Major plant nutrient-releasing patterns in the leachates from the soil incorporated rice hull biochar adjusted pH with dry fish powder (산도를 조절한 왕겨 바이오차와 어분 혼합물을 처리한 토양 침출수의 양분용출 패턴)

  • This batch experiment was conducted to investigate the patterns of major plant nutrients in the leachates from the soil that was incorporated with rice hull biochar adjusted pH with dry fish powder utilizing rice hull biochar for loading the soil microorganisms. The rice hull biochar adjusted pH between 6.0 and 7.0, and the mixture ratio of rice hull biochar and dry fish powder was 4:6. The treatments consisted of three; the soil incorporated with rice hull biochar non-adjusted pH with dry fish powder as control (RB + DF), the soil incorporated with rice hull biochar adjusted pH by pyroligneous acid solution and dry fish powder (RBP+DF), and the soil incorporated with rice hull biochar adjusted pH by citric acid solution and dry fish powder (RBC+DF). NH4-N, NO3-N, PO4-P, and K concentrations in the leachates were analyzed during incubation. The accumulated NH4-N and PO4-P concentrations in the leachates from the RBC+DF treatment were the highest during leaching periods. The highest accumulated NO3-N and K concentrations in the leachates from the RBP+DF treatment were observed. It observed that NH4-N and PO4-P were more released in the adjusted citric acid solution, but NO3-N and K were less released than those in the pyroligneous acid solution due to their low absorption capacity. Furthermore, it is necessary to investigate crop growth responses to the soil incorporated with adjusted pH rice hull biochar and dry fish powder for loading soil microorganisms.

Liquefaction Resistance of Gravel-Sand Mixtures (자갈-모래 혼합토의 액상화 거동)

  • In this research, the effects of the gravel content on the liquefaction behavior for both of the isotropically and $K_0-anisotropically$ consolidated gravel-sand mixtures are investigated. for this purpose, the cyclic triaxial tests for the specimens with the same relative density (Dr=40%) and variations of gravel content were performed. On the other hand, a series of undrained cyclic triaxial tests were carried out on the isotropically consolidated gravel-sand mixtures with the same void ratio (e=0.7) and from 0% to 30% gravel contents. Void ratios of gravel-sand mixtures with the same relative density (Dr=40%) are found to decrease significantly with the increase of the gravel content from 0% to about 70% and increase thereafter. But the void ratio of the sand matrix among the gravel skeleton increases with the increase of the gravel contents. Test results are as follows : for the isotropically consolidated specimen with 40% of relative density and low gavel contents (GC=0%, 20%, 40%), pore water pressure development and axial strain behavior during undrained cyclic loading show similar behavior to those of the loose sand because of high void ratio, and the specimens with high gravel content (70%) both pore pressure and strata behaviors are similar to those of dense sand. And the isotropically consolidated specimens with the same void ratio (e=0.7) and higher gravel contents show the same behavior of pore water pressure and axial strain as that of the loose sand, but for the lower gravel content this behavior shows similar behavior to that of dense sand. The liquefaction strength of the isotropically consolidated specimens with the same relative density increases with gravel content up to 70%, and the strength decreases with the increase of the gravel content at the same void ratio. Thus, it is confirmed that the liquefaction strength of the gravel-sand mixtures depends both on relative density and void ratio of the whole mixture rather than the relative density of the sand matrix filled among gravels. On the other hand, the behavior of pore water pressure and axial strain for the $K_0-anisotropically$ consolidated gravel-sand mixtures shows almost the same cyclic behavior of the sand with no stress reversal even with some stress reversal of the cyclic loading. Namely, even the stress reversal of about 10% of cyclic stress amplitude, the permanent strain with small cyclic strain increases rapidly with the number of cycles, and the initial liquefaction does not occur always with less than maximum pore water pressure ratio of 1.0. The liquefaction resistance increases with the gravel contents between 0% and 40%, but tends to decrease beyond 40% of gravel content. In conclusion, the cyclic behavior of gravel-sand mixtures depends on factors such as gravel content, void ratio, relative density and consolidation condition.

