재배지역에 따른 농산물의 phytochemical 함량 (Phytochemical contents of agricultural products cultivated by region)

  • 최근 건강 증진을 위해 소비가 증가하고 있는 농산물을 대상으로 phytochemical 성분을 정량하고, 다양한 phytochemical 성분을 동시에 분석하는 방법에 대해 조사하였다. Lycopene, ${\alpha}$, ${\beta}$-carotene, cryptoxanthin과 lutein을 대상으로 동시분석을 실시한 결과 회수율은 각각 lycopene $120.7{\pm}4.1%$, lutein $89.1{\pm}3.5%$, ${\alpha}$-carotene $91.2{\pm}2.9%$, ${\beta}$-carotene $99.1{\pm}4.4%$, cryptoxanthin $100.0{\pm}5.3%$로 나타나 향후 다양한 phytochemical 성분을 동시에 분석하는데 유효하 방법이 될 것으로 판단된다. 농산물을 대상으로 4종의 phytochemical(${\beta}$-carotene, lycopene, lutein 및 cryptoxanthin)을 분석한 결과, lutein은 시금치>참다래>토마토>블루베리>메론 순이었으며 시료별 분석 결과 시금치(나주, 신안산), 참다래(해남, 보성산)와 블루베리(담양산)가 타 시료에 비해 lutein 함량이 더 높은 결과를 보였다. Lycopene은 토마토와 시금치에서 검출되었으며, 분석 시료 중 흑 토마토($56.66{\pm}7.48mg/kg$)와 장성 토마토($50.28{\pm}5.42mg/kg$)의 lycopene 함량이 가장 높았다. ${\beta}$-carotene의 경우 시금치와 토마토 시료에 가장 많이 함유되어 있었으며 특히, 나주 시금치($65.03{\pm}4.83mg/kg$)와 신안 시금치($37.67{\pm}5.49mg/kg$)에 다량 함유되어 있었다. Quercetin 분석 결과 블루베리에서 가장 높게 검출되어 담양산 블루베리가 $1,054.06{\pm}80.54mg/100g$으로 수입산($986.57{\pm}67.85mg/100g$) 보다 높았으며, kaempferol의 경우도 비슷한 경향을 보였다. 총 폴리페놀의 경우 블루베리(213.60~229.96 mg/100 g)가 가장 높은 결과를 보였고, 시금치(112.50~141.67 mg/100 g), 참다래(46.49~70.44 mg/100 g) 에서도 높게 검출되었다. 총 플라보노이드 함량을 분석한 결과 블루베리, 시금치 시료에서 타 시료에 비해 함량이 높게 검출되었으며, 비타민 C 함량은 참다래(39.45~86.79 mg/100 g), 한라봉(38.65~50.96 mg/100 g), 토마토(5.90~15.97 mg/100 g), 블루베리 순으로 나타났고, 블루베리에서 총 안토시아닌 함량을 측정한 결과 담양산 블루베리>수입산 블루베리 순으로 나타났다. 이상의 결과로 보아 유통 중인 농산물에 다양한 phytochemical 성분이 존재하는 것으로 확인되었으며, 이는 재배지역, 품종, 숙성 정도 등에 따라 조금씩 차이가 나는 것으로 판단된다.

  • SH2 domains and their associated catalytic or noncatalytic proteins constitute critical signal transduction targets for drug discovery. Grb2 associates with phosphotyrosine sites of the activated receptors or Shc via their SH2 domain to link receptor tyrosine kinases to ras signalling. Blocking of the Grb2-Shc complex may be to intervene the oncogenic signal transduction pathways and to develop a new antitumor drug. In the search for blockers of Grb2 SH2-Shc interaction, Lutein, a family of carotenoids, was isolated from the extract of the leaf of Agastache rugosa O. Kuntze as SH2 domain antagonists. The $IC_{50}$ of Lutein against Grb2-Shc binding was $6.8\;{\mu}M$.

