Effects of Blends of Low-Protein Winter Wheat Flour and Barley Byproducts on Quality Changes in Noodles

  • The physicochemical characteristics of fresh noodles made with blends of low-protein wheat flour and barley byproduct (BBP, $250{\mu}m$) were investigated. The crude protein contents (PC) of flour from Goso and Backjoong cultivars were 7.91% and 7.67%, respectively. PC and ${\beta}$-glucan contents from the BBP were 14.10% and 3.11%, respectively, which were higher than those in wheat flour. The water-holding capacity (WHC) of various blends was increased as a function of BBP but not gluten contents. Goso flour had the highest starch content (78.68%), with peak and final viscosities of 3,099 and 3,563 cp, respectively. Peak and final viscosities, trough, breakdown, and setback of the blends were decreased with the addition of BBP. Noodles made with Backjoong had the highest thickness score, while the hardness of noodles made with blends of Goso or Backjoong and 20% BBP were similar to those made from wheat flour only. The WHC of the samples was strongly correlated with PC, crude fiber, and ${\beta}$-glucan. The PC was not correlated with final viscosity, setback, thickness, hardness, gumminess, or chewiness.

Effect of a Bacterial Laccase on the Quality and Micro-Structure of Whole Wheat Bread

  • The gluten protein content in whole-wheat flour is low, which affects the elasticity and viscosity of the dough. Enzymatic modification of the protein may result in a network that mimics gluten, which plays an important role in the processing of whole-wheat foods. In this study, the effects of Halomonas alkaliantartica laccase (LacHa) on the quality parameters of whole-wheat bread were investigated. The optimum dosage of LacHa was 4 U/100 g of whole-wheat flour. At this dosage, whole-wheat bread exhibited the best specific volume and optimum texture parameters. Laccase also extended the storage duration of whole-wheat bread. We analyzed the micro-structure of the dough to determine its gluten-free protein extractable rate and free sulfhydryl group content, and verify that LacHa mediates cross-linking of gluten-free proteins. The results demonstrated that the cross-linking of gluten-free protein by LacHa improves the texture of whole-wheat bread. As a flour improver, LacHa has great developmental and application potential in baked-food production.

Effect of Nitrogen Fertilizer Application on Yield and Quality of Korean Soft Wheat Cultivar 'Goso'

  • Wheat flour can be categorized into bread, all-purpose, cake flour according to its protein content. Since optimal wheat flour protein content is different for each end use, it is necessary to diversify the nitrogen fertilizer methods depending on the end use and cultivar. Optimal wheat flour protein content of soft wheat (for cake flour) is lowest (<=10%) among all end use, it is necessary to develop nitrogen fertilizer methods for high yield and low protein content. In order to analyze the yield and quality changes of soft wheat as nitrogen fertilizer amount and splitting timing, soft wheat cultivar 'goso' was sown on paddy soil in jeunju, Republic of Korea ('21.10). the amount of nitrogen fertilizer was divided into 4 levels by adjusting 2kg/10a increments from 5.1 to ll.lkg/lOa, and in the N 7.1 and 9.1 kg/1 Oa(standard) treatment, N amount divided into sowing date:regrowing stage=3:7,4:6(standard), 5:5. In regrowing stage, Tiller number and N fertilizer amount at sowing date showed a correlation; y=-121.14x2+792.66x-525.41 (R2=0.77*, y: Tiller number/m2, x: N amount at sowing date(kg/10a)). Tiller number in regrowing stage was the highest when the nitrogen fertilizer amount at sowing date was 3.23kg/10a. spike number per m2 was the highest when N fertilizer was divided into sowing date:regrowing stage=3:7(N amount: 9.1kg/10a). If N fertilizer amount was fixed, grain yield was also the highest when N fertilizer was divided into sowing date :regrowing stage=3:7. Also, N amount at sowing date and grain yield showed no correlation, but N amount at regrowing stage and grain yield showed significant correlation. As N amount increased, protein content also showed a tendency to increase.

