고지방급여 흰쥐의 혈청과 간의 지질 농도 및 조직 과산화지질 농도에 미치는 홍삼분말의 영향 (Effect of Korean Red Ginseng Powder on the Lipid Concentrations and Tissue Lipid Peroxidation in the Rats Fed High Fat Diet)

  • 생리활성 물질을 함유하고 있는 홍삼분말의 섭취수준에 따른 고지방식 급여 흰쥐의 지질 농도 및 과산화지질 농도에 미치는 영향을 검토하기 위하여 200 mg/rat및 400 mg/rat 수준으로 홍삼분말을 첨가하여 3주간 자유 섭취시켰다. 혈청 총 콜레스테롤 농도는 대조군에 비해 KRG I군과 KRG II군에서 유의적으로 감소(p<0.05)경향을 나타내었다. 혈청 HDL-cholesterol 농도는 대조군에 비해 KRG I군 및 KRG II군에서 각각 69.75% 및 39.l5% 증가하였고, 동시에 동맥경화 지수는 각각 74.76% 및 37.3n% 감소하였다 혈청 중성지질 농도는 대조군에 비해 KRG II군에서만 유의적으로 감소(p<0.05)하였다. 혈청 지질의 과산화물 생성지표인 TBARS는 대조군에 비해 KRG I군과 KGB II군에서 유의적으로 감소(p<0.05)하였다. 과산화지질 촉매물질로 작용하는 혈중 비헴철 함량을 측정한 결과, 대조군에 비해 홍삼분말 첨가군에서 유의성 있게 감소하여 혈청 과산화지질 농도와 상관관계를 나타내었다. 간장에서의 MDA 함량은 대조군에 비해 KRG I군 및 KRG II군에서 유의적으로 감소(p<0.05)하였다. 간장으로부터 분획한 microsome의 세포막 과산화지질 농도는 대조구에 비해KRG I군 및 KRG II군에서 유의적으로 감소(p<0.05)하고, nitochondria 분획에서는 효과가 없었다. 홍삼분말을 0, 2, 4, 및 6 mg/mL농도로 간장 세포 분획의 반응액에 첨가하여 MDA 함량을 측정하였을 때, homogenate 및 microsome 분획에서는 6 mg/mL 농도에서만 유의적으로 감소(p<0.05)하고, mitochondria 획분에서는 2, 4 및 6 mg/mL 첨가구에서 모두 유의적으로 감소(p<0.05)하였다. 이상의 실험에서 고지방 식이를 급여한 흰쥐에서 홍삼분말 첨가는 혈중의 콜레스테롤 농도와 과산화지질 농도를 낮추는 효과를 나타내었다.

천마 분말, 에탄올 및 열수추출물이 고지방식이를 급여한 흰쥐의 혈청지질과 체지방에 미치는 영향 (Effect of Whole Powder and Extracts of Gastrodiae Rhizoma on Serum Lipids and Body Fat in Rats Fed High-Fat Diet)

  • 천마의 분말과 50% 에탄올 및 열수추출물이 돼지기름(lard 10%)을 첨가한 고지방식이를 급여한 흰쥐의 체중증가율, 혈청지질과 테스토스테론농도, 혈당치 및 정소상체지방패드(EFP) 등에 미치는 영향을 조사하였다. 시험기간 중 체중증가율은 식이군간 통계적인 차이는 없었지만 대조군(A, B)과 열수추출물군(G)이 높았고, 그 외 천마식이군은 비슷하였다. 장기무게는 식이군간에 통계적인 차이가 있었다(p<0.05). 혈청지질중 TC와 TG농도는 두 대조군이 가장 높았고, 열수추출물군(H)이 유의하게 낮았다(p<0.05). HDL농도는 천마식이군중 50% 에탄올추출물군(E)이 가장 높았고 A군이 가장 낮았다(p<0.05). LDL+VLDL농도는 A군이 가장 높았고 천마식이군에서는 50% 에탄올추출물군(E)와 열수추출물군(G, H)이 유의하게 낮았다(p<0.05). 테스토스테론농도는 식이군간에 통계적인 차이는 나타나지 않았지만 천마군중에서는 E군, D군 및 H군이 높았고 대조군을 포함한 다른 천마군은 비슷하였다. 혈당치는 B군과 G군이 식이군 중 통계적으로 가장 높았고 H군은 통계적으로 가장 낮았다(p<0.05). 체지방의 축적정도를 나타내는 EFP무게는 두 대조군이 천마식이군 보다 유의하게 높았고 분말군(C, D)과 열수추출물군(H)이 유의하게 낮았다(p<0.05). 이상의 결과로 볼 때 천마 분말 보다 50% 에탄올추출물 2 brix군(E)과 열수추출물 10 brix군(H)이 고지방식이를 급여함에도 혈청지질과 체지방의 축적을 감소시키는 것으로 나타났고 혈청 테스토스테론농도도 E군이 통계적인 유의성은 없었지만 식이군 중 가장 높았다.

