Effect of Lotus (Nelumbo nucifera) Leaf Extract on Serum and Liver Lipid Levels of Rats Fed a High Fat Diet (고지방 식이와 병행 섭취한 연잎 추출물이 흰쥐의 혈청 및 간 조직 지질 함량에 미치는 영향)

  • Lotus (Nelumbo nucifera) leaf is known to be effective for 'overcoming body heat' and stopping bleeding. It is commonly used as a traditional curing plant for the treatment of hematemesis, epistaxis, hemoptysis, hematuria, and metrorrhagia in traditional Chinese medicine. This study investigated on the effect of oral administration of lotus (Nelumbo nucifera) leaf extract on the serum and liver lipid levels of rats fed a high fat diet. Experimental rats were divided into five different experimental groups, including the general diet group (Cont), high fat diet with lotus leaf extract groups (HL40, HL80, HL120), and high fat diet group (HFG). Body weight significantly decreased in the HL120 sample compared to that of Cont. The weights of the livers and kidneys of rats corresponded to the increase in body weight. Total cholesterol and triglyceride contents in liver tissues of rats were lowest in the sample HL120 sample. The levels of total lipids, total cholesterol, and triglycerides in serum were lower in the HL120 sample compared to the HFG.

A Study on the Change of Dyeability and Colorfastness of Lotus Leaf Extract with Mordanting Treatment (매염처리에 따른 연(蓮)잎 추출물의 염색성 및 염색견뢰도 변화에 관한 연구)

  • The purpose of this study was to investigate the colorfastness of cotton and silk fabric dyed with extract of Lotus leaf princeps. The experimental items were classified into mordanting treatments, fabric components and kind of mordants. The experimental test was done by laundering, abrasion(dry/wet), perspiration(acid/alkali) , light fastness, color difference by C.C.M(computer color matching) system and K/S test. The summarized findings resulting from the experiment and investigation are as follows: In the C.C.M and K/S test on mordanting method and kind of fabrics, co-mordanting treatment method and protein fabric (silk) were the most effective. Especially, Fe mordanting treatment was higher than Al, Cu, Cr and none-mordanting. In colorfastness on the mordants, perspiration and laundering fastness showed high grade (4-5) but grades of perspiration(2-3/4) and light fastness(1-3) were low.

Stability Evaluation of the Cosmetics Containing Lotus Leaf Extract (연잎 추출물을 함유한 화장품의 안정성 평가)

  • The stability of cream containing lotus leaf extract (LLE) was evaluated. Stability pH and viscosity were measured at 3 different temperatures ($25^{\circ}C$, $45^{\circ}C$ and $65^{\circ}C$), and at room temperature under the sun light at 2 day intervals during 12 days. The cleansing cream and foam cleansing containing the extract did not show a significant pH change at the different temperatures. However, the pH of the cleansing cream and foam cleansing containing LLE slightly decreased at $25^{\circ}C$. They showed a high stability at temperature conditions ($45^{\circ}C$ and $65^{\circ}C$) and under the sun light condition. The cleansing cream and foam cleansing containing LLE did not show a significant viscosity change at $25^{\circ}C$, but had a tendency of decrease at $45^{\circ}C$ and $65^{\circ}C$. There was no significant offflavor and discoloration as well as physical changes such as creaming and cohesion at $25-65^{\circ}C$ and under the sun light condition. And this LLE could be used as a stable functional cosmetic material.

Antioxidative Activity of Ethanol Extract from Lotus (Nelumbo nucifera) Leaf (연잎 에탄올 추출물의 항산화 효과)

  • This study was investigated on the antioxidative activity of $70\%$ ethanol extracts from lotus (Nelumbo nucifera) leaf. The extraction yields of $70\%$ ethanol extract was $8.16\%$. The extract was further fractionated subsequently by hexane, chloroform, ethyl acetate, butanol and water. Antioxidative activity was examined by electron donating ability (EDA) using DPPH, TBA value, acid value (AV), and peroxide value (POV), in comparison with commercial antioxidants. Butanol fraction showed the highest extraction yields but ethyl acetate fraction showed the highest phenol content. The ethyl acetate fraction showed the highest EDA. The antioxidative activity of ethyl acetate fraction determined by AV and POV in accelerated oxidation condition of lard was similar to that of BHA.