해조류 중의 Anti-Tumor Initiator 및 Promoter의 해석-4: 발암성 Heterocyclic Amine에 대한 곰피 추출물 중의 돌연변이원성 억제인자 (Elucidation of Anti-Tumor Initiator and Promoter Derived from Seaweed-4: Desmutagenic Principles of Ecklonia stolonifera Extracts against Carcinogenic Heterocyclic Amines)

  • The present study was performed to elucidate desmutagenic principles from Ecklonia stolonifera extracts against 2-amino-1-methyl-6-phenylimidazo[4,5-b] pyridine(PhIP) and 2-amino-3,8-dime-thylimidazo[4,5-f]duinoxaline(MeIQx) with Salmonella/mammalian-microsome mutagenicity test. Alginate, phenols, chlorophyll and carotenoids from Ecklonia stolonifera were extracted and their desmutagenicities were assayed. Alginate hydroysates showed desmutagenic activities against PhIP and MeIQx at high level dose. Phenol fractions and bromophenol showed desmutagenic activity of about MeIQx at high level dose. Phenol fractions and bromophenol showed desmutagenic activity of about 90% per 0.5mg against PhIP and MeIQx. Chlorophyllin among chlorophyll derivatives exhibited remarkable desmutagenic activities of 92.9% and 82.7% at 20uM against PhIP and MeIQx, respectively. Carotenoids, such as lutein and $\alpha$-cryptoxanthin isolated from Ecklonia stolonifera exerted also high desmutagenic activity. Major desmutagenic substances from Ecklonia stolonifera are considered to be chlorophyllin, phenols, lutein, $\alpha$-cryptoxanthin and low molecular alginates.

달걀.메추리알.오리알의 Retinol Equivalent 측정과 Carotenoids 분석 (Determination of Retinol Equivalent and Identification of Carotenoids in Hen, Quail and Duck Eggs)

  • 시중에서 판매되는 달걀, 메추리알, 오리알의 총 carotenoid, provitamin A carotenoids, retinol함량을 측정하여 Retinol Equivalent를 산출하고, 이들 난황의 carotenoid 조성을 비교분석하였다. 각 난황에서 단위 g당 총 carotenoid함량은 달걀>메추리알>오리알의 순서로, retinol은 메추리알>오리알>달걀의 순서로, Retinol, Equivalent는 메추리알>오리알>달걀의 순서로 높았다. 간 난황의 carotenoid 분석결과, carotenoid 조성은 거의 비슷했으나, 달걀과 메추리알의 경우 lutein과 zeaxanthin이 총 carotenoid의 75% 정도였고, 오리알의 경우는 60% 정도였다. 거의 대부분을 차지하고 있는 carotenoids임을 보였다.

여대생의 Carotenoids 식품 보충 섭취에 따른 혈청 비타민 A 함량 변화 (Serum Vitamin A Response to SHort-term Carotenoids Foods Supplementation in Female University Students)

  • Korean intakes of Vitamin A have not reached RDA for a long time according to the National Nutrition Survey. The pupose of this study was to examine the serum response to short-term carotenoids-rich vegetable juices supplementation. A total of 31 female students were recruited for this study. Their average serum retinol content before supplementation began was 14.74$\mu\textrm{g}$/100ml, and 87% of them had lower than adequate levels. Serum $\beta$-carotene, $\alpha$-carotene, lycopene and lutein contents were 0.77, -, 0.74 and 8.29$\mu\textrm{g}$/100$m\ell$ respectively. The serum responses to 3 kinds of juice supplementation for 3 weeks are summarized as follows. A small but steady increase in serum retinol concentration was detected with the supplementation (p<.05), but the kinds of juice made no significant difference. Serum $\beta$-carotene contents also increased with the supplementation(p<.01), but the kinds of juice again made no significant difference. Serum lycopene increased with the lycopene-containing vegetable juice and tomato juice supplementation, but the increase wasn't significant. There was no change in serum lycopene levels with the lycopene free carrot juice supplementation. Finally, there were no signifnicant changes in serum lutein and $\alpha$-carotene levels with the 3 kinds of juice supplementation.

Change of growth and carotenoid concentration in Korean fir with varied annual temperature on Mt. Halla

  • Deforestation and conservation of coniferous forest have been recognized as critical issues in Korea due to climate change. The main purpose of this research was to monitor changes of secondary metabolite contents and growth of Korean fir (Abies koreana) according to the temperature change in Mt. Halla. The Korean fir located at three different regions, Yeongsil, Witseoreum, and Jindallaebat, was monitored in April, July, and October from 2016 to 2018 and secondary metabolites, specifically lutein, α-carotenoid, and β-carotenoid, were analysed with high performance liquid chromatography. The results showed that average concentrations of lutein, α-carotenoid, and β-carotenoid were 0.82 - 23.30, 0.02 - 2.01, and 0.11 - 2.84 ㎍·g-1 and the highest concentration of secondary metabolite was observed in October compared to April and July. The average length and width of Korean fir in the three regions were 11.84 - 20.70 and 1.78 - 2.41 mm from 2016 - 2018. A correlation analysis showed that the concentrations of all three secondary metabolites were negatively correlated with temperature and a significant difference was observed between temperature and lutein concentration in Korean fir. Overall, growth and production of secondary metabolites in Korean fir highly depended on the temperature, and global warming thus might have an adverse effect on the growth and physiological changes of Korean fir in Mt. Halla.