Lipid Metabolism and Peroxidation in Broiler Chicks under Chronic Heat Stress

  • The effects of taurine supplementation on growth performance, serum and liver concentrations of lipid, fatty acid composition and lipid peroxidation in the livers of broilers under chronic heat exposure conditions were investigated. The chicks with a similar body weight were equally assigned to one of three controlled-environment chambers. The brolier chicks, which were kept at $34^{\circ}C$ were fed either with a control diet or the control diet supplemented with 0.8% taurine, whereas broiler chicks kept at $22^{\circ}C$ were fed a control diet. Both of the BW and BW gains of broilers maintained at a temperature of $34^{\circ}C$ were significantly lower than those of the control group, which was maintained at a temperature of $22^{\circ}C$ (p<0.05). However, taurine addition in the diet of birds submitted to heat stress siginficantly improved BW gain (p<0.05). The feed intake of chicks declined with increases in temperature. The relative liver and gall bladder weights of chicks fed the control diet and maintained at $34^{\circ}C$ were significantly lower than those measured in the control birds (p<0.05). However, dietary taurine was found to compensate for these reductions in liver and gall bladder weights. Relative weights of abdominal fat did not differ significantly among the three groups. Serum triglyceride concentrations were significantly lower in the chicks fed the control diet and maintained at $34^{\circ}C$ compare to those measured in the chicks fed the control diet at $22^{\circ}C$ (p<0.05). Heat stress resulted in a significant reduction in total lipid and triglyceride levels, but also increased the levels of total cholesterol in the liver (p<0.05). However, dietary taurine supplementation under the heat stress condition resulted in the recovery, to control levels, of serum triglyceride concentrations, as well as the amounts of total lipids, triglycerides, and cholesterol in the liver. The livers of chicks fed on taurine diets at $34^{\circ}C$ showed significantly higher proportions of C14:0, C16:1, C18:1, C18:2, and 20:3, and lower C18:0 and C20:4 proportions than those of chicks fed on control diets at the same temperature (p<0.05). The total levels of saturated fatty acids decreased, but monounsaturated fatty acids and unsaturated fatty acid levels increased in chicks fed the taurine diet, as compared to chicks fed the control diet at $34^{\circ}C$ (p<0.05). Peroxidizability indices were significantly lower in the heat-exposed chicks fed the taurine diet than in the non-taurine heat-exposed groups (p<0.05). In conclusion, dietary taurine results in an increase in the growth performances of chicks under heat stress conditions via improvements in lipid absorption and metabolism, as well as an induced reduction in lipid peroxidation.

흰쥐의 간장 및 혈청 지질농도와 임상생화학적 지표에 미치는 식이섬유 및 식이지방 급원을 달리한 식이 투여의 영향 (Effects of Different Dietary Fats and Fibers on the Lipid Concentrations of Liver and Serum and Biochemical Index in Rats)

  • 본 실험에서는 Sprague-Dawley계 수컷 흰쥐의 간장 및 혈청 지질 농도와 임상생화학적 지표에 미치는 식이지방원 및 식이섬유원을 달리한 식이를 급여한 영향을 검토하였다. 실험 식이군은 5% corn oil +5% cellulose 함유 CO군, 10% corn oil +10% 차전자피 함유 CO +PH군 및 10% lard+10% 차전자피 함유 LA+PH군인 3군으로 나누었으며 실험식이와 물은 6주간 자유급여 시켰다. 체중 증가량 및 식이효율은 CO군에 비해 LA+PH군에서 유의적으로 증가하였고, CO+PH군에서는 유의적인 증가경향을 보였다. 식이 섭취량 및 장기 중량은 실험군간에 차이가 없었다. 혈청총 콜레스테롤 농도는 CO군에 비해 CO+PH군에서 유의적으로 감소하였으나, LA + PH군과는 차이가 없었다. 혈청 중성지방 및 인지질 농도는 CO군에 비해 CO +PH군 및 LA+ PH군에서 유의적으로 감소하였다. 간장 콜레스테롤 농도는 LA +PH군에서 유의적으로 감소하였다. 혈청 임상생화학적 지표 중 glutamic pyruvic transaminase 활성 은 LA+PH군에서 유의적으로 높았으며, lactate dehydrogenase 활성 및 blood urea nitrogen은 오히려 LA + PH군에서 유의적으로 감소하였다. 다른 혈청 임상생화학적 지표중 총 단백질, 알부민, 포도당, glutamic oxaloacetic transaminase 및 lactate dehydrogenase활성은 각 실험군 간에 차이가 없었다. 이상의 결과로부터 식이 차전자피에 의한 혈중 지질 농도의 저하효과는 식이지방원 lard 첨가군에 비해 corn oil 첨가군에서 더욱 현저하였다.

