카르복시베타인 유도체(誘導體)의 계면활성(界面活性) (Surface Activities of Carboxybetaine Derivatives)

  • Isoelectric point and surface tension of twelve of the alkylcarboxy betaines such as 2-(trimethylammonio) dodecanoate, 2-(trimethyammonio) tetradecanoate, 2-(trimethylammonio) hexadecanoate, (dimethyldodecylammonio)ethanoate, (dimethyltetradecylammonio)ethanoate, (dimethylhexadecylammonio) ehtanoate, 2-(dimethylododecylammonio) propanoate, 2-(dimethyltetradecylammonio)proopanoate, 2-(dimethylhexadecylammonio)propanoate, 2-(dimethyltetradecylammonio)-3-phenyl propanoate, 2-(dimethyltetradecylammonio)-3-phenyl propanoate are tested. From the measurement of the isoelectric point, it was found that the isoelectric point were leaned toward the alkaline zone for c-alkylarbrxybetaines, and toward the acidic zone for N-alkylcarboxybetaines. At the range of the carboxybetaine concentration $2\;{\times}\;10^{-2}{\sim}2\;{\times}\;10^{-5}$mole/l, the surface tension of the aqueous solution were decreased to 30-38 dyne/cm, showing the tendency that the ability of lowering the surafce tension was depending on the increase of carbon atom number in the lipophilic alkyl chain. The critical micelle concetration measured by the surface tension and concentration curves have been found at the range of $10^{-2}{\sim}10^{-5}$mole/l.

  • Four amphoteric surfactants, 1-(N-alkyl-N,N-dimethyl ammonio)-4-naphthalene sulfonates, were prepared by the alkylation of 1-(N,N-dimethylamino)-4,naphthalene sulfonic acid with chloroalkanes such as 1-decylchloride, 1-tetradecyl chloride and 1-hexadecyl chloride. These quaternary ammonium compounds such as 1-(N-decyl-N, N-dimethylammonio)-4-naphthalene sulfonate, 1-(N-dodecyl-N,N-dimethylammonio)-4-naphthalene sulfonate, 1-(N-tetradecyl-N,N-dimethylammonio)-4-naphthalene sulfonate and 1-(N-hexadecyl-N,N-dimethylammonio)-4-naphthalene sulfonate could be separated by means of thin layes chromatography and column chromatography. The surface chemical properties such as surface tension, foaming power, foam stability, wetting efficiency and solubilizing effect for these four compounds were measured. Also critical micelle concentration and hydrophilic-lipophilic balance(HLB) were evaluated. These compounds showed good surface as O/W type emulsifying agent and detergent.

  • 1-(carboxyalkyl) trimethyl ammonium chlorides such as 1-(carboxyundecyl) trimethyl ammonium chloride and 1-(carboxytridecyl) trimethyiammonium chloride were synthesized by the reaction of ${\alpha}-bromoalkanoic$ acid with trimethyl amine hydrochloride. In other hand, (carboxymethyl) alkyl dimethyl ammonium chlorides such as (carboxymethyl) dodecyl dimethyl ammonium chloride and (carboxymethyl) tetradecyl dimethyl ammonium chloride were synthesized by the reaction of alkyl dimethylamine with sodium chloroacetate. The four kinds of alky carboxy betaine such as 2-(trimethylammonio) dodecanoate, 2-(trimethyl ammonio) tetradecanoate, (dodecyl dimethylammonio) ethanoate and (tetradecyl dimethyl ammonio) ethanoate were prepared from 1-(carboxyalkyl) trimethyl ammonium chlorides or (carboxymethy1) alkyl dimethyl ammonium chlorides. The surface activities including surface tension, emulsifying power, foaming power, foam stability, deflocculating effect, effectiveness of wettability and solubilizing effect were measured and also critical micelle concentration and hydrophilic-lipophilic. balance(HLB) were evaluated. These carboxy betaines show good surface activities as O/W type emulsifing agent and detergent.