미세조류로부터 색소물질 생산 증대를 위한 추출 조건 최적화 (Optimization of Extraction Conditions to Enhance Production of Bioactive Compounds from Microalgae)

  • We optimized ultrasound-assisted extraction to improve the extraction efficiency of bioactive compounds from the microalgae Acutodesmus reginae. To optimize this extraction process, we investigated the effects of solvent type, solvent concentration, extraction time, extraction number, and extraction power on the production of lutein, α-carotene, β-carotene, and chlorophylls a, and b. After sequential optimization of these main variables, the maximum amount of each compound was extracted at 30℃ with an ultrasound power of 80 W and using 99.5% methanol. Under these optimum conditions, the amount of lutein, α-carotene, β-carotene, and chlorophylls a, and b, were measured as 10.43, 8.66, 3.76, 15.43, and 6.39 mg/g dry matter respectively.

메기과에 속하는 메기와 미유기의 Carotenoid 색소 성분의 비교 (Comparision of Carotenoid Pigments in Catfish, Parasilurus asotus and Slender catfish, Parasilurus microdorsalis in the Family Siluridae)

  • 미이용자원인 어류 carotenoid에 관한 비교 생화학적 연구의 일환으로서, 메기과의 자연산 및 양식산 메기와 한국 특산의 담수어류인 미유기의 표피 carotenoid를 TLC, column chromatography 및 HPLC로 분리하고 NaBH$_4$에 의한 환원반응, I$_2$에 의한 이성화반응 및 UV-visible spectrophotometer로 비교하고 IR 및 Mass spectrum을 측정하여 carotenoid를 동정한 결과는 다음과 같다. 자연산 메기 표피의 총 carotenoid 함량은 0.27 mg($\pm$0.7)%였으며 carotenoid의 조성은 zeaxanthin 25.8%, diatoxanin 9.7% 및 cynthiaxanthin 9.1%로서 주성분을 이루고 있었으며 그 외 isocryptoxanthin 6.5%, $\beta$-carotene 5.1%, $\beta$-cryptoxanthin 4.3%, lutein 3.4% 및 $\alpha$-cryptoxantin 2.4%의 순으로 함유하고 있었다. 양식산 메기 표피의 총 carotenoid 함량은 0.07 mg($\pm$0.03)%로서 자연산 메기에 비해 다소 낮았으며, carotenoid의 조성은 lutein 48.5%, zeaxanin 13.9% 및 isocryptoxanin 13.3%로서 주성분을 이루고, 그 외 $\beta$-carotene 9.5%, $\beta$-cryptoxantin 1.4% 및 $\alpha$-cryptoxanthin 4.6%의 순으로 함유하고 있었다. 한편 미유기 표피의 총 carotenoid 함량은 0.75 mg($\pm$0.25)%로서 메기과의 다른 어종에 비해 함량이 높았으며, carotenoid 조성은 zeaxanthin 24.5%, 7'8'-dihydro-$\beta$ -carotene-4-ol 24.1%, 7'8'-dihydro-$\beta$-carotene 17.9% 및 7'8'-dihydro-$\beta$-carotene-3-ol 10.8%로서 주성분을 이루고 있었으며 그 외 diatoxanthin 8.7%, cynthiaxanthin 6,7% 및 lutein 5.0%의 순으로 함유하고 있었다. 이러한 결과는 parasiloxanthin과 7',8'-dihyo-parasiloxanthin을 주성분으로 함유하는 biwa호수 메기, P. asotus 표피 carotenoid 조성과 비교하여, 미유기에서는 7'8'-dihydro-$\beta$-carotene의 구조를 가진 carotenoid가 존재하였으나 자연산 및 양식산 메기에서는 발견되지 않아 특이하였다.

