Zinc Oxide Nanoparticles Exhibit Both Cyclooxygenase- and Lipoxygenase-Mediated Apoptosis in Human Bone Marrow-Derived Mesenchymal Stem Cells

  • Nanoparticles (NPs) have been recognized as both useful tools and potentially toxic materials in various industrial and medicinal fields. Previously, we found that zinc oxide (ZnO) NPs that are neurotoxic to human dopaminergic neuroblastoma SH-SY5Y cells are mediated by lipoxygenase (LOX), not cyclooxygenase-2 (COX-2). Here, we examined whether human bone marrow-derived mesenchymal stem cells (MSCs), which are different from neuroblastoma cells, might exhibit COX-2- and/or LOX-dependent cytotoxicity of ZnO NPs. Additionally, changes in annexin V expression, caspase-3/7 activity, and mitochondrial membrane potential (MMP) induced by ZnO NPs and ZnO were compared at 12 hr and 24 hr after exposure using flow cytometry. Cytotoxicity was measured based on lactate dehydrogenase activity and confirmed by trypan blue staining. Rescue studies were executed using zinc or iron chelators. ZnO NPs and ZnO showed similar dose-dependent and significant cytotoxic effects at concentrations ${\geq}15{\mu}g/mL$, in accordance with annexin V expression, caspase-3/7 activity, and MMP results. Human MSCs exhibited both COX-2 and LOX-mediated cytotoxicity after exposure to ZnO NPs, which was different from human neuroblastoma cells. Zinc and iron chelators significantly attenuated ZnO NPs-induced toxicity. Conclusively, these results suggest that ZnO NPs exhibit both COX-2- and LOX-mediated apoptosis by the participation of mitochondrial dysfunction in human MSC cultures.

녹두추출물의 자극완화 효과에 관한 임상 연구 (Clinical Studies on the Anti-Irritation Effects of Mung Bean (Phaseolus aureus) Extract in Cosmetics)

  • 본 연구는 화장품에서 빈번히 사용되고 있는 다양한 자극원에 대한 녹두추출물의 자극완화 효과를 알아보기 위한 임상연구에 관한 것이다. 녹두는 해독 작용이 탁월하여 예로부터 민간요법이나 화장료의 재료로 사용되어 왔다. 그러나, 녹두의 생물학적인 효능 및 구성 성분을 구체적으로 밝혀낸 연구는 전무한 실정이다.. 우리는 기존의 연구를 통하여 녹두의 에탄을 추출물이 항산화 및 항염증 효과가 우수한 것을 확인하고 이러한 효능을 제공하는 유효물질인 비텍신(vitexin)과 이소비텍신(isovitexin)을 분리해낸 바 있다. 본 연구에서는 녹두추출물의 항염증 기작을 살펴보고자 흰쥐 복강 비만세포로부터의 항원 유도성 히스타민 유리 억제능 및 5-lipoxygenase 활성 억제능을 측정하였다. 그 결과 녹두추출물은 농도 의존적으로 히스타민 유리를 억제시켰지만 5-lipoxygenase 활성을 억제하는 효과는 나타내지 않았다. 또한, 현재 화장품에 널리 사용되고 있는 자극 유발 물질인 lactic acid, retinol, 방부제 등에 대한 녹두추출물의 자극완화 효과를 알아보기 위하여 다양한 임상 연구를 수행하였다. 20명의 피험자를 대상으로 실시한 인체 첩포시험 결과, 5.0% lactic acid, 4000 IU retinol, 1.0% Preservative mixture가 들어간 제형에 2.0% 녹두추출눌을 첨가할 경우 각각 60%, 30%, 50% 정도의 우수한 자극 억제 효과를 보여주었으며, 주관적인 자극을 평가하는 피부 자극감시험 결과도 2.0% 녹두추출물을 함유하는 제형에서 약 50∼30%의 자극완화 효과를 보여주었다. 마지막으로, 30명의 피험자를 대상으로 4주간 실시한 double-blind usage test 결과 역시 녹두추출물을 화장품에 사용하였을 경우 우수한 항자극 효과를 보임을 확인하였다.

