Natural Occurrence of Fumonisins $B_1$ and $B_2$ in Korean Corns (한국산 옥수수의 Fumonisin $B_1$$B_2$ 오염현황)

  • One hundred and ninety seven corn samples, which collected from each local province in Korea between 1995 and 1996, were analyzed for fumonisin $B_1\;(FB_1)$ and $B_2\;(FB_2)$ contamination using fluometric HPLC. $FB_1$ and $FB_2$ were detected in 143 (72.6%) and 128 (62.4%) samples from 197 samples at the concentrations ranging from 0 to $224.2\;{\mu}g/g$ for $FB_1$ and from 0 to $60.0\;{\mu}g/g$ for $FB_2$, respectively. The total fumonisin levels analyzed from 143 samples were 15.4, 37.5 and 14% for concentrations below $10\;{\mu}g/g$, above $10\;{\mu}g/g$ and above $30\;{\mu}g/g$, respectively. The total fumonisin levels containing above $10\;{\mu}g/g$ were detected the highest concentration with 53.2% in samples collected from Kangwon, and followed by Kyeongki (45.0%), Cheonbuk (40.0%), Chungbuk (26.9%) and Kyeongbuk (11.1%). The total fumonisin level containing above $10\;{\mu}g/g$ in samples collected in 1996 was higher than that in 1995. Both fumonisin $B_1$ and $B_2$ from samples collected in 1995 were detected higher than that in 1996 and $FB_1$ was detected the highest level in samples collected from Kyeongki province and followed by Kangwon, Cheonbuk, Chungbuk and Kyeogbuk (11.1%). Also, similar results were observed in $FB_2$.

  • The effects of solvent type and concentration, solid/liquid ratio, extraction time and repeated extraction on recovery of keratinase from solid-state fermentation (SSF) of chicken feather by a local Streptomyces sp. NRC 13S were investigated in order to establish the experimental conditions for keratinase yield. Among solvents tested, 0.5% (v/v) glycerol was the best. Box-Behnken design was used to investigate the effect of relevant variables on keratinase recovery. The factors investigated were solid/liquid ratio (1:1.66-1:6.66 g/mL), glycerol concentration (0.5-5% v/v) and repeated extraction (1-5 cycle). The results showed that the maximum recovery of keratinase (6933.3 U/gfs) was obtained using 0.5 (v/v) glycerol as extracting solvent, in a solid/liquid ratio of 1:5 and three extraction cycles.

Somatic Embryogenesis and Plant Regeneration in Shoot Apical Meristem Cultures of an African Local Variety Cassava (Manihot esculenta Crantz) (아프리카 재래종 카사바의 경단분열조직 배양에 의한 체세포배발생과 식물체 재분화)

  • Shoot apical meristem dome explants from cassava plants (Ghanaian local variety) produced somatic embryos at a frequency of 32% when cultured on MS medium supplemented with 2 mg/L 2,4-D. Somatic embryo segments formed secondary embryos at frequencies of up to 93% when cultured on medium containing 1 mg/L 2,4-D for 2 to 3 weeks. Since the somatic embryos were not capable of converting into plantlets, adventitious shoot were induced from the sliced embryo segments by culturing them on medium containing 0.1 to 5 mg/L BA. After 8 weeks of culture, numerous shoots formed on the segments at frequencies up to 100%. The shoots were rooted and successfully transplanted to potting soil.

Pain alleviation in patients undergoing cardiac surgery; presternal local anesthetic and magnesium infiltration versus conventional intravenous analgesia: a randomized double-blind study

  • Background: Magnesium is one of the effective, safe local anesthetic adjuvants that can exert an analgesic effect in conditions presenting acute and chronic post-sternotomy pain. We studied the efficacy of continuous infusion of presternal magnesium sulfate with bupivacaine for pain relief following cardiac surgery. Methods: Ninety adult patients undergoing valve replacement cardiac surgery randomly allocated into three groups. In all patients; a presternal catheter was placed for continuous infusion of either 0.125% bupivacaine and 5% magnesium sulfate (3 ml/h for 48 hours) in group 1, or 0.125% bupivacaine only in the same rate in group 2, versus conventional intravenous paracetamol and ketorolac in group 3. Rescue analgesia was iv $25{\mu}g$ fentanyl. Postoperative Visual Analog Scale (VAS) and fentanyl consumption during the early two postoperative days were assessed. All patients were followed up over two months for occurrence of chronic post-sternotomy pain. Results: VAS values showed high significant differences during the first 48 hours with the least pain scale in group 1 and significantly least fentanyl consumption ($30.8{\pm}7{\mu}g$ in group 1 vs. $69{\pm}18{\mu}g$ in group 2, and $162{\pm}3$ in group 3 respectively). The incidence of chronic pain has not differed between the three groups although it was more pronounced in group 3. Conclusions: Continuous presternal bupivacaine and magnesium infusion resulted in better postoperative analgesia than both presternal bupivacaine alone or conventional analgesic groups.

