Determination of Isotopic Ratios for Ca in Inductively Coupled Plasma Mass Spectrometry (ICPMS) by Removing Water Related Molecules

  • Calcium isotopic ratios are precisely measured by removing isobaric interferences originated from water in the plasma. Liquid Ar cryogenic trap combined with membrane desolvator could eliminate backgrounds at m/z 42 and 44. Slow drift of ICP-MS is corrected by the frequent running of the standards. It is found necessary to separate Ca from the sample matrix using Ca oxalate precipitation technique. Currently, the RSD is 0.5-1.0% for 2 minutes of measurement but is expected to be improved if the measurement time is increased. The technique was applied to 42Ca enriched baby fecal samples and successfully determined 42Ca/44Ca ratio changes.

A Study on a Morphological Identification of Notoginseng Radix (삼칠근(三七根)의 형태(形態)에 관한 연구(硏究))

  • Objectives : In order to distinguish morphological characteristics of trunk bark and root bark of Ulmus davidiana var. japonica (Rehder) Nakai and the trunk bark and root bark of Hemiptelea davidii Planchon were sampled and compared in terms of their external and internal features with flour states according to their medical use, through microscopic examination. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and the flour of it was mounted by the liquid made by the same ratio of each of glycerin, acetic acid, and water, and then observed and photographed by olymphus-BHT. Results : 1. Internal Features 1) A large parenchymatous cell was observed in the phloem of the slice of both trunk bark and root bark of Ulmi Cortex, However, both of the trunk bark and root bark of Hemipteleae Cortex did not have parenchymatous cell in the phloem; instead, stone cells including much square crystal of calcium oxalate were distributed around fiber bundle, and the parenchymatous cell included much druse crystal of calcium oxalate. 2) In both the Ulmi Cortex and Hemipteleae Cortex, rhytidome was observed in trunk bark, but not in root bark, but in the parenchymatous cell of the root bark of the Ulmi Cortex contained starch grain. 2. Flour States 1) In the flour of root bark of the Ulmi Cortex, a large parenchymatous cell was observed. However, in the flour of trunk bark and root bark of Hemipteleae Cortex, no parenchymatous eel was found; instead, stone cell including square crystal of calcium oxalate and druse crystal of calcium oxalate were observed. 2) There was no remarkable difference between the trunk bark and root bark of Hemipteleae Cortex. However, starch grain was contained in the parenchymatous cell of the root bark of Ulmi Cortex but not in the trunk bark of it. Conclusions : There were some morphological differences in external, internal, and flour parts of Ulmi Cortex and Hemipteleae Cortex. In particular, there was a morphological difference in flour states between the trunk bark and root bark of Ulmi Cortex, it is possible to use microscope to distinguish their flour states.

  • Silanized-hydroxypropyl methylcellulose (Si-HPMC)/phase transformed calcium phosphate (PTCP) composites are prepared to purpose application of injectable bone cements with enhanced biocompatibility. The crystal structure and chemical state of the synthesized PTCP and Si-HPMC as solid and liquid phase of the composite cements were measured by XRD and FT-IR. The handling and mechanical properties of cements were measured by injectability tests and three-point bending tests. The in-vitro mechanical properties, XRD, and SEM results of bone cements were showed that enhanced hardening behavior was an inherent function of bone cements after in-vitro test. The cytotoxicity result of bone cements also was showed enhanced biocompatibility. Therefore, these injectable cements had potential be used as calcium phosphate cements for biomedical applications.

Bioremediation of Heavy Metal Contaminated Mine Wastes using Urease Based Plant Extract (요소분해효소 기반 식물추출액을 이용한 광산폐기물 내 중금속 오염 저감)

  • Acid mine drainage occurrence is a serious environmental problem by mining industry, it usually contains high levels of metal ions, such as iron, copper, zinc, aluminum, and manganese, as well as metalloids of which arsenic is generally of the greatest concern. An indigenous plant extract was used to produce calcium carbonate from Canavalia ensiformis as effective biomaterial, and its ability to form the calcium carbonate under stable conditions was compared to that of purified urease. X-ray diffraction and scanning electron microscopy were employed to elucidate the mechanism of calcium carbonate formation from the crude plant extracts. The results revealed that urease in the plant extracts catalyzed the hydrolysis of urea in liquid state cultures and decreased heavy metal amounts in the contaminated soil. The heavy metal amounts were decreased in the leachate from the treated mine soil; 31.7% of As, 65.8% of Mn, 50.6% of Zn, 51.6% of Pb, 45.1% of Cr, and 49.7% of Cu, respectively. The procedure described herein is a simple and beneficial method of calcium carbonate biomineralization without cultivation of microorganisms or further purification of crude extracts. This study suggests that crude plant extracts of Canavalia ensiformis have the potential to be used in place of purified forms of the enzyme during remediation of heavy metal contaminated soil.

