Historical Record of Mushroom Research and Industry in Korea

  • Two kinds of mushrooms, Gumji (金芝; Ganoderma) and Soji, were described in old book of Samguksagi (History of the three kingdoms; B.C 57~A.D 668; written by Bu Sik Kim in 1145) in Korea-dynasty. Many kinds of mushrooms were also described in more than 17 kinds of old books during Chosun-dynasty (1392~1910) in Korea. Nowadays, mushroom cultivation has been increased through out the world last decade years. Production of mushrooms has also been increased 10-20% and many varieties have been cultivated. Similar trends were also observed in Korea. Approximately two hundred commercial strains of 37 species in mushrooms were developed and distributed to cultivators. Somatic hybrid variety of oyster mushroom 'Wonhyeong-neutari' were developed by protoplast fusion, and distributed to grower in 1989. The fruiting body yield index of somatic hybrids of Pleurotus ranged between 27 and 155 compared to parental values of 100 and 138. In addition, more diverse mushroom varieties such as Phellinus baumi, Auricularia spp., Pleurotus ferulae, Hericium erinaceus, Hypsizigus marmoreus, Grifola frondosa, Agrocybe aegerita and Pleurotus cornucopiae have been attempted to cultivate in small scale cultivation. Production of mushrooms as food was 190,111 metric tons valued at 800 billion Korean Won (one trillion won if include mushroom factory products; 1dollar = 1,040 Won) in 2011. Major cultivated species are Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Lentinula edodes, Agaricus bisporus, and Ganoderma lucidum, which cover 90% of total production. Since mushroom export was initiated from 1960 to 1980, the export and import of mushrooms have been increased in Korea. Technology developed for liquid spawn production and automatic cultivation systems lead to the reduction of the production cost resulting in the increasement of mushroom export. However some species were imported because of high production cost for these mushrooms requiring the effective cultivation methods. Developing of effective post-harvest system will be also directly related to mushroom export. In academic area, RDA scientists have been conducting mushroom genome projects. One of the main results is the whole genome sequencing of Flammulina velutipes for molecular breeding. An electrophoretic karyotype of of F. velutipes was obtained using CHEF with 7 chromosomes, with a total genome size of approximately 26.7 Mb. The mususcript of the genome of F. velutipes was published in PLOS ONE this year. For medicinal mushrooms, we have been conducting the genome research on Cordyceps and its related species for developing functional foods using this mushroom. In 2013, Korea Food and Drug Administraion (KFDA) approved Cordyceps mushroom for its value as an immune booster.

Inhibitive Effects of Edible Mushrooms Extracts on Pathogenic Bacteria and Proliferation of Cancer Cells (식용버섯 추출물의 식중독균 및 암세포 증식에 대한 저해 효과)

  • The antibacterial effect of methanol and water extracts from edible mushrooms on the growth of pathogenic bacteria (Bacillus cereus, Bacillus subtilis, Staphylococcus aureus, Echerichia coli O-157, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhi) were investigated. The Lyophyllum cinerascens and Pleurotus ostreatus2 methanol extracts treated with 5.0 mg/disc showed the highest antimicrobial activity against 7 kinds of pathogenic bacteria. And methanol extracts of edible mushrooms showed higher antimicrobial activity against gram positive and gram negative microorganisms than water extracts. The methanol extracts of mushrooms revealed high inhibitive activites in cytotoxicity on human cancer HepG2 and HT-29 cells. The growth of cancer HepG2 and HT 29 cells which treated with 1 mg/mL of Cordyceps militaris and Sarcodon aspratus methanol extracts were strongly inhibited to $67\%$ and $81\%$ respectively. And most of the methanol extracts exhibited the stronger effects against these cells, at the same concentration, comparing with water extracts. Particularly, the methanol and water extracts of Cordyceps militaris, Agaricus blazei, Lyophyllum ulmarium, Ganoderma lucidum and Sarcodon aspratus have the strongest antitumoral effects on HepG2 and HT-29 cells. From these results, it is considered that wild mushrooms have stronger antimicrobial and in vitro cytotoxic effects.

Major Components of Mushroom Mycelia Cultivated with Citrus Juice Processing Wastes (감귤 주스 착즙박을 이용하여 재배된 버섯균사체의 성분)

  • This study investigated the major components of citrus juice processing wastes (CJPW) as well as mushroom mycelia that were cultivated with the CJPW, such as Pycnoporus coccineus (PC), Lentinus edodes (LE), Pleurotus eryngii (PE), Hericium coralloides (HC), Panellus serotinus (PS) and Ganoderma lucidum (GL). The organic acid contents of the mushroom mycelia were similar to or less than those of the CJPW, but the free sugar contents of the mycelia were lower than those of the CJPW. The narirutin contents of the mushroom mycelia ranged from 448.67-932.98 mg% and were similar to or less than those of the CJPW. However, the hesperidin contents of the mycelia ranged from 3019.94-4980.94 mg% (except for the PC mycelium) and were 17.81-52.61% greater than the CJPW. The dietary fiber contents of the mycelia were similar to or more than those the CJPW. With the exception of PE, the electron donating abilities (EDA) and nitrite scavenging abilities (NSA) showed a tendency to decrease.

