정상 성인의 혈청 인슐린 농도에 따른 비만지수, 혈청 지질, 혈압 및 영양소 섭취상태 비교 (Comparisons of BMI, Blood Lipid Profile, Blood Pressure and Nutrient Intakes by Serum Insulin Concentrations)

  • This study was conducted to figure out the differences of the BMI, blood lipids, blood pressure and nutrient intakes by serum insulin concentration among adults in Korea. Ninety-three subjects(male : n=37, female : n=56) participated in this study. The subjects were grouped by serum insulin concentrations into two groups-hyperinsulinemia group(n=17) and normoinsulinemia group(n=76). Anthropometric measurements, blood lipid profiles, blood pressure, oral glucose tolerance test, and daily nutrient intakes were analyzed. And serum glucose and insulin secretion pattern as shown through and oral glucose tolerance test were performed. BMI (p<0.05) and WHR(p<0.01) were significantly higher in hyperinsulinemic group than in normoinsulinemia group. There was no differences in fasting blood glucose level between groups, but total glucose area (p<0.001) and insulin glucose resistance(p<0.001) were significantly higher in hyperinsulinemia group than in normoinsulinemia higher in hyperinsulinemia group, but HDL-cholesterol(p<0.05), and systolic blood pressure(p<0.05) were significantly higher in hyperinsulinemia group, but HDL-cholesterol(p<0.001) was significantly lower in hyperinsulinemia group. They showed significant differences in energy, carbohydrate, potassium, riboflavin, niacin and dietary fiber intakes(p<0.05), the intake of those nutrients were low in normoinsulinemia group. Further investigation is necessary to determine the effects of amounts and types of carbohydrate and dietary fiber on serum insulin concentrations.

Photochemical Response in 0-Year-Old and 1-Year-Old Needles of Picea glehnii during Cold Acclimation and Low Temperature

  • P. glehnii, an evergreen conifer found in northern areas, is known as a cold-resistant species. In this experiment, we measured the water content, PSⅡ efficiency, chlorophyll fluorescence, pigments of the xanthophyll-cycle and activity of enzymes of the ascorbate-glutathione cycle during cold acclimation and at subsequent low-temperature conditions to examine the importance of acclimation to cold tolerance. P. glehnii showed a decrease in PSⅡ efficiency (especially in Fv) during cold acclimation and at subsequent low temperatures. However, cold-acclimated needles showed higher PSⅡ efficiency at low temperatures than nonacclimated needles. In addition, 0-YON (first-year needles) showed an increase in $\beta$-carotene and lutein, while 1-YON (one-year-old needles) immediately developed an antioxidant mechanism in the ascorbate-gluthathione cycle as soon as they were exposed to low temperature and both 0-YON and 1-YON showed increased zeaxanthin and de-epoxidation ratios at continuous low temperature. Based on our results, we suggest that P. glehnii maintain PSⅡ efficiency at low temperature by effectively protecting the photosynthetic apparatus from photo-damage by rapid induction of an antioxidant mechanism in 1-YON and dissipation of excess energy by $\beta$-carotene and lutein in 0-YON.

Functional Properties of Yogurt Containing Specific Peptides derived from Whey Proteins

  • The purpose of this study was to investigate the acid tolerance, bile acid tolerance, and fermentation activity of lactic acid bacteria isolated from Kimchi in the presence of hydrolysates of whey protein concentrate. Kimchi isolates DK109, DK119, DK121, DK128, DK211, DK212, and DK215, which were identified as Lactobacillus sp., and L. casei DK128 showed the highest acid and bile acid tolerance. To produce whey hydrolysates, enzymes were added to a 10% (w/v) whey protein concentrate (WPC) solution at 1:50 (w/v, protein). The viabilities of the DK strains were determined in the presence of low pH and bile salts. Then, yogurt was produced via fermentation with L. casei DK128, an isolate from Kimchi, in the presence of the following additives: CPP, WPC, and WPC hydrolysates (WPCH) generated by alcalase (A) or neutrase (N). The produced yogurts were subjected to various analyses, including viable cell counts (CFU/mL), pH, titratable activity, and sensory testing. After 8 h of fermentation, the pH and titratable activity values of all test samples were 4.2 and 0.9, respectively. The viable counts of LAB were $3.49{\times}10^8$, $5.72{\times}10^8$, $7.01{\times}10^8$, and $6.97{\times}10^8$, for the Control, CPP, A, and N samples, respectively. These results suggest that whey proteins have potential as dietary supplements in functional foods and that WPCH could be used in yogurt as a low-cost alternative to CPP.