Physicochemical Properties of Ground Pork with Lotus Leaf Extract during Refrigerated Storage (연잎추출물 첨가가 분쇄돈육의 냉장저장 중 이화학적 품질에 미치는 영향)

  • This study was carried out to investigate the effect of lotus leaf extract on the physicochemical characteristics of ground pork stored at $4^{\circ}C$ for 10 days. Four types of ground pork were evaluated: ice water added (T0), 3% lotus leaf extract added (T1), 7% lotus leaf extract added (T2), and 10% lotus leaf extract added (T3). The pH was increased during storage, but decreased after 10 days of storage (p<0.05). The TBARS increased with a longer storage period (p<0.05), and those of T0, T1, T2 and T3 were 0.777, 0.244, 0.185, 0.182 mg MA/kg, respectively, after 10 days of storage. The VBN content of T0 and T1 increased with a longer storage period (p<0.05), but those of T2 and T3 were not significantly changed. In the case of surface color, the $L^*$ increased with a longer storage period, and that of T0 was the highest (p<0.05). The $a^*$ decreased with a longer storage period, and the $b^*$ was the highest after 1 day of storage (p<0.05). In the case of internal color, $L^*$ was not significantly changed, and $a^*$ was decreased with a longer storage period (p<0.05). The $b^*$ increased with a longer storage period (p<0.05). Water holding capacity increased with a longer storage period, and those of T2 and T3 were the highest (p<0.05). Cooking loss decreased with a longer storage period, and those of T0 and T1 were higher than those of T2 and T3 (p<0.05). Hardness and chewiness were not significantly different with a longer storage period, or among any samples.

Melanogenesis-Inhibitory Effects of Ethyl Acetate Fraction from Diospyros lotus L. Leaf Extract (고욤(Diospyros lotus L.)잎 Ethyl Acetate 분획물의 멜라닌형성 억제 효과)

  • Diospyros lotus has been cultivated for its edible fruits and leaves which are considered for its medicinal importance. The aim of this study was to evaluate the anti-melanogenesis of ethyl acetate (EA) fractions from D. lotus leaves in B16 cells. The order of the total polyphenol content with regard to the different solvent fractions from D. lotus leaves was EA>butanol>metahanol>chloroform>n-hexane. The major compounds of EA fraction from D. lotus leaves by HPLC analysis were myricitrin and myricetin. Cellular TYR activity and melanin content in response to treatment with 100 mg/mL of EA fraction was inhibited more strongly than group treated with arbutin. Further, EA fraction exhibited significant anti-melanogenesis effects by reducing the levels of microphthalima-associated transcription factor (MITE), inhibiting the synthesis of TYR, tyrosinase-related protein-1 (TRP-1) and TRP-2. Therefore, EA fractions from D. lotus leaves may be a good source of skin-whitening agents in the future development of medicine-based trouble skin therapy.

Quality Characteristics and Palatability of Ground Pork Meat Containing Lotus Leaf and Root Extracts (연잎 및 연근 추출물을 함유한 분쇄돈육의 품질 특성 및 기호성)

  • This study was conducted to investigate the effects of addition of lotus (Nelumbo nucifera) leaf and root extracts on the quality and sensory characteristics of ground pork meat. Four types of ground pork were evaluated: 5% ice water added (T0), 5% lotus leaf extract added (T1), 2.5% lotus leaf extract and 2.5% root extract added (T2), and 5% lotus root extract added (T3). There were no significant differences in moisture, protein, fat, ash, cooking yield, moisture retention, water holding capacity, reduction in diameter, a-value (redness), b-value (yellowness), VBN content (volatile basic nitrogen), hardness, springiness, cohesiveness or chewiness. The fat retention was highest in T0 (p<0.05). The L-values (lightness) of T2 and T3 were higher than those of T0 and T1 (p<0.05). The pH was lowest in T1 (p<0.05). The TBARS (2-thiobarbituric acid reactive substances) values of T0, T1, T2, and T3 were 0.47, 0.17, 0.21, and 0.32 mgMA/kg, respectively, with that of T1 being significantly lower than those of the other samples (p<0.05). The contents of free amino acids related to sweet taste was 642.5 ppm for T1, which was highest among the samples (p<0.05). The flavor was highest in T1 (p<0.05). These results suggest that lotus leaf extracts improved the lipid oxidation and flavor of ground pork meat.