Cyanobacterial Development and Succession and Affecting Factors in a Eutrophic Reservoir (부영양 저수지에서 남조류의 발달과 천이 및 영향 요인)

  • This study was conducted to evaluate the causes and effects of cyanobacterial development and succession in a shallow eutrophic reservoir from March 2003 to February 2004. Phytoplankton succession, sedimentation rate, and sediment composition were analyzed. Algal bioassay also was conducted with the consideration of light, water temperature and nutrients. Cyanobacteria dominated throughout the year, except for spring season (March${\sim}$April) in which diatoms and flagellates dominated. Total cell density increased in July and November when P loading through inflows was high. Oscillatoria spp. and Aphanizomenon sp. were dominant in May and June, respectively, but replaced with Microcystis spp. in July. Thereafter, Microcystis spp. sustained until December, and again shifted to Oscillatoria spp. and Aphanizomenon sp. The dominance of Oscillatoria spp. in May was accompanied with high TN/TP ratio and the increase of water temperature and light intensity. While the dominance of Microcystis spp. was related with relatively low TN/TP ratio, ranging from 46 to 13 (average: 27). The sedimentation rate was highest in March (0.6 m $day^{-1}$) when diatoms dominated. During the period of cyanobacterial dominance, relatively high sedimentation rate was observed in May (0.4 m $day^{-1}$) and October (0.36m $day^{-1}$). C/N ratio of the sediment ranged $6{\sim}8$. Inorganic P concentration in the pore water was low when DO concentration was < 2 mg $O_2$ $L^{-1}$ in the hypolimnion, reflecting the P release from the sediment. Cyanobacterial growth rate depended on phosphorus concentration and water temperature, and high P concentration compensated for the low temperature in the growth rate. Our results suggest that the potential of cyanobacterial development and substantiality in eutrophic reservoirs be high throughout the year, as being supplied with enough P, and emphasize the consideration of sediment man. agement for the water quality improvement and algal bloom control.

A Study on the Floating Island for Water Quality Improvement of a Reservoir (저수지 수질개선을 위한 인공식물섬 조성에 관한 연구)

  • Three floating islands have been constructed for water quality improvement for a polluted irrigation reservoir. Each floating island consists of 10 segments. Each segment hay an area of $16m^2$(4×4m) and is made of wood frames and floats(polystyrene foam). We planted three species of aquatic macrophytes(Typha angustifolia, Zizania latifolia, and Phragmites australis) in floating island on June, 1998. They grew very well without death. We would like to evaluate Phragmites australis is the most suitable aquatic macrophyte that could be planted in a floating island because it maintained the best balance of its root and shoot among them. During their grown period, net primary productivity of Typha angustifolia was $962gDM/m^2$, Zizania latifolia was $1,115gDM/m^2$, and Phragmites australis was $523gDM/m^2$. From these data, it would be estimated to 5.0Kg uptake of nitrogen by aquatic macrophytes and phosphorus 0.8Kg in 3 floating islands. The floating islands worked well as a habitat of fish and prawns. Many kinds of insect lived on the floating islands. The floating island has not only the function of water quality treatment but also several advantages: improvement of landscape and species diversity; low cost of maintenance; low technology; unnecessary of energy; less susceptible to variations in pollutant loading. It could be evaluated a good measure of water quality improvement for an irrigation reservoir. However, it should be intensively studied to develop more light, strong, durable and low-priced frames for efficient floating islands.

Review of Production, Husbandry and Sustainability of Free-range Pig Production Systems

  • A review was undertaken to obtain information on the sustainability of pig free-range production systems including the management, performance and health of pigs in the system. Modern outdoor rearing systems requires simple portable and flexible housing with low cost fencing. Local pig breeds and outdoor-adapted breeds for certain environment are generally more suitable for free-range systems. Free-range farms should be located in a low rainfall area and paddocks should be relatively flat, with light topsoil overlying free-draining subsoil with the absence of sharp stones that can cause foot damage. Huts or shelters are crucial for protecting pigs from direct sun burn and heat stress, especially when shade from trees and other facilities is not available. Pigs commonly graze on strip pastures and are rotated between paddocks. The zones of thermal comfort for the sow and piglet differ markedly; between 12-22$^{\circ}C$ for the sow and 30-37$^{\circ}C$ for piglets. Offering wallows for free-range pigs meets their behavioural requirements, and also overcomes the effects of high ambient temperatures on feed intake. Pigs can increase their evaporative heat loss via an increase in the proportion of wet skin by using a wallow, or through water drips and spray. Mud from wallows can also coat the skin of pigs, preventing sunburn. Under grazing conditions, it is difficult to control the fibre intake of pigs although a high energy, low fibre diet can be used. In some countries outdoor sows are fitted with nose rings to prevent them from uprooting the grass. This reduces nutrient leaching of the land due to less rooting. In general, free-range pigs have a higher mortality compared to intensively housed pigs. Many factors can contribute to the death of the piglet including crushing, disease, heat stress and poor nutrition. With successful management, free-range pigs can have similar production to door pigs, although the growth rate of the litters is affected by season. Piglets grow quicker indoors during the cold season compared to outdoor systems. Pigs reared outdoors show calmer behaviour. Aggressive interactions during feeding are lower compared to indoor pigs while outdoor sows are more active than indoor sows. Outdoor pigs have a higher parasite burden, which increases the nutrient requirement for maintenance and reduces their feed utilization efficiency. Parasite infections in free-range pigs also risks the image of free-range pork as a clean and safe product. Diseases can be controlled to a certain degree by grazing management. Frequent rotation is required although most farmers are keeping their pigs for a longer period before rotating. The concept of using pasture species to minimise nematode infections in grazing pigs looks promising. Plants that can be grown locally and used as part of the normal feeding regime are most likely to be acceptable to farmers, particularly organic farmers. However, one of the key concerns from the public for free-range pig production system is the impact on the environment. In the past, the pigs were held in the same paddock at a high stocking rate, which resulted in damage to the vegetation, nutrient loading in the soil, nitrate leaching and gas emission. To avoid this, outdoor pigs should be integrated in the cropping pasture system, the stock should be mobile and stocking rate related to the amount of feed given to the animals.

