카로티노이드 $^1O_2$ 퀜칭효과

  • $^1O_2$ quenching abilities of several carotenoids which contain hydroxy, carbonyl and ester groups were compared quantitatively with $\beta$-carotene, and the capacity of the quenching was interpreted in the light of electronic effects. The rate constans of $^1O_2$ quenching of lutein diester and astaxanthin diester in MeOH solution were shown to be $1.9\times10^{10}M^{-1}Sec^{-1}$, $2.3\times10^{10}M^{-1}Sec^{-1}$ respectively. Under the experimental conditions, and within the carotenoid tested results, the larger the resonance energy is, the larger becomes the rate constant and consequently the lower the transition energy is, the better becomes the quencher.

Some Aspects of functional foods and their perspective (건강기능성 식품의 현황 및 전망)

  • The purpose of this paper is to explore some aspects of functional foods and to give an prospective view of the foods. Recently the increasing rate of old ages is very rapid so that the functional foods is demanded as the treatment of cure and health foods for the old ages. Those are the foods which includes such elements as polyphenol, vitamin E & C, 𝛽-carotene, selenium, lycopene, lutein, isoflavon, capsaicin, DHA, lecithin, peptide, dietry fiber, alginic acid, lactic ferments, caffeine, kitin and kitosan, taurine, and phenylalanine. The total amounts of those foods are $ 112 billion in 2001, the American market marks $36.3 billion (32%) and Europe marks $32.6 billion (29%) and Japan marks $27.1 billion (24%), however, Korea marks $11 billion which equivalent to 1%. We are in need to develop the new foods for the antioxidant effect, anti-cancer, the prevention of cardio vascular disease, the control of intestine vacillius, control of diabetes, with those functional elements in order to secure the world market for the functional foods.

Effects of Various Light Sources on the Carotenoid and Glucosinolate Contents in Chinese Cabbage (Brassica rapa L. ssp. pekinensis) (다양한 광원이 배추 내 Carotenoid와 Glucosinolate 함량에 미치는 영향)

  • BACKGROUND: Chinese cabbage biosynthesizes various phytochemicals including carotenoids and glucosinolates. Environmental stress has a major effect on the growth and yields of vegetables, and can significantly affect nutritionally important phytochemicals. Phytochemicals of plants are influenced by light, temperature, carbon dioxide, and growing conditions. The aim of this study was to investigate the effect of various light sources on carotenoid and glucosinolate contents in Chinese cabbage. METHODS AND RESULTS: [Experiment I] Set the control (field control, FC) on the ground. Using acrylic sunlight, experiments were set up transparency box (field transparency, FT), red box (field red, FR) and blue box (field blue, FB). [Experiment II] Set the control (chamber control, CC) in the greenhouse. Using plant growth chamber with artificial light, experiments were set up LED red (chamber red, CR), LED blue (chamber blue, CB), LED mixed red+blue (chamber red+blue, CRB) and fluorescent (chamber fluorescent, CF). After plant growth, Chinese cabbage was harvested at 110 days after sowing (DAS). The status of plants growth (leaf length, width, fresh weight etc.) was immediately investigated. Carotenoid and GSL contents were analyzed by HPLC. [Experiment I] Results documented that the ranges of total carotenoid contents were 25.39 ~ 58.80 mg/kg dry wt for lutein, 0.84~ 4.22 mg/kg dry wt for zeaxanthin, and 3.85~18.71 mg/kg dry wt for ${\beta}$-carotene. Lutein was the highest for the content and the largest for the variation as well. [Experiment II] Results documented that the ranges of total carotenoid contents were 24.66~137.96 for lutein, 2.51~20.65 for zeaxanthin, and 8.40~49.80 mg/kg dry wt for ${\beta}$-carotene. The total carotenoid contents of CR (156.62) and CB (115.90) were 1.6~2.3 times larger than the other treatments, and ${\beta}$-carotene content was about twice as high as that of the other treatments on the CR (38.74 mg/kg dry wt.). [Experiment I] Total GSL content was the highest in FT (19.76) that was higher 1.7 times than the lowest treatment ($11.39{\mu}mol/g\;dry\;wt$.). [Experiment II] The total content of GSL was highest in CRB (4.19) and lowest in CF ($2.88{\mu}mol/g\;dry\;wt$.). In the CRB, total GSL contents ($4.19{\mu}mol/g\;dry\;wt$.) was the highest. CONCLUSION: Total and individual carotenoid and GSL contents in Chinese cabbage show significant differences under different light sources. Red and blue lights contribute to significant carotenoids expression and antioxidant activity for nutrition and health benefits. These results concluded that the introduction of varying lights affected the synthesis of important nutrient compounds in Chinese cabbage. It is predicted that the application of good light source enhances the accumulation of functional compounds.