난포낭종 및 황체난종우의 혈장과 낭종내강액내 성호르몬의 농도에 관한 연구 (Studies on the Concentrations of Sex Hormone in the Blood Plasma and Antrum Fluid of Follicular and Lutein Cystic Ovaries of Holstein Cows)

  • The study was carried out to find out the changes of the sex hormone concentrations in the blood plasma and antrum of follicular and lutein cystic ovaries of Holstein cows. The progesterone, estradiol-17$\beta$, testosterone, FSH and LH from samples of the blood plasma and antrum of cystic ovaries of cows assayed by radioimmunoassay method. The results of this study were summarized as follows : 1. The concentrations fo progesterone and estradiol-17$\beta$ in the blood plasmaat estrous and luteal phase were 0.95$\pm$0.18ng/ml, 11.45$\pm$3.12pg/ml and 4.25$\pm$0.27ng/ml, 6.27$\pm$0.82pg/ml respectively. The concentrations of progesterone and estradiol-17$\beta$ in the antrum fluid of follicles at estrous and luteal phase were 24.8$\pm$4.12ng/ml, 54.3$\pm$7.25pg/ml and 21.7$\pm$3.79ng/ml, 14.3$\pm$2.72pg/ml respectively, and showed significant changes among the estrous and luteal phase and blood plasma and antrum fluid of follicles. 2. The concentrations of progesterone, estradiol-17$\beta$, testosterone and LH in the blood plasma of follicular cystic ovareis of cows were 0.85$\pm$0.25ng/ml, 9.23$\pm$2.72pg/ml, 17.12$\pm$3.26pg/ml and 3.78$\pm$1.02mIU/ml respectively. And the concentrations of progesterone, estradiol-17$\beta$, testosterone and LH in the antrum fluid of follicular cystic ovareis of cows were 284$\pm$48.21ng/ml, 389$\pm$67.23ng/ml, 12.84$\pm$0.29ng/ml and 1.84$\pm$0.17mIU/ml respectively, and showed significant changes between in the blood plasma and antrum fluid of cystic ovaries. 3. The concentrations of progesterone, estradiol-17$\beta$, testosterone and LH in the blood plasma of lutein cystic ovaries of cows were 3.40$\pm$0.78ng/ml, 4.02$\pm$0.42pg/ml, 10.72$\pm$2.74pg/ml and 0.76$\pm$0.12mIU/ml respectively. And the concentrations of progesterone, estradiol-17$\beta$, testosterone and LH in the antrum fluid of lutein cystic ovareis of cows were 427$\pm$35.79ng/ml, 0.76$\pm$0.07ng/ml, 3.45$\pm$0.57ng/ml and 0.29$\pm$0.07mIU/ml respectively, and showed significant changes between the blood plasma and antrum fluid of cystic ovaries. 4. Accordingly, the diagnosis of follicular and lutein cystic ovareis of cows from progesterone, estradiol-17$\beta$ and LH levels in the blood plasma and antrum of cystic ovaries of cows is thought to be possible a diagnostic means.

Variation in Carotenoid Composition in Carrots during Storage and Cooking

  • High-performance liquid chromatography (HPLC) was applied to determine the carotenoid composition of carrots during storage and cooking. Analyses were conducted immediately after harvest and 1, 2, 4, and 8 weeks after harvest. During the course of the storage, the carotenoid levels generally decreased, and this decrease was found to be greater during the first week for $\beta$-carotene (all-trans-$\beta$-carotene) and lutein, and during the second week for $\alpha$-carotene. Additionally, the amount of the $\alpha-$ and $\beta$-carotenes in carrot leaves changed slightly within the first 2 weeks of harvest when stored at $4^{\circ}C$. Specifically, the level of lutein, the main component of carrot leaves, increased from 233.8$\pm$11.7 to $346.2\pm26.7{\mu}g$/g DW during the first 2 weeks. In addition, the change in carotenoid contents was observed during the home-processing of one Korean cultivar. Carrots fried in oil showed the highest amount of $\beta$-carotene ($164.3\pm6.6{\mu}g$/g DW) and $\alpha$-carotene ($50.1\pm0.4{\mu}g$/g DW), while carrots that were prepared by sauteing, pressure-cooking in water and microwaving had the second highest levels. The greatest loss of in carotenoids occurred in response to boiling in water containing 1% NaCl, braising and baking. The content of lutein increased slightly after boiling in water containing 1% NaCl ($9.3\pm0.4{\mu}g$/g DW), while a loss in lutein occurred after preparation using other home-processing methods. A cis-isomer of all-trans-$\beta$-carotene, 13-cis-$\beta$-carotene, was present in detectable amounts in all processed samples, but not in raw roots. Another isomer, 9-cis-$\beta$-carotene, was detected in carrots that were prepared by boiling, frying and pressure-cooking.
