Transformation of Cell Wall-weakened Perilla Seedlings Using Phenolic Compound-treated Agrobacterium Cells and Recombinant Protein Expression (페놀화합물 처리 Agrobacterium 및 세포벽 약화 들깨새싹을 이용한 형질전환과 재조합 단백질 발현)

  • Perilla [Perilla frutescens (L.) Britt] seedlings are easy to grow and eaten as the health vegetable sprout. Two day old perilla seedlings since germination were given a mild wounding using cell wall lytic NaOH/SDS solution for infiltration with recombinant Agrobacterium cells treated with phenolic compounds. In the analysis of fluorometric GUS gene expression for the transformed perilla seedlings, GUS enzyme activity was the highest by the combined treatments of 50 mM acetosyringone and 0.5% NaOH solution containing 0.01% SDS implying a synergic effect. This result could be successfully applied for demonstrating hepatitis B virus antigen (HBsAg) protein expression.

A Case Report: Diagnostic Imaging of a Chondrosarcoma of the Rib in a Dog (개에서 늑골 연골육종에 대한 영상학적 진단증례)

  • An 11-year-old female, Golden retriever dog with a history of solid mass on the chest wall was referred to Veterinary Medical Teaching Hospital, Seoul National University. The mass was firm on palpation. A soft tissue opacity mass with calcified foci around the right 7th rib and extrapleural patterns around the right 5th rib and 7th rib was shown on thoracic radiographs. The mass of the 7th rib has a mixed-echo pattern with a strong acoustic shadowing and internal vascular signals on ultrasonography. On CT scan, the mass showed contrast enhancement effect, expansive pattern of intrathoracic and extrathoracic legion around costochondral junction. The 7th rib appeared mildly lytic. The mass of the 5th rib had a soft tissue swelling without bone lysis. The mass was diagnosed as a sarcoma by fine needle biopsy. Therefore, the mass was surgically removed. Subsequent histopathological study found the mass was chondrosarcoma.

Notch Signal Transduction Induces a Novel Profile of Kaposi's Sarcoma-Associated Herpesvirus Gene Expression

  • Kaposi's sarcoma-associated herpesvirus (KSHV) RTA transcription factor is recruited to its responsive elements through interaction with RBP-Jk that is a downstream transcription factor of the Notch signaling pathway that is important in development and cell fate determination. This suggests that KSHV RTA mimics cellular Notch signal transduction to activate viral lytic gene expression. Here, I demonstrated that unlike other B lymphoma cells, KSHV -infected primary effusion lymphoma BCBL1 cells displayed the constitutive activation of ligand-mediated Notch signal transduction, evidenced by the Jagged ligand expression and the complete proteolytic process of Notch receptor I. In order to investigate the effect of Notch signal transduction on KSHV gene expression, human Notch intracellular (hNIC) domain that constitutively activates RBP-Jk transcription factor activity was expressed in BCBL1 cells, TRExBCBL1-hNIC, in a tetracycline inducible manner. Gene expression profiling showed that like RTA, hNIC robustly induced expression of a number of viral genes including KS immune modulatory gene resulting in downregulation of MHC I and CD54 surface expression. Finally, the genetic analysis of KSHV genome demonstrated that the hNIC-mediated expression of KS during viral latency consequently conferred the downregulation of MHC I and CD54 surface expression. These results indicate that cellular. Notch signal transduction provides a novel expression profiling of KSHV immune deregulatory gene that consequently confers the escape of host immune surveillance during viral latency.

Effects of Juglans regia Complex Extract on Osteoclast Differentiation from Bone Marrow Derived Macrophage (호두복합추출물이 골수유래대식세포의 파골세포 분화에 미치는 효과)

  • The purpose of this study was to evaluate the inhibitory effects of Juglans regia complex extract(JCE) consisted of Juglans regia, Eucommia ulmoides, Eleutherococcus senticosus and Zingiber officinale on osteoclast differentiation. Cell toxicity test by using CCK-8, TRAP activity and TRAP positive multi-nucleated cell counting were performed to evaluate inhibitory effect on differentiation of osteoclast from bone marrow derived macrophages(BMMs) induced by receptor activator of nuclear $factor-{\kappa}B$ ligand(RANKL). As a result, JCE inhibited RANKL-induced osteoclast differentiation in BMMs dose-dependently without cytotoxicity. These results suggest that JCE may have a potential role for treating bone lytic diseases such as osteoporosis.

