NUMERICAL SOLUTIONS OF AN UNSTEADY 2-D INCOMPRESSIBLE FLOW WITH HEAT AND MASS TRANSFER AT LOW, MODERATE, AND HIGH REYNOLDS NUMBERS

  • In this paper, we have proposed a modified Marker-And-Cell (MAC) method to investigate the problem of an unsteady 2-D incompressible flow with heat and mass transfer at low, moderate, and high Reynolds numbers with no-slip and slip boundary conditions. We have used this method to solve the governing equations along with the boundary conditions and thereby to compute the flow variables, viz. u-velocity, v-velocity, P, T, and C. We have used the staggered grid approach of this method to discretize the governing equations of the problem. A modified MAC algorithm was proposed and used to compute the numerical solutions of the flow variables for Reynolds numbers Re = 10, 500, and 50000 in consonance with low, moderate, and high Reynolds numbers. We have also used appropriate Prandtl (Pr) and Schmidt (Sc) numbers in consistence with relevancy of the physical problem considered. We have executed this modified MAC algorithm with the aid of a computer program developed and run in C compiler. We have also computed numerical solutions of local Nusselt (Nu) and Sherwood (Sh) numbers along the horizontal line through the geometric center at low, moderate, and high Reynolds numbers for fixed Pr = 6.62 and Sc = 340 for two grid systems at time t = 0.0001s. Our numerical solutions for u and v velocities along the vertical and horizontal line through the geometric center of the square cavity for Re = 100 has been compared with benchmark solutions available in the literature and it has been found that they are in good agreement. The present numerical results indicate that, as we move along the horizontal line through the geometric center of the domain, we observed that, the heat and mass transfer decreases up to the geometric center. It, then, increases symmetrically.

Long-term exposure to gefitinib differentially regulates the endosomal sorting complex required for transport machinery, which accelerates the metastatic potential of oral squamous cell carcinoma cells

  • Oral squamous cell carcinoma (OSCC), which accounts for approximately 90% of oral cancers, has a high rate of local recurrence and a poor prognosis despite improvements in treatment. Exosomes released from OSCC cells promote cell proliferation and metastasis. Although it is clear that the biogenesis of exosomes is mediated by the endosomal sorting complex required for transport (ESCRT) machinery, the gene expression pattern of ESCRT, depending on the cell type, remains elusive. The exosomal release from the human OSCC cell lines, HSC-3 and HSC-4, and their corresponding gefitinib-resistant sub-cell lines, HSC-3/GR and HSC-4/GR, was assessed by western blot and flow cytometry. The levels of ESCRT machinery proteins, including Hrs, Tsg101, and Alix, and whole-cell ubiquitination were evaluated by western blot. We observed that the basal level of exosomal release was higher in HSC-3/GR and HSC-4/GR cells than in HSC-3 and HSC-4 cells, respectively. Long-term gefitinib exposure of each cell line and its corresponding gefitinib-resistant sub-cell line differentially induced the expression of the ESCRT machinery. Furthermore, whole-cell ubiquitination and autophagic flux were shown to be increased in gefitinib-treated HSC-3 and HSC-4 cells. Our data indicate that the expression patterns of the ESCRT machinery genes are differentially regulated by the characteristics of cells, such as intracellular energy metabolism. Therefore, the expression patterns of the ESCRT machinery should be considered as a key factor to improve the treatment strategy for OSCC.

Reciprocal regulation of SIRT1 and AMPK by Ginsenoside compound K impedes the conversion from plasma cells to mitigate for podocyte injury in MRL/lpr mice in a B cell-specific manner

  • Background: Deposition of immune complexes drives podocyte injury acting in the initial phase of lupus nephritis (LN), a process mediated by B cell involvement. Accordingly, targeting B cell subsets represents a potential therapeutic approach for LN. Ginsenoside compound K (CK), a bioavailable component of ginseng, possesses nephritis benefits in lupus-prone mice; however, the underlying mechanisms involving B cell subpopulations remain elusive. Methods: Female MRL/lpr mice were administered CK (40 mg/kg) intragastrically for 10 weeks, followed by measurements of anti-dsDNA antibodies, inflammatory chemokines, and metabolite profiles on renal samples. Podocyte function and ultrastructure were detected. Publicly available single-cell RNA sequencing data and flow cytometry analysis were employed to investigate B cell subpopulations. Metabolomics analysis was adopted. SIRT1 and AMPK expression were analyzed by immunoblotting and immunofluorescence assays. Results: CK reduced proteinuria and protected podocyte ultrastructure in MRL/lpr mice by suppressing circulating anti-dsDNA antibodies and mitigating systemic inflammation. It activated B cell-specific SIRT1 and AMPK with Rhamnose accumulation, hindering the conversion of renal B cells into plasma cells. This cascade facilitated the resolution of local renal inflammation. CK facilitated the clearance of deposited immune complexes, thus reinstating podocyte morphology and mobility by normalizing the expression of nephrin and SYNPO. Conclusions: Our study reveals the synergistic interplay between SIRT1 and AMPK, orchestrating the restoration of renal B cell subsets. This process effectively mitigates immune complex deposition and preserves podocyte function. Accordingly, CK emerges as a promising therapeutic agent, potentially alleviating the hyperactivity of renal B cell subsets during LN.