Suppression of Powdery Mildew Using the Water Extract of Xylogone ganodermophthora and Aqueous Potassium Phosphonate Solution on Watermelon under Greenhouse Conditions (Xylogone ganodermophthora 배양체 추출물 및 아인산칼륨 수용액을 이용한 시설수박 흰가루병 발생 억제효과)

  • Xylogone ganodermophthora (Xg) is an ascomycetous fungus that causes yellow rot on cultivated Ganoderma lucidum. Previously, we reported in vitro antifungal activities of a Xg culture extract against several watermelon pathogens. In 2014, we conducted greenhouse experiments to evaluate the control efficacy of a water extract of cultured Xg on watermelon powdery mildew (WPM). The test material (stock solution, ca. $4,000{\mu}g/ml$) was prepared by an autoclaved Xg culture in water at a ratio of 800 g of culture per 6 liter of water, and then filtering it through filter paper. Six foliar applications of the solutions (diluted 100- and 1,000-fold) significantly suppressed the formation of conidiophores and conidia. The inhibitory effect of aqueous potassium phosphonate solution on the disease and its phytotoxicity was tested. Phytotoxicity on watermelon plants was observed at concentrations of 1,000 and $2,000{\mu}g/ml$ as irregular brownish spots. The control efficacies against WPM were 91.9% at $2,000{\mu}g/ml$, 64.9% at $1,000{\mu}g/ml$, and 62.2% at $500{\mu}g/ml$.

Variations of leaf thickness in the Chrysanthemum zawadskii complex and in two related Korean species: C. boreale and C. indicum (Asteraceae) (국화속 구절초무리와 근연종인 산국 및 감국 에서 보이는 잎의 해부학적 특징)

  • The Chrysanthemum zawadskii complex is demarcated from other species by having the white and pink ligulate flowers. Its morphological characters are greatly diversified, so that various classification systems have been suggested. The character of leaf thickness has been mentioned as the characteristic for recognizing some of infra-specific taxa within this complex. In this study, we used longitudinal leaf sections to investigate the leaf thickness and cell number of leaf blades of 13 populations including those of the members of the C. zawadskii complex, as well as 4 populations of the related species of C. boreale and C. indicum. From the result, it was clear that the leaves were thicker in populations of C. boreale, C. indicum and C. zawadskii complex (diminishing in that order), and that the leaves were composed of about 9 cell layers in all populations. Within the C. zawadskii complex, leaf shape and thickness varied among the populations. It was very interesting that the taxa with restricted distribution, like C. zawadskii var. tenuisectum, C. zawadskii var. alpinum, C. zawadskii var. lucidum, and C. zawadskii subsp. coreanum had a thicker leaves than found among widely occurring taxa. From this, leaf thickness is supposed to be an adaptation to the unique habitat of each population.

Quality Characteristics and Antioxidant Activity of Fermented Milk containing Mushroom Extracts (버섯 추출물을 첨가한 발효유의 품질특성 및 항산화 활성)

  • This study was carried out to investigate the quality characteristics and antioxidant activity of fermented milk containing mushroom (Phellinus baumii, Ganoderma lucidum, and Lentinus edodes) extracts. As the ratio of the mushroom extract increased, the pH of the fermented milk decreased proportionally and titratable acidities increased significantly. The number of lactic acid producing bacteria was the highest in the fermented milk sample containing 1.0% Lentinus edodes extract. The DPPH and ABTS radical scavenging activities of the fermented milk containing mushroom extracts were higher than that of the controls. The quality characteristics, such as pH, titratable acidity, and the number of lactic acid producing bacteria were not remarkably different between the milk samples subjected to treatments with and without the addition of mushroom extracts during the storage period. From the sensory evaluation of the fermented milk samples containing mushroom extracts, the color, flavor, taste, texture, and overall acceptability of the fermented milk sample containing 1.0% Lentinus edodes extract was found to be considerably better than those of the other groups. In conclusion, the present study indicated that the fermented milk containing mushroom extracts could be used as a functional antioxidant containing food.