Evaluation of the quality stability on the soybean flour depending on storage conditions

  • Legumes are good sources of various nutrients. Among legumes, soybean and its flour are accessible foods to consumers. However, in case of soybean flour, there is a disadvantage of easily going rancid. Thus, this study aimed to investigate how the quality of soybean flour changes during storage according to the packaging materials and storage temperatures. The raw and roasted soybean flours were packed in two types of packaging, polyethylene (PE) and polypropylene (PP) film bags respectively, and stored at three different storage temperatures (4, 20, and $45^{\circ}C$) for 1 year. The acid value, conjugated diene value, peroxide value, p-anisidine value, thiobarbituric acid (TBA) value, lipoxygenase activity, and fatty acid content of raw and roasted soybean flours were measured at the point of starting storing, and after 1, 2, 4, 8, and 12 weeks of storage. The acid value of soybean flour was increased for 4 weeks and thereafter significantly decreased (p < 0.0001). The conjugated diene value was significantly increased after 4 weeks storage at $45^{\circ}C$ with PE and PP film bags (p < 0.0001). The peroxide value had no changes during 4 weeks storage at $45^{\circ}C$ with PE and PP film bags, and then those was dramatically increased after 8 weeks (p < 0.0001). The p-anisidine values in all of storage conditions were decreased after 4 weeks. Lipoxygenase activity was decreased at 12 weeks storage at $20^{\circ}C$ and $45^{\circ}C$ with PP film bags. Acid value had positive correlations with p-anisidine value (r = 0.30 and p < 0.0001) and lipoxygenase activity (r = 0.36 and p < 0.0001), and had negative correlations with conjugated diene value (r = -0.45 and p < 0.0001) and peroxide value (r = -0.25 and p < 0.001). Conjugated diene value had a high positive correlation with peroxide value (r = 0.76 and p < 0.0001), but that had a negative correlation with lipoxygenase activity (r = -0.51 and p < 0.0001). Peroxide value had negative correlations with p-anisidine value (r = -0.20 and p < 0.01) and TBA value (r = -0.15 and p < 0.05). The degree of reduction in fatty acid content of raw soybean flour was higher than the roasted soybean flour during the storage. Total fatty acid content had positive correlations with acid value (r = 0.45 and p < 0.0001) and p-anisidine value (r = 0.58 and p < 0.0001), but had a weak negative correlation with conjugated diene value (r = -0.19 and p < 0.01). This study showed how the rancidity of the raw and roasted soybean flours progressed during storage. Thus, our findings can be used as base data to do a further study of finding and developing more stable storage conditions of the soybean flour.

Lipoxygenase 결여 콩 두부의 품질 특성 및 항산화 활성 (Quality characteristics and antioxidant activity of tofu made from lipoxygenase-free genotypes)

  • LOX 결여 콩(개척#1, 개척#2 및 진양)과 일반콩(태광)으로 만든 두부의 이화학적 특성 및 항산화 활성을 비교하였다. 두부의 무기물 함량은 대조구에 비해 진양 두부에서 훨씬 높은 함량이었다. 경도 및 검성은 대조구에 비해 LOX 결여 콩 두부에서 유의적으로 높았다. 두부의 맛, 풍미를 포함한 전체적인 기호도는 개척#2 두부가 다소 높게 평가되었다. 두부의 이소플라본 함량은 대조구에 비해 LOX 결여 콩 두부에서 높았다. 두부의 총 페놀 함량은 대조구와 개척#1 및 개척#2 두부에서 비슷한 수준이었으며, 플라보노이드 함량은 대조구에 비해 개척#1 두부는 2.6배, 개척#2 두부는 1.4배 높았다. 두부의 항산화 활성은 개척#1 두부에서 가장 높았으며, 다음으로 개척#2 두부였다. $4^{\circ}C$에서 15일 저장하는 동안 두부 침지액의 탁도는 점차 증가하였는데, 저장 15일 경과 후 개척#1 및 #2 두부는 태광 두부에 비해 유의적으로 낮았다. LOX가 결여된 개척#1 및 #2 두부는 이소플라본 및 총 페놀 함량이 태광 두부에 비해 높아 항산화 활성 및 저장 안정성에 효과적이었으며, 특히 개척#2 두부는 전체적인 기호도가 높아 LOX 결여 콩 품종이 두부 가공에 적합할 것으로 판단된다.