  • Salt tolerances of Chinese cabbage and cabbage were observed by means of the sand culture and field experiment. The point of 50% yield reduction of Chinese Cabbage was 1% of salt concentration in sand culture. The Na absorption in the salty upland conditions was increased but the absorption of Ca, Mg were interrupted as the salt concentration was raised and there were no differences in the absorption of N and P. The Si absorption was increased at low salty conditions, but the salt concentration was raised, the absorption was interrupted drastically. The cabbage was more stronger salt tolerance than Chinese cabbage, and it was possible to prevent the salt damage significantly by planting on sloping beds instead of planting on the double-row beds in field condition.

  • P-V 곡선법(曲線法)을 적용(適用)하여 20종(種)의 낙엽활엽수(落葉闊葉樹)에 대한 내건성진단(耐乾性診斷)을 하였다. 엽(葉)의 수분특성인자(水分特性因子) 중 내건성판별(耐乾性判別)에 적합(適合)한 인자(因子)는 최대포수시(最大飽水時)의 삼투(滲透)포텐셜 (${\Psi}_0{^{sat}}$), 초기원형질분리점(初期原形質分離點)의 삼투(滲透)포텐셜(${\Psi}_0{^{tlp}}$), 세포막(細胞膜)의 최대탄성계수(最大彈性係數)($E_{max}$), 초기원형질분리점(初期原形質分離點)의 상대함수율(相對含水率)($RWC^{tlp}$)였으며, 그 밖에 상대함수율(相對含水率)(FWC)와 워터포텐셜(${\Psi}_L$)과의 관계, 팽압(膨壓)($P_{vat}$)과 ${\Psi}_L$과의 관계, H$\ddot{o}$fler diagram등의 그림을 내건성진단(耐乾性診斷)에 사용하였다. 이와같은 수분특성인자(水分特性因子)로 고찰(考察)할 때 물푸레나무, 상수리나무, 졸참나무, 갈참나무, 현사시등은 비교적 내건성(耐乾性)이 높은 수종(樹種)으로, 들메나무, 자작나무, 이태리포플러, 음나무, 서어나무, 까치박달, 산벚나무, 개벚나무, 층층나무등은 비교적 내건성(耐乾性)이 약(弱)한 수종(樹種)으로, 그리고 신갈나무, 고로쇠, 복자기, 당단풍, 느릅나무, 느티나무등은 중간(中間) 수종(樹種)으로 판별(判別)되었다.

저온 충격에 노출된 참돔 Pagrus major 치어의 임계 저 수온 및 행동 내성 반응 (Critical Low Temperature and Response of Behavioral Tolerance in Red Seabream Pagrus major fingerlings Exposed to Cold Shock)

  • 본 연구는 연속행동모니터링시스템(CBMS)을 사용하여 저온 충격에 노출된 참돔 Pagrus major 치어의 행동반응에 대한 임계 수온 및 행동 내성 반응을 규명하였다. 실험생물의 행동은 8.0~20.0℃ 수온구간에서 12시간과 24시간 마다 각각 2.0℃와 4.0℃씩 하강시키며 관찰하였다. 참돔 치어는 12.0℃ 이하로 수온이 하강한 경우 수온 노출시간과 변동 폭에 관계없이 유영활동이 감소하고 정지가 반복되는 불안정한 행동패턴이 관찰되었다. 또한 8.0~10.0℃ 수온에 노출된 생물의 유영능력은 급격히 저하되어 수조 바닥에서 머무르는 아치사 반응 행동을 보였다. 실험어류는 저 수온 스트레스 영향을 받아 생물의 50 %만 생존하였으며, 저온 충격(cold shock) 후 6시간 이내에 모든 개체가 사망하였다. 또한 행동지수(BI)는 급격히 감소하였으며 변이계수(CV)의 진폭변화는 타 수온보다 변동 폭이 큰 것으로 분석되었다(p<0.05). 참돔 치어의 저온 스트레스는 12.0℃를 경계로 촉진되며, 8.0~10.0℃의 저온 충격에 노출된 생물의 아치사 반응을 유발시키는 내성한계인 것으로 해석된다.

Downregulation of PyHRG1, encoding a novel secretory protein in the red alga Pyropia yezoensis, enhances heat tolerance

  • An increase in seawater temperature owing to global warming is expected to substantially limit the growth of marine algae, including Pyropia yezoensis, a commercially valuable red alga. To improve our knowledge of the genes involved in the acquisition of heat tolerance in P. yezoensis, transcriptomes sequences were obtained from both the wild-type SG104 P. yezoensis and heat-tolerant mutant Gy500. We selected 1,251 differentially expressed genes that were up- or downregulated in response to the heat stress condition and in the heat-tolerant mutant Gy500, based on fragment per million reads expression values. Among them, PyHRG1 was downregulated under heat stress in SG104 and expressed at a low level in Gy500. PyHRG1 encodes a secretory protein of 26.5 kDa. PyHRG1 shows no significant sequence homology with any known genes deposited in public databases to date. However, PyHRG1 homologs were found in other red algae, including other Pyropia species. When PyHRG1 was introduced into the single-cell green alga Chlamydomonas reinhardtii, transformed cells overexpressing PyHRG1 showed severely retarded growth. These results demonstrate that PyHRG1 encodes a novel red algae-specific protein and plays a role in heat tolerance in algae. The transcriptome sequences obtained in this study, which include PyHRG1, will facilitate future studies to understand the molecular mechanisms involved in heat tolerance in red algae.