Early Changes after Death of Plaice, Paralichthys olivaceus Muscle -5. Effect of Storage Temperature on Morphological Changes of Myofibrils and Histological Changes of Muscle- (넙치(Paralichthys olivaceus)육의 사후 조기 변화 -5. 저장 온도가 근원섬유의 형태학적 및 육의 조직학적인 변화에 미치는 영향-)

  • To clarify the effect of storage temperature on the morphological and histological changes of plaice, Paralichthys olivaceus muscle at early stages after killing, the changes in breaking strength of muscle, morphological observation of myofibrils and histological observation of extracellular spaces during storage at $0^{\circ}C\;and\;10^{\circ}C$ were studied. The maximum breaking strength of samples stored at $0^{\circ}C$ was reached within 10hrs and then it dropped significantly (p<0.05) from 10hrs to 25hrs of storage. However, breaking strength was not increased in fresh muscle stored at $10^{\circ}C$ and gradually decreased after 10hrs storage. In myofibrils prepared from dorsal muscle immediately after death, A-band, I-band, H-band and Z-line in sarcomere were clearly distinguishable from each other. Due to muscle contraction, it was not easy to distinguish H-band from I-band observed in sarcomere stored at $0^{\circ}C$ after 10hrs storage. But, in the case of samples stored at $10^{\circ}C$, H-band could be observed dimly until 15hrs of storage. The changes in morphological myofibrils were closely related to increase of breaking strength. No extracellular space was observed among muscle cells immediately after killing. Stored samples at $0^{\circ}C$ showed extracellular spaces after 15hrs storage. On the other hand, samples stored at $10^{\circ}C$ didn't show any extracellular spaces until 15hrs storage and showed extracellular spaces after 24hrs storage. It was thought that the post-mortem tenderization of plaice muscle was closely related to the gradually disintegration of the extracellular matrix structure after killing.

Isolation and Evaluation of Anti-oxidative Constituents from the Extracts of Ficus erecta var. sieboldii King Leaves (좁은잎천선과 잎 추출물 유래 항산화 활성 성분의 동정 및 효능 확인)

  • In this study, we investigated identification of anti-oxidative constituents from Ficus erecta var. sieboldii King (F. erecta) leaves. DPPH and $ABTS^+$ radical scavenging activities were screened for the ethanol extract and solvent fractions, and ethyl acetate fraction showed the most potent scavenging activities. Five constituents were isolated from the ethyl acetate fraction of F. erecta leaves; monoolein (1), oleic acid (2), lutein (3), afzelechin (4), catechin (5). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including NMR spectra, as well as comparison of the data to the literature values. As far as we know, all of the compounds 1 ~ 5 were isolated for the first time from this plant. Studies on DPPH and $ABTS^+$ radical scavenging activities were conducted for the isolated compounds. Among them, afzelechin (4) and catechin (5) showed strong DPPH and $ABTS^+$ radical scavenging activities, whose activities were comparable to a positive control vitamin C. Also, the content of catechin isolated from this plant was determined by HPLC and it was about 3.8 mg/g for the 70% ethanol extract and 20.8 mg/g for the ethyl acetate fraction. From these results, F. erecta leaves extract could be potentially applicable as anti-oxidant ingredients in cosmetic industries.

Effects of dietary marigold extract supplementation on growth performance, pigmentation, antioxidant capacity and meat quality in broiler chickens

  • Objective: This experiment was conducted to investigate the effects of dietary supplementation with marigold extract on growth performance, pigmentation, antioxidant capacity and meat quality in broiler chickens. Methods: A total of 320 one-day-old Arbor Acres chickens were randomly divided into 5 groups with 8 replicates of 8 chickens each. The chickens of control group were fed with basal diet and other experimental groups were fed with basal diet supplemented with 0.075%, 0.15%, 0.30%, and 0.60% marigold extract respectively (the corresponding concentrations of lutein were 15, 30, 60, and 120 mg/kg). Results: The results showed that marigold extract supplementation increased the yellowness values of shank, beak, skin and muscle and the redness ($a^*$) value of thigh muscle (linear, p<0.01). Marigold extract supplementation significantly increased the total antioxidant capacity, and the activities of superoxide dismutase in liver and thigh muscle (linear, p<0.01) and significantly decreased the malondialdehyde contents of liver and thigh muscle (linear, p<0.01). Marigold extract supplementation significantly decreased the drip loss and shear force of thigh muscles (linear, p<0.01). There was no significant effect on growth performance with marigold extract supplementation. Conclusion: In conclusion, dietary supplementation of marigold extract significantly increased the yellowness values of carcass, antioxidant capacity and meat quality in broiler chickens.