왕느릅나무 추출물에 의한 설치류 혈중지질 개선 효과에 대한 연구 (Study of in Vivo Serum Lipid Regulation with Ulmus macrocarpa Hance Extract in Rats)

  • 이전 연구에서 Ulmus macrocarpa Hance의 열수추출물(UME)이 고지질혈증 상태에서 지질대사를 향상시켰으며, AMPK 경로를 통한 지질합성 억제에 관해 제안된 바 있다. 이 연구의 목표는 UME가 혈청 지질 수치를 어떻게 조절할 수 있었는지에 대한 가능성을 확장하기 위해 간과 말초 조직 사이의 관련 지질 대사를 밝히기 위한 것이다. 고지질혈증 유발 설치류에서 6주 동안 UME를 경구 투여한 후 혈청 중 트리글리세라이드(TG)와 총콜레스테롤(TC) 수치가 감소했고, 혈청 HDL콜레스테롤 수치는 증가했다. 간 분석에서도 UME 처리시 대조군에 비해 TG와 TC 함량은 감소하였다. 단, UME에 의한 AMPK 경로 조절은 간의 지방생성 억제를 통해 고지질혈증 상태를 부분적으로 개선하였다. 실제로, UME에 의한 간지질 함량 감소는 훨씬 적은 반면, 아포리포프로틴 B (apoB)와 아포리포프로틴 A (apoA)의 혈청 농도는 UME 투여에 의해 정상 그룹 정도의 수치로 돌아왔다. 이것은 간지방생성의 억제만으로 일어날 수 있는 현상은 아니라고 판단하였다. 따라서, UME 투여의 효과는 지질산화의 개시 자극과 간 지질합성의 억제 모두를 통해 혈청 지질 수치를 조절함으로써 혈액 순환을 향상시킬 수 있을 것이다.

Ameliorating Effect of Mycoleptodonoides aitchisonii on High-fat Diet-induced Obese Mice

  • The present study investigated the anti-obesity effects of Mycoleptodonoides aitchisonii (MA) in mice fed a high-fat (HF) diet. Two groups were fed either a normal control diet or an HF (45% kcal fat) diet for 12 weeks and three groups were fed an HF diet supplemented with powdered MA (MAP, 1%, 3%, and 5%) for 12 weeks. The anti-obesity effects of MAP supplementation on body weight, fat mass development, and lipid-related markers were assessed. Consumption of an HF diet resulted in increased body weight, serum lipids, relative adipose tissues weight, and liver fat accumulation. However, administration of MAP significantly decreased body weight gain, food intake, food efficiency ratio, hepatic cholesterol level, and adipose tissue weight in a dose-dependent manner. In addition, treatment with MAP significantly reduced the occurrence of fatty liver deposits and steatosis, and inhibited an HF diet-induced increase in adipocyte size. These results suggest that dietary supplementation with MAP exerts anti-obesity effects and indicate that MAP could be used as a functional food to control obesity.

The Effects of Carnitine and/or Gamma-Aminobutyric Acid (GABA) Supplementation on the Recovery of Chronic Ethanol Administered Rats

  • To investigate the effects of the supplementation of carnitine and/or ${\gamma}$ -aminobutric acid (GABA), Sprague-Dawley male rats were orally treated with either an AIN-76 diet (control), a control diet plus ethanol (CE, 4 g ethanol/kg bw), CE plus L-carnitine (CEC, 0.5 g/kg bw), CE plus GABA (CEG, 0.5 g/kg bw), or CE plus L-carnitine plus GABA (CECG, 0.25 g/kg bw each) for 6 weeks. Serum triglyceride levels were increased in the CE group and were decreased significantly in the CEC, CEG and CECG groups. HDL-cholesterol was increased and LDL-cholesterol was decreased in the CEG and CECG groups compared with the CE group. Serum GOT and GPT levels increased by the chronic ethanol administration were decreased in the CEC group. In addition, we have evaluated the mRNA levels of carnitine palmitoyltransferase-I in those groups. Supplementation of carnitine/GABA had some recovery effects on the liver CPT-I mRNA levels which decreased by chronic ethanol administration. These results may suggest that supplementations of either L-carnitine or GABA aye effective on the recovery of chronic ethanol-related symptoms, but no combined effects were shown.