Differential induction of allergy responses by low molecular weight wheat proteins from six wheat cultivars

  • Although wheat is a common staple food in the world, some people suffer from a variety of wheat allergies. For example, wheat-dependent exercise-induced anaphylaxis is induced in the gastrointestinal tract by wheat proteins. Relatively high molecular weight proteins that are salt-insoluble induce many wheat allergies. In the present study, we investigated the induction of an allergy response using crude wheat proteins, which are relatively low molecular weight, salt-soluble proteins. The crude antigen used in this study was extracted using phosphate buffered saline. When the antigen extracts from various wheat cultivars were orally administered, differentiable degrees of allergy responses were observed as measured by serum IgE and histamine secretion compared to the control. Serum IgE levels increased following administration of three of the wheat extracts. This evidence suggests that a combination of salt-soluble wheat proteins could be antigens for the induction of various allergy responses.

Isolation of Cold Acclimation-related Genes in Wheat Chromosome Substitution Line 5D

  • To identify low temperature-induced genes of wheat chromosome substitution line 5D, suppression subtractive hybridization (SSH) was performed with mRNAs from leaf samples that treated with low temperature ($4^{\circ}C$). A cDNA library was constructed using mRNA isolated from wheat chromosome substitution line 5D leaves treated with low temperature ($4^{\circ}C$). The nucleotide and deduced amino acid sequences of the putative gene products were compared. wfr-9 and wfr-32 showed identity over 90% related to vernalization gene. Other two genes, wfr-77 and wfr-83 which is related to freezing-resistant gene have also identity over 90%. This result suggest that those genes may be transcribed into antifreeze proteins which are accumulated within leaf apoplasts, when wheat chromosome substitution line 5D is acclimated during low temperature treatment.

The Effect of Variety and Growing Conditions on the Chemical Composition and Nutritive Value of Wheat for Broilers

  • The aim of this study was to examine the effect of variety and growing conditions of wheat on broiler performance and nutrient digestibility. One hundred and sixty-four wheat samples, collected from a wide range of different sources, locations, varieties and years, were analyzed for a range of chemical and physical parameters. Chemical and physical parameters measured included specific weight, thousand grain weight (TG), in vitro viscosity, gross energy, N, NDF, starch, total and soluble non-starch polysaccharides (NSP), lysine, threonine, amylose, hardness, rate of starch digestion and protein profiles. Ninety-four of the wheat samples were selected for inclusion in four bird trials. Birds were housed in individual wire metabolizm cages from 7 to 28 d and offered water and feed ad libitum. Dry matter intake (DMI), live weight gain (LWG) and gain:feed were determined weekly. A balance collection was carried out from 14 to 21 d for determination of apparent metabolizable energy (AME), ME:gain, DM retention, oil and NDF digestibility. At 28 d the birds were sacrificed, the contents of the jejunum removed for determination of in vivo viscosity and the contents of the ileum removed for determination of ileal DM, starch and protein digestibility. The wheat samples used in the study had wide-ranging chemical and physical parameters, leading to bird DMI, LWG, gain:feed, ME:GE, AME content and ileal starch and protein digestibility being significantly (p<0.05) affected by wheat sample. A high level of N fertilizer application to the English and NI wheat samples tended to benefit bird performance, with increases of up to 3.4, 7.2 and 3.8% in DMI, LWG and gain:feed, respectively. Fungicide application also appeared to have a positive effect on bird performance, with fungicide treated (+F) wheat increasing bird DMI, LWG and gain:feed by 6.6, 9.3 and 2.7%, over the non-fungicide treated (-F) wheats. An increase (p<0.1) of 9.3% in gain:feed was also observed at the low seed rate of 40 compared to 640 seeds/$m^2$. It was concluded that the type of wheat sample and environmental growing conditions significantly affects bird performance when fed wheat-based diets.

