Detection of rare point mutation via allele-specific amplification in emulsion PCR

  • It is essential to analyze rare mutations in many fields of biomedical research. However, the detection of rare mutations is usually failed due to the interference of predominant wild-type DNA surrounded. Herein we describe a sensitive and facile method of detecting rare point mutation on the basis of allele-specific amplification in emulsion PCR. The identification and selective amplification of rare mutation are accomplished in one-pot reaction. The allele-specific primers coupled on magnetic beads allow the exclusive amplification and enrichment of the mutant amplicons. The productive beads bearing mutant amplicons are subsequently stained with the fluorescent dyes. Thus, the rare point mutations with a percentage as low as 0.1%, can be detected by fluorescent analysis. The relative percentages of mutation among different samples can be roughly accessed by counting the fraction of fluorescent positive beads through flow cytometry.

마찰저항 저감을 위한 고분자 수지 합성 및 PIV 유동장 계측 (Synthesis of FDR-SPC Resin and PIV Measurement for Frictional Drag-reduction)

  • In this study, a novel FDR-SPC is first synthesized in this study. The drag reducing functional radical such as PEGMA (Poly(ethylene) glycol methacrylate) has been utilized to participate in the synthesis process of the SPC. The types of the baseline SPC monomers, the molecular weight and the mole fraction of PEGMA were varied in the synthesis process. The resulting SPCs were coated to the substrate plates for the subsequent hydrodynamic test for skin friction measurement. In a low-Reynolds number flow measurement using PIV (Particle Image Velocimeter), a significant reduction in Reynolds stress was observed in a range of specimen, with the maximum drag reduction being 15.9% relative to the smooth surface.

  • 액상 이소프렌 고무에 무수 말레간(MAH)을 그라프팅시킨 후 2,2,6,6-tetramethyl-4-piperidinol (TMPO)를 반응시켜 hindered amine light stabilizer (HALS)가 함유된 새로운 고분자형 광안정제를 합성하고, SBR에 대한 광안정제로서의 특성을 관찰하였다. 새로운 고분자형 광안정제의 조성과 물리적 성질은 적정법, GPC 그리고 열중량 분석에 의하여 분석하였다. SBR의 광산화 반응에 대한 고분자형 광안정제의 효과를 254nm 자외선 조사에 따른 자외선 흡광도의 변화, 관능기 변화 그리고 잔막율로서 관찰한 결과, 새로운 고분자형 광안정제를 첨가함으로써 광산화 반응이 효과적으로 억제되었다. 또한, 고부자형 광안정제의 추출내성은 저분자량의 모델 화합물에 비하여 훨씬 우수하였으며, 고분자형 광안정제와 SBR의 상용성은 TMPO가 SBR 필름 표면에 균일하게 고루 분산되어 있어서 매우 우수하였다.

Progression of Apoptotic Cells by Pretreatment of Proteinase K

  • Apoptosis can be difficult to detect in routine histological sections. Since extensive DNA fragmentation is an important characteristic of this process, visualization of DNA breaks could greatly facilitate the identification of apoptotic cells. Several techniques for the qualitative and quantitative detection of this process have been established; recently, an in situ nick end-labelling technique based on the detection of DNA fragmentation, which is a molecular characteristic of apoptotic cell death, was described. Applying this method to paraffin sections of rat tissues, sensitivity was observed to be inconsistently low with regard to the expected number of apoptotic cells. I describe a new modified method for formalin-fixed, paraffin-embedded tissue sections, pretense pretreatment to permeate the tissue sections that involves an TUNEL (terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling) is acknowledged as a method of choice in the rapid identification and quantification of the apoptotic cell fraction in paraffin tissue preparations. TUNEL was performed without apoptosis and with apopotosis samples to each of the three concentrations of proteinase K (10, 25, 40 mg/ml) pretreatments. In this study, I show that chemical pretreatments of the tissue sections in proteinase K (25 mg/ml for 15 min at room temperature) considerably enhances the sensitivity of this nick end labelling technique.

플라즈마 중합법에 의해 제작된 PHENYL ISOTHIOCYANATE 막의 화학적 구조 (The Chemical Structure of Phenyl Isothiocyanate Thin Films Fabricated by Plasma Polymerization Method)

  • The Thin films were obtained by plasma polymerization of phenyl isothiocyanate. Polymerizations were carried out in rf(13.56 [MHz]) glow discharge generated in an inter-electrode capacitively coupled gas flow system. It was fecund that this monomer produces uniform films with a wide range of thicknesses, from hundreds of nanometers to tens of micrometers. The deposition rate appeared to be dependent on the substrate distance from the monomer inlet. The IR data revealed significant decrease in -NCS groups content in the polymer as compared with the monomer spectrum and indicated for the appearance of new absorption bands corresponding to the -CN and C-H aliphatic groups. The soluble fraction by GC was found to be composed of numerous low molecular-weight compounds.

