Development of the Method Allowing DNA Size Markers to be Ambient Storage with Lyophilized Type (상온보관이 가능한 건조체 명태의 DNA size marker)

  • Gel electrophoresis of DNA is a well known technique in molecular biology. This technique is simple, rapid to perform, and capable of adequately separating fragments of DNA. A number of mixtures of DNA fragments ("DNA size markers") are frequently employed in a purpose of extrapolating the sizes or the amount of DNA molecules during gel electrophoresis. DNA size markers are constructed by digesting plasmid DNA, bacteriophage DNA, or recombinant DNA molecules with one or more restriction enzymes. However, liquid suspension containing DNA size marker needs to be kept at a low temperature during storage and shipping. In an attempt to maintain the DNA samples at room temperature for extended period of time, lyophilization of DNA with addition of nuclease inhibitor was studied. Gel loading buffer was also added to the lyophilized DNA to provide additional convenience such that DNA size marker was the "ready-to-use" followed by simply reconstituting with distilled water.

An important factor for the water gas shift reaction activity of Cu-loaded cubic Ce0.8Zr0.2O2 catalysts

  • The Cu loading of a cubic $Ce_{0.8}Zr_{0.2}O_2$-supported Cu catalyst was optimized for a single-stage water gas shift (WGS) reaction. The catalyst was prepared by a co-precipitation method, and the WGS reaction was performed at a gas hourly space velocity of $150,494h^{-1}$. The results revealed that an 80 wt% $Cu-Ce_{0.8}Zr_{0.2}O_2$ catalyst exhibits excellent catalytic performance and 100% $CO_2$ selectivity ($X_{CO}=27%$ at $240^{\circ}C$ for 100 h). The high activity of 80 wt% $Cu-Ce_{0.8}Zr_{0.2}O_2$ catalyst is attributed to the presence of abundant surface Cu atoms and the low activation energy of the resultant process.

  • This paper has been presented the dynamic effect by the journal speed. eccentricity and source positions in order to overcome the defects of air bearing such as low stiffness and damping coefficient. Choosing the two row source position of air bearing is different from previous investigations in the side of pressure distribution of air film by the wedge effects. These optimal chooses of the two row source positions enable us to improve the performance of the film reaction force and loading force as making the high speed spindle. The results of investigated characteristics may be applied to precision devices like ultra-precision grinding machine and ultra high speed milling.

Performance Change according to the Catalyst Intrusion Rate in the MEA for the PEM Water Electrolysis (고분자전해질 수전해용 MEA의 촉매침투도에 따른 성능변화)

  • The performances of proton exchange membrane (PEM) water electrolysis depend on many factors such as materials, geometries, fabrication methods, operating conditions, and so forth. The fabrication method is concerned, membrane electrode assemblies (MEA) are a most important part to show different performances by different fabrication methods. The performance change of PEM water electrolysis was experimentally measured according to the fabrication differences of the anode electrodes. One point of view is the catalyst intrusion rate to the anode gas diffusion layer (GDL), and the other point of view is the catalyst loading distribution in depth of the anode GDL. Results show that the performances of MEA with deep intrusion of the catalysts are better in the range of low current densities but worse at higher current densities. The catalyst loading distribution does not affect significantly to the performance of PEM water electrolyser.

Study on seismic behavior of fabricated beam-column bolted joint

  • To better promote the development of fabricated buildings, this paper studies the seismic behavior of precast concrete beam-column bolted joint under vertical low cyclic loading. The experimental results show that cracks appear in the beam-column joint core area. Meanwhile, the concrete and the grade 5.6 bolts are damaged and deformed, respectively. Specifically, the overall structure of the beam-column joint remains intact, and the bolts have good energy dissipation capacity. Based on the experimental study, a new method of beam-column bolted connection is proposed in simulation analysis. The simulation results show that the bolts deform in the core area of the new beam-column joint, which enhances the concrete shear capacity legitimately and protects the T-end of the beam against shear failure. To summarize, both the experimental joint and the simulated joint prolong the service life by replacing the bolts under the seismic loading. The research results provide a reference for applications of the fabricated beam-column joint.

Influence of Loading Procedure of Liquid Butadiene Rubber on Properties of Silica-filled Tire Tread Compounds

  • Low molecular weight liquid butadiene rubber (LqBR) is a processing aid that can resolve the migration problem of tire tread compounds. Various studies are being conducted to replace the petroleum-based processing oil with LqBR. However, the effect of the loading time of LqBR in the compounding process on silica dispersion and vulcanizate properties is not well known. In this study, we analyzed silica dispersion, vulcanizate properties, and viscoelastic properties of silica-filled tire tread compound according to the processing aid type (TDAE oil, non-functional LqBR) and, silane terminated LqBR) and input timing. In the non-functional LqBR compounds, the 'with TESPT' mixing procedure showed excellent dynamic viscoelastic properties while silane-terminated LqBR compounds showed that the 'after TESPT' mixing procedure was good for 300% modulus and abrasion resistance.