흰쥐에서 우절(藕節) 추출물의 국소 뇌혈류량 조절 효과 (Effect of Nodus Nelumbinis Rhizomatis Extract on the Regulation of Regional Cerebral Blood Flow in Rats)

  • Objectives : In brain disorders such as ischemic stroke, the final outcome depends largely on the duration and the degree of the ischemia as well as the susceptibility of various cell types in the affected brain region. In the present study, the effects of Nodus Nelumbinis Rhizomatis Extract(NNRe) were tested for the anti-oxidative action of rCBF. Methods : Regional cerebral blood flow(rCBF) were determined by LDF methods. LDF allows for real time, noninvasive, continuous recordings of local CBF. The LDF method has been widely used to trace hemodynamic changes in the superficial or the deep brain structures in experimental stroke research. Results : NNRe treatment showed no change on rCBF in methylene blue, ODQ and L-NNA pretreated rats. 120 minutes of MCAO and followed reperfusion, 0.1% concentration of NNR treatment improved the altered cerebral hemodynamics of cerebral ischemic by increasing rCBF. Conclusions : The ischemia/reperfusion induced oxidative stress may have contributed to cerebral damage in rats, and the present study provides clear evidences for the beneficial effect of NNR on ischemia/reperfusion induced brain injury.

T cell phenotype and intracellular $IFN-{\gamma}$ production in peritoneal exudate cells and gut intraepithelial lymphocytes during acute Toxoplasma gondii infection in mice

  • Although there are many reports on the splenic (systemic) T cell response after Toxoptasma gondii infection, little information is available regarding the local T cell responses of peritoneal exudate cells (PEC) and gut intraepithelial Iymphocytes (IEL) following peroral infection with bradyzoites. Mice were infected with 40 cysts of the 76K strain of T. gondii, and then sacrificed at days 0, 1, 4, 7 and 10 postinfection (PI). The cellular composition and T cell responses of PEC and IEL were analyzed. The total number of PEC and IEL per mouse increased after infection, but the ratio of increase was higher in IEL. Lymphocytes were the major component of both PEC and IEL. The relative percentages of PEC macrophages and neutrophils/eosinophils increased signiflcantly at day 1 and 4 PI, whereas those of IEL did not change significantly. The percentage of PEC NK1.1 and ${\gamma\delta}T$ cells peaked at day 4 PI (p < 0.0001), and CD4 and $CD8{\alpha}T$ cells increased continuously after infection. The percentages of IEL $CD8{\alpha}$ and ${\gamma\delta}T$ cells decreased slightly at first, and then increased. CD4 and NK1.1 T cells of IEL did not change significantly after infection. $IFN-{\gamma}-producing$ PEC NK1.1 T cells increased significantly from day 1 PI, but the other T cell subsets produced $IFN-{\gamma}$ abundantly thereafter. The proportion of IEL $IFN-{\gamma}-producing$ $CD8{\alpha}$ and ${\gamma\delta}T$ cells increased significantly after infection, while IEL NK1.1 T cells had similar $IFN-{\gamma}$ production patterns. Taken together, CD4 T cells were the major phenotype and the important $IFN-{\gamma}$ producing T cell subsets in PEC after oral infection with T. gondii whereas $CD8{\alpha}T$ cells had these roles in IEL. These results suggest that PEC and IEL comprise different cell differentials and T cell responses, and according to infection route these factors may contribute to the different cellular immune responses.

TOPLATS 지표해석모형 기반의 고해상도 수문성분 평가 (Evaluation of High-Resolution Hydrologic Components Based on TOPLATS Land Surface Model)

  • High spatio-temporal resolution hydrologic components can give important information to monitor natural disaster. The objective of this study is to create high spatial-temporal resolution gridded hydrologic components using TOPLATS distributed land surface model and evaluate their accuracy. For this, Andong dam basin is selected as study area and TOPLATS model is constructed to create hourly simulated values in every $1{\times}1km^2$ cell size. The observed inflow at Andong dam and soil moisture at Andong AWS site are collected to directly evaluate the simulated one. RMSEs of monthly simulated flow for calibration (2003~2006) and verification (2007~2009) periods show 36.87 mm and 32.41 mm, respectively. The hourly simulated soil moisture in the cell located Andong observation site for 2009 is well fitted with observed one at -50 cm. From this results, the cell based hydrologic components using TOPLATS distributed land surface model show to reasonably represent the real hydrologic condition in the field. Therefore the model driven hydrologic information can be used to analyze local water balance and monitor natural disaster caused by the severe weather.

