Preparation of Novel Quaternary Plastomers with Extremely Low Glass Transition Temperature

  • In this study, novel quaternary plastomers consisting of ethylene, 1-hexene, high ${\alpha}$-olefin, and divinylbenzene were prepared using Zr metallocene catalyst, borate type cocatalyst, and tri-isobutylaluminium. The molar ratio changes of 1-hexene and high ${\alpha}$-olefin (1-octene, 1-decene, and 1-dodecene) had an effect on the properties of the quaternary plastomers. The structure of the quaternary plastomers was characterized using $^1H$ NMR. Molecular weight properties, crystallinity, and thermal properties of the plastomers were determined by GPC, WAXS, and DMA, respectively. Compared with the terpolymers prepared in our previous study, molecular weight and molecular weight distribution of the quaternary polymers were very similar, whereas glass transition temperature ($T_g$) was very low. Also, hardness and tensile properties of the quaternary plastomers were measured.

  • Alkaline bacterium producing a high pro-tease activity at low temperature was isolated by using enrichment culture from soil samples and identified as Bacil-lus sp. WRD-1 Cell growth was maximal at 10 hours and the optimal initial pH and culture time of culture condition for enzyme production was pH 7 and 10 hours, respectively. Temperature range of high enzyme activity were $10~40^{\circ}C$. The optimal pH and temperature for the enzyme activity were pH9 and $30^{\circ}C$.

Effect of Low Temperature Pretreatment on Pollen Dimorphism and Embryo Formation in Anther Culture of Platycodon grandiflorum (도라지 (Platycodon grandiflorum) 약배양에서 저온처리가 화분 2형현상 및 배형성에 미치는 영향)

  • In order to investigate the effect of low temperature pretreatment on pollen dimorphism and embryo formation in anther culture of Platycodon grandiflorum, the anthers with microspore at the uninucleate stage were cultured on Murashige and Skoog medium supplemented with 0.5mg/L NAA and 1.0 mg/L BA. The low temperature pretreatment have clear effect on the frequencies of S pollen grains, symmetrical binucleate microspores (B type of S pollen), multinucleate and multicelled pollen grains. Especially, after low temperature pretreatment at 8$^{\circ}C$ for 5 days increased the frequency of S pollen grain (20.6%) in vivo. In addition, the highest frequency of callus induction (54.9%) and embryo formation (9.9%) were obtained from the anther pretreatment at 8$^{\circ}C$ or 5 days. Three distinct pathways could be recognized in the androgenesis, one involving mainly the vegetative cell, the second starting with the vegetative and the generative cell, respectively, and the third accompaning with two equal vegetative type cells in the pollen grains.

Characterization of Low Temperature-adapted Leuconostoc citreum HJ-P4 and Its Dextransucrase for the Use of Kimchi Starter

  • Leuconostoc citreum HJ-P4 is a strain isolated for kimchi fermentation with its low temperature-adapted growth feature and its high dextransucrase activity. The detailed characteristics of cell growth and dextran sucrase activities were investigated at various environmental conditions such as temperatures, pHs, salts, and raw ingredients. This strain showed almost 2-fold higher maximal cell concentration ($X_{max}$) than that of the type culture Leuconostoc mesenteroides B-512F at $10^{\circ}C$. The $X_{max}$ of the strain was maximum at pH 7 and the cell growth was inhibited by salts in a dose-dependent mode up to 7%. Addition of pepper (<6%), garlic (<10%), and ginger (<2%) in kimchi gave no inhibition effect on the growth of HJ-P4. Dextransucrase synthesized by this strain retained over 80% of its maximum activity at $10^{\circ}C$ showing a comparable cold-adapted feature to its host microbe. This culture can be used as a starter culture in the industrial kimchi production giving desirable functions and predominance at low temperature.

Variation of Immature Kernel as Affected by Growth Temperature and Grain -filling Period, and of Seedlings Obtained from Barley Embryo Culture for Shortening Generation (보리 생육온도조건과 등숙기간에 따른 미숙종자의 특성 및 세대촉진을 위한 배배양시 유묘 생육의 변이)

  • In relation to shortening generation by using embryo culture of barley immature seed, it is important to find out profitable embryo age for embryo culture and to understand the relationship among embryo and other characters. Two varieties(Olbori and Dusan #12) were cultivated under two growth conditions(15/10 and 25/15$^{\circ}C$). The embryos were aseptically excised when immature seeds were collected 9, 13, 17, 21, 25 and 29 days after heading and cultured on $B_5$ medium. On the 21st day after heading, the length of embryos from top or middle part of spike was longer than that from bottom part. Embryos from bottom part under low temperature condition had the shortest length. Shoot length, root length and root number after embryo culture were little difference among three parts of spike under high temperature condition. Under low temperature, seedlings from bottom part of spike were inferior to those from top or middle part. Length of 29-day-old embryos under low temperature condition was similar to that of 17-day-old ones under high. Under high temperature condition, the length of 17-day-old embryo had positive correlation with kernel width, shoot length, root length and root number, but that of 21-day-old one didn't have. Seventeen-day-old embryos obtained from 25/15$^{\circ}C$ growth condition seem to be efficient to shortening generation by using embryo culture.