  • Due to the intermittency of renewable energy sources, a need to store and transport energy will increase. Hydrogen production through water electrolysis will provide an excellent way to supplement the intermittency of renewable energy sources. While alkaline water electrolysis is currently the most mature technology, it has drawbacks of low current density, large footprint, gas crossover, etc. The PEM water electrolysis has potential to replace the alkaline electrolysis. However, expensive catalyst material used in the PEM electrolysis has been the bottleneck of widespread use. In this review, we have reviewed recent efforts to reduce catalyst loading in PEM water electrolysis. In core-shell nanostructures, the precious metal catalyst forms a shell while heteroatoms form a core. In this way, the catalyst loading can be significantly reduced while maintaining the catalytic activity. In another approach, a corrosion-resistant support is utilized, which provides a stable platform to impregnate precious metal catalyst.

Enhancement of in-plane load-bearing capacity of masonry walls by using interlocking units

  • This paper presents a comparative experimental study on structural behavior of the interlocking masonry walls under in-plane cyclic loading. The main purpose of this study is to increase lateral load-bearing capacities of masonry walls by using interlocking units. The interlocking units were designed by considering failure modes of masonry walls and produced using lightweight foamed concrete. To this end, three masonry walls which are hollow, fully grouted, and reinforced were constructed with interlocking units. Also, a traditional masonry brick wall was built for comparison reasons. The walls were tested under in-plane cyclic loading. Then, structural parameters of the walls such as lateral load bearing and total energy dissipation capacities, ductility, stiffness degradation as well as failure modes obtained from the tests were compared with each other. The results have shown that the walls with the interlocking units have better structural performance than traditional masonry brick walls and they may be used in the construction of low-rise masonry structures in rural areas to improve in-plane structural performance.

Microstructure and Mechanical Properties of High-Strength Low-Carbon Bainitic Steels with Enhanced Deformability (높은 변형능을 갖는 저탄소 베이나이트계 고강도강의 미세조직과 기계적 특성)

  • Recently, steel structures have increasingly been required to have sufficient deformability because they are subjected to progressive or abrupt displacement arising from structure loading itself, earthquake, and ground movement in their service environment. In this study, high-strength low-carbon bainitic steel specimens with enhanced deformability were fabricated by varying thermo-mechanical control process conditions consisting of controlled rolling and accelerated cooling, and then tensile and Charpy V-notch impact tests were conducted to investigate the correlation between microstructure and mechanical properties such as strength, deformability, and low-temperature toughness. Low-temperature transformation phases, i.e. granular bainite (GB), degenerate upper bainite(DUB), lower bainite(LB) and lath martensite(LM), together with fine polygonal ferrite(PF) were well developed, and the microstructural evolution was more critically affected by start and finish cooling temperatures than by finish rolling temperature. The steel specimens start-cooled at higher temperature had the best combination of strength and deformability because of the appropriate mixture of fine PF and low-temperature transformation phases such as GB, DUB, and LB/LM. On the other hand, the steel specimens start-cooled at lower temperature and finish-cooled at higher temperature exhibited a good low-temperature toughness because the interphase boundaries between the low-temperature transformation phases and/or PF act as beneficial barriers to cleavage crack propagation.

A Study on Low-Velocity Impact Characterization of Various Sandwich Panels for the Korean Low Floor Bus Application (초저상 버스 차체 적용을 위한 샌드위치 패널들의 저속충격 특성 연구)

  • In this paper, a study on low-velocity impact response of four different sandwich panels for the hybrid bodyshell and floor structure application of the Korean low floor bus vehicle was done. Square samples of 100mm sides were subjected low-velocity impact loading using an instrumented testing machine at six energy levels. Impact parameters like maximum force, time to maximum force, deflection at maximum force and absorbed energy were evaluated and compared for four different types of sandwich panels. The impact damage size and depth of the permanent indentation were measured by 3-Dimensional Scanner. Failure modes were studied by sectioning the specimens and observed under optical microscope. The impact test results show that sandwich panel with composite laminate facesheet could not observe damage mode of a permanent visible indentation after impact and has a good impact damage resistance in comparison with sandwich panel with metal aluminum facesheet.