Protective effect of ethanolic extract of antler-shaped Ganoderma lingzhi against oxidative stress in PC12 neuronal cell line (PC12 신경세포주에서 녹각영지버섯 주정 추출물의 산화 스트레스 개선 효과)

  • This study was carried out to identify medicinal mushrooms with protective effects against oxidative stress in PC12 neuronal cell line, followed by evaluation of their antioxidant property. Extracts of medicinal mushrooms, including Ganoderma lucidum extract (GLE), antler-shaped Ganoderma lingzhi extract (AGLE), Hericium erinaceus extract (HEE), and Sanghuangporus baumii extract (SBE), were screened for cytotoxicity using MTT assay. None of the extracts up to $10{\mu}g/ml$ concentration affected cell viability. These extracts were further checked for their protective effect against oxidative stress-induced reactive oxygen species (ROS) production. Exposure to $50{\mu}M$ $H_2O_2$ induced ROS generation in PC12 cells, which was inhibited only by treatment with AGLE. In addition, inhibition of $H_2O_2-induced$ ROS generation by AGLE was found to be in a dose-dependent manner (2.5, 5, and $10{\mu}g/ml$). Microscopic examination of DCF fluorescence for detection of ROS showed a similar pattern. Further, antioxidant activity of AGLE was determined by ABTS radical cation assay, and its $IC_{50}$ was found to be $46.90{\pm}0.31{\mu}g/ml$. Taken together, these results suggest that AGLE may help to alleviate oxidative stress in PC12 neuronal cells.

Preparation of fermented vinegars using mushroom mycelial cultures and characterization of their physicochemical properties (버섯 균사체를 이용한 발효 식초 제조 및 이화학적 특성)

  • This study was conducted to prepare healthy vinegars using the mycelia of various mushrooms (Cordyceps militaris, Phellinus baumi, Pleurotus cornucopiae, and Ganoderma lucidum). Analyses of the bioactive components of these vinegars revealed that those prepared using mushroom mycelial cultures on fermented brown rice had increased bioactive component levels compared to those cultured on unfermented brown rice. The highest beta-glucan contents (78.7 mg/g) were found in P. baumi cultures and 0.34mg/g of cordycepin was detected in C. militaris cultures. Regarding the chemical properties of these mushroom mycelia vinegars (MMVs), those prepared using C. militaris and P. cornucopiae cultures showed higher acidity and reduced sugar content. Sensory assessments of C. militaris and P. cornucopiae MMVs yielded scores of 7.3 and 6.6, respectively. Analyses of bioactive components revealed that C. militaris MMV contained high levels of polyphenols and 786 mg/L of cordycepin. P. cornucopiae MMV contained the highest amount of beta-glucan (20.9 mg/g).

A Case of Podostroma Cornu-Damae Intoxication Induced Pancytopenia and Skin Desquamation: Successful Treatment with Granulocyte Colony Stimulation Factor (G-CFS) (과립구집락자극인자 투여로 치료한 범혈구감소증과 피부 박리를 보인 붉은사슴뿔버섯 중독 1례)

  • Podostroma cornu-damae is a rare species of fungus belonging to the Hyocreaceae family. Its fruit body is highly toxic, as it contains trichothecene mycotoxins. The morphology is similar to that of immature Ganoderma lucidum, making identification difficult for non-experts. We experienced such a case of a 56- year-old male who picked and consumed podostroma cornu-damae, and consumed. Later that day, he developed digestive system symptoms, including nausea, vomiting, and abdominal pain. He presented to the emergency room (ER), there were no abnormal physical findings, symptoms improved after gastric lavage, and the patient voluntarily discharged himself on the same day. The following day, as the symptoms gradually deteriorated, he was admitted via the ER. He was presented with severe pancytopenia, alopecia, desquamation of skin, and acute renal failure. He recovered without any complications after conservative care, antibiotics therapy, and granulocyte colony stimulating factor administration. The most commonly reported complications of podostroma cornu-damae intoxication were reported pancytopenia, infection, disseminated intravascular coagulation, acute renal failure, etc. since Prevention is especially important because its toxicity can be lethal and there is no particular treatment to date, prevention is especially important. Promotion and education for the public are needed.

Analysis of Fatty Acid Compositions and Biodiesel Properties of Seeds of Woody Oil Plants in Korea (국내 목본 유지식물 종자의 지방산 조성 및 바이오디젤 특성 분석)

  • In order to evaluate their potential as sources of biodiesel, oil content and fatty acid composition of seeds and fatty acid methyl ester (FAME) properties from seven woody oil plants in Korea were analysed. The oil content of seed of all woody plant species ranged from 15.1 (Ligustrum lucidum) to 70.3% (Camellia japonica) by dry weight. Fatty acid composition consisted mainly of oleic acid, linoleic acid, linolenic acid, palmitic acid and stearic acid, with oleic acid being the most abundant. The content of unsaturated fatty acids of all species was higher than saturated fatty acids. Oxidation stability of seed oils of all woody plants ranged from 2.25 to 8.62 hours/$110^{\circ}C$. Fatty acid methyl ester of Styrax japonica has been found to have the highest iodine value, indicating that unsaturated fatty acid content is higher than other seed oils. Cold filter plug point(CFPP) was varied over a wide range from $0^{\circ}C$ to $-13^{\circ}C$. The cold fluidity of FAME of Chionanthus retusa were excellent.