Anti-inflammatory Compounds from the Leaves of Ailanthus altissima Meihua JIN

  • In our ongoing search for biological components from the Korea endemic plants, the MeOH extract of Ailanthus altissima leaves (Simaroubaceae) showed cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) dual inhibitory activity by assessing their effects on the production of prostaglandin $D_2$ ($PGD_2$) and leukotriene $C_4$ ($LTC_4$) in mouse bone marrow-derived mast cells (BMMCs). In further study, eight compounds, squalene (1), ${\beta}$-sitosterol (2), scopoletin (3), quercetin (4), luteolin (5), astragalin (6), scopolin (7), and daucosterol (8) were isolated, the chemical structures were elucidated on the basis of physicochemical and spectroscopic data and by comparison with those of published literatures. Among the compounds, 2, 4, and 5 strongly inhibited both the COX-2-dependent PGD2 generation with $IC_{50}$ values of 2.6, 7.3 and 2.5 ${\mu}M$, respectively and the generation of $LTC_4$ in the 5-LOX dependent phase with $IC_{50}$ values of 2.0, 5.1 and 1.8 ${\mu}M$, respectively, which suggest that the anti-inflammatory activity of A. altissima might occur in part via the inhibition of both $PGD_2$ and $LTC_4$ generation by 2, 4 and 5.

Effect of Multiple Freeze-Thaw Cycles on Lipid Degradation and Lipid Oxidation of Grass Carp Surimi Containing Different Amounts of Pork Back Fat

  • Fresh grass carp was used to produce surimi samples that were supplemented with 50 g/kg, 100 g/kg, or 150 g/kg pork back fat. The lipid composition, lipase activity, lipid oxidation index, and lipoxygenase activity of samples subjected to repeated freezethaw process were determined to assess the effects of the added fat on lipolysis and lipid oxidation of grass carp surimi. Freeze-thaw treatment increased free fatty acid content, mainly due to the decomposition of phospholipids and some neutral lipids by lipase. With repeated freeze-thaw treatment, the levels of free fatty acids and phospholipids were correlated with the lipid oxidation indexes and lipoxygenase activity, indicating that lipid degradation can promote lipid oxidation. In the same freeze-thaw cycle, surimi products with high fat content are more vulnerable to oxidative damage, neutral lipids are the main source of free fatty acids in the early stage of freeze-thaw, and phospholipids are the main source of free fatty acids in the late stage.

초피나무 추출물의 항산화, 항염증 및 항혈전 효능에 관한 연구 (A Study on the Antioxidative, Anti-inflammatory and Anti-thrombogenic Effects of Zanthoxylum piperitum DC. Extract)

  • 본 연구에서는 초피나무의 생리활성 물질로서의 효능을 알아보고자 초피나무 뿌리, 줄기 및 잎의 부위별 및 분획물을 이용하여 흰쥐 간 microsome의 지질과산화 억제 작용, DPPH radical 소거작용, soybean lipoxygenase 활성 및 activated partial thromboplastin times(APTT)을 관찰하였다. 초피나무의 뿌리, 줄기 및 잎의 부위별 및 분회별 추출물을 이용하여 흰쥐 간 microsome의 지질과산화 억제 활성을 1.00 mg/mL농도에서 1차 screening한 결과 뿌리, 줄기 및 잎 모두 ethyl acetate층 및 methylene chloride층에서 높은 활성을 나타내었다. 부위별 지질과산화 억제 활성을 비교한 결과 잎의 활성이 가장 높았다. 초피나무 뿌리, 줄기 및 잎의 ethyl acetate층 및 methylene chloride층은 $0.05\~0.25$ mg/mL의 범위에서 농도 의존적으로 지질과산화 억제활성이 증가하였다. 지질과산화 억제활성의 차이가 가장 큰 0.05 mg/mL 농도에서 선별된 물질의 지질과산화 억제활성을 비교한 결과 초피나무 잎의 ethyl acetate층 및 methylene chloride 층에서 높았다. DPPH에 의해 생성된 라디칼 소거 활성은 지질과산화 억제활성과 같은 경향이었다. 초피나무의 SLO 활성 및 APTT활성을 위에서와 같이 1.00 mg/mL 농도, 농도별 및 활성의 차이가 큰 농도에서 screening한 결과 잎의 n-butanol층에서 가장 높은 활성을 나타내었다. 이상의 결과로부터 초피나무 뿌리, 줄기 및 잎의 분획물 중에서 잎의 ethyl acetate층의 높은 지질과산화 억제효과 및 DPPH radical소거 작용을 관찰할 수 있었으며, 잎의 n-butanol층에서의 항염증 및 항응고 효능을 관찰할 수 있었다.