  • Leaf dise explants of Solanum tuberosum cultivar. Desiree and Atlantic, were infected with a Agrobacterium MP90 strain containing chimeric gene construct, consisting of antibiotic and low temperature regulated gene (H28) for transformation. regenerated multiple shoots were selected on a medium containing kanamycin and carbenieillin after exposure to Agrobacterium. Both PCR analysis of NPT Ⅱ, H28 genes and northern blot analysis indicated that the genes coding for the enzyme were successfully integrated into the potato genome and could be expressed in potato plants.

  • The promotive effect of Ca in semi-vitro culture of Oenothera biennis pollen was by and large identical to that of in vitro systems. This promotive effect became additive when boron was supplemented to the media and even more so if K was further supplemented. No such Ca action was observed then performed under the conditions of relatively low temperature. An uncoupling agent of ATP in respiration, DNP, inhibited the promotive action of B, but not that of Ca. The inhibitory effect of DNP was greater at low temperatures. IAA was rather inhibitory on pollen growth in semi-vitro culture, even much greater than it if DNP was supplemented to the IAA-containing media.

Analysis of Locomotor Activity and Body Temperature Rhythms in the Process of Daily Torpor in Djungarian Hamsters (Phodopus sungorus)

  • Djungarian hamsters show distinct seasonal rhythms in several physiological parameters. One of them is daily torpor that occurs in winter with decreased body temperature (about 1O-20$^{\circ}$C) during daytime. Daily torpor is induced by short-day photoperiod, food restriction and castration. But the mechanism to induce daily torpor has not been clarified. In the present study, we tried to clarify the process of daily torpor induction in detail. Adult male hamsters were kept in long photoperiod and high temperature (LP-HT) before the experiment and, thereafter, the animals were transferred to short photoperiod and low temperature (SP-LT), and they were kept in this condition for about six months. The daily rhythms of locomotor activity and body temperature were recorded every three-minutes by using the Minimitter telemetry system. Locomotor activity and body temperature showed very closely synchronized rhythms. All animals under LP-HT showed daily rhythms with higher locomotor activity and body temperature in nighttime than in daytime. Under SP-LT, there were two types of animals with and without showing daily torpor. Thus, they have individual differences in the response to SP -LT.

  • The effects of ambient air temperature on the clothing ventilation were investigated numerically by a finite difference method. Numerical analysis using a 2-dimensional model comprising the air space between the skin and the clothing was conducted under the assumption that the clothing ventilation occurred only through the openings not through the fabric. The larger the temperature difference between the skin and the surroundings, the more apparent the thermal boundary layer As the ambient air temperature decreased, the air flow and the rate of the return of oxygen concentration to the atmosphere level in the clothing increased. Convection was dominant under low ambient air temperature, whereas conduction was dominant under high ambient air temperature. The ventilation rate was faster in the clothing microenvironment of the body part than that of the arm part.

Antioxidant Activity of Yogurt Fermented at Low Temperature and Its Anti-inflammatory Effect on DSS-induced Colitis in Mice

  • This study was performed to evaluate the antioxidant activity of yogurt fermented at low temperature and the anti-inflammatory effect it has on induced colitis with 2.5% dextran sodium sulfate (DSS) in Balb/c mice. Yogurt premix were fermented with a commercial starter culture containing Lactobacillus acidophilus, Bifidobacterium lactis, Streptococcus thermophilus, and Lactobacillus delbrueckii subsp. bulgaricus at different temperatures: $22^{\circ}C$ (low fermentation temperature) for 27 h and $37^{\circ}C$ (general fermentation temperature) for 12 h. To measure antioxidant activity of yogurt samples, DPPH, $ABTS^+$ and ferric reducing antioxidant potential (FRAP) assays were conducted. For animal experiments, inflammation was induced with 2.5% DSS in Balb/c mice. Yogurt fermented at low temperature showed higher antioxidant activity than that of the yogurt fermented at general temperature. In the inflammatory study, IL-6 (interleukin 6) was decreased and IL-4 and IL-10 increased significantly in DSS group with yogurt fermented at general temperature (DYG) and that with yogurt fermented at low temperature (DYL) compared to that in DSS-induced colitic mice (DC), especially DYL had higher concentration of cytokines IL-4, and IL-10 than DYG. MPO (myeloperoxidase) tended to decrease more in treatments with yogurt than DC. Additionally, yogurt fermented at low temperature had anti-inflammatory activity, although there was no significant difference with general temperature-fermented yogurt (p>0.05).