Microbial Production of Carotenoids: Biological Functions and Commercial Applications (미생물에 의한 카로티노이드 생산; 생물학적 기능성 및 상업적 적용)

  • Carotenoids are isoprenoids with a long polyene chain containing 3 to 15 conjugated double bonds, which determines their absorption spectrum. They typically consist of a $C_{40}$ hydrocarbon backbone often modified by different oxygen-containing functional groups, to yield cyclic or acyclic xanthophylls. Much work has also been focused on the identification, production, and utilization of natural sources of carotenoid (plants, microorganisms and crustacean by-products) as an alternative to the synthetic pigment which currently covers most of the world markets. Nevertheless, only a few carotenoids (${\beta}-carotene$, lycopene, astaxanthin, canthaxanthin, and lutein) can be produced commercially by fermentation or isolation from the small number of abundant natural sources. The market and demand for carotenoids is anticipated to increase dramatically with the discovery that carotenoids exhibit significant anti-carcinogenic activities and play an important role in the prevention of chronic diseases. The increasing importance of carotenoids in the feed, nutraceutical food and pharmaceutical markets has renewed by efforts to find ways of producing additional carotenoid structures in useful quantities. Because microorganisms and plants synthesize hundreds of different complex chemical carotenoid structures and a number of carotenoid biosynthetic pathways have been elucidated on a molecular level, metabolic and genetic engineering of microorganisms can provide a means towards economic production of carotenoid structures that are otherwise inaccessible. The aim of this article is to review our current understanding of carotenoid formation, to explain the perceived benefits of carotenoid in the diet and review the efforts that have been made to increase carotenoid in certain microorganisms.

Physicochemical Properties of Shade-cultivated Powdered Green Teas (차광재배 가루녹차의 이화학적 품질 특성)

  • The Daecha-12 and Yabukita varieties of powdered green teas were grown under 85% shade-cultivated condition, and their physicochemical properties were analyzed. Total catechin content was not significantly different but, non-gallated catechin content in Daecha-12 was significantly higher than that in Yabukita. Theanine (32%), caffeine (14%), lutein (15%) and total chlorophyll (28%) levels were significantly higher in Daecha-12 than those in Yabukita. The results of a color analysis showed that the lightness L-value of Yabukita was higher than that of Daecha-12 but, that the greenness negative a-value and b-values of Daecha-12 were higher than those of Yabukita. The Daecha-12 cultivar had lower catechin content but higher content of theanine, caffeine, theobromine, lutein, chlorophyll, and a negative a-value than those of the Yabukita cultivar. Thus, the Daecha-12 cultivar is suitable to prepare a high-quality powdered green tea product.

Studies on the carotenoids in the viscera of abalone (Haliotis discus hannai) (전복(Haliotis discus hannai) 내장(內臟)의 Carotenoid에 관(關)한 연구(硏究))

  • The studies on the carotenoids in the viscera of abalone (Haliotis discus hannai) have been-carried out. The pigments were extracted with acetone-methanol mixture (4 : 1) from the viscera of abalones which were caught around the coastal water of Korea from March to August. The individual carotenoid in the extracts was separated and purified by the silica gel TLC, $Mg(OH)_2$ impregnated paper chromatography and $Mg_2(OH)_2CO_3$ TLC. The isolated eleven carotenoids were investigated and identified by epoxide test, partition test, reduction with sodium borohydride, alkaline hydrolysis, co-chromatography and comparative test with reference carotenoids and electronic and IR absorption spectrophotometry. ${\alpha}$-Carotene, ${\beta}$-carotene. lutein, zeaxanthin, siponaxanthin, siponein, fucoxanthin, loroxan-thin-like and fucoxanthinol-like have been identified among the eleven carotenoids isolated. It has been found that fucoxanthin, on alkaline treatment, was transformed to the product of which chromophore was the same one as fucochrome and semifucoxanthol. Among the identified nein carotenoids siphonaxanthin, siponein, fucoxanthin, loroxanthin-like and fucoxanthinol-like have not been reported previously to be contained in the shellfish.

Oxidative Stability of Wheat germ Lipid and Changes in the Concentration of Carotenoid and Tocopherol during Oxidation (밀배아 지방질의 산화 안정성과 카로티노이드 및 토코페롤의 변화)

  • The changes of the lipid composition and of the contents of carotenoid and tocopherol in wheat germ were studied during the storage at $30^{\circ}C$. The contents of triglyceride and free fatty acid were changed from 66% and 7% to 49% and 24% respectively after 30 days. The predominant free fatty acids were lauric acid (29%), palmitic acid (21%) and linoleic acid (20%), however, linoleic acid increased to 30%, lauric acid reduced to 21% after storage of 30 days. The carotenoids in the wheat germ were ${\beta}-carotene,\;{\alpha}-carotene$, lutein and taraxanthin, and the contents of these were 306, 59, 383 and 356 ng/g wheat germ, respectively. Their contents, however, were reduced to 36, 4, 203 and 149 ng respectively after 20 storage days. Especially, degradation rate of ${\beta}-carotene$ was 22.5 ng/day. The tocopherol isomers in wheat germ were ${\alpha}-,\;{\beta}-\;and\;{\gamma}-tocopherol$, and they reduced from $55,\;48\;and\;38\;{\mu}g/g$ wheat germ to 35, 32 and $32\;{\mu}g$ respectively after 20 storage days. The ${\alpha}-tocopherol$ was degraded by $1.26\;{\mu}g/day$ at this storage condition.