마늘의 가공 조리 방법에 따른 lipoxygenase 활성도 저해 효과 (Inhibition of Lipoxygenase Activity by the Extract of Various Processed Garlic)

  • Bioactivity of the extract from various processed garlic was evaluated based on the inhibition of lipoxygenase(LPO), and the effect of some stabilizers on the bioactivity was investigated. Water, ethanol or chloroform extract of 30 min boiled garlic showed 75%, 76% or 70% inhibition, respectively, compared to extracts of fresh garlic. In pickled garlic, LPO inhibition decreased gradually during storage. Chloroform extract of 40 day-stored pickled garlic inhibited LPO by 77%, and even on 60th day storage it still retained inhibitory effect of 73%, compared to that of fresh garlic. Meanwhile, the powdered (freeze-dried) garlic showed more bioactivity(80%) than the other processed garlics, and moreover, the jrreversible/unstable components seem to be stabilized by freeze-drying. The optimum pH for stabilization of bioactive components in garlic macerate was pH 3 for 48 hr incubation and pH 11 for 6 hr incubation. In addition, the effect of NaCl was not so great, although but maximal stabilization was observed at 150 mM. Stabilizing effect of $\alpha$-tocopherol was markedly great, and at 6mM it showed 308% stabilizing effect after 48hr incubation. More stabilizing effect was observed at lower concentrations of ascorbic acid($\leq$0.6mM) than higher concentrations. The stabilizing effect of soybean oil was found to be remarkable only during initial period(6 hr) of incubation.

냉동저장동안 풋콩의 품질에 영향을 미치는 Blanching 조건 (Effects of Blanching Conditions on the Quality of Immatured Soybeans during Frozen Storage)

  • Blanching conditions for immatured soybeans were optimized by analyzing the effects of various time/temperature blanch treatment on the activities of peroxidase and lipoxygenase, the stability of vitamin C and color, and moisture content in immatured soybeans for the purpose of minimizing quality deterioration during frozen storage. Blanching at 96$^{\circ}C$ for 70 sec led to maximum inactivation of lipoxygenase in the immatured soybeans, while my blanching conditions tested in this study were not enough to inactivate peroxidase. Blanching at 82$^{\circ}C$ for 60 sec resulted in the highest amount of vitamin C remaining in the immatured soybeans after blanching. Hunter -a/b ratios of immatured soybeans blanched at 82$^{\circ}C$ for 60 sec and hue values (ΔE) of the immatured soybeans blanched at 76$^{\circ}C$ for 60 sec showed the closest values to those of fresh products. The changes in moisture content of immatures soybeans was not so significant after blanching. In conclusion, it was suggested that immatured soybeans be blanched at 96$^{\circ}C$ for 70 sec to minimize lipoxygenase activity and resulting quality deterioration, while blanching at 82$^{\circ}C$ for 60 sec was recommended to stabilize vitamin C and color.

CoMFA vs. Topomer CoMFA, which One is better a Case Study with 5-Lipoxygenase Inhibitors

  • Quantitative structure-activity relationships (QSAR) have been applied for two decades in the development of relationships between physicochemical properties of chemical substances and their biological activities to obtain a reliable statistical model for prediction of the activities of new chemical entities. The fundamental principle underlying the QSAR is that the structural difference is responsible for the variations in biological activities of the compounds. In this work, we developed 3D-QSAR model for a series of 5-Lipoxygenase inhibitors, utilizing comparative molecular field analysis (CoMFA) and Topomer CoMFA methodologies. Our developed models addressed superiority of Topomer CoMFA over CoMFA. The CoMFA model was obtained with $q^2$=0.593, $r^2$=0.939, $Q^2$=0.334 with 6 optimum number of components (ONC). Higher statistical results were obtained with the Topomer CoMFA model ($q^2$=0.819, $r^2$=0.947, ONC=5). Further robustness of developed models was checked with the ANOVA test and it shows F=113 for CoMFA and F=162.4 for Topomer CoMFA model. Contour map analysis indicated that the more requirement of electrostatic parameter for improved potency.