Effects of Functional Extracts Made from Fermented Plants on Serum Glucose and Lipids Level in Streptozotocin-Induced Diabetic Rats

  • This study was performed to explore the effects of the fermented functional extracts (FE) on blood glucose and lipid levels in diabetes. FE were created by mixing 9 kinds of plants with sea water and then allowing the mixture to ferment for 1 year. FE were supplemented in the feed of streptozotocin (STZ)-induced diabetic rats at 1%, 3% and 5%. The 1% feeding group showed the lowest weight loss of the three experimental groups. The blood glucose and glycosylated hemoglobin level were significantly decreased in the FE fed rats compared to the diabetic control (DMC) group. The lipid levels in serum were decreased in 1% and 3% FE fed rats in comparison to the DMC group, and there was no significant difference in triglyceride levels due to the FE concentration. The HDL-C level was significantly increased in rats with FE supplemented diets, compared to the DMC group. The levels of lipid peroxides in liver tissue were significantly decreased in FE fed diabetic rats, and the hepatic glycogen content was increased in rats receiving supplements. As a result of these studies, we believe 1% FE may be the optimum level for controlling blood glucose and alleviating hyperlipidemia in STZ-induced diabetic rats.

키토산의 효소분해물질이 흰쥐의 당 및 지방대사에 미치는 영향 (Effects of Chitosan Treated with Enzymatic Methods on Glucose and Lipid Metabolism in Rats)

  • This study was conducted to investigate the effets of chitosan and beef tallow at different level on glucose and lipid metabolism in rats. Dietary fot level was 20% and 40%, and chitosan was given at levels of 0%, 3%, and 5%(wt/wt) of diet. Chitosan supplement tended to decrease the serum total lipids, total cholesterol, and triglycerides. HDL cholesterol and HDL cholesterol : total cholesterol ratio tended to increase with 5% chitosan supplementation. LDL cholesterol and VLDL triglyceride tended to decrease with chitosan supplementation. Lipid concentration of liver and epididymal fat pad(EEP) tended to decrease with medium dietary fat and chitosan treatment. fecal excretion of total lipid and triglyceride exhibited a tendency to increase with high fat levels and chitosan. Length of small intestine and gastrointestinal transit time were not affected by dietary fit levels or chitosan supplements. Therefore, it could be suggested that chitosan supplement had beneficial effects on lipid metabolism. (Korean J Nutrition 31(7) : 1112-1120, 1998)

Conjugated Linoleic Acid Changes fatty Acid Composition by Decreasing Monounsaturated fatty Acids in Rabbits and Hep G2 Cells

  • Conjugated dienoic derivatives of linoleic acid(CLA) are a mixture of positional and geometric isomers of linoleic acid(LA). We previously found that CLA changes the fatty acid profile in chicken eggs and serum by decreasing monounsaturated fatty acids. Studies were conducted to explore the effects of CLA on fatty acid composition. Rabbits were fed a semisynthetic diet with or without CLA(0.5g CLA/rabbit/day) for 22 weeks. Compared to the control, rabbits fed CLA had significantly lower monounsaturated fatty acid levels(palmitoleic acid Cl6 : 1 by 50% and oleic acid Cl8 : 1, by 20%) in plasma lipids. We found similar differences in fatty acid composition in the liver and the aorta. The inhibitory effect of CLA on $\Delta$9 desaturation was confirmed in a human hepatoma cell line, Hep G2. CLA significantly decreased $\Delta$9 desaturation in 4-5 hours as shown by an increase in the ratio of Cl6 : 0 to C 16 1, This is apparently due to a decrease in $\Delta$9 desaturase(stearoyl-CoA desaturase, SCD) activity ; it was decreased more than 50%. These results, along with our previous findings, indicate that CLA is an inhibitor of $\Delta$9 desaturase in the liver.
