Using high-fat-fed C57BL/6 mice, therapeutic effects of oral administration of aqueous extracts of Platycodon grandiflorum and Acanthopanax senticosus on obesity were evaluated by comparing body and liver weights, feed intake, adipose tissue mass, blood lipid profile, and triglyceride and total cholesterol levels in liver and feces. Obesity was successfully induced in high-fat diet group. P. grandiflorum and A. senticosus were effective in reducing food intake, serum lipid levels, adipose tissue accumulation, and weight. P. grandiflorum extract reduced triglyceride and cholesterol levels in liver by excluding them in feces. A. senticosus extract increased triglyceride level in liver but decreased in feces, and decreased total cholesterol in liver and feces, indicating active ingredient of A. senticosus exert antiobesity effect through mechanism different from that of P. grandiflorum extract. These results suggest aqueous extracts of P. grandiflorum and A. senticosus have synergistic effect for prevention of hyperglyceridemia and obesity.
$\beta$-Sitosterol, campesterol and stigmasterol have been known to the phytosterols the most frequently found in plants. Metabolism of phytosterols was investigated using rat feces and liver microsomes. Feces were collected after phytosterols (a well characterized mixture of $\beta$-sitosterol 40%, campesterol 30% and dihydrobrasicasterol) were administered orally (0.5 ${g/kg$) to rats. Metabolites of phytosterols were identified using GC/MS. Three peaks were eluted at 12.47, 12.65, 12.87 min and had characteristic molecular ions m/z 428, 430, 432, respectively. Three fecal metabolites were identified as androstadienedione, androstenedione, and androstanedione. No metabolites could be detected in the rat liver microsomal reaction mixture. The results suggest that the metabolites of phytosterols in rat feces are formed by oxidation at 3- position, saturation at 5- and 6- position, and 17- side chain cleavage in the rat large intestine.
We investigated the effect of a green seaweed Capsosiphon fulvescens extract (CFE) on the serum, liver tissue, and fecal cholesterol levels in rats. Male Sprague-Dawley rats (four weeks old) were given on of three diets for four weeks: basal, high cholesterol, and CFE, The total serum and liver tissue cholesterol levels in the CFE group were significantly decreased compared to those in the cholesterol group. The CFE group showed increased amounts of feces, total fecal bile acid and dietary fiber as compared to basal and cholesterol group. Hematoxylin and eosin staining revealed fat droplets in the livers of the rats in the cholesterol group; however, a decreased number of droplets was observed in the rats fed the CFE diet. Our results suggest that CFE supplementation may improve lipid metabolism, by controling serum and liver tissue cholesterol levels, and by increasing the total amounts of bile acid, dietary fiber and cholesterol excretion in feces.
Folate coenzymes are involved in one-carbon transfer reactions needed for the synthesis of nucleic acids, amino acids, and proteins which are very important for cell proliferation and differentiation. To investigate the effects of dietary folate content on plasma and tissue folate concentrations and on folate excretions in urine and feces, male Sprague-Dawley rats were raised for 4-10 weeks on semi-purified experimental diets containing 0mg, 2 mg, 8mg folate/kg diet. Folate concentrations were determined microbiologically using Lactobacillius casei (ATCC 7469). When compared to the folate adequate diet, the folate deficient diet decreased folate levels in plasma, liver and kidney , and the values were further decreased with experimental period. In rats reviving folate supplemented diets, plasma , liver and kidney folate adequate or supplemented diets, folate concentrations weer increased compared to animals on the folate adequate diet. In the folate adequate or supplemented diets, folate concentrations in the plasma and kidney were maintained at essentially the same level for 10 weeks . Folate concentrations in the liver, however, continued to increase with experimental period. Dietary folate intake seems to influence plasma and liver folate concentrations more than kidney folate concentrations. Folate excretions unrine and feces were significantly increased with dietary foalte intakes and experimental period. Folate excreted via urine was consideerably greater than that via feces. These resutls indicated that the foate supplemented diet improved plasma and tissue foalte status. Whether folate supplmentation improves foalte-dependent reactions remains to be researched.
This study was designed to obseve the effect of defatted perilla as a dietary fiber on lipid components of serum feces and liver in rats. Serum cholesterol and triglyceride were signfican-ly decreased in defatty perilla added groups. total lipid cholesterol and triglyceride of feces were significantly increased in defatted perilla added groups. Liver phospholipid content was higher in the defatted perilla added groups. The other lipid components of the liver were not affected by the defatted perilla administration. These results suggest the possibitlity that defatted perilla have a reducing effect of serum cholesterol and triglyceride through the incre-ment of feacal excretion of lipid component and phospholipid in liver.
Polymannuronate를 첨가한 사료를 흰쥐에 각각 기초식이군, 콜레스테롤식이군과 5$\%$ polymannuronate가 함유된 콜레스테롤식이군을 4주간 급이한 후 혈청, 간장조직 및 분변에 미치는 영향을 살펴보았다. 콜레스테롤식이군에 비해 polymannuronate식이군에서는 혈청 및 간장조직의 총콜레스테롤 함량이 유의적으로 감소하였으며, 간장조직 중의 지방분포도 동일한 경향을 보였다. Polymannuronate식이군은 콜레스테롤식이군보다 일일 분변량이 유의적으로 증가하였는데, polymannuronate가 답즙산 및 콜레스테롤과 결합하여 분변으로 배출되어 분변 중의 총담즙산과 총콜레스테롤 및 총식이섬유 함량이 증가한 것으로 보여진다. 이상의 결과에서, polymannuronate는 체내에서 담즙산 및 총콜레스테롤과 결합하여 분변 중으로 배출함으로써 흰쥐의 혈청 및 간장조직 중의 총콜레스테롤 함량을 감소시켜 지질개선의 효과를 나타내는 것으로 보여 진다.
