Co-DNA (n-doping) and $Gd^{3+}$-based Co-DNA (p-doping), due to the reduction of effective electron and hole barrier heights after the doping. In terms of optoelectronic device performance (photoresponsivity and detectivity), the $Tb^{3+}$ or $Er^{3+}$-Co-DNA (n-doping) and the $Eu^{3+}$ or $Gd^{3+}$-Co-DNA (p-doping) improve the $MoS_2$ and $WSe_2$ photodetectors, respectively.