구강편평세포암종 이식 누드마우스에서의 혈액 점도 변화 (BLOOD VISCOSITY CHANGE IN ORAL SQUAMOUS CELL CARCINOMA XENOTRANSPLANTED NUDE MICE)

  • It is well known that malignant tumor have hypoxic cell fraction, which is radio resistant and is one of the most important cause of local recurrence after radiotherapy. One of the causes of hypoxia in tumor is blood flow decrease due to increase in blood flow resistance and one of the causes of increased blood flow resistance could be attributed to the increase in blood viscosity. For the evaluation of the change of blood viscosity in oral cancer, experiments were carried out to test the change of blood viscosity among the normal control and xenografted oral cancer nude mice. Relative viscosity measured against distilled water was $3.30{\pm}0.14$ for normal control, and $3.67{\pm}0.62$ for tumor bearing mice at the first time of blood sampling in experimental period ($100mm^3$ $200mm^3$). There was no statistically significant difference between the control group and experimental group (p>0.05). However, as the tumor grew, significant difference of blood viscosity was detected at the third time of blood sampling (control group:$3.37{\pm}0.59$, and experimental group: $4.31{\pm}0.41\;300mm^3$

바나듐 레독스 흐름전지용 열가소성 탄소 복합재료 하이브리드 분리판 개발 (Development of Thermoplastic Carbon Composite Hybrid Bipolar Plate for Vanadium Redox Flow Batteries (VRFB))

  • 스택 체결압에 의해 접촉되는 분리판(BP)과 탄소펠트전극(CFE) 사이의 전기적 접촉저항은 상대적으로 낮은 바나듐 레독스 흐름전지(VRFB) 스택의 체결압 때문에 스택 효율에 큰 여향을 미친다. 본 연구에서는 이러한 접촉저항을 줄이고 셀 성능을 향상시키기 위해 국부 가열 접합 공정을 통해 폴리에틸렌(PE) 복합재료-CFE 하이브리드 BP 구조를 개발하였다. 탄소섬유 복합재료 BP의 PE 매트릭스를 국부적으로 녹여 CFE의 탄소 섬유와 BP의 탄소 섬유의 직접 접촉 구조를 만들어 전기 접촉 저항을 감소시겼다. PE 복합재료-CFE 하이브리드 BP의 성능을 평가하기 위해 면적비저항(ASR)과 기체투과도를 측정하였다. 또한 스택 신뢰성을 측정하기 위해 내산성 시험을 수행하였다. 최종적으로, 개발된 PE 복합재료-CFE 하이브리드 BP와 기존의 BP의 성능을 비교 분석하기 위하여 VFRB 단위셀 충/방전 시험을 수행하였다.

  • In this paper a hybrid grid generation program for general 2-D region is introduced. The program is developed by using JAVA programming language, and it can be used either as an application program on a local computer or as an applet in the network environment. The hybrid grid system for a 2-D problem means a combination of triangular cells and quadrilateral cells, and it can offer both of the high flexibility of triangular cells and the high accuracy and efficiency of structured-type quadrilateral cells. To accommodate a quadrilateral-cell region and a triangular-cell region into one computational domain, it is importance to take good care of the interface between two different regions so that overall good grid quality can be maintained. In this research advancing layer method(ALM) augmented by elliptic smoothing method is used for the quadrilateral-cell region and advancing front method(AFM) is used for the triangular-cell region. A special treatment technique for the interface between those two regions is also developed. The interface treatment technique is basically to prevent the propagation of small cell size due to ALM method into the triangular region and maintain the smooth transition of cell-size scale between two different regions. By applying current technique high-quality hybrid grids for general 2-D regions can be easily generated, and typical grid generation results and flow solutions are demonstrated.

Delayed Human Neutrophil Apoptosis by Trichomonas vaginalis Lysate

  • Neutrophils play an important role in the human immune system for protection against such microorganisms as a protozoan parasite, Trichomonas vaginalis; however, the precise role of neutrophils in the pathogenesis of trichomoniasis is still unknown. Moreover, it is thought that trichomonal lysates and excretory-secretory products (ESP), as well as live T. vaginalis, could possibly interact with neutrophils in local tissues, including areas of inflammation induced by T. vaginalis in humans. The aim of this study was to investigate the influence of T. vaginalis lysate on the fate of neutrophils. We found that T. vaginalis lysate inhibits apoptosis of human neutrophils as revealed by Giemsa stain. Less altered mitochondrial membrane potential (MMP) and surface CD16 receptor expression also supported the idea that neutrophil apoptosis is delayed after T. vaginalis lysate stimulation. In contrast, ESP stimulated-neutrophils were similar in apoptotic features of untreated neutrophils. Maintained caspase-3 and myeloid cell leukemia-1 (Mcl-1) in neutrophils co-cultured with trichomonad lysate suggest that an intrinsic mitochondrial pathway of apoptosis was involved in T. vaginalis lysate-induced delayed neutrophil apoptosis; this phenomenon may contribute to local inflammation in trichomoniasis.
