Effect of Glycyrrhizin on the Rat Liver Glycogen Storage (Glycyrrhizin이 Liver Glycogen Storage에 미치는 영향(影響))

  • Many investigators reported the mineral corticoid hormon-like action of Glycyrrhizin. The other investigators suggested concerning the glucocorticoid hormon-like action. Authors had been observed the glycogen content of the liver to make sure the relationship between glucocorticoid like action of Glycyrrhizin and glucose metabolism in the liver. There was significant increase in the liver glycogen content in rat fed Glycyrrhizin for two weeks, however it was confirmed that Glucuronic acid combined with Glucuronic was unable to make any influence to glycogen content. The relation concerning the effect of Glycyrrhizin on liver glycogen and its mechanism of action might be needed followup study.

  • Introduction: Liver fibrosis is defined unbalance of collagen metabolism, especially a stimulation of collagen synthesis and inhibition of collagen degradation, and antifibrotic effect is delaying or inhibition of new collagen synthesis and deposition in liver tissue. In this study, we investigated the antifibrotic and antioxidative effect of Solanum lycopersicum (SL) in liver fibrosis induced rats. Methods: : Rats were randomly divided in three groups (normal, CCl$_4$ and CCl$_4$-SL group) and were received 0.6 ml mixture of CCl$_4$ and olived oil (1:1 v/v) 3 times/week for 4 weeks except of the normal group. Changes of Activities and Isozymes of Lactate Dehydrogenase in Coreoperca herzi and Pseudogobio esocinus Acclimated to Rapid Increase of Dissolved Oxygen (급격한 용존산소량 증가에 순응한 꺽지(Coreoperca herzi)와 모래무지(Pseudogobioesocinus) 젖산탈수소효소 활성과 동위효소의 변화)

  • The metabolism of lactate dehydrogenase (EC 1.1.1.27, LDH) and $C_4$ isozyme were studied in tissues of Coreoperca herzi and Pseudogobio esocinus acclimated to rapid increase of dissolved oxygen (DO). In C. herzi the LDH activity was changed $35-39\%$ in brain and liver tissues, and within $20\%$ in other tissues. The $B_4$ isozyme was increased and isozyme containing subunit C was decreased in muscle tissue. The $B_4$ isozyme was increased in heart and kidney. In P. esocinus, the LDH activity in liver tissues was largely increased to $150\%$ for 30 minute and $70\%$ in other tissues. The $A_4$ isozyme was increased in muscle and $B_4$ isozyme was increased in other tissues. Especially, the metabolism of liver tissue in P. esocinus was regulated by increasing liver-specific $C_4$ and decreasing $A_4$ isozyme. But the metabolism of eye tissue in C. herzi was regulated by decreasing LDH activity and eye-specific $C_4$ isozyme. The LDH activity and LDH isozyme in P. esocinus were largely increased than C. herzi acclimated to rapid increase of DO. And eye-specific $C_4$ and liver-specific $C_4$ isozymes played role as lactate oxidase. Therefore, the response of species acclimated to rapid increase of DO seems to be variable, perhaps due to prior exposure to environmental conditions.

Influence of Squid Liver Powder on Accumulation of Cadmium in Serum, Kidney and Liver of Mice

  • In this study, the effect of squid liver powder intake on accumulation of cadmium in mice was investigated. Subjects were divided into 4 groups including the control group (CON), squid liver powder group with lipids not removed (SLP100), and squid liver powder groups with lipids removed (LFSLP50 and LFSLP100). Feed intake and food efficiency ratio of squid liver powder groups was significantly higher than the CON. As a result of investigating cadmium content in hair, serum, liver, and kidney during intake of squid liver powder, all groups showed increase in cadmium accumulation through consistent, long-term intake. Especially, cadmium content in liver and kidney of LFSLP100 was significantly higher than the content of SLP100 and CON. As a result of pathological observation on liver and kidney tissues according to squid liver powder diet, LFSLP100 showed most serious pathological symptoms. In case of kidney tissues, degeneration was significantly more severe in LFSLP100 compared to other groups. Such results suggest that cadmium concentration in human body can be increased by ingestion of whole squid including internal organs and that tissues can be damaged by increased cadmium concentration. More specific and systematic studies are deemed necessary.

