The Antioxidant Effect of Lactobacillus gasseri KACC 91155 Isolated from Korean Infant in Jurkat T Cells (유아의 분변에서 분리한 Lactobacillus gasseri KACC 91155의 Jurkat T Cells에서 항산화 효과)

  • In the present study, we investigate the protective effect of antioxidant strain Lactobacillus gasseri KACC 91155, isolated from Korean infant feces(Obstetrics & Gynecology, Suwon, Korea) on the oxidative stress damage on the Jurkat T cells. To estimate the extent of cellular lipid peroxidation inhibition, MDA(malondialdehyde) production was measured Furthermore, cell viability was detected by the MTT assay, DNA damage was tested by the comet assay. Cell grown in medium with or without L gasseri lysate$(100\~1,000{\mu}g)$ were treated with $H_2O_2,\;Fe^{2+}$ as an oxidative stimulus. From the result obtained, the supplementation of Jurkat T cells with L. gasseri lysate significantly decreased in MDA production (1,250 vs. 835 nmol/mg protein), and DNA damage(31.6 vs. 22.6 tail moment). Also L gasseri increase cell viability against oxidative damage. We concluded that the L. gasseri KACC 91155 showed a protective effect against oxidative stress.

Application Effects of Organic Fertilizer Utilizing Livestock Horn Meal as Domestic Organic Resource on the Growth and Crop Yields (국내산 유기자원 우각을 활용한 유기질비료의 작물 생육 및 수량에 미치는 영향)

  • Objective of this study was to develop an organic fertilizer utilizing domestic livestock horn meal and to investigate the application effect of rice and eggplant. The possibility of utilization of livestock horn meal as an organic resource to replace imported expeller cake fertilizer was examined. In order to select domestic organic resources with high nitrogen content, 8 kinds of organic matter such as chicken manure, fish meal, soybean meal, sesame meal, perilla meal, blood meal, livestock horn meal, and beer sludge were analyzed and organic resources with high nitrogen content were selected. In addition, the conditions for the production of organic fertilizers that can be used in organic agriculture were established by mixing of the rice husk biochar and the rice bran as the supplements with the raw materials for mixing ratios. The content of total nitrogen (T-N) in the livestock horn meal was 12.0 %, which was the next low in 13.5 % blood meal. The content of total nitrogen was 5.9 ~ 7.9 % in fish meal and oil cakes. Total nitrogen content of non-antibiotic chicken manure for organic farming was 3 % and nitrogen content in beer sludge was 3.5 %. Organic fertilizer was produced by using biochar, rice bran as a main ingredient of non-antibiotic chicken manure, livestock horn meal and beer sludge. Compared to nitrogen content (4.0 to 4.2 %) of imported expeller cake fertilizer (ECF), the nitrogen content of organic fertilizer utilizing domestic livestock horn meal is as high as 7.5 %. The developed organic fertilizer is met as Zn 400 mg/kg, Cu 120 mg/kg the quality of organic agricultural materials such as or less. To investigate the effect of fertilizer application on the crops, prototypes of developed organic fertilizer were used for the experiment under selected conditions. As a result of application the developed organic livestock horn meal fertilizer (LHMF) for cultivation of the rice and eggplant, the application quantity of the developed organic LHMF 100 % was decreased by 40 % compared to that of the mixed expeller cake fertilizer (MECF). The application of LHMF, which refers to the application rate corresponding to the nitrogen fertilization recommended by the soil test, was reduced by 40% compared to the application rate of MECF, but the same results were obtained in crop growth and yield. The selection of a new high concentration nitrogen source utilizing domestic organic resources and the development of organic fertilizer is the starting point of the research for substitution of imported ECF using domestic local resources at the present time that the spread of eco-friendly agriculture is becoming increasingly important. If it is expanded in the future, it is expected to contribute to the stable production of eco-friendly agricultural products.

