Experimental infection of atypical Aeromonas salmonicida in Nile tilapia Oreochromis niloticus and its treatment with carvacrol and cymene mixture

  • The pathogenecity of atypical Aeromonas salmonicida was studied in healthy Oreochromis niloticus. Inoculum at concentration of $1.5{\times}10^8CFU/ml$ and $3{\times}10^8CFU/ml$ was injected into healthy fish through intramuscular and intraperitoneal injections. Experimentally infected Oreochromis niloticus showed ulceration at the dorsal musculature and trunk region in addition to black coloration, congested gills, exophthalmia, and ocular hemorrhage. Congested liver and kidney were recorded in post-mortem examination. Mortality of the experimentally infected Oreochromis niloticus reached 100% after intramuscular injection at concentration of $3{\times}10^8CFU/ml$. Histopathological investigation of infected organs was also performed. There was a focal area of bundles of skeletal musculature showing hyalinization. In addition, hyperplasia, congestion, and fusion were noticed in the gill lamellae. There was also congestion in the blood vessels in the ocular chamber. Severe congestion was also noticed in the central vein of liver associated with focal aggregation of the melanin pigmented cells in the parenchyma. Degenerative changes were noticed in the epithelial cells lining of kidney tubules. Plant extracts carvacrol and its biological precursor cymene were found to be effective in treating experimentally infected Oreochromis niloticus at concentration of 100 or 200 ppm.

The Cytotoxicity of Rat Hepatocytes on Viscum album Lectin (겨우살이 lectin의 흰쥐 간세포 독성에 미치는 영향)

  • In the present study, the author investigated to the cytotoxocity in cultured rat hepatocytes of Viscum album lectin. The cytotoxcity effect in Viscum album lectin on the activity of LDH was also investigated. Viscum album lectin significantly increased LDH leakage into medium of hepatocytes treated or untreated with $CCl_4$ (p<0.001). However, Viscum album lectin significantly increased LDH leakage from $CCl_4$-induced hepatocyte (p<0.001). There was a significant increase in LDH levels relative to the control group. Histological observation basically supported the result obtained from LDH assay. The livers of rats challenged with $CCl_4$ produced a marked increased cytoplasmic vacuoles and inflammatory cells in number, while the number of necrotic cells and swollen hepatocytes did not change significnatly. Rats administered DMSO alone did not alter the normal hepatic architecture. Histological observation of liver section in rat treated 72 hrs with either Viscum album lectin $CCl_4$-induced liver damage showed number of cytoplasmic vaculoe and necrotic cell. The number of inflammatory cell increased markedly. This results suggest to the conclusion that Viscum album lectin has a effect of hepatotoxicity activator.

  • The mutagenicity of 22 food and cosmetic dyes had been evaluated. Two different short-term mutagenicity tests were used: (1) Salmonella typhimurium preincubation assay (Ames test) (2) Chromosome aberration test with cultured Chinese hamster lung (CHL) fibroblast cells. Orange No. 203 was mutagenic in Salmonella typhimurium with and without rat liver microsomal activation, and Red No. 204 was mutagenic in Salmonelhl typhimurium with rat liver microsomal activation. Red No. 104-1 and Red No. 215 showed slight increase of chromosomal aberration in CHL cells.

  • Kamiondamtang(KODT) has been used in oriental medicine as stress by many medical practitioner for the study of KODT, we had fed mice divided to three groups(basal diet, hyperlipidemic diet, hyperlipidemic diet + KODT), and observed the change of weight, total cholesterol, triglyceride, SGOT, SGPT, HDL-cholesterol, LDL-cholesterol on the serum per every 7days for 6weeks. To help comparison with the results above, we had tested endothelial cells and liver. The results of this Study were obtained as fallows ; 1. Total cholesterol, triglyceride and LDL-cholesterol were decreased significantly by KODT. 2. HDL-cholesterol was increasd significantly by KODT. 3. SGOT was intended to decrease by KODT, 4. %W was decreased significantly by KODT. 5. In case of supplying KODT, WBC attatched on endothelial cells and vacuoles in muscular layer were not observed. 6. In case of supplying KODT, fatty degeneration was not observed in liver portal area.

Pancytopenia Improves with Compromised Liver Function in A Hepatocellular Carcinoma Patient Treated by Modified Sipjeondaebo-tang (십전대보탕가감방을 투여한 간세포암종 환자에서 범혈구감소증 호전 및 간수치 상승 증례)

  • Objectives : To observe the therapeutic effects of an oriental herbal prescription in a hepatocellular carcinoma patient with splenomegaly and pancytopenia. Methods : Modified Sipjeondaebo-tang was prescribed three times a day to a hepatocellular carcinoma patient with splenomegaly and pancytopenia. Laboratory tests were carried out regularly to observe the therapeutic effects of the oriental herbal prescription for pancytopenia. Results : When treated with modified Sipjeondaebo-tang including Cinnamomi Cortex, the levels of white blood cells, red blood cells, hemoglobin, and hematocrit increased. However, aspartate transaminase, alanine transaminase, alkaline phosphatase, gamma-glutamic transpeptidase, total bilirubin, direct bilirubin levels also increased. Conclusions : Administering modified Sipjeondaebo-tang with Cinnamomi Cortex showed effect in improving pancytopenia but an increase in liver enzyme levels was also observed.

