A survey of Newcastle disease virus antibody titers on slaughtered chickens (도계장 출하 닭에 대한 뉴캐슬병 항체가 조사)

  • To investigate serum antibody titers of Newcastle disease (ND) virus, serum samples were collected from 19,774 slaughtered chickens from 862 farms in Geoje, Goseong, Tongyung and other province during the period from January 2001 to December 2005. The results obtained are summarized as follows; 1. The negative percentage of ND antibody titer with below 1.0 levels decreased from 46.7% in 2001 to 40.5% in 2004 but those of it increased to 47.0% in 2005 and the positive percentage of ND antibody titer with higher 5 levels increased from 15.8% in 2001 to 26.2% in 2005. 2. The season levels of ND antibody titer showed various levels according to seasons but the general trend of ND antibody titer decreased in the later half of the year than those of the first half of the year. 3. The negative percentage of ND antibody titer on provinces decreased from 52.0% (2001) to 36.2% (2004) in Goseong but those of it increased to 51.4% in 2005. The negative percentage of ND antibody titer of Tongyung and other provinces increased from 51.0%, 28.5% (2001) to 55.0% and 44.3% (2005), respectively. 4. The year average levels of ND antibody titer increased from 2.2 to 2.6 during 2001- 2004 but those of it decreased to 2.4 in 2005. The provinces average levels of ND antibody titer increased from 2.0 to 2.8 during 2001 - 2005 and others provinces except Gyeongnam decreased from 3.3 to 2.6 during the 2001-2005. 5. The farm average levels of ND antibody titer with below 1.0 levels decreased from 26.3% in 2001 to 10.2% in 2004 but those of it increased to 17.6% in 2005 and those of below 1.0 or 2.0 levels increased from 17.9% (2001) to 27.7% (2005). The protective levels of ND antibody titer with higher 5.0 levels increased from 2.1% in 2001 to 8.8% in 2005.

  • Purpose: Classical swine fever (CSF) reemerged on CSF-free Jeju Island where vaccination is not practiced by the unintentional injection of a live attenuated vaccine (modified live attenuated vaccines-low-virulence Miyagi [MLV-LOM]) in 2014. Since the Jeju provincial authority is considering adopting a voluntary immunization policy using a CSF-E2 subunit vaccine to combat LOM-derived CSF endemic, this study aimed to evaluate in Jeju herds. Materials and Methods: Two vaccination trials using the Bayovac CSF-E2 vaccine licensed for use in South Korea assessed the safety and humoral immunity of the CSF-E2 vaccine in breeding (trial 1) and nursery animals (trial 2) under farm application conditions. Results: Neither local nor systemic (including reproductive) adverse effects were objectively observed in pregnant sows and young piglets following a respective vaccination regime at pregnancy or weaning, respectively. Trial 1 showed that sows immunized with the CSF-E2 vaccine possessed high and consistent E2-specific and neutralizing antibody levels. The CSF-E2 vaccine-immunized pregnant sows subsequently conferred appropriate and steady passive immunity to their offspring. In trial 2, a double immunization scheme of the CSF-E2 vaccine in piglets at 40 and 60 days of age could elicit a consistent and long-lasting adequate antibody response. Additionally, the two trials detected no Erns-specific antibody responses, indicating that CSF-E2 vaccine can differentiate infected from vaccinated animals (DIVA). Conclusion: Our trial data collectively provide invaluable information on applying the CSF-E2 subunit vaccine to circumvent the possible drawbacks associated with the MLV-LOM concerning the safety, efficacy, and DIVA, in the LOM-endemic field farms and contribute to advanced CSF eradication on Jeju Island.

Diagnosis and seroprevalence of porcine respiratory coronavirus disease (돼지호흡기코로나바이러스 감염증의 감별진단과 항체분포 조사)

  • Porcine respiratory coronavirus (PRCV) is antigenically related to transmissible gastroenteritis virus (TGEV). Differential serological diagnosis between PRCV and TGEV infection is not possible with the classical sero-neutralization test. Infection with PRCV or TGEV induces antibodies which neutralize both viruses to the same titer. However, the enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) can differentiate between PRCV and TGEV infection. This study was carried out to investigate the prevalence of PRCV infection of swine in Gyeongnam province. A total of 391 serum samples from 37 herds in Gyeongnam were examined for antibody to PRCV using blocking ELISA. All serum samples were collected from 130- to 150-day-old pigs between August and December 2006. By ELISA, 182 out of 391 sera tested (46.5%) and 29 out of 37 sample herds (78.4%) were positive against PRCV. Our data suggested that seropositive herds for PRCV are distributed diffusely throughout Gyeongnam. The PCR methods were established to diagnose PRCV spike protein (S) gene. PCR were conducted to identify the PRCV genome against 150 pigs in PRCV antibody positive herds.