Influences of Calving Year, Calving Season and Parity on the Lactation Curve of Korean Cattle (분만년도, 계절 및 산차가 한우의 비유곡선에 미치는 영향)

  • This study was conducted to investigate the effect of environmental factor on milk yield and to estimate lactation curve in Korean cattle. The data for milk yields were collected from 118 cows from 1997 to 2000 at National Livestock Research Institute in Daekwanryoung, Kangwon-do. Average daily milk yields for 1st, 2nd, 3rd and 4th month after calving were 3.74kg, 3.64kg, 3.26kg and 2.99kg. Average daily milk yield for the four months was 3.52kg. The milk yields for cows calved in spring were larger than those calved in fall. Lactation curve of Korean cattle was $y_n$=$2.4845n^{0.1734}e^{-0.0060n}$. Peak milk yield was 3.75kg on 29.03 day after calving. The peak milk yields for multi-parous cows were larger than those of primiparous cows. The peak milk yields for multi-parous cows reached later than those for primiparous cows. The cows calved in spring had higher and earlier peak milk yields than those calved in fall had.

Increased of the Red Blood Cell in Peripheral Plasma of Transgenic Pigs Harboring hEPO Gene

  • The present study were performed to analysis the hematocrit and the red blood cells content into the blood plasma of the transgenic pigs harboring recombinent human erythropoietin gene (rhEPO). Mouse whey acidic protein (mWAP) linked to rhEPO gene was microinjected into pronuclei of porcine one-cell zygotes. After delivered of offspring, PCR analyses identified one mWAP-rhEPO transgenic founder offspring(F$_{0}$). The first generation of transgenic pig (F$_{0}$) harboring mWAP-hEPO appeared to be a male, and the second generation (F$_1$) pigs were made by natural mating of F$_{0}$ with domestic swine, and male and female transgenic pigs (F$_1$) were identified by PCR. The blood samples from transgenic and normal pigs were collected for 50 days during lactation and were counted the red blood cell (RBC) numbers and Hematocrit (HCT) content into the blood. The transgenic pigs expressing rhEPO in their blood gave rise to higher RBC numbers and HCT contents than control animals. rhEPO was secreted both in the blood and milk of genetically engineered pigs harboring rhEPO gene. Therefore, this study provides a model regarding the production of transgenic pig carrying hEPO transgene for biomedical research.earch.

  • Human-induced soil salinity is becoming a major threat to agriculture across the world. This salinisation occurs in both irrigated and rain-fed agricultural zones with the highest proportions in the arid and semi-arid environments. Livestock can play an important role in the management and rehabilitation of this land. There are a range of plants that grow in saline soils and these have been used as animal feed. In many situations, animal production has been poor as a result of low edible biomass production, low nutritive value, depressed appetite, or a reduction in efficiency of energy use. Feeding systems are proposed that maximise the feeding value of plants growing on saline land and integrate their use with other feed resources available within mixed livestock and crop farming systems. Salt-tolerant pastures, particularly the chenopod shrubs, have moderate digestible energy and high crude protein. For this reason they represent a good supplement for poor quality pastures and crop residues. The use of salt-tolerant pasture systems not only provides feed for livestock but also may act as a bio-drain to lower saline water tables and improve the soil for growth of alternative less salt tolerant plants. In the longer term there are opportunities to identify and select more appropriate plants and animals for saline agriculture.

A Study on Life Style of Rural Homemakers (농촌주부의 가정생활양식에 관한 연구)

  • The purpose of this study is to offer basic data of education to improve farm lives though investigating rural homemakers' life style. The results of this study are as follows : 1. The life structure of rural homemakers was classified into two structures as a material resources and human resources. The life style of rural homemakers was classified into systematical life style, traditional value style, social active style, and progressive active style. 2. The life styles of rural homemakers according to social population variance were as follows. The lower of age, homemakers whose family incomes more 30 million won or below 10 million won, who has religions, who employed in livestock were showed at very systematical life style. The homemakers who live in mountain village, higher the much age becomes, and the education level become lower, have showed traditional value style. Rural homemakers the lower of age, the higher incomes, composed of single parent + children who remain single were appeared social active style. Rural homemakers the lower of age, the more shortened farm life periods, who employed in livestock were appeared progressive active style. 3. Systematical life style was more affected by material resources structure, traditional value style was affected by material resources. And social active style was more affected by human resources, progressive active style was affected by human resources.