Distribution Status, Physicochemical Composition, and Physiological Activity of Spergularia marina Cultivated in the Western Region in Jeon-Ra-Nam-Do (전남 서부 해안가에서 생산되는 세발나물의 유통실태, 이화학적 성분 및 생리활성)

  • This study was conducted to examine the distribution status, the physicochemical composition, and the physiological activity of Spergularia marina grown in the western region in Jeon-Ra-Nam-Do, looking for its potential as a local specialty goods. The amount sold of Spergularia marina to different areas from a rural market place was examined during April, 2008. The proportions of Spergularia marina are as follows; Yeongam(30.5%), Muan(24.3%), Naju(20.3%), Hampyeong(18.0%), and Haenam(7.0%), which were shown higher than those of Pimpinella brachycarpa. Total vitamin C and total chlorophyll contents in Spergularia marina were 68.61mg/100g DW and 58.11mg/g DW, respectively. Total amino acid contents were 331.94mg/100g DW and 20 kinds of amino acids were separated and classified. The contents of inorganic matters in Spergularia marina were as follows; Na(64.23mg/L), Mg(15.88mg/L) K(9.99mg/L), Ca(3.83mg/L). The total phenol and the total flavonoid contents of methanol extracts from Spergularia marina were 41.7mg/L and 18.9mg/L, respectively, with an extracting concentration of 2,000mg/L. The nitrite scavenging activity was highest (83.4%) in hot water extract. These results may be useful as basic data for using Spergularia marina as a local specialty goods.

Effects of Silkworm (Bombyx mori) Pupa Extract on the Function of Osteoblastic MC3T3-E1 Cells

  • Osteoporosis is recognized as one of the major hormonal deficiency diseases, especially in menopausal women and the elderly. When the estrogen level is reduced in the body, local factors, which are known to be related with bone resorption, are increased and promote osteoclastogenesis. In our previous study, we validated the estrogenicity of silkworm pupa. In this study, we investigated the effect of silkworm pupa extract (SPE) on the function of osteoblastic MC3T3-E1 cells. SPE (10 and $50\;{\mu}g/mL$) significantly elevated cell viability, alkaline phosphatase (ALP) activity, and collagen content in the cells. The effect of SPE ($50\;{\mu}g/mL$) in increasing cell viability, ALP activity, and collagen content was completely inhibited by the presence of $10^{-6}\;M$ of cycloheximide and $10^{-6}\;M$ of tamoxifen, suggesting that SPE's effect results from a newly synthesized, protein component and that it might be partly involved in estrogen action. Furthermore, we examined the effect of SPE on the $H_2O_2-induced$ apoptosis and production of local factors in osteoblasts. Treatment with SPE ($50\;{\mu}g/mL$) decreased the 0.2 mM $H_2O_2-induced$ apoptosis and the production of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6 and nitric oxide (NO) in osteoblasts. Our data indicate that the enhancement of osteoblast function by silkworm pupa may prevent osteoporosis and inflammatory bone diseases.

Radiation Therapy in The Treatment of $T_1$ Glottic Cancer (병기 $T_1$ 성대종양 환자의 방사선치료성적에 관한 고찰)

  • Radiation therapy is generally considered to be the treatment of choice in $T_1$ glottic cancer, maninly because of preservation of voice function and its local control rate is comparable to that of surgery. Failures from radiation therapy can be ultimately salvaged by surgery. A retrospective analysis of the treatment of 25 patients with $T_1$ glottic cancer seen at the Yonsei Cancer Center from 1980 to 1984 is presented. Radiation dose to the target volume varied from 6400 to 7000 cGy in 6-7 weeks. The local control rate is 84%. Four patients had primary failure and three of these patients had salvage surgery. Of the 3 patients who had salvage surgery, 2 were cured and aonther one was died with progression of the disease. 5-year acturial and disease free survival rate are 91.1%,78.0% respectively.

Influence of Sugar Cane Diets and a High Fibre Commercial Diet on Growth and Carcass Performance in Local Caribbean Pigs