Fabrication of PLGA/Dextran Double-Layered Microspheres by Oil-in-Water Solvent Evaporation Method (O/W 용매 증발법을 이용한 PLGA와 덱스트란의 이중층 미립구 제조)

  • Double-layered spheres play an important role in controlling drug delivery for pharmaceutical application, because of the low initial burst compared with single-layered spheres and targetable delivery to specific organ. But it has drawback in loading drug and controlling size. In this study, we developed double-layered spheres using relatively simple oil-in-water (O/W) solvent evaporation method witw/without ultrasonication and investigated the size variation of the double-layered microspheres on the contents of poly(lactide- co-glycolide) (PLGA). Double - layered spheres were char-acterized by scanning elecron microscope (SEM), camscope, and confocal fluorescence laser microscope (CFLM). Double-layered spheres showed smooth surfaces and obvious difference between core and corona by SEM observation and camscope. We observed the fluorescent core in the double-walled spheres composed of FlTC-dextran and PLGA using CFLM. It was found that the core of the microsphere was dextran and the corona of the fabricate microsphere was PLGA. Also, the more PLGA concentration, the more the size of the fabricating double-layered sphere observed.

Estimation of the $T_4$ Binding Capacity of Serum Thyroxine Binding Globulin (혈중(血中) Thyroxine-결합(結合)-globulin(TBG)의 $T_4$ 결합능(結合能) 측정(測定)에 관한 고찰(考察))

  • The most commonly used methods for determining thyroxine binding globulin(TBG) concentration as the total thyroxine-binding capacity utilize electrophoretic seperation of serum. Although technically simple, the electrophoretic method is time consuming and is limited in the number of samples which can be run in a single assay. The author presented a single $T_4$ load ion exchange resin method as an approach to simplify the technique as with clinical practicability and results were analyzed. For construction of the standard curves, serum mixtures were diluted with barbital buffer.which effectively blocked $T_4$-binding to TBPA. For each serum dilution, a constant amount of $T_4-^{125}I$ and increments of unlabelled $T_4$ were added. After incubation in water bath, resin beads were dispensed to the samples which binded all $T_4$ not bound to TBG. The radioactivity in the supernatant was counted in the gamma scintillation counter. Each standard curve was plotted from the percent counts in the supernatant and total $T_4$ in each tube. Unknown samples were diluted to 1:40 and ran at a single $T_4$ loading concentration, and the TBG capacity of the samples was able to be read on the standard isobars. The following results were obtained. 1) Mean and standard deviation for TBG capacity in normal population was $28.6{\pm}5.09{\mu}g\;T_4/100ml$. 2) $24.9{\pm}3.87{\mu}g\;T_4/100ml$ in hyperthyroidism showed low TBG capacity comparing to normal population.(p<0.025) 3) $31.0{\pm}2.40{\mu}g\;T_4/100ml$ in hypothyroidism showed high TBG capacity tendency comparing to normal population. 4) Reversed correlationship existed between TBG capacity and $T_3$ resin uptake(r=-0.624), TBG capacity and serum $T_4$ value (r=-0.859), and TBG capacity and free thyroxine index(r=-0.623). The author assumes that this method of assay is considerably simpler in instrumentation and technique than any other assays traditionally being used, and seems to be more practical for routine clinical laboratory use.