Dohaekseungkitang extract induced apoptosis in Human Cervical carcinoma HeLa cells (도핵승기탕(桃核承氣湯) 자궁경부암세포(子宮經部癌細胞)(HeLa cell)의 apoptosis에 미치는 영향(影響))

  • Purpose : To address the ability of Dohaekseungkitang (DST: a commonly used herb formulation in Korea, Japan and China to have anti-cancer effect on cervical carcinoma), we investigated the effects of DST on programmed cell death (apoptosis) in HeLa human cervical carcinoma cells. Methods : We cultured HeLa cell which is human metrocarcinoma cell in D-MEM included 10% fetal bovine serum(Hyclone Laboratories) below $37^{\circ}C$, 5% CO2. Then we observed apoptosis of log phage cell which is changed cultivation liquid 24 Hours periodically. Results : After the treatment of DST for 48 hours, apoptosis occurred in a dose-dependent manner. In this study, we have shown that DST induces calpain and the associated caspase-8 and -9 activations. Apoptosis was prevented by pre-incubation of the cells with the calcium cHeLator-BAPTA-AM, calcium channel blocker-Nif edipine or Ryonidine agonist-Ryonidine peptide, implicating calcium in the apoptotic process. Ubiquitous calpains (mu- and m-calpain) have been repeatedly implicated in apoptosis, especially in calcium-related apoptosis. However this study showed 1hat either calpain inhibitor-calpastin or caspase-3 inhibitor-DEVD- did not blocked the herb formulation-induced apoptosis in HeLa human cervical carcinoma cells. D ST initiates a cell death pathway that is partially dependent of caspases. DST-induced apoptosis requires caspase-independent mechanism. Conclusion : We conclude that DST-induced calpain activation triggers the intrinsic apoptotic pathway in which caspase-independent mechanism is also involved.

Recovery of Lactic Acid from Fermentation Broth Using Precipitation and Reactive Distillation (발효액으로부터 침전과 반응증류를 이용한 젖산의 회수)

  • Precipitation and reactive distillation were employed to recover lactic acid from fermentation broth. Lime was initially added to fermentation broth in order to convert soluble lactic acid to an insoluble calcium lactate form. Drowning-out crystallization was used to decrease the solubility of calcium lactate by adding ethanol as a co-precipitant. In the ideal solution of organic acids as well as fermentation broth, precipitation experiments were performed with varying amounts of ethanol. Precipitation process was followed by reactive distillation. Carboxylate salts formed in the previous precipitation process were mixed with carbon dioxide and triethylamine to precipitate as calcium carbonate. The remaining liquid was distilled for 1 hr at different temperatures. Triethylamine and water were recovered from the top of the distiller, while organic acids, inducing lactic acid as a main component remained in feeding bottle. The yield of recovered lactic acid was 67.5% with the purity of 99.7%.

Calcium Solubilization Ability and Anti-Inflammatory Effects of Hydrolyzed Casein

  • This study performed to evaluate the applicability of functional dairy food materials by comparing the calcium solubilization ability and anti-inflammatory effects of hydrolyzed casein protein. Commercial enzyme (Alcalase®; Neutrase®; Protamex®; Flavourzyme®) was added to the 10% casein solution to prepare the casein hydrolysates. Samples obtained every hour [1:200 (w/v)]. According to results of measuring the degree of hydrolysis (DH), all of four enzymatic hydrolysates increased rapidly from 30 to 40 min, and after 150 min, there were no change. Protamex® and Neutrase® had the highest DH compared to others enzymatic hydrolysates. After that, peptides obtained throughout a preparative liquid chromatography system. In the calcium solubility experiments, neutrase fraction (NF) 4 and NF7 showed similar activities with casein phosphopeptide (CPP). In vitro cell experiments showed that no cytotoxicity except for NF6. Also, the production of nitric oxide (NO) inhibited as the concentration of fraction samples increased. The cytokine (IL-1α, IL-6, and TNF-α) production was lower than lipopolysaccharide (+) group significantly. Therefore, the possibility of anti-inflammatory activity found in the hydrolyzed samples. According to the above experiments, NF3 and Protamex Fraction (PF) 3 selected. Amino acids selected throughout an AccQ-Tag system. As a result, 17 species of amino acids and several species of unknown amino acids identified. Both fractions had the highest content of phenylalanine. This study identified the potential of biologically active and functional peptides derived from casein that affect the food and dairy industry.