  • One of the methods to increase the solubility of a drug is to use complexation with a cyclodextrin. Due to the hydrophobic nature of the interior cavity of the cyclodextrin, it has been known that undissociated lipophilic drugs can be included within the cyclodextrin by hydrophobic interaction. Recently, inclusion of hydrophilic or dissociated form of a drug has been investigated. In this study, the synergism of pH and complexation with $hydroxypropy-{\beta}-cyclodextrin\;(HP\;{\beta}\;CD)$ to increase the solubility of two oxicam derivatives was investigated. In addition, the effect of partition coefficient of dissociated and undissociated form of the drug on the extent of complexation with HP ${\beta}$ CD was studied. The solubility was measured by equilibrium solubility method. The solubility of tenoxicam and piroxicam increased exponentially with an increase in solution pH above the pKa of the drug in the presence and absence of HP ${\beta}$ CD. The solubility of the drugs increased linearly as a function of HP ${\beta}CD$ concentration at fixed pH. Although the stability constant of ionized species is less than that of the unionized species, the concentration of the ionized drug complex is greater than that of the unionized drug complex due to higher concentration of ionized species at pH 7.3.

떡쑥 추출물로부터 분리된 Dicaffeoylquinic Acid 유도체들의 항산화 및 타이로시네이즈 저해 활성 (Antioxidant and Tyrosinase Inhibitory Activities of Dicaffeoylquinic Acid Derivatives Isolated from Gnaphalium Affine D. DON)

  • 본 연구에서는 떡쑥 추출물로부터 3가지 dicaffeoylquinic acid (DCQA)를 분리 정제하여 구조를 결정하고, 항산화 활성, 세포보호효과, 그리고 tyrosinase 저해 활성을 평가하였다. 떡쑥 추출물의 에틸아세테이트 분획에 대하여 크로마토그래피, $^1H$-NMR 및 MS 분석을 한 결과, 분리된 화합물은 총 3가지 DCQA 유도체 : 3,5-dicaffoylquinic acid (3,5-DCQA, 1), 4,5-dicaffeoylquinic acid (4,5-DCQA, 2), 1,5-dicaffoylquinic acid (1,5-DCQA, 3)로 나타났다. 분리된 compound 1-3의 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical 소거활성($FSC_{50}$)은 각각 3.70, 5.80 및 $5.50{\mu}M$로 지용성 항산화제로 알려진 (+)-${\alpha}$-tocopherol ($FSC_{50}=21.90{\mu}M$)과 비교하여 더 큰 free radical 소거활성을 나타내었다. compound 1-3의 $^1O_2$로 유도된 사람 적혈구의 광용혈에 대한 세포보호효과는 (+)-${\alpha}$-tocopherol과 유사한 세포보호활성을 나타내었다. Compound 1-3의 tyrosinase 저해활성($IC_{50}$)은 각각 0.15, 0.16, 및 0.13 mM로 미백성분으로 알려진 arbutin ($IC_{50}=0.33mM$)과 비교하여 높은 tyrosinase 억제효과를 나타내었다. 이상의 결과들은 세 가지 DCQA가 식품 또는 화장품 산업에서 항산화 및 미백 기능성 소재로서 응용가능성이 있음을 시사한다.

자초 뿌리의 함유성분 및 아토피피부염 관련 연구현황 (Progress on Phytochemical and Atopic Dermatitis-related Study of the Root of Lithospermum erythrorhizon)