Metabolism and Disposition of Myristicin in the Isolated Perfused Rat Liver

  • To investigate the hepatic metabolism of myristicin isolated rat livers were perfused under single-pass conditions with perfusate containing myristicin. In outflow perfusate and bile, 5-allyl-1-methoxy-2,3-dihydroxybenzene (M1), M1-sulfate, and M1-glucuronide conjugates were identified as the metabolites of myristicin. HPLC method with UV detection was applied to investigate the hepatic disposition of the compounds. The concentration of myristicin, M1, and M1-conjugates in the outflow perfusate reached steady-state levels within 20 min after commencing the perfusion of $4.5{\mu}M$ myristicin. At steady-state, the mean (${\pm}S.D.$) extraction ratio of myristicin was $0.49({\pm}0.16)$ and clearance was $13.7({\pm}4.5)ml/min$. M1 accounted for $44.0{\pm}5.3%$ of eliminated myristicin and was recovered as unchanged M1, M1-sulfate, and M1-glucuronide in the bile and outflow perfusate.

Effects of Phellinus spp. Extract on Alcohol Metabolic Enzymes in Alcohol-treated Rats

  • Alcoholism is a significant health problem in the world. The liver is the first and primary target organ for alcohol metabolism. Alcohol dehydrogenase and aldehyde dehydrogenase play important roles in the metabolism of alcohol and aldehyde. In this study, I aimed to investigate the eliminatory effects of a Phellinus spp. extract on alcohol metabolism in drunken Sprague-Dawley (SD) rats. Male SD rats were given Phellinus spp. extract at 30 min after 40% (5 g/kg) alcohol ingestion. To assay the effect of Phellinus spp. extract on blood alcohol concentration, blood samples were taken from the tail vein at 1, 3 and 5 h after alcohol ingestion. The concentrations of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase in Phellinus spp. extract treated rat were significantly lower than that of the control with a time-dependent manner. In addition, the alanine aminotransferase and aspartate aminotransferase activities of Phellinus spp. extract-treated groups were altered compared to those of the control group. These results suggest that Phellinus spp. extract intake can have a positive effect on the reduction of alcohol, alcohol dehydrogenase, and aldehyde dehydrogenase concentrations in the blood and may alleviate acute alcohol-induced hepatotoxicity by altering alcohol metabolic enzyme activities. Phellinus spp. extract is thus a good nutraceutical candidate.

Evaluation of the role of Lactobacillus casei on alcohol metabolism and liver functions of rats

  • Alcohol consumption causes numerous consequences on the health of the human body. Heavy drinking on a daily base has caused liver diseases. Furthermore, some products such as acetaldehyde produced from alcohol metabolism are more toxic than alcohol itself. This study was carried out to evaluate the role of Lactobacillus casei on alcohol metabolism, especially, the removal of the toxic effect of alcohol. The maximum alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities from L. casei were observed at 4 hr of culture. L. casei was confirmed to produce the ADH and ALDH by the SDS-PAGE. From in vivo test using SD rats with 22% alcoholic drink, blood alcohol concentration (BAC), glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) of the rats feeding the medium containing L. casei were lower than those of the rats feeding the medium containing an alcoholic drink only This demonstrates that the ADH and ALDH produced by L. casei have virtual functions to detoxicate the alcohol in vivo and the fermentation broth of L. casei can be used as an alcohol detoxification drink.