Investigation of atrophic piglets diseases in northern area of the Gyeongnam province, Korea (경남 북부지역 위축자돈에 대한 질병조사)

  • This study was investigated to diagnose pathogenic organisms of atrophic piglets in northern area of the Gyeongnam province, Korea. Samples such as feces, blood and necropsy specimens of 42 atrophic piglets (${\leq}10$ weeks old) were taken from May to December 2013 for this survey. Samples were examined by reverse transcriptase-polymerase chain reaction (RT-PCR) assay and bacteria isolation for detection of pathogenic agents. 93 pathogens were isolated from 42 samples can be classified into ll groups. We identified bacterial agents in 56 cases (60.2%) and viral agents in 31 cases (33.3%). However, 6 cases (6.5%) were undetected. Among these pathogens, the most prevalent disease were porcine reproductive and respiratory syndrome (PRRS) in 22 cases (23.7%). The major diseases were Colibacillosis in 15 cases (16.1%), Glasser's disease in 12 cases (12.9%), and porcine epidemic disease (PED) in 9 cases (9.7%). Mixed infections were accounted for 77.8% of atrophic piglets. In particular, the rate of mixed infections with PRRS virus showed the highest frequency (71.4%). In addition, there is a seasonal variation. Viral pathogens were dominantly detected in winter, but in the rest of the season bacterial agents were mainly detected. Gastrointestinal diseases occurred mainly in the pre-weaning piglets, the respiratory diseases and wasting diseases occurred mainly in the post-weaning piglets.

Monitoring for prevalence of mosquitoes collected and pathogen infection (세종지역 질병매개모기 분포 및 병원체 감염 실태 조사)

  • This survey is performed for the vector mosquitoes including of Japanese encephalitis, Malaria and West niles virus. Black light traps were installed in 5 sites (Sejong lake, Janggun-myeon, Jeonui-myeon, Yeongi-myeon, Bukang-myeon). Mosquitoes were collected twice every week from five collection sites using a black light trap from May to September. A total of 10 species belonging to 4 genera, 14,234 female mosquitoes were collected and identified as vector mosquitoes. Among ten mosquito species, Culex pipiens complex was the most commonly mosquitoes (6,163 individuals, 43.3%). The greatest number of Culex pipiens pallens was trapped in the first week of July. A total of 115 pools from 14,234 mosquitoes were prepared according to their species, collection date and site. We have screened infection of virus in mosquitoes. All of virus were not detected in mosquitoes. Our findings confirm that population and first appearance time of vectors mosquitoes and virus infection in Sejong city.

Histopathological features and viral genome detection in caprine arthritis encephalitis virus infected dairy goats in Korea

  • Caprine arthritis encephalitis (CAE) virus is a causative agent of caprine arthritis-encephalitis. In our previous study we reported a prevalence of CAE. In this study, we described the further detailed pathological features of CAE and examined the detection of virus by in situ hybridization (ISH). Histopathologically, interstitial pneumonia and bronchopneumonia in lung, focal inflammation in mammary glands, perivascular cuffing in brain, arthritis, and focal necrosis, mild steatosis, inflammatory cell infiltration of liver were noted. CAEV proviral-DNA was identified by nested polymerase chain reaction (PCR) in blood cells, brain, synovial fluid, and lymph node. Confirmation by nested PCR involved amplification of a 296 bp ($1^{st}$ PCR) and 185 bp ($2^{nd}$ PCR) fragments corresponding to a conserved region on the gag gene of CAEV. Positive ISH signals were detected in the brain and liver. In conclusion, significant histopathological findings included parenchymal infection in various organs, including the lung, liver, brain, joint, and mammary gland were noted in the CAEV infected dairy goat. ISH can help confirm the diagnosis of CAE in formalin-fixed samples.