Establishment of Database and Distribution Maps for Biomass Resources (바이오매스 자원 DB 구축과 분포도 작성)

  • This study was carried out to understand the national and regional distribution of the biomass resources produced in Korea annually via establishing database (DB) and distribution maps of biomass resources data including as livestock manures, food wastes and agricultural by-product. The information of the annual production of each biomass resources was obtained from Ministry for Food, Agriculture, Forestry and Fisheries (MIFAFF), Ministry of Environment (MOE) and National Statistical Office (NSO). Based on biomass resources data, we established database architecture table about livestock manures and food wastes. The distribution maps for the total amount of manures produced from each livestock animal were built up in both national and regional scales and used for analysis of the space-based and time-based distribution of the manure resources. Distribution maps for food wastes and agricultural by-product were also produced, respectively. It was shown that the analysis through resource mapping can be used to identify the sources of collectable biomass feasibly determining suitable region for establishment of a biomass-energy production. The biomass distribution maps graphically provide the information regarding biomass resources to policy-makers, farmers, general users and it was expected to be utilized for policy-making of environmental-friendly agriculture and bio-energy.

Confirming Single Nucleotide Polymorphisms from Expressed Sequence Tag Datasets Derived from Three Cattle cDNA Libraries

  • Using the Phred/Phrap/Polyphred/Consed pipeline established in the National Livestock Research Institute of Korea, we predicted candidate coding single nucleotide polymorphisms (cSNPs) from 7,600 expressed sequence tags (ESTs) derived from three cDNA libraries (liver, M. longissimus dorsi, and intermuscular fat) of Hanwoo (Korean native cattle) steers. From the 7,600 ESTs, 829 contigs comprising more than two EST reads were assembled using the Phrap assembler. Based on the contig analysis, 201 candidate cSNPs were identified in 129 contigs, in which transitions (69%) outnumbered transversions (31%). To verify whether the predicted cSNPs are real, 17 SNPs involved in lipid and energy metabolism were selected from the ESTs. Twelve of these were confirmed to be real while five were identified as artifacts, possibly due to expressed sequence tag sequence error. Further analysis of the 12 verified cSNPs was performed using the program BLASTX. Five were identified as nonsynonymous cSNPs, five were synonymous cSNPs, and two SNPs were located in 3'-UTRs. Our data indicated that a relatively high SNP prediction rate (71%) from a large EST database could produce abundant cSNPs rapidly, which can be used as valuable genetic markers in cattle.

Sperm Fertility of Transgenic Boar Harboring hEPO Gene is Decreased

  • This study was conducted to compare the reproduction ability of the wild type boar and recombinant human erythropoietin (hEPO) transgenic boar semen. Ejaculated boar semen was analyzed by flow cytometry, Elisa and IVF methods. In experiment 1, flow cytometric analysis showed that the live sperm ratio of transgenic boar sperm significantly lower (P<0.05) than that of wild type boar after incubation at 20, 22, 24 and 26 hr. In experiment 2, the presence and levels of various cytokines (IL-6, IL-10 and $TNF-{\alpha}$) to related animal reproduction in the seminal and blood plasma were examined using specific enzyme immunoassay. There was no significant difference between both groups. In experiment 3, the fertilizing capacity and developmental ability of both boar sperm were compared. The transgenic boar sperm had a significantly low capacity of penetration, sperm-zona binding, embryo development, and blastocyst formation compared to wild type sperm (P<0.05). These results suggest that transgenic boar sperm harboring hEPO gene has low sperm viability than wild type boar, and it is a reason to decrease of fertility and litter size.

Association of functional sequence variants of the myosin heavy chain 3 gene with muscle collagen content in pigs

  • This study examined the association between functional sequence variants (FSVs) of myosin heavy chain 3 (MYH3) genotypes and collagen content in a Landrace and Jeju native pig (JNP) crossbred population. Four muscles (Musculus longissimus dorsi, Musculus semimembranosus, Musculus triceps brachii, and Musculus biceps femoris) were used for the analysis of meat collagen content, and the same animals were genotyped for the FSVs of the MYH3 gene by using PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism). Three FSVs of MYH3 genotypes were identified and had genotype frequencies of 0.358, 0.551, and 0.091 for QQ, Qq, and qq, respectively. QQ animals for the FSVs of the MYH3 genotypes showed higher collagen content in their M. longissimus dorsi (p < 0.001), M. semimembranosus (p < 0.001), M. triceps brachii (p < 0.001), and M. biceps femoris (p < 0.001) than qq homozygous animals. After the validation of this result in other independent populations, the FSVs of MYH3 genotypes can be a valuable genetic marker for improving collagen content in porcine muscles and can also be applied to increase the amount of collagen for biomedical purposes.