Evaluation of Grain Zinc and Iron Contents of Wheat Germplasm

  • Wheat is the staple food crop in the word, but wheat products have a low bioavailability of iron and zinc. So in the developing world, where wheat is a staple food, it suffers from micronutrients deficiency. This study was conducted to generate wheat varieties with enhanced grain Zn and Fe contents. Sixty wheat resource were cultivated over 2 years (2019-2021) in the field of NICS, Jeonju, Republic of Korea, to identify agronomic traits. Wheat grains were ground using grinder and analyzed whole wheat flour protein contents and Fe and Zn contents using ICP-OES. The average contents of Zn and Fe grain were 4.6 mg/100g (2.4~8.8 mg/100g) and 4.5 mg/100g (2.4~7.9 mg/100g), respectively. The contents of Fe and Zn in the wheat grain had a positive correlation with the protein content of whole wheat flour, but there was no correlation with heading date (4.22~5.27) and the thousand kernel weight (21.3~57.5 g). Although there was year variation, six resources with high contents of Fe (>5.2 mg/100 g) and Zn (>5.3 mg/100 g) grain in 2 years were selected. These results provide information for selecting breeding materials for biofortified wheat, and further studies on germplasms genetic variations and bioavailability are needed.

환경 변동에 따른 경ㆍ연질 소맥의 등숙 및 품질의 변화에 관한 연구 (Studies on Grain Filling and Quality Changes of Hard and Soft Wheat Grown under the Different Environmental Conditions)