Effect of Grape Intakes on Lipid Metabolism of Rats during Aging (포도의 섭취가 흰쥐의 노화 과정 중 지방 대사에 미치는 영향)

  • This study was performed to see effects of whole grape, grape pomace or grape juice intakes on lipid metabolism during aging in old Sprague-Dawely male rats. One hundred twenty rats of 13 months old Sprague-Dawely were blocked into eight groups according to body weight and raised for 3, 5 or 7 months with diets containing 2% (w/w) dried powders of three different pars of grape and 0.02% (w/w) CdCl₂. Body weights of Cd groups were lower than Cd free groups. Kidney and spleen weights were incre ased with age, and EEP weights of Cd groups were lower than those of Cd free groups. Total lipid, triglyceride and total cholesterol concentration in plasma increased with age. Whole grape, grape pomace or grape juice intake lowered plasma total lipid triglyceride and total cholesterol concentrations, and especially grape pomace lowered them markedly. Whole grape, grape pomace or grape juice intake decreased liver total lipid, triglyceride and total cholesterol concentrations and increased fecal lipid excretion. Grape diets decreased and Cd administration increased TBARS concentration in LDL fraction. In conclusion, grape diets were effective in decreasing lipid levels of liver and plasma, TBARS in LDL, and in increasing HDL cholesterol. The grape pomace was most effective among three grape parts. It is plausible that grape might be recommended for the treatment or prevention of cardiovascular disease and delaying aging. (Korean J Nutrition 35 (7): 713∼728, 2002)

Metabolic aspects of the toxicology of mixtures of diazinon, toxaphene and/or endrin in mice (마우스에서 diazinon, toxaphene 과 endrin 단독 혹은 그 혼합물 독성의 대사)

  • The effects of mixtures of diazinon(DA;5mg/kg), toxaphene(TOX;40mg/kg) and/or endrin(END; 5mg/kg) on the hepatic mixed-function oxygenase(MFO) system were stuided in ICR mice(18~22g) by oral intubation daily for 7 days. In general, TOX and TOX-containing mixtures were found to induced the metabolism of aminopyrine(22~60%), aniline(42~85%), phenacetin(145~194%) and benzo [a]pyrene(158~210%), and pentobaribtal biotransformation in the 9,000g liver supernatants and to increased the hepatic cytochrome p-450 contents(47~89%). Results of these may be, at least in part, associated with the MFO system. TOX pretreatment increased the aliesterase activity in the serum and liver homogenates and supernatants by 23~145%. The toxicity of TOX and TOX-containing mixtures would be lower than that of diazinon because of TOX-induced increase in the metabolism of diazinon(DA) or diazioxon(DO) and capability of TOX to stimulate the metabolism of diazinon and diazioxon and provide a pool of non-critical enzymes. These results suggest that this information might be helpful in the evaluation of the potential hazard due to occupational and/or environmental exposures to pesticides and their mixtures.

Studies on the Metabolism of Sinigrin in Rat (흰쥐에서 sinigrin 대사에 관한 연구)

  • The detoxifying properties of cruciferous vegetables components have been the subject of several recent investigations. Evidences from many biochemical and pharmacological studies indicated that higher consumption of cruciferous vegetables is associated with lower incidence of harmful actions such as hepatotoxicity and oxidative stress in animal and human populations. Recently, it has been reported that drug metabolizing and detoxifying enzyme activities were increased by cruciferous vegetable extract in which sinigrin is known to be a main active component, accounting for about 2 to 3 percents of total extract. The detoxifying effect of sinigrin has been well reported in several literatures. The metabolism of sinigrin in animal, however, has not been reported yet. That led us to study the metabolism of sinigrin in rat. Sinigrin is nown to be metabolized into three compounds, i.e., allyl isothiocyanate, glucose and potassium phosphate in cruciferous vegetables. Allyl isothiocyanate was formed in rat hepatic mitochondrial fraction in dose and incubation time dependent manner, that was confirmed by HPLC. Glucose formation was came up with results similar to that of allyl isothiocyanate. Three hours after i.p. administration of sinigrin to rat, allyl isothiocyanate appeared in rat liver, and five hours later it was detected in liver and blood. The above results suggested that sinigrin might be metabolized into allyl isothiocyanate, glucose and potassium phosphate in rat.
