Tetrachloroauric Acid Depresses the Activation Processes of Phagocytic Cells

  • Gold compounds depress phagocytic cell responses, including chemotaxis, and respiratory burst. However, the effects of gold compounds on the function of phagocytic cells are variable according to the preparation of medicine. In this study, effect of tetrachloroauric acid on activated neutrophil responses, including respiratory burst, lysosomal enzyme release and change of intracellular $Ca^{2+}$ level and on the synthesis of interleukin-8 and granulocyte-macrophage colony stimulating factor by macrophages was studied. This study further examines how gold compounds affect the activation processes. The respiratory burst stimulated by complement C5a, degraded IgG and PMA in neutrophils was inhibited by tetrachloroauric acid. In contrast to C5a and degraded IgG, PMA-stimulated superoxide production was weakly inhibited by tetrachloroauric acid. Staurosporine, genistein, EGTA and verapamil inhibited superoxide and $H_2O_2$ production caused by C5a and degraded IgG. PMA-stimulated superoxide production was inhibited by staurosporine but was not affected by genistein. Tetrachloroauric acid, genistein, EGTA and verapamil inhibited the release of acid phosphatase and myeloperoxidase, while the effect of staurosporine was not detected. The synthesis of interleukin-8 and granulocyte-macrophage colony stimulating factor by $interleukin-1{\beta}$ in macrophages was inhibited by tetrachloroauric acid. Preincubation with tetrachloroauric acid, genistein, EGTA and verapamil, the elevation of [$Ca^{2+}_i$] evoked by C5a was inhibited. Store-regulated $Ca^{2+}$ entry in thapsigargin-pretreated neutrophils was decreased by the addition of tetrachloroauric acid and genistein. The effect of staurosporine on intracellular $Ca^{2+}$ mobilization was not observed. In conclusion, tetrachloroauric acid may suppress neutrophil responses through its inhibitory action on elevation of intracellular $Ca^{2+}$ level and protein kinase C. It might exhibit an inhibitory effect on the action of protein tyrosine kinase. Tetrachloroauric acid depresses cytokine production by macrophages.

Protein Kinase 억제제 첨가 후 Platelet-Activating Factor에 의하여 자극된 호중구반응의 변경 (Alteration of the Activated Responses in Platelet-Activating Factor-Stimulated Neutrophils by Protein Kinase Inhibitors)

  • Platelet-activating factor (PAF)에 의하여 자극된 호중구 respiratory burst, 탈과립과 세포질 칼슘농도의 증가에 있어 protein kinase C와 protein tyrosine kinase의 역할을 관찰하였다. PAF에 의하여 자극된 호중구에서 superoxide 및 $H_2O_2$의 생성과 myeloperoxidase와 acid phosphatase의 유리는 protein kinase C 억제제인 staurosporine과 H-7 그리고 protein tyrosine kinase 억제제인 genistein과 tyrphostin에 의하여 억제되었다. PAF에 의한 호중구 세포내 칼슘농도의 증가는 staurosporine, genistein과 methyl-2,5-dihydroxycinnamate에 의하여 억제 되었다. Staurosporine은 PAF에 의하여 자극된 호중구에서 세포내 칼슘유리와 망간유입을 억제 하였다. Genistein과 methyl-2,5-dihydroxycinnamate는 PAF에 의한 망간유입을 억제하였으나, 세포내 칼슘유리에 대한 이들의 효과는 관찰되지 않았다. PMA에 의하여 활성화된 호중구에서 세포내 칼슘농도의 증가에 대한 PAF의 자극효과는 감소되었다. Protein kinase C와 protein tyrosine kinase는 PAF에 의하여 자극된 호중구에서의 respiratory burst, lysosomal enzyme유리와 칼슘동원에 관여할 것으로 제시된다. 세포내 칼슘농도의 증가는 protein kinase의 영향을 다르게 받는 세포내 칼슘유리와 세포외부로 부터의 칼슘유입에 의하여 이루어질 것으로 추정된다. Protein kinase C가 활성화되어 있는 상태에서 세포내 칼슘동원에 대한 PAF의 자극작용은 감소될 것으로 시사된다.

