  • Over the past few years, hydrogen sulfide (H2S) has been shown to exert several biological functions in mammalian. The endogenous production of H2S is mainly mediated by cystathione β-synthase, cystathione γ-lyase and 3-mercaptopyruvate sulfur transferase. These enzymes are broadly expressed in liver tissue and regulates liver function by working on a variety of molecular targets. As an important regulator of liver function, H2S is critically involved in the pathogenesis of various liver diseases, such as non-alcoholic steatohepatitis and liver cancer. Targeting H2S-generating enzymes may be a therapeutic strategy for controlling liver diseases. This review described the function of H2S in liver disease and summarized recent characterized role of H2S in several cellular process of the liver.

  • To evaluate an effect of pathological liver damage on the cyclohexane (CH) metabolism, rats were pretreated with 50% carbon tetrachloride $(CCl_4)$ dissolved in olive oil (0.1ml/100g body weight) 10 or 17 times intraperitoneally at intervals of every other day. To these liver damaged animals, CH (a single dose of 1.56g/kg body weight, i.p.) was administered at 48hr after the last injection of $CCl_4$. The CH metabolites; cyclohexanol (CH-ol), cyclohexane-l,2-diol (CH-l,2-diol) and cyclohexane-l,4-diol (CH-l,4-diol) and cyclohexanone (CH-one) were detected in the urine of CH treated rats. After CH treatment, the serum levels of CH-ol and CH-one were remarkably increased at 4 hr and then decreased at 8hr in normal group. Whereas in liver damaged rats, these CH metabolites were higher at 8hr than at 4hr. The excretion rate of CH metabolites trom serum into urine was more decreased in liver damaged animals than normal group, with the levels of excretion rate being lower in $CCl_4$ 17 times injected animals than 10 times injected ones. It was interesting that the urinary concentration of CH metabolites was generally more increased in liver damaged animals than normal ones, and the increasing rate was higher in $CCl_4$ 17 times injected rats than 10 times injected ones. Taken all together, it is assumed that reduced urinary excretion rate of CH metabolites in liver damaged rats might be resulted from deteriorated hepatic and renal blood flow, and an increased urinary excretion amount of CH metabolites in liver damaged rats might be caused by reduced expiration amount of the metabolites due to lung damage.

  • In order to study the effect of source and level of the commonly used dietary fats on growth and metabolism of rats fed on low protein diet (rice diet) the weaning white rats were fed on various different experimental diets (see tables 1 and 2) during 11 weeks. The observations were made as follows : 1. Growth: (see table 3 and figures 1-9) In all dietary fats, among the 3 levels, 5% fat level is the best. Especially, the perilla oil group was remarkably good. 10% and 20% fat levels impaired the growth, consequently the growth rates of both 10% and 20% fat level groups were worse than those of Basal group (no fat added). However, 10% and 20% fat levels did not impaired the growth of VII group (10% soy flour added) In 5% fat level, the growth was good in sequence of perilla oil, tallow, sesame oil, soy oil and lard. 2. Feed consumption: (see table 3) In 20% fat level, the feed consumption was lowered. Generally, the feed consumption rate was proportional to the growth rate. In feed efficiency, 5% fat level was the best. 3. Liver weight: (see table 4) In liver weight per 100 G body weight, 20% fat level was the largest. This may be due to the poor body growth and liver fat accumulation. 4. Liver nitrogen: (see table 4) Generally, lower fat level groups showed liver nitrogen. Liver nitrogen is low in the groups of 20% fat level. 5. Liver fat: (see table 4) Generally, higher fat level groups showed higher liver fat. 6. Serum cholesterol: (see table 5) Generally, higher fat level groups showed higher serum cholesterol. Lard, sesame oil, and tallow groups showed higher level and soy oil and perilla oil groups showed lower level. Especially, perilla oil group showed remarkably lower level and VII group (10% soy flour added) showed lower level than VI group (same fat but no soy flour added).

  • ${\beta}$-Lapachone has drawn increasing attention as an anti-inflammatory and anti-cancer drug. However, its oral bioavailability has not been yet assessed, which might be useful to develop efficient dosage forms possibly required for non-clinical and clinical studies and future market. The aim of the present study was thus to investigate pharmacokinetic properties of ${\beta}$-lapachone as well as its first-pass metabolism in the liver, and small and large intestines after oral administration to measure the absolute bioavailability in rats. A sensitive HPLC method was developed to evaluate levels of ${\beta}$-lapachone in plasma and organ homogenates. The drug degradation profiles were examined in plasma to assess the stability of the drug and in liver and intestinal homogenates to evaluate first-pass metabolism. Pharmacokinetic profiles were obtained after oral and intravenous administration of ${\beta}$-lapachone at doses of 40 mg/kg and 1.5 mg/kg, respectively. The measured oral bioavailability of ${\beta}$-lapachone was 15.5%. The considerable degradation of ${\beta}$-lapachone was seen in the organ homogenates but the drug was quite stable in plasma. In conclusion, we suggest that the fairly low oral bioavailability of ${\beta}$-lapachone may be resulted from the first-pass metabolic degradation of ${\beta}$-lapachone in the liver, small and large intestinal tracts and its low aqueous solubility.