The Effect of Refractory Crucible on Microstructure of Duplex Stainless Steel Cast with Gadolinium during Air Induction Melting (대기용해 시 내화물 도가니의 종류가 가돌리늄(Gadolinium)을 함유한 듀플렉스 스테인레스 강의 미세조직에 미치는 영향)

  • This paper reports the effect of a refractory crucible type on the microstructure of duplex stainless steel (DSS) cast with the addition of gadolinium using air-induction melting. Grade 4A DSSs with 1 wt% of gadolinium (Gd) were fabricated in various crucibles including alumina ($Al_2O_3$), magnesia (MgO), calcia (CaO) coated with yttria ($Y_2O_3$) and graphite. The standard free energies of the formation of calcium and yttrium oxide were lower than those of gadolinium oxide and other crucible elements based oxide. The yield of Gd in DSS using $Al_2O_3$, MgO, CaO-coated $Y_2O_3$ and graphite was 5, 19, 83 and 96%, respectively. As Gd yield increased, the amount of Gd-based inclusions increased, the size of the inclusions were reduced, and the inclusions became evenly distributed.

Effect of Heat-Treat Methods on the Soluble Calcium Levels in the Commercial Milk Products

  • Milk is well known to be rich in some nutrients such as protein, calcium, phosphorus, and vitamins. In particular, absorption and bioavailability of calcium receive lots of attention because calcium is very little absorbed until it is changed to the ionized form in the intestine. In this study, concentration of the soluble calcium was determined in the commercial bovine milk products, which were processed by different heat-treatment methods for pasteurization. As for general constituents, lactose, fat, protein, and mineral were almost same in the liquid milk products by different processors. Ultrafiltration of the skimmed milk caused little change in the permeate as for lactose content but both fat and protein decreased. pH values ranges from 6.57-6.62 at room temperature and slightly increase after centrifugation, 10,000 g, 10 min. Rennet-coagulation activity was the lowest in the ultra high temperature (UHT-)milk compared to the low temperature long time (LTLT-) and high temperature short time (HTST-)milk products. Each bovine milk products contains 1056.5-1111.3 mg/kg of Ca. The content of sulfhydryl group was the lowest in raw milk compared to the commercial products tested. For the skimmed milks after ultrafiltration with a membrane (Mw cut-off, 3 Kd), soluble Ca in the raw milk was highest at 450.2 mg/kg, followed by LTLT-milk 336.4-345.1 mg/kg, HTST-milk 305.5-313.3 mg/kg, UHT-milk 370.3-380.2 mg/kg in the decreasing order. After secondary ultrafiltration with a membrane (Mw cut-off, 1 kD), total calcium in raw milk had a highest of 444.2 mg/kg, and those in the market milk products. As follow: UHT-milk, 371.3 to 378.2 mg/kg; LTLT-milk, 333.3 to 342.2 mg/kg; HTST-milk 301.9 to 311.2 mg/kg in a decreasing order.

The Content of Macrominerals in Beverages, Liquid Teas, and Liquid Coffees (유통 음료, 액상차 및 액상커피의 다량무기질 함량)

  • The aims of this study were to investigate some macrominerals (Na, Ca, P, K, Mg) in 207 beverages, 19 liquid teas, and 24 liquid coffees. The samples were digested by microwave and determinations of macrominerals were carried out by an Inductively Coupled Plasma Optical Emission Spectrometer (ICP-OES). The elements, listed in order of mean value of macromineral content, were potassium $208.4{\pm}298.2mg/L$ ($72.2{\pm}169.8mg/container$)> calcium $89.0{\pm}161.0mg/L$ ($26.0{\pm}57.7mg/container$)> sodium $71.2{\pm}75.0mg/L$ ($20.9{\pm}27.9mg/container$)> phosphorus $55.6{\pm}91.9mg/L$ ($17.9{\pm}33.8mg/container$)> magnesium $6.1{\pm}18.4mg/L$ ($2.4{\pm}10.1mg/container$). All 250 samples contained sodium and potassium, and the detection rate of calcium, phosphorus and magnesium was 88.4%, 93.2%, and 20.4%. The mean ratio of phosphorus to calcium in beverages, liquid teas, and liquid coffees was $4.2{\pm}16.0$ (ND~164.4), and sports drinks showed the highest mean ratio ($48.5{\pm}75.6$) significantly (p<0.05). In case of sodium, detected content exceeding labeling regulations (less then 120%) was observed in 12 samples (5.5%).