Establishment of reverse transcription polymerase chain reaction for detection of Getah virus infection in livestock

  • Getah virus (GETV) infection causes sporadic outbreaks of mild febrile illness in horses and reproductive failure in pigs. In this study, we established a reverse transcription polymerase chain reaction (RT-PCR) method to detect GETV from suspected virus-infected samples. The reaction conditions were optimized and validated by using RNA extracted from GETV propagated in cell culture. A GETV-specific GED4 primer set was designed and used to amplify a 177 bp DNA fragment from a highly conserved region of the E1 glycoprotein gene in the GETV genome. RT-PCR performed with this primer set revealed high sensitivity and specificity. In the sensitivity test, the GED4 primer set detected GETV RNA at the level of $10^{2.0}\;TCID_{50}/mL$. In the specificity test, the GED4 primer set amplified only a single band of PCR product on the GETV RNA template, without non-specific amplification, and exhibited no cross-reactivity with other viral RNAs. These results suggest that this newly established RT-PCR method is useful for accurate identification of GETV infection in animals.

  • “DK 537”has been selected by forage crop breeding team at the National Livestock Research Institute(NLRI), RDA and has been determined by the Deliberative Council of the National Agricultural Cooperative Federation(NACF) in 2002, as a new recommended hybrid which is early maturing, good quality and hi호 yield of corn for silage. The characteristics of this hybrid are as follows ; 1. The seed coat is yellow and the mean of tasseling date is 5th of July, included in early maturing hybrid which is one day delayed than check cultivar, DK 501. The culm length is 246cm. It is resistant to lodging because of its low height from surface to ear. 2. DK 537 shows resistance to H. maydis and Maize Black-Streaked Dwarf Virus(MBSDV). It also shows strength to corn borer as much as DK 501 does. 3. Fresh yield. dry matter yield. and TDN per a ha are 50 tons, 16.9 tons, and 11.4 tons respectively. which are almost same yielding level of DK 501. Its percent ear to total dry matter is 50.8% at the same time. Through all these tests, we could make sure at DK 537 hybrid will be recommended as good forage crop.

Investigations of bacterial contamination level and prevalence of major viral disease for fresh-extended porcine semen (인공수정용 돼지 액상정액 세균오염도 조사 및 정액유래 주요 바이러스성 질병 감염률 조사)

  • Bacteroiospermia is a frequently finding in fresh raw and extended porcine semen and can results in detrimental effects on semen quality and longevity. This study aims to evaluate the type of bacterial contaminants in raw and extended porcine semen and the reducing effect of antibiotic test. To investigate bacterial contaminants, out of 387 sample (raw semen 201, extended semen 186) were collected from 6 artifical insemination centers in Gyeongsangnam-do, were inoculated onto blood agar and MacKonkey agar, respectively. Bacterial colonies were selected after culturing for 48 hours, at $37^{\circ}C$, followed by Gram staining, KOH test, oxidase test, catalase test and eventually identified using VITEK System. Total 15 genus and 24 species of bacteria were isolated from these semen samlpes. In raw semen, the most prevalent contaminants were Pseudomonas aeruginosa, Escherichia coli, Proteus mirabilis, Staphylococcus auricularis, Delftia acidovorans, Acinetobacter lowffii, S. aureus and others. And in extended porcine semen, A. lowffii, S. aureus, S. auricularis and other bacteria were identified. Most of them was G(-), which is nonpathogenic bacteria. It seems that bacterial contaminants in fresh raw and extended porcine semen originated from multiple sources at the farms/stud, and were from animal origin and non-animal origins. Whereas, the 7 virus which is known to be detected in porcine semen in 75 cases was not detected. This results showed that removal of bacterial contamination in raw and extended porcine semen is essential and farms were kept for biosecurity and individual hygienes.

Cost Analysis for the Carcass Burial Construction (가축매몰지 조성비용 평가)

  • A lot of burial sites for the disposal of the contagious livestock slaughtered by foot and mouth disease were constructed in a short time for preventing the rapid spread of the virus. More than 4,700 carcass burial sites were constructed in 2011. Approximately 7 million poultry and 3.5 million livestock including head of cattle and swine were buried in farm land. However, the available cost data for a burial site was not provided. This study was to performed in order to understand the economic issues related to carcass burial disposal. The data from a local government was analyzed and the assumption data web based was constructed to evaluate the cost for constructing a carcass burial. The results showed that the local government paid KRW 5,386 for the burial disposal of a mortality (swine). It was estimated that the cost could be reduced through an appropriate measures.