The Effect of Lotus (Nelumbo nucifera) Leaf, Stem, and Yeonjabang Powder Extract on the Biochemical Factors in Serum in Mice Fed a High-Fat Diet (고지방식이 병행섭취 시 연잎, 연 줄기, 연자방 분말가루가 흰쥐의 혈중 생화학적 인자에 미치는 영향)

  • In this study, we analyzed the biochemical factors in lotus (Nelumbo nucifera) leaf, stem, and yeonjabang and their effects on serum factor levels in mice fed a high-fat (HF) diet. The loutus leaf showed $9.47{\pm}0.30%$ moisture content, $8.25{\pm}0.39%$ ash, $21.45{\pm}1.25%$ crude protein, and $2.21{\pm}0.13%$ crude fat content; the lotus stem showed $11.84{\pm}0.43%$ moisture, $10.21{\pm}0.64%$ ash, $17.55{\pm}0.92%$ crude protein, and $4.16{\pm}0.23%$ crude fat content; and the lotus yeonjabang showed $11.86{\pm}0.50%$ moisture, $6.81{\pm}0.51%$ ash, $18.71{\pm}1.02%$ crude protein, and $3.95{\pm}0.15%$ crude fat. Blood triglyceride levels were higher in the HF group ($146.43{\pm}38.81mg/dL$), and lower in the HF+yeonjabang groups ($98.00{\pm}17.18mg/dL$). In particular, blood triglyceride levels were significantly lower in the groups that had 10% dry yeonjabang powder added to the high-fat diet. The inclusion of excessive high-fat diet increased concentrations of serum insulin and leptin. Serum leptin concentrations were highest in the HF group mice ($3.00{\pm}1.35ng/dL$), whereas they were significantly lower in the HF+yeonjabang groups by $1.34{\pm}0.52ng/dL$ (p<0.05). Thus, addition of dry yeonjabang powder to the high-fat diet was more effective in regulating the levels of serum triglycerides and leptin in mice. Additional studies would help in the development of yeonjabang as a functional food.

Effect of Green Tea and Lotus Leaf Boiled Water Extracts Treatment on Quality Characteristics in Salted Mackerel during Storage (녹차 및 연잎 열수추출물 처리가 염장고등어의 저장 중 품질특성에 미치는 영향)

  • This study was carried out to analyze the quality change of mackerel treated of green tea and lotus leaf boiled water extracts. And investigate the antioxidant effects of them on the lipid peroxidation of mackerel during storage at $4^{\circ}C$ for 28 days. It was estimated periodical measurements of proximate composition, pH, carbonyl, volatile basic nitrogen, trimethylamine-N, acid, peroxide and thiobarbituric acid values. It had no effect on proximate composition compared with non- treated control nearly but, pH of all the samples was increase during 28 days continuously. The contents of volatile basic nitrogen and trimethylamine-N lower than control during storage. Also, acid, peroxide and thiobarbituric acid values of mackerel treated of green tea and lotus leaf boiled water extracts were significantly lower than control throughout storage period. Especially, mackerel treated of 2% green tea and lotus leaf boiled water extracts shows that more effective than 1% of things in antioxidant during storage. Results indicate that the application of green tea and lotus leaf boiled water extract on the surface of mackerel may be useful to lower the rancidity degree and fish odor during storage.

Anti-Allergy and Anti-Pruritic Effects of Diospyros lotus L. Leaf Extract (고욤(Diospyros lotus L.)잎 추출물의 항알레르기 및 항가려움 효과)

  • The worldwide prevalence and severity of allergic diseases including atopic and contact dermatitis has increased dramatically over the past decade, especially in developed countries. Mast cells are important effector cells in allergic reactions. The purpose of this study was undertaken to investigate the anti-allergic and anti-pruritic effects of Diospyros lotus leaf extract (DLE). DLE was prepared by extracting with distilled water. In the present study, we investigated the effect of DLE on the production of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\alpha}$) and histamine in rat peritoneal mast cells (RPMCs), and on the skin lesion, leukocyte infiltration and scratching behavior in mice. Phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 significantly increased TNF-${\alpha}$ and IL-$1{\beta}$ production compared with media control. However, TNF-${\alpha}$ and IL-6 production increased by PMA plus A23187 treatment were significantly inhibited by DLE in a dose-dependent manner. DLE also inhibited the histamine release from RPMCs stimulated by compound 48/80, which promotes histamine release. Moreover, DLE administration had an inhibitory effects on the scratching behavior induced by pruritogen (compound 48/80, histamine) in ICR mice. Furthermore, DLE inhibited the skin lesions, inflammatory and mast cells in hairless mice sensitized by 2,4-dinitrofluorobenzene (DNFB). DLE administration reduced the IL-4 and IgE production induced by DNFB sensitization in hairless mice. These results suggest that DLE has a potential use as a herb medicine for treatment against allergy and pruritus-related disease.