Involvement of Protein Tyrosine Kinase in Stimulated Neutrophil Responses by Sodium Fluoride

  • In this study, during the activation of neutrophil responses by sodium fluoride. involvement of protein tyrosine kinase was studied. Respiratory burst lysosomal enzyme release and elevation of $[Ca^{2+}]_i$stimulated by sodium fluoride in neutrophils were inhibited by protein kinase inhibitors, genistein and tyrphostin. The inhibitory effect of genistein and tyrphostin on superoxide and $H_{2}O_{2}$ production was less than that of protein kinase C inhibitors, staurosporine and H-7. Staurosporine and H-7 had little or no effect on the release of myeloperoxidase and acid phosphatase stimulated by sodium fluoride. EGTA and verapamil inhibited the elevation of $[Ca^{2+}]_i$ evoked by sodium fluoride. The inhibitory effect of staurosporine on the elevation of $[Ca^{2+}]_i$ was less than that of genistein. Phorbol 12-myristate 13-acetate (PMA)-stimulated superoxide production, which is sensitive to staurosporine, was further enhanced by genistein, whereas the stimulatory action of PMA on myeloperoxidase release was inhibited by genistein. A pretreatment of neutrophils with PMA signifcantly attenuated sodium fluoride-evoked elevation of $[Ca^{2+}]_i$ These results suggest that protein tyrosine kinase may be involved in the activation process of neutrophil responses due to direct stimulation of guanine nucleotide regulatory proteins. In neutrophil responses, PMA-stimulated neutrophils appear to show a different type of inhibition of protein tyrosine kinase.

Antitumor and Immunostimulating Activities of Acanthopanax sessiliflorus Fruits

  • The antitumor and immunostimulating activities of Acanthopanax sessiliflorus fruits were investigated. Polysaccharide isolated from this plant, when administered consecutively for 9 days at 50 and 100 mg/kg i.p. in mice, caused a significant increase in the life span and a significant decrease in the tumor weight and volume in mice inoculated with Sarcoma-180 tumor cells. Polysaccharide was also demonstrated to exhibit phagocytosis-enhancing activity as measured by the carbon clearance in mice. Polysaccharide, when administered i.p. at 50 and 100 mg/kg/day for 3 consecutive days, exhibited a significant RCtr/RCc [the rate of regression coefficient of the animals teated (RCtr) to that of the control (RCc)], being 1.44 (PI = 1), 1.52 (PI = 2) which was approximately the same with that of enhancement of phagocytosis, its potency as expressed by the regression coefficient ratio of zymosan (RCtr/RCc = 1.55, PI = 2), a typical phagocytosis enhancer. Polysaccharide also caused a significant increase in the acid phosphatase activity representing lysosomal enzymes in macrophages at 1-100 ig/ml in vitro in compliance with in vivo results. These results suggest that the antitumor activity of polysaccharide might be related to the immunostimulating function.

Substrate reduction therapy in three patients with Gaucher disease

  • Purpose: Gaucher disease (GD) is the most common lysosomal storage disease caused by beta-glucocerebrosidase (GBA) deficiency. Oral substrate reduction therapy with miglustat ($Zavesca^{(R)}$) was approved for the treatment of adults with GD type 1, for whom enzyme replacement therapy (ERT) is unsuitable or not a therapeutic option. In this study, we report the effect of miglustat ($Zavesca^{(R)}$) in three Korean GD patients. Materials and Methods: Clinical findings comprising age at diagnosis, presenting signs, laboratory findings at diagnosis, GBA activity and mutations, and clinical courses of the three patients were reviewed. Results: Miglustat was administered to three patients who reported allergic reactions during intravenous imiglucerase infusions. One patient withdrew after 15 months of miglustat administration owing to continuous elevation of disease biomarker levels (chitotriosidase, acid phosphatase, and angiotensin-converting enzyme). Poor adherence to medication was suspected but was denied by the patient. In the other two patients, platelet count and levels of hemoglobin and other biomarkers remained stable during miglustat administration. However, they suffered from severe diarrhea and weight loss, which led to miglustat discontinuation after 1 and 12 months of administration. Conclusion: Our study shows that although miglustat is suggested to GD patients as an alternative treatment to ERT, significant adverse reactions may lead to discontinuation of miglustat. In addition, it is difficult to monitor the drug adherence.