Scrophulariae Radix Aqueous Extracts Ameliorate the Pressure Overloaded Heart Failure by Transverse Aortic Constriction in Mice

  • Objectives: The purpose of this study was to investigate the cardioprotective effect of the potent antioxidant properties of Scrophulariae Radix (SR) aqueous extracts by examining pressure overload (PO) heart failure (HF) induced by transverse aortic constriction (TAC) of C57BL/6 mice. Methods: SR (500, 250, 125 mg/kg) and resveratrol (10 mg/kg) were administered orally once a day for 14 days, after the TAC operation. Changes in mortality, the body and heart weights, histopathology of the heart, and antioxidant defense system of the heart were analyzed. Results: After the TAC operation, increases were observed in mortality, heart weights, left ventricular hypertrophy, and lytic and focal fibrotic histological change, and destruction of the heart antioxidant defense system. However, the HF signs showed dose-dependent inhibition following 14 days of continuous oral treatment with SR. A SR dose of 125 mg/kg gave a similar inhibition to that obtained with resveratrol at 10 mg/kg. Conclusions: Oral administration of SR beneficially improves PO-induced HF following TAC surgery by increasing the activity of the heart antioxidant defense system. The overall effect of SR at 125 mg/kg is similar to the effect of resveratrol at 10 mg/kg. However, more detailed mechanistic studies should be performed by screening of the biologically active compounds in SR.

Biological Control of Diamondback Moth (Plutella xylostella L.) by Lysobacter antibioticus HS124 (Lysobacter antibioticus HS124를 이용한 배추좀나방 (Plutella xylostella L.)의 생물학적 방제)

  • Lysobacter antibioticus HS124 was isolated from rhizosphere soil in previous experiments, which produced lytic enzymes such as chitinase, gelatinase, lipase and protease. In addition, HS124 released an antibiotic compound, 4-hydroxyphenylacetic acid (4-HPAA). When larvae of P. xylostella was treated with HS124 culture broth, its body was destroyed, and degraded with the increase of incubation time, yielding glycine which was detected from HS124 culture broth. When 4-HPAA produced from HS124 was sprayed, larvae mortality increased with increasing concentration of 4-HPAA. When HS124 culture supplemented with Tween 80 was sprayed, its insecticidal activity against larvae was approximately 1.4 times higher compared to the culture without Tween 80. Insecticide (IS), HS124 culture broth (HS124), Magic-pi (MP) and HS124 culture broth+Magic-pi (HS124+MP) were each treated against larvae of P. xylostella to investigate their insecticidal effect where sterile diluted water (SDW) was used as a control. The highest mortality of larvae was found in HS124+MP, followed by IS, MP, HS124 and SDW respectively. Mortality of larvae in HS124 was 31% higher than that in SDW, but 41% lower than that in HS124+MP, meaning that both enzymes and antibiotics produced from HS124 may synergistically act as active agents with plant extract containing neem oil and turmeric in HS124+MP treatment. These results suggested that L. antibioticus HS124 together with plant extract can be one of candidates for biocontrol agents against Plutella xylostella.

Inhibition of Clostridium perfringens using Bacteriophages and Bacteriocin Producing Strains

  • In this study, we isolated and characterized a bacteriocin-producing strain and two bacteriophages (P4, A3), showing antimicrobial effects against Clostridium perfringens, from chicken and swine feces by the spot-on-the lawn antagonism method. The selected strain was identified as Streptococcus hyointestinalis by 16S rRNA gene sequencing. The bacteriocin from the isolated strain exhibited strong inhibitory activity against four strains of C. perfringens and all the tested strains of Listeria monocytogenes, and the bacteriocin were highly heat- and pH-stable even at pH 2, pH 10 and $121^{\circ}C$ for 15 min. We also evaluated the combined effects of the isolated bacteriocin and phages. Combining the phage treatments and bacteriocin resulted in a synergetic effect compared with the phage or the bacteriocin alone. In addition, during the probiotic test, the bacteriocin-producing S. hyointestinalis B19 strain reduced the population of C. perfringens significantly. Treatment with S. hyointestinalis B19 and a cocktail of lytic bacteriophages eradicated the C. perfringens KCTC $3269^T$, completely. Consequently, the isolated bacteriocin and bacteriophages represent candidates for effective biocontrol of C. perfringens, and bacteriocin-producing S. hyointestinalis B19 is a potential probiotic candidate for use in domestic animals.