  • This study was performed to investigate effect of dietary fibers in rice and barley on glucose, lipid and cadmium (Cd) metabolism in the rat. Fifty-six male Spague-Dawley rats weighing 244.6$\pm$2.7g were blocked into eight groups according to body weight and raised for four weeks with diets containing 0 or 0.04%(w/w) CdCl2 and four different carbohydrate sources, starch, rice flour, barley flour and mixture of rice and barley flour(7 : 3, w/w). Total dietary fibers and $\beta$-glucan contents of barley were about three times higher than those of rice (10.75% vs. 3.94%, 3.11% vs. 1.06%, respectively). Food intake, weight gain, food efficiency ratio, liver and kidney weights were lower in Cd exposed groups, and barley group among Cd exposed animals showed highest weight gain, food efficiency ratio and organ weights. Fasting serum glucose levels were not significantly different among groups, Serum cholesterol level was lowest in Cd exposed barley group. Serum HDL-cholesterol level was higher in none-Cd exposed starch and barley groups, and HDL-cholesterol : total cholesterol ratios were higher in none-Cd rice and mixed flour groups than other groups. Liver total lipid and triglyceride levels were lowest in barley groups regardless of Cd administration. Fecal total lipid, cholesterol and triglyceride excretions were high in barley and mixed flour groups. Liver Cd concentrations were low in Cd exposed barley and mixed flour groups. In Cd exposed barley group, fecal weight and Cd excretion were highest and Cd retention ratio was lowest among groups. Small intestine metallothionein(MT) concentration was highest in Cd exposed rice group, and kidney MT concentration was highest in Cd exposed barley group. In conclusion, cereals showed different effects on lipid and Cd metabolism that might be mediated by dietary fibers in cereals. Especially $\beta$-glucan-rich barley group showed greatest lipid and Cd lowering effects by increasing fecal lipids and Cd excretions.

  • In order to minimize the occurrence of fatty liver, this study investigated how adding a high level of dietary choline or methionine affected the lipid metabolism of Tsaiya ducks. Feeding trials were conducted with sixty Tsaiya ducks during their growing period, when they were 8-12 weeks old, and during their laying period, 10-14 weeks after the onset of laying when they were 26-30 weeks old. The ducks were randomly assigned to one of three groups: a control group (basal diet), a group in which methionine was added to the basal diet, and a group in which choline was added to the basal diet. The levels of methionine and choline added were twice as high as the levels recommended by the NRC for layer-type chickens. Experimental results indicated that adding choline to the basal diet of growing ducks significantly (p<0.05) increased their body weight, while adding methionine significantly (p<0.05)reduced their body weight. Adding either choline or methionine reduced the ducks' liver fat content in both the growing and the laying periods (p<0.05). Ducks receiving added methionine or choline in their diets displayed enhanced egg production (p<0.05). Adding choline increased serum triacylglycerol (TG) in the laying period (p<0.05). Adding either choline or methionine did not significantly (p>0.05) affect the t-globulin level. Adding methionine increased the activity of liver malic dehydrogenase in both the growing and the laying periods, and increased fatty acid synthetase in the laying period (p<0.05). While adding choline markedly (p>0.05) increased VLDL and apo B in both the growing and the laying periods, it decreased HDL and apo A in the laying period (p<0.05). This study found that adding a high level of choline to the basal diets of Tsaiya ducks in both the growing and the laying periods had beneficial effects. Furthermore, adding methionine affected the lipid metabolism of Tsaiya ducks to a lesser extent than adding choline.

  • Objectives : Alcohol hangover is a common phenomenon which basically occurs after heavy drinking. Moreover, heavy alcohol consumption leads to acute and chronic diseases. We investigated the effect of herbal mixture (SJ) on alcohol metabolism in serum or/and liver. Methods : 5W-old Sprague Dawley rats were used for the study. To overnight fasted rats, 0.9% saline or SJ extract was administrated per os before alcohol treatment. Then, 40% alcohol was orally administrated to all rats in 30 mins. Ethanol, acetaldehyde concentrations, alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH) activities were measured by assay kits. Aspartate aminotransferase (AST) and alanine transaminase (ALT) were measured by analyzer. Results : Ethanol and acetaldehyde concentrations lowered in SJ groups compared with CON group. Especially, acetaldehyde concentration significantly decreased in SJ groups compared with CON group. AST and ALT levels tended to increase in SJ groups compared with CON group but there was no significant difference between CON group and SJ groups. ADH activity in serum was higher in SJ groups than CON group but no significant difference in liver. ALDH activity in both serum and liver showed significantly increased in SJ groups compared with CON group. Conclusions : Treatment of SJ extract showed not only reducing concentration of alcohol and acetaldehyde but also increasing activities of ADH and ALDH. These results suggest SJ may influence in alcohol metabolism via control of metabolic enzyme activities and metabolite. Therefore, SJ, herbal mixture, might have a function of preventing hangover after drinking alcohol.