탄소/탄소 복합재 매트릭스 전구체로서의 메조페이스 핏치의 제조 및 특성에 관한 연구 (Preparation and Characterization of Mesophase Pitches as a Matrix Precursor of Carbon Fiber Reinforced Carbon Composite)

  • A study on mesophase pitch as a matrix precursor of carbon fiber reinforced carbon (C/C) composite has been recently presented. This study is concerned with the production of mesophase pitch as matrix precursors for C/C composite from coal tar pitch. A commercial coal tar pitch was heat-treated at 25$0^{\circ}C$ for 2 hours to remove low molecular weight fraction from the pitch then increasing the temperature of the pitch to between 350~45$0^{\circ}C$ to produce mesophase pitch. The pitch was continuously stirred during this time and nitrogen gas was continuously bubbled through the pitch. Spherical and bulk mesophases were formed in the pitch after heat-treatment,. Parent and mesophae pitches were characterized by elemental analysis coke yield solubi-lity in tetrahydrofuran and hexane and an optical microscopy to measure the mesiophase content. It was neces-sary to produce C/C composite that a mesophase pitch with about 30-40 vol% mesophase spherulites can be infiltrated into a fiber preform without a filter effect as a matrix precursor conditions. This condition was satisfied with mesophase pitch heat treated at 40$0^{\circ}C$ for 2 hours. The other heat treatment conditions showed the nuclei of mesophase or bulk mesophae which were not satisfied with the matix precursor condition.

칠황버섯으로부터 분리한 Lanostane-type Triterpenoid의 암세포성장 억제효과 (Cancer Cell Growth Inhibition of Lanostane-type Triterpenoids Isolated from Ganoderma gibbosum)

  • The CHCl3 fraction of the MeOH extract of Ganoderma gibbosum (Ganodermataceae) exhibited cytotoxic activity on five cancer cell lines (MDA-MB-231, SK-hep1, A549, HCT116, and SNU638). Six highly oxygenated lanostane-type triterpenoids (lanostanoids) were isolated by column chromatography to test cytotoxicity on cancer cells. The five known lanostanoids were identified as gibbosic acids A, B, D, G, and H by comparison of molecular ion peaks with the literature data. The structure of a new lanostanoid, gibbosic acids I, was identified to be 3β,8β,15β,20S-tetrahydroxy-7,12,23-trioxolanost-9(11)-en-26-oic acid on the basis of NMR and MS spectroscopy. The three lanostanoids of gibbosic acids A, H, and I of the six isolates significantly suppressed the growth of cancer cells. In particular, the IC50 of gibbosic acid H was prominently low ranging from 2.64-6.56 μM.

Cellulase 처리가 쌀의 이화학적 특성 및 밥의 텍스쳐 특성에 미치는 영향 (The Effect of Cellulase Treatment on the Physicochemical Properties of Rice and the Texture of Cooked Rice)

  • 추청벼, 조생통일벼, IR 36등 세 품종의 쌀에 셀룰라제(cellulase) 처리가 팽윤력, 용해도, 아밀로그래프, differential scanning calorimetry (DSC), 가용성 탄수화물 용출 및 밥의 경도 등에 미치는 효과에 대하여 시험하였다. 효소처리에 의하여 세 품종의 쌀에서 모두 팽윤력과 용해도가 증가하였으며 아밀로그램 점도는 모두 감소하였다. 시차 주사 열량계(DSC)를 이용한 밥의 노화 시험에서는 효소 처리의 뚜렷한 효과가 없었다. 효소처리한 쌀의 가용성 탄수화물의 겔 크로마토그래피에서 분자량 10,000미만의 세포벽 분해산물이 확인되었다. Rheometer로 측정한 밥의 경도는 효소처리한 모든 품종의 쌀에서 감소하였다.

  • 비대칭 블록 공중합체와 단일고분자로 구성된 블렌드의 미세 도메인 구조와 결정화 거동을 소각 X-선 산란, 광학 현미경 및 DSC를 사용하여 조사하였다. 본 연구에서는 폴리(메틸 메타크릴레이트) 블록의 무게 분률이 0.53인 폴리(메틸 메타크릴레이트)-폴리스티렌 이종 블록 공중합체 (PMMA-b-PS)를 저분자량 폴리(비닐리덴 플루오라이드) (PVDF)와 혼합하였다. PVDF 함량 증가에 따라 블렌드 미세구조가 라멜라에서 실린더 구조로 전이하였으며, PVDF의 결정화는 결정화 이전에 형성된 미세구조의 배열을 교란시켜 질서도가 낮은 형태를 야기시켰다. 또한, PVDF 결정화 거동은 PMMA 블록과의 혼화성 및 미세 도메인에 의해 부가되는 공간적 제한에 큰 영향을 받았다.