The Hemolytic Characteristics of Amphotericin B-Containing Egg PC Liposomes (Amphotericin B가 함유된 Egg PC 리포솜의 용혈 특성)

  • The hemolytic characteristics of amphotericin B-containing liposomes have been investigated in vitro. From the hemolysis of human erythrocytes against free and liposomal amphotericin B, the marked reduction in the toxicity of amphotericin B was observed by incorporating the drug in egg PC liposomes. For 45 min, free amphotericin B at $9.6\;{\mu}g/ml$ could completely lyse 2 wt% human erythrocytes. However, liposomal amphotericin B had essentially no lytic effect even in the range over $9.6\;{\mu}g/ml$. In the 66 hr-hemolysis experiment, liposomal amphotericin B showed the slowly hemolysing chracteristics during the experimental period regardless of the concentration of amphotericin B but rapid hemolysis only for 12 hr was observed in the case of free amphotericin B and the degree of hemolysis for 12 hr was maintained after that time. Also the hemolysing ability of liposomal amphotericin B at $4\;{\mu}g/ml$ was lower than that of free amphotericin B at the same concentration for 66 hr. On the other hand, the dependence of hemolysis on amphotericin B contents in egg PC liposomes was significant between 1.64 mole% amphotericin B-containing liposomes and 15.79 or 27.27 mole% amphotericin B-containing liposomes. But no marked difference in hemolysis was observed between 15.79 and 27.27 mole% amphotericin B-containing liposomes. Especially, cholesterol as an excipient in amphotericin B-containing liposomes significantly reduced the hemolysis of human erythrocyte. The degree of hemolysis in 5 mole% amphotericin B-containing liposomes was reduced to approximately 50% of value in the cholesterol-free liposomes by adding 50% cholesterol.

Protective Effect of Lonicerae Flos Aqueous Extracts on a Pressure Overload-induced Heart Failure Model

  • Objectives: Lonicerae flos (LF), a dried flower part of Lonicera japonica Thunb., has been widely used in Korean medicine as anti-inflammatory and antioxidative agent. The purpose of this study was to determine the cardioprotective effects of LF, through potential antioxidant effects, on the pressure overload (PO)-induced heart failure (HF) in C57BL/6 mice after transverse aortic constriction (TAC) surgery. Methods: Resveratrol (10 mg/kg body weight) or LF (125, 250 or 500 mg/kg body weight) was orally administered, once daily for 14 days, starting 14 days after TAC surgery. Changes in the mortality, body weights, heart weights, histopathology of the heart, and antioxidant defense systems of the heart were analyzed. Results: Marked and noticeable increases of heart weights, mortalities, and hypertrophic, focal, and lytic fibrotic histological changes in the LVs were observed, with destruction of heart antioxidant defense systems after surgery. However, HF signs, induced by TAC surgery through PO, and destruction of heart antioxidant defense systems were significantly and dose-dependently inhibited by 14 days of maintained oral treatment with LF 500, 250 or 125 mg/kg. Treatment with 250 mg/kg LF was comparable to treatment with 10 mg/kg resveratrol. Conclusions: The results in this study suggest that oral administration of LF favorably relieves PO-induced HF following TAC, through increase of heart antioxidant defense systems. The overall effects of 250 mg/kg LF were similar to those of 10 mg/kg resveratrol. More detailed mechanistic studies should be conducted in the future, with screening of the biologically active compounds in LF.

Effects of Substitutions of Gln16 and Asp18 with Phe or Tyr in HP(2-20) on its Structure and Antimicrobial Activity

  • HP (2-20), a 19-residue peptide derived from the N-terminus of Helicobacter pylori Ribosomal Protein L1, has antimicrobial activity but is not cytotoxic to human erythrocytes. Previously, we have synthesized several analogue peptides to investigate the effects of substitutions on the structure and antimicrobial activity. Substitution of $Gln^{16}$ and $Asp^{18}$ with Trp (Anal 3) caused a dramatic increase in bacterial and fungal lytic activities. In this study, analogue peptides were synthesized to investigate the effects of substitution of Gin and Asp with Phe (Anal 6) or Tyr (Anal 7) in HP (2-20) on its structure and antimicrobial activity. Substitution of Gin and Asp with hydrophobic aromatic residues at position 16 and 18 of HP (2-20) caused increase in antibiotic activity without hemolytic effect. Substitution of Gin and Asp with Trp and Try increased antibiotic activity of HP (220) twice more compared to substitution with Phe. The tertiary structures of Anal 6 and Anal 7 in SDS micelles has been investigated using NMR spectroscopy. The structures revealed that substitutions of the aromatic residues at C-terminus resulted in longer and well defined alpha-helix and improved their antibacterial activities