This study was conducted to investigate the effect of Platycodi radix Saponin(PRS) on the reduction of lipid status in rats fed on high fat diet for 6 weeks after which lipid contents were measured in serum , liver and feces. The results obtained from this study are as follows : That the levels of total lipid, total cholesterol and triglyceride in serum and liver were significantly lower in the PRS group as compared with the control group. The contents of total lipid and total cholesterol excreted in the feces were tended to be slightly increase in PRS group compared to the control group, which were not significant.
율무쌀이 흰쥐의 혈장과 조직의 콜레스테롤, 중성지방, 이지질 및 총담즙산 수준에 미치는 영향을 알아보자 흰쥐에 1%의 콜레스테롤을 첨가시킨 실험식이를 자유급식하여 사양한 결과를 요약하면 다음과 같다. 1) 식이섭취량, 식이효율, 증체량은 각 군간에 유의한 차이가 없었다. 2)형장콜레스테롤 함량은 라이드 급여시 현저히 높았으며 라이드-ㅠ율무쌀을 급여시에는 가장 낮았다. 혈장의 중성지방 역시 라이드 급여시 높았다. 혈장의 인지질 역시 라아드 급여서 현저히 높았고 대두유군이 가장 낮았다. 혈장의 총담즙산 함량은 각군간에 차이가 없었으며 혈장 담즙산은 glycoursodeoxy cholicacid, tauroursodeoxy choilic acid와 cholic acid이었다. 3) 간의 단위무게당 총지질, 콜레스테롤, 중성지방역시 라이드- 율무쌀 급여시 현저히 낮았다. 4) 소장의 단위무게당 총지질, 콜레스테롤, 중성지방 함량도 역시 라이드-율무쌀을 급여시 낮았다. 5)실험식의 소화율을 모든식이 처리간에 차이가 없었으나 콜레스테롤 보유율은 라이드-전분군이 가장 높았고 대두유-율무쌀군이 가장 낮았다.

The large-scale preparation method for black soybean peptide (BSP; about 70% less than 10,000 Da; highly enriched with glutamic acid, aspartic acid, and arginine) was developed, and its effect on weight reduction and lipid profiles in rats was investigated. Sprague-Dawleymale rats were assigned to four dietary groups (high-fat diets containing 0, 2, 6, and 10% BSP) and fed four weeks to examine the effects of BSP. During the experiment, food intake was measured every two days and body weight was monitored two times a week. After the supplementation of BSP, liver and adipose tissues (epididymal, retroperitoneal and perirenal adipose tissue) in the rats were weighted and the lipid profiles in serum, liver, and feces were analyzed. At the results of body weight gain, liver and epididymal adipose tissue weight, BSP groups were more decreased than HF group (0% BSP), with greater decreases at higher BSP levels. The same patterns were shown in lipid profiles of serum, as BSP was increased, triglyceride and total cholesterol concentration decreased. The serum HDL-cholesterol level was increased with increasing at BSP levels. Total cholesterol concentration of liver and feces were decreased and increased, respectively, as BSP increased. The results confirm that BSP is involved in reducing the body weight and the improvement of lipid composition in serum and liver of rats and that BSP can be applied in weight reduction in the food products industry.
This study has been investigated the potenial of increased dietary cysteine to alter the effects of cadmium and lead on tissure and bone metal concentrations, excretion and tissue metallothionein(MT) concentrations. Fifty-four male rats of Sprgue-Dawley strain weighing 149$\pm$17g were divided into 9 groups according to body weight. Nine experimental diets with different cadmium (0ppm, 400ppm), lead(0ppm, 710ppm) and cysteine (0.06%, 0.45%, 0.90%) levels were given to rats for 30 days ; Food intake, weight gain, F.E.R, and weights of liver, kidney and femur were decreased in cadmium supplied groups than in cadmium free groups. Urinary and fecal cadmium excretions were increased and MT synthesis we induced in liver, kidney and small intestine in cadmium supplied groups. In lead supplied groups, weight gain and F.E.R were decreased. With cysteine supplementation in cadmium supplied groups, weight gain and F.E.R, and weights of liver, kidney and femur were increased. Cadmium excretion in feces and MT concentrations in liver and kidney were also increased with cysteine supplementation. In lead supplied groups, there was no significant increase in food intake, weight gain and F.E.R with cysteine supplementation. Lead excretion in feces was increased in cysteine supplemented groups. In conclusion, effect of cadmium administration was more toxic than lead adminstration. Cysteine alleviated cadmium and lead toxicity by increasing metallothionein concentration and fecal excretions of heavy metals. Especially, effect of cysteine supplementation was more effective in cadmium groups than in lead groups. Effect of cysteine supplementation was not different with level of cysteine supplementation in both cadmium and lead groups.