Identification of Bovine Pregnancy-Specific Whey Proteins using Two-Dimensional Gel Electrophoresis

  • The early diagnosis of bovine pregnancy is an essential component of successful reproductive planning on farms, because lack of bovine pregnancy over the long term results in reproductive failure and low milk yield-the latter of which is a special concern on dairy farms. This study was designed to identify early pregnancy-specific whey proteins in bovine, by comparing milk samples collected from cattle during pregnancy (Days 30 and 50) and from non-pregnant cattle. In this study, differentially expressed proteins in five pregnant and five non-pregnant Holstein dairy cattle were investigated and compared, using proteomics analysis. The first dimension was applied to a pH $3.0{\sim}10.0$ strip, by loading a 2-mg milk protein sample. After the second-dimension separation was performed, the gels were stained with colloidal Coomassie brilliant blue. The stained gels were scanned and the images were analyzed, to detect variations in protein spots between non-pregnant and pregnant cattle milk protein spots, using ImageMaster, this was followed by analysis with MALDI TOF-MS. Analysis of the 2-DE gel image resulted in a total of approximately $500{\sim}600$ protein spots, of which 12 spots were differentially expressed, six spots were up-regulated, and four spots were down-regulated; two spots were identified as pregnancy-specific proteins. These proteins were identified as lactoferrin, NA-DH dehydrogenase subunit 2, albumin, serum albumin precursor and transferrin. Our results via 2-D PAGE analysis revealed composite profiles of several milk proteins related to early bovine pregnancy, implying the possible use of these milk proteins in the early detection of bovine pregnancy.

A Study on the Foodservice Quality of Japanese Restaurants using the Importance-Performance Analysis (IPA) (중요도-만족도 분석을 활용한 일식 레스토랑의 품질특성에 관한 연구)

  • Japanese restaurant owners seek to secure their competitive advantage by implementing a unique competition strategy that may overcome hostile environment. Brand power, aggressive expansion and the partnership strategy with other businesses may be suggested. The present study is to find out the competitive advantages of Japanese restaurants, to compare the importance and satisfaction by different customer factors of choices and then, to identify what kinds of factors affect customer satisfaction. Total 205 copies of questionnaire, out of 230 distributed, were used for analyses. Cronbarch's alpha coefficient of all 20 items was 0.911. In terms of importance, Cronbarch's alpha coefficients were 0.887(food), 0.796(menu), 0.851(employee), 0.815(service), and 0.730(physical environment). For satisfaction, the coefficients were 0.876(food), 0.860(menu), 0.880(employee), 0.851(service), 0.730(physical environment). In addition, the exploratory factor analysis showed an acceptable factor loading, supporting 20 items' validity. The result showed that customers' perceptions of the importance of Japanese restaurant service quality was higher than their level of satisfaction in general. In particular, IPA analysis revealed that food and service factors were well operated as located in the first quadrant. However, service factor located in the second quadrant was perceived to be important yet perceived as being satisfied poorly. So this study suggests that Japanese restaurant owners should concentrate on the attributes included in this quadrant to improve the quality of foodservice. Lastly, menu and physical environment factors were located in the third quadrant showing low level of importance and satisfaction. On the basic of the IPA analysis, Japanese restaurant owners should identify their strengths and weaknesses and draw up a plan to improve service quality and to maximize their profits.

Kinetic of Catalytic CO2 Gasification for Cyprus Coal by Gas-Solid Reaction Model (기-고체 반응모델을 이용한 Cyprus탄의 CO2 저온촉매가스화 반응거동)

  • In general, the coal gasification has to be operated under high temperature ($1300{\sim}1400^{\circ}C$) and pressure (30~40 bar). However, to keep this conditions, it needs unnecessary and excessive energy. In this work, to reduce the temperature of process, alkali catalysts such as $K_2CO_3$ and $Na_2CO_3$ were added into Cyprus coal. We investigated the kinetic of Cyprus char-$CO_2$ gasification. To determine the gasification conditions, the coal (with and without catalysts) gasified with fixed variables (catalyst loading, catalytic effects of $Na_2CO_3$ and $K_2CO_3$, temperatures) by using TGA. When catalysts are added by physical mixing method into Cyprus coal the reaction rate of coal added 7 wt% $Na_2CO_3$ is faster than raw coal for Cyprus char-$CO_2$ gasification. The activation energy of coal added 7 wt% $Na_2CO_3$ was calculated as 63 kJ/mol which was lower than raw char. It indicates that $Na_2CO_3$ can improve the reactivity of char-$CO_2$ gasification.