  • An inverse finite element (FE) model parameter estimation algorithm can be used to characterize mechanical properties of biological tissues. Using this algorithm, we can consider the influence of material nonlinearity, contact mechanics, complex boundary conditions, and geometrical constraints in the modeling. In this study, biomechanical experiments on macro and micro samples are conducted and characterized with the developed algorithm. Macro scale experiments were performed to measure the force response of porcine livers against mechanical loadings using one-dimensional indentation device. The force response of the human liver cancer cells was also measured by the atomic force microscope (AFM). The mechanical behavior of porcine livers (macro) and human liver cancer cells (micro) were characterized with the algorithm via hyperelastic and linear viscoelastic models. The developed models are suitable for computing accurate reaction force on tools and deformation of biomechanical tissues.

Effects of Zinc Deficiency on Immune Response in Mouse (식이 아연이 Mouse의 면역 반응에 미치는 영향에 관한 연구)

  • The purpose of this study was to investigate the effects of dietary zinc on immune response in mice. Weanling male mice was placed individually in stainless steel cages and fed a zinc dificient diet and control diet. All mice were given deionized water ad libitum. The introduction of extraneous zinc was minimized in all cage by washing feed jars and water bottles sequentially with 4mM EDTA and conc-nitiric acid followed by deionized water. After 4 and 5 weeks of the diets, mice were immunized with lx 106 Naegleria fowleri intraperitoneally. Mice were weighed once a week. The results from this study are summarized as followed ; 1) Mice fed the zinc dificient diet showed growth retardation. After 3 weeks of diets, mean body weight of zinc deficient mice was 21.4g and that of control was 25.0g. This difference is singnificant statistically (p<0.01). The more time passed, the more remarkable difference was found. 2) The weigth of organs were measured on liver, kidney, spleen, thymus, heart, lung, brain. Difference in weight were observed only in liver and spleen. 3) Proliferative response of spleen cells of zinc deficient mice to con A was lower than that of control mice after one week on immunization(p<0.005). 4) Stimulation index was lower in zinc deficient mice to phytohemagglutinin after two weeks on immunization (p<0.05). 5) Blastogenesis of speen cells of zinc deficient mice to Naegleria fowleric lysate was lower after 10 days on immunization (p<0.05). 6) Immunoglobulin G antribody titers of zinc deficient mice sera by ELISA was lowered to control mice after 5 weeks on immunization (p<0.005).

Cholangiocarcinoma and Cloacal Prolapse in a Yellow-crowned Amazon Parrot (Amazona ocrocephala) (아마존앵무에서 병발한 담관암종과 총배설강 탈출증)

  • This report deals with a 3-year-old male Yellow-crowned Amazon (Amazona orcrocephala) that died after twenty five days history of cloacal prolapse. On necropsy, multiple to occasionally coalescing well-demarcated tan nodules were scattered throughout the entire liver lobes. The nodules were firm and slightly elevated from the surface. The neoplastic nodules were also recognized on cut sections of the liver. Histologically, the nepatic nodules consisted of cuboidal to low columnar cells forming tubular to acinar pattern with marked desmoplasia. The neoplastic cells had small to moderate amounts of eosinophilic cytoplasm and round nucleus with one to two prominent nucleoli. Based on these results, hepatic nodules was diagnosed as primary cholangiocellular carcinoma.

Multicentric T cell lymphoma in a Maltese dog

  • A case of multicentric high grade T cell lymphoma is reported in a 5-year-old male Maltese dog with generalized lymphadenopathy. The dog showed depression, anorexia, blindness, jaundice, arrhythmia, and hematuria for 8 months. Complete blood count and chemistry profile revealed anemia and increased alanine transferase, alkaline phosphatase, total bilirubin, and total cholesterol. Grossly, most of lymph nodes, spleen, and liver were enlarged and neoplastic masses were occupied in these tissues. Histologically, massive accumulation of small noncleaved neoplastic lymphocytes with high mitotic figures was observed in all lymph nodes and spleen. Infiltration of neoplastic lymphocytes was also noted in the lung, liver, kidney, eye, skin, muscle, and bone marrow of femur. Immunohistochemistry revealed that tumor cells were CD3-positive and but CD79a-negative, consistent with T-cell lineage. In our best knowledge, this is the first report of multicentric lymphoma clarified the origin of tumor cells in Korea.

Pathological and immunohistochemical studies in Syrian hamsters experimentally infected with encephalomyocarditis virus isolated in Korea (국내 분리 encephalomyocarditis virus의 실험적 감염 Syrian hamster에 대한 병리학적 및 면역조직화학적 연구)

  • To elucidate pathogenicity to hamster of encephalomyocarditis virus $K_3$ strain that was isolated in Korea from the swine with reproductive failures, adult male syrian hamsters were experimentally infected intraperitoneally with the virus at $10^{7.0}\;TCID_{50}/0.1ml$ and pathological and immunohistochemical studies were performed. The results obtained through the experiment were as follows. 1. Clinical signs such as depression, unkempt hair and bilateral parlysis of hind limbs were observed. 2. At necropsy, mild congestion was observed in the cerebrum, liver, kidney and lung, and atrophy was evident in testis. 3. In microscopic observation, degeneration and necrosis of the nervous cells and perivascular mononuclear cell infiltration were manifested in central nerve system, and various degrees of degeneration and necrosis of parenchymal cells were detected in pancreas, lacrimal gland, liver, kidney and testis. 4. In immunohistochemical observation, strong positive reactions were observed in degenerated parenchymal cell of testis, and weak positive reactions, in hepatocytes.