수삼추출물 첨가 배지에서 배양된 노루궁뎅이버섯 균사체 심부발효물 조다당획분의 면역 및 암전이 억제활성 (Immune Stimulation and Anti-Metastasis of Crude Polysaccharide from Submerged Culture of Hericium erinaceum in the Medium Supplemented with Korean Ginseng Extracts)

  • 수삼과 버섯의 새로운 용도를 개발하기 위하여, 수삼추출물이 첨가된 액체배지에서 배양된 노루궁뎅이 균사체 심부발효물의 열수추출물로부터 조다당획분을 조제하였다. 수삼추출물이 액체배지 전체양의 1%, 3%와 5% 첨가된 노루궁뎅이버섯 균사체 심부발효물의 열수추출물로부터 조다당획분(HE-GE-CP-1, 3과 5)이 분획되었을 때, 수율은 각각 5.7, 5.1과 4.8%이었다. 이러한 조다당획분 중 HE-GE- CP-5(saline 대조군의 1.89배)는 HE-GE-CP-1(1.64배)과 HE-GE-CP-3(1.76배)보다 유의적으로 높은 비장세포의 마이토젠 활성을 보여주었다. 또한 HE-GE-CP-5(1.83배)는 소장의 국소 면역조직인 Peyer's patch를 경유한 골수세포 증식활성에서도 HE-CP(1.59배), HE-GE-CP-1(1.44배)과 HE-GE-CP-3(1.69배)보다 높은 활성을 나타내었으며, 항암효과로서의 암전이 억제활성 측정에서는 수삼추출물이 5% 첨가된 조다당획분에서 72.4%의 높은 암전이 억제활성도 보여주었다. 그러나 복강 내 마크로파지의 lysosomal phosphatase의 활성측정에서는 HE-GE-CP-3에서 2.03배의 가장 높은 마크로파지 자극활성을 확인할 수 있었다. 한편, 이러한 면역 및 암전이 억제활성을 갖는 조다당획분인 HE-GE-CP-5는 중성당(63.2%)과 함께 상당량의 산성당(19.3%)을 함유하였으며 단백질은 8.8%가 구성분으로 분석되었다. 이러한 결과로부터 HE-GE-CP-5는 암전이를 억제하기 위한 면역계를 자극할 수 있음이 확인되었으며, 이러한 암전이 억제의 항종양 활성은 마크로파지, 비장세포 및 Peyer's patch cell 등의 활성화 기작과 연계되어 있음을 추정할 수 있었다.

당원병과 뮤코다당체침착증이 동시에 발견된 증례 1예 (A case of simultaneously identified glycogen storage disease and mucopolysaccharidosis)

  • 당원병(GSD)과 점액다당류증(MPS)은 각각 독립된 유전 질환으로, 각기 다른 대사에 관여하는 효소의 결핍으로 인해 전구물질이 축적되어 임상 증상을 나타낸다. 간비대는 두 질환 모두에서 나타나는 중요한 임상 양상이다. 저자들은 이 드문 두 질환이 한 환자에서 동시에 발견되어 이에 보고하고자 한다. 16개월 남아가 2개월 동안 반복적인 중이염을 앓았고, 인근 병원을 다니던 중 우연히 간비대가 발견되었다. 환아는 이마가 튀어나온 조악한 얼굴 모양을 가지고 있었다. 환아의 간은 4횡지 정도로 촉지되었고, 그 성상은 부드럽고 무른 느낌이었으나 경계가 예리하게 만져졌다. 혈액 검사상, AST와 ALT가 85 IU/L, 77 IU/L로 약간 상승되어 있었다. 공복 저혈당은 보이지 않았고, 혈청 젖산, 요산, 콜레스테롤 모두 정상 소견을 보였다. 복부 초음파에서는 간의 메아리 발생 정도(echogenecity)가 증가되어 있었다. 간 생검을 실시하였고, 현미경 검사상 둥근 간세포들이 관찰되었는데, 그 간세포 내에는 Periodic Acid-Schiff 염색에서 진하게 염색되는 거대한 당원 입자들 다수와 이와 동반된 지방 공포(lipid vacuole)가 포함되어 있었다. 전자현미경에서 간세포의 핵과 세포질 모두에 당원 입자들이 포함되어 있었고 거대한 미토콘드리아도 관찰할 수 있었다. 생검을 통해 채취한 간 조직을 분석한 결과, 간의 당원 함량은 9.3%로 정상수치에 비해 증가되어 있었으나, 간의 glucose-6-phosphatase 활성도는 정상이어서 Ia형이 아닌 당원병으로 진단하게 되었다. 한편, X 선상 척추 앞면의 부리모양(beak-shape)와 J 모양의 안장(sella)과 같은 여러 골격의 이상 소견을 보였으며, 소변의 점액다당류 농도 또한 증가되어 있었다. Iduronate sulfatase 활성도가 0.1 nmol/hr/mg protein로 낮은 소견을 보여 헌터 증후군(제2형 점액다당류증, MPS type II)으로 진단할 수 있었다. 이에 저자들은 상기 환자가 당원병과 헌터증후군이 동시에 발견된 첫 환자로 생각되어 보고하는 바이다.