  • 본연구는 1972년에 수원에서 그리고 1973년에는 다시 수원, 매리 및 이리 광주의 3개 지역에서 경질 소맥인 NB 68513, Caprock와 중간질 소맥인 영광, 연질 소맥인 수계 169호를 공시하여 시비량 및 재배시기를 각각 3수준의 처리를 두어 등숙 및 품질의 변화에 관한 실험을 수행하였다. 한편 1973년 및 1974년에는 수원에서 중간질 소맥인 영광과 경질 소맥인 NB 68513을 공시하고 온실에서 온도, 습도 및 일사량을 달리하여 이들이 소맥의 등숙과 품질에 미치는 영향을 추구하였으며, 얻어진 결과는 다음과 같다. 1. 소맥립의 등숙: 1) 1립중의 변이폭은 대립종이 소립종에 비하여 크고, 립중별 소맥립의 분포는 대립종이 평균치에 가까이 분포된 율이 높았고, 소립종은 넓은 분산을 보였다. 2) 립중의 증가에 미치는 립장, 립후 및 립폭의 영향의 정도는 립장보다 립후와 립폭이 컸다. 3) 등숙 시기별 립중의 변화에 있어서 영광은 개화후 14일부터 35일까지, NB 68513은 14일부터 28일까지에 급증하였고, 대립종인 영광은 소립종인 NB68513에 비하여 등숙기간이 길었으며 배란비율도 완만한 증가를 보였다. 4) 1000립중은 대체로 저온에서보다 고온에서, 다습보다 건조한 공중습도 조건에서 가벼웠고, 저온이나 고온에서라도 다습 조건하에서는 무거운 경향을 보였다. 또한 서광의 영향은 저온에서보다 고온에서 컸으며, 다습한 경우에는 일정한 경향이 없었다. 5) 등숙기간중 온도, 습도 및 서광의 영향은 소립종인 NB 68513보다 대립종인 영광에서 컸으며, 1000립중의 증감과 등숙일수 간에는 높은 정의 상관이 있었다. 6) 1000립중과 1$\ell$중은 시비량의 증가에 따라 무거워졌으나, 보비와 다비구간에는 그 증가율이 비교적 낮았고, 조숙재배는 만파재배보다 무거웠으며, 이와 같은 경향은 수원에서 현저하였고 광주 및 이리에서는 미미하였다. 2. 제분성: 7) 동일 품종에서 1000립중이 무겁게 나타난 저온다습 조건 및 조숙재배가 고온, 건조, 조건 및 만파재배에 비하여 제분율이 높았고, 지역간 차이는 일정한 경향이 없었다. 8) 제분율은 영광이 가장 높고 수계 169호가 가장 낮았으며, 경질 소립종인 Caprock, NB 68513은 연질 소립종인 수계 169호보다 높았다. 9) 회분 함량은 지역, 시비량 및 재배시기에 따른 차이는 작았고, 품종간 차이는 컸으며, NB 68513 및 Caprock는 영광 및 수계 169호보다 현저히 높았다. 3. 단백질 함량: 10) 단백질 함량은 1000립중이 가벼웠든 고온, 건조 및 서광 조건에서 저온, 다습 조건에서보다 높았으며, 이러한 경향은 영광에서 현저하고 NB 68513에서 적었다. 11) 종질의 단백질 함량은 개화후 1~2주일 사이에 높았는데, 이는 배란에 대한 배와 종피의 비율이 크기 때문이었고, 등숙이 진전됨에 따라 배, 종피의 영향은 감소되며, 달서 단백질 함량도 감소되었으나 개화후 3~4주부터는 다시 단백질함량이 증가되어 개화후 7주에 최대에 달하였다. 등숙 중기(개화후 3~4주) 이후의 단백질함량 증가는 대체로 1000립중의 증가와 비슷하였으나, 성숙된 종실의 단백질 함량의 증감은 등숙기간의 장단의 영향보다는 역일상에 따른 기상적 환경, 특히 기온에 의한 영향을 많이 받은 것으로 보였다. 12) 소맥분의 단백질 함량은 종실의 단백질 함량의 변화와 같아서 이 양자간에는 높은 정의 상관이 있었다. 13) 단백질 함량은 각 품종 모두 시비량이 증가할수록, 또 재배시기가 늦어질수록 증가하는 경향이었고, 수원에서는 광주, 이리에서보다 그 경향이 현저하였다. 14) 품종별로 존 단백질 함량은 영광, NB 68513 및 Caprock은 비슷하였으며 비교적 높았고, 수계 169호는 낮았는데 이 품종은 재배법에 따른 단백질 함량의 변이도 적었다. 15) 단위 면적당 단백질 수량은 시비량이 많을수록, 재배시기가 빠를수록 높은 경향이었고, 조숙재배에서는 질소질 비료의 이용율이 높았으며, 영광은 시비량 증가에 따른 단백질 수량 증가가 비교적 컸다. 4. 분의 물리화학적 특성: 16) Sedimentation value는 저온, 다습, 소비 조건에서보다 고온, 건조, 다비 조건에서 컸고, NB 68513 및 Caprock는 그 경향이 현저하였는데 영광 및 수계 169호는 뚜렷하지 않았었고, 광주, 이리보다는 수원에서 Sedimentation value의 변이가 컸다. Sedimentation value의 증감은 단백질 함량의 증감과 관계가 깊으나, 다습조건에서는 Sedimentation value가 감소되었다. 한편 Sedimentation value는 단백질 함량의 증가에 따라 증가되었으며, 성숙기에 최대에 달하였다. 17) Pelshenke value는 재배방법 및 지역 간의 차이가 Sedimentation value의 경우와 대체로 같은 경향을 보였다. 18) Mixing time은 NB 68513이 4~6분, Caprock가 5~7분 소요되었으며, 영광 및 수계 169호는 NB 68513 및 Caprock보다 지역 및 재배법에 따른 변이가 컸다. Mixing height와 Mixing area는 NB 68513 및 Caprock에서 컸고 영광 및 수계 169호에서는 작았으며, 재배방법에 따른 변이는 일정한 경향이 없었고, 지역에 따른 차이는 이리, 광주에서 낮고 수원에서 높았다. 19) NB 68513 및 Caprock의 품질에 있어서 제분성은 고온, 건조 조건, 또는 다비, 만파재배에서 떨어졌으나 분의 물리화학적 특성은 양호하여 제빵 적성이 좋고, 조숙재배는 물리화학적 특성이 다소 불량하였으나 제분성이 높고 단위 면적당 단백질 수량인 높은 경향이었다. 지역간에서 보면 이리, 광주보다는 수원이 NB 68513 및 Caprock의 재배에 적합한 것으로 판단되었다. 5. 분의 물리화학적 특성의 상호관계: 20) 제분율 및 회분함량과 분의 물리화학적 특성과는 직접적인 상관이 없었으며, 1000립중이 가벼운 것이 단백질 함량이 높았고 분의 물리화학적 특성도 양호하였다. 21) NB 68513 및 Caprock에 있어서 단백질 함량과 Sedimentation value, Pelshenke value 및 mixing height와는 정의 상관이 있었으며, 단백질 함량이 높으면 분의 Gluten strength도 강하고 따라서 제빵 적성도 양호하였다. 영광 및 수계 169호에 있어서는 단백질 함량과 Sedimentation value와는 정의 상관이 있으나 Pelshenke value와 Mixing height와는 상관이 없었다. 따라서 단백질 함량 증가에 따라 Gluten strength 양자와 연결됨이 크고, Pelshenke value와 Mixogram은 Gluten strength와 연관됨으로 경질과 연질의 품종 구분에는 Mixogram, Pelshenke value의 검정이 유리하고, Sedimentation valuer검정은 같은 품종내에서 재배법 차이에 따른 품질 평가에 알맞은 것으로 보았다.