  • The aim of this study was to evaluate the effect of a milling by-product diet and two sugar cane diets on the local Creole pig breed (CR). A total of 48 CR pigs (24 females and 24 castrated males) were randomly assigned to four different groups of 12 animals. Pigs were allotted to one of 4 dietary treatments: fed with a control soya-bean meal-corn diet containing 19.1% crude protein (CP) and 15.4 MJ DE/kg (diet 1), with an experimental milling by-product diet (soya-bean meal and wheat by-products) containing 19.4% CP and 13.0 MJ DE/kg (diet 2), with ground cane stalks (GCS) or with fresh sugar cane juice (SCJ). Both GCS and SCJ were supplemented with soya-bean meal complement (400 g/d of a 48.7% CP and 16.1 MJ DE/kg diet) in order to obtain diets 3 and 4, respectively. Pigs were fed close to ad libitum level and had free access to water. All the pigs were slaughtered at 65 kg BW. Between 30 and 65 kg BW, growth performance was significantly (p<0.001) affected by dietary treatments: average daily BW gain was 657, 530, 546 and 200 g/d for diets 1, 2, 4, and 3, respectively. Average daily DM intake was 1.8, 1.9, 2.5 and 1.4 kg/d for diets 1, 2, 4, and 3, respectively. Fat cuts (backfat+leaf fat) and backfat thickness were significantly lower on diet 3 than for other treatments (127 vs. 192, 166 g/kg of left half-carcass weight and 24.6 vs. 39.0, 35.3 mm for diet 3 vs. diets 1 and 4, and diet 2, respectively; p<0.001). The dressing weight was significantly lower on diets 2 (82.7 vs. 84.0%; p<0.001). The entire empty digestive tract (DT) weight was higher on diet 2 (73.1 vs. 66.7 g/kg empty BW). However, stomach and large intestine were more developed on diet 3: 12.8 vs. 9.3 g/100 g empty DT (p<0.001) and 26.4 vs. 23.8 g/100 g empty DT (p<0.05), respectively. In conclusion, this study suggests the CR pig has the ability to reach rather good growth and carcass performance with a well-formulated sugar cane meal and/or with a milling by-product diet refined according to its low requirements.

An Assessment of Dietary Fiber Intake in Preschool Children in Busan (부산지역 학령전 아동의 식이섬유섭취 상태평가)

  • To assess the dietary fiber intake of preschool children in Busan and to evaluate the relationship between of the intake of dietary fiber and nutrient intake. Nutrient intake using 24 hour recall, and total dietary fiber (TDF) intake based on tables of TDF of common Korean floods developed by the modified Prosky Method, were estimated for 176 preschool children. The mean daily intakes of TDF, and TDF after adjusting energy intake, were 10.20 g and 7.69 g/1,000 kcal, respectively. The mean daily intakes of TDF for children aged 1-3 and 4-6 years were 9.20 g and 11.08 g, respectively. The range of TDF intake was 1.86 to 22.16 g. The major sources of TDF were cereals (31.0%), vegetables (18.9%) and fruits (11.9%). The TDF intake showed positive correlations with nutrient adequacy ratios (NAR) of iron and Vitamin $B_1$, (p < 0.05, p < 0.05). The TDF intake per 1,000 kcal showed negative correlations with the NARs of protein, calcium, phosphate, iron, Vitamin A, vitamin $B_1$, Vitamin $B_2$, and niacin (p < 0.001, p < 0.001, p < 0.001, p < 0.05, p < 0.001, p < 0.001, p < 0.001, p < 0.001) and with the mean adequacy ratio (MAR, p < 0.001). When children were stratified into quartiles ($Q_1-Q_4$) on the basis of their fiber intake per 1,000 kcal, their NARs for calcium, phosphate, iron, Vitamin A, Vitamin $B_1, Vitamin $B_2$ and niacin (p < 0.05, p < 0.05, p < 0.001, p < 0.05, p < 0.05, p < 0.001, p < 0.001), and their MAR (p < 0.001) were significantly lower in the children with higher fiber intake per 1,000 kcal (the upper quartile). The NARs fur calcium (0.63), iron (0.60), Vitamin A (0.66), Vitamin $B_2$(0.74), niacin (0.64), Vitamin C (0.65) and the MAR (0.74) were lower than 0.75 in the children with fiber intakes of more than 9.25 g per 1,000 local (0,), the highest fiber intake per 1,000 kcal. Based on these results, the mean TDF intake of children was higher than the age (yr)+5g , the minimum recommended level for American children. Meals with a fiber intake of more than 9.25 g per 1,000 local ($Q_4$) could cause a decreased nutritional status for minerals and vitamins. The result of this study could contribute to the establishment of Recommended Dietary Allowances (RDA) for dietary fiber for Korean Preschool children.

Characteristics of Shear Strength of Coarse Grained Materials Depending on Experimental Densities (시험밀도에 따른 조립재료의 전단강도 특성)

  • The density of coarse grained materials which is used in rock-fill dam or the piling the ground are used as $1.85\sim2.10g/cm^3$. Hereupon, the effect of variation of density on shear strength of ones was analyzed from the results of large scale shear test. The sample for the test was obtained from the local quarry sites. The test conditions are that density(1.85 versus $2.10g/cm^3$), material size range$(76.3\sim2.0\;mm)$, water content(air dry condition) and uniformity coefficient(5.0) Test result shows that the shear strength of $2.10g/cm^3$ is relatively larger than that of $1.85g/cm^3$.