Responses to Starch Infusion on Milk Synthesis in Low Yield Lactating Dairy Cows

  • The effect of starch infusion on production, metabolic parameters and relative mRNA abundance was investigated in low yield lactating cows from 86 days in milk. Six Holstein cows fitted with permanent ruminal cannulas were arranged into one of two complete $3{\times}3$ Latin squares and infused with a starch solution containing 800 grams starch for 16 days. The three treatments were: i) ruminal and abomasal infusion with water (Control); ii) ruminal infusion with cornstarch solution and abomasal infusion with water (Rumen); iii) ruminal infusion with water and abomasal infusion with cornstarch solution (Abomasum). There were no significant differences (p>0.05) among the three treatments with low yield lactating cows in feed and energy intake, milk yield and composition, plasma metabolism, or even on gene expression. However, cows receiving starch through rumen performed better than directly through the abomasum during the glucose tolerance test procedure with a higher area under the curve (AUC; p = 0.08) and shorter half-time ($t^{1/2}$; p = 0.11) of plasma insulin, therefore, it increased glucose disposal, which stated a lipid anabolism other than mobilization after energy supplementation. In conclusion, extra starch infusion at concentration of 800 g/d did not enhance energy supplies to the mammary gland and improve the lactating performance in low yield lactating cows.

Genetic improvement of potato plants

  • Genetic improvement in potato can be carried out through several approaches, as sexual crosses, somatic hybridization, mutation and genetic engineering. Although the approach is different, but the goal is the same, to get a superior cultivar. Mutation and genetic engineering are very interesting methods for genetic improvement of potato plants. Mutation by gamma-ray irradiation have been performed to get some new potato cultivars which are more resistant to disease and have higher productivity. We have carried out a mutation of some potato cultivars and obtained some excellent clones to be potentially released as new superior cultivars. By the mutation method, we have released one potato cultivar for the French fries industry, and we registered one cultivar of potato for chips, and two cultivar for vegetable potatoes. Actually we are doing multi-location trial for three clones to be released as new cultivars. Through genetic engineering, several genes have been introduced into the potato plant, and we obtained several clones of transgenic potato plants. Transgenic potato plants containing FBPase gene encoding for fructose bisphosphatase, have a higher rate of photosynthesis and higher tuber productivity than non-transgenic plants. This result suggests that FBPase plays an important role in increasing the rate of photosynthesis and potato tuber productivity. Some transgenic potatoes containing the Hd3a gene are currently being evaluated for their productivity. Over expression of the Hd3a gene is expected to increase tuber productivity and induce flowering in potatoes. Transgenic potato plants containing MmPMA gene encoding for plasma membrane ATPse are more tolerant to low pH than non-transgenic plants, indicating that plasma membrane ATPase plays an important role in the potato plant tolerance to low pH stress. Transgenic potato plants containing c-lysozyme genes, are highly tolerant of bacterial wilt diseases caused by Ralstonia solanacearum and bacterial soft rot disease caused by Pectobacterium carotovorum. Expression of c-lyzozyme gene plays an important role in increasing the resistance of potato plants to bacterial diseases.

Impact of Position on Efficacy of Caudal Epidural Injection for Low Back Pain and Radicular Leg Pain Due to Central Spinal Stenosis and Lumbar Disc Hernia

  • Objective : This study was performed to evaluate and compare the efficacies of caudal epidural injections performed at prone and lateral decubitus positions. Methods : A total of 120 patients suffering from low back pain and radicular leg pain were included and patients were randomly distributed into 2 groups according to the position during injection. In Group 1 (n=60; 32 women, 28 men), caudal epidural injection was performed at prone position, whereas it was implemented at lateral decubitus position in Group 2 (n=60; 33 women, 27 men). Visual analogue scale, Oswestry Disability Index (ODI), walking tolerance (WT) and standing tolerance (ST) were compared in 2 groups before and after injection. Results : In Group 1, ODI values were higher at 30th minute (p=0.007), 3rd week (p=0.043) and 6th month (p=0.013). In Group 1, ODI, VAS and ST values were improved significantly at all follow-up periods compared to initial values. In Group 1, WT scores were better than initial values at 30th minute, 3rd week and 3rd month. In Group 2, ODI scores at 30th minute, 3rd week, 3rd month and 6th month were improved while VAS and ST scores were improved at all periods after injection. WT scores were better at 30th minute, 3rd week and 3rd month compared to initial WT scores. Conclusion : Our results indicated that application of injection procedure at lateral decubitus position allowing a more concentrated local distribution may provide better relief of pain.