곡류 및 두류 단백질이 흰쥐의 성장 발육에 미치는 영향 (A study of the Body Growth and Development in Albino Rats Fed by the Cereals and Legumes Proteins)

  • This study was designed to compare the Biological effects of the cereals and legumes Proteins on albino rats. Fifty weaning albino rats weighing $49\;gr{\pm}2$ in both sexes were divided into 5 groups and fed corn crude protein diet, wheat crude protein diet, soybean protein diet, yeast diet and 30% sugar-casein diet for the control group. The protein contains same levels with isocaloric values each diet. After 12 weeks the rats were sacrificed for chemical analysis and the results are as follow ; 1. The highest food consumption was found in the groups of yeast and soybean, and the lowest was found in the groups of wheat diet and corn diet. 2. The groups of yeast & soybean protein showed the highest body weight increase, while the groups of wheat diet and corn diet showed the lowest. 3. Highly significant difference was found between the standard group and the groups of corn protein and soybean protein in Food Efficiency Ratio. (F. E. R.) (p<0.01). 4. Protein Efficiency Ratio showed a similar pattern as the F.E.R., however, there was no significant differences among the groups. 5. The kind of diets did not influence the hematology of the subjects. 6. The rate of nitrogen retention of male standard group was lower compared with all the experimental groups (p<0.01), and that of female soybean group was higher than any other groups (p<0.01). 7. Female corn diet group had the lowest organ weights, as found in the growth rate, which was significant (p<0.01). In the male corn diet group only the kidney showed significantly low in weight (p<0.01), and the spleen of male yeast group was also shown low with significance (p<0.01). 8. Nitrogen retentions of the liver and muscle in male corn diet group were lower than any other groups (p<0.05), and the brain nitrogen content of female standard group was high with significance (p<0.01). According to the results above, yeast and soybean protein can be regarded as a nutritious and also inexpensive protein sources.

고농도 소맥 글루텐의 효소적 가수분해와 약산에 의한 전처리 효과 (Effect of Weak Acid Pretreatment on the Enzymic Hydrolysis against Wheat Gluten of High Concentration)

  • To determine the optimum conditions for the enzymic hydrolysis against wheat gluten of high con centrations (6~14%, w/w, protein), a hydrolysis system combining weak acid pretreatment and enzymic hydrolysis was investigated. Alcalase showed the highest DH(degree of hydrolysis) of the tested proteases. After hydrolysis by alcalase, subsequently peptidases were applied for the better DH of the wheat gluten hydrolyzate. Peptidase NP2 showed the highest DH of the tested peptidases, but flavour zyme was shown for the lowest bitter taste of the resulting hydrolyzate. In order to minimize aggregation or gelling at higher initial substrate concentration during heat treatment, wheat gluten suspension was pretreated with possibly low concentrations of hydrochloric acid at 105oC for 1 hour, and then enzy matically hydrolysed with alcalase and subsequently with flavourzyme. Each required minimum concen tration of hydrochloric acid in the wheat gluten suspension of 6, 8, 10, 12, and 14%(w/w, protein) was 0.10, 0.15, 0.20, 0.225, and 0.275N, respectively. After the subsequent enzymic treatment by alcalase and peptidase NP2 for 24 hrs, the nitrogen solubility in the final wheat gluten hydrolysates was increased to 94.9, 86.4, 85.3, 89.3 and 95.0%, and their amino nitrogen content was increased to 2.87, 5.68, 7.34, 9.71 and 12.50mg/m, respectively.