  • Traditionally, the root of Lithospermum erythrorhizon Sieb. et Zucc(L.E) has been used as efficacious therapy for inflammation, burns, frostbite and skin ailments (e.g eczema and psoriasis). It contains isohexenylnaphthoquinone derivatives (shikonin and its esters) and furylhydroquinones (shikonofurans) in lipophilic fractions and caffeic acid oligomers (rosmarinic acid, lithospermic acid B) in polar fractions. Recently, new preparative isolation and analysis procedures of shikonin along with its oligomers from the extract of L. erythrorhizon by the combination of high-speed counter-current chromatography with high-performance liquid chromatography-diode array detection have also been introduced. Although there have been many reports on the wound healing, antiinflammatory, and anticancer effects, the research on the effects of anti-atopic dermatitis of the root of L. erythrorhizon were relatively scarce. However, in recent years, new information gathered from research efforts, on the anti-atopic dermatitis properties of the extract or constituents of L. erythrorhizon has been accumulated. In this paper, the findings and advance on the in vitro and in vivo activities of L. erythrorhizon and its constituents especially focused on antiinflammatory and anti-atopic dermatitis effects are summarized. The phytochemical constituents of L. erythrorhizon or its tissue cultures are also presented. Although there are few to verify or refute its activity in human, one result of clinical study of the extract of L. erythrorhizon on the atopic dermatitis patients was introduced to assess the possibility of its clinical use. The reported mechanisms of action and in vivo pharmacological studies in different animal models for the various types of extracts or constituents of L. erythrorhizon are supportive of its therapeutic potential or dietary supplement, however, more evidence from clinically relevant models, as well as systemic studies on the active constituents or the various types of standardized extracts at the cellular and molecular level, are required.

In silico Analysis on hERG Channel Blocking Effect of a Series of T-type Calcium Channel Blockers

  • Human ether-a-go-go related gene (hERG) potassium channel blockade, an undesirable side effect which might cause sudden cardiac death, is one of the major concerns facing the pharmaceutical industry. The purpose of this study is to develop an in silico QSAR model which uncovers the structural parameters of T-type calcium channel blockers to reduce hERG blockade. Comparative molecular similarity indices analysis (CoMSIA) was conducted on a series of piperazine and benzimidazole derivatives bearing methyl 5-(ethyl(methyl)amino)-2-isopropyl-2-phenylpentanoate moieties, which was synthesized by our group. Three different alignment methods were applied to obtain a reliable model: ligand based alignment, pharmacophore based alignment, and receptor guided alignment. The CoMSIA model with receptor guided alignment yielded the best results : $r^2$ = 0.955, $q^2$ = 0.781, $r^2_{pred}$ = 0.758. The generated CoMSIA contour maps using electrostatic, hydrophobic, H-bond donor, and acceptor fields explain well the structural requirements for hERG nonblockers and also correlate with the lipophilic potential map of the hERG channel pore.

뇌혈류 영상용 방사성의약품 PRODD의 $^{99m}Tc$ 표지 및 생체내분포 ($^{99m}Tc$ Labeling and Biodistribution of PRODD as a Cerebral Blood Flow Imaging Radiopharmaceutical)

  • $^{99m}Tc$ labeled PnAO(propylene amine oxime) derivatives have been widely studied as brain perfusion agents. We synthesized and characterized a PnAO derivative, (RR/SS/ meso)-4,8-diaza-3,9-dimethylundecane-2, 10-dione bisoxime(PRODD). Proton-NMR spectroscopy and thin layer chromatography(silica gel) were performed for its characterization. PRODD was labeled with $^{99m}Tc$ using stannous chloride as reducing agent. The labeling efficiency was determined to be about 85%. Brain uptakes of $4.16{\pm}0.42$ %ID/g and $3.24{\pm}0.13$ %ID/g were found after 10min and 30min after intravenous injection. Brain-to-blood ratios were 1.17 and 0.75 at 10 and 30 minutes, which were lower than 1.3 and 1.9 of the ratios with commercial ${\pm}$-HMPAO. Autoradiographs of rat brain sections showed the gray matter to white matter ratios of $1.13{\pm}0.10$ at 30 min after intravenous injection, which was lower than $1.94{\pm}0.19$ of commercial $^{99m}Tc$-HMPAO. With the above findings, we concluded that the lipophilic $^{99m}Tc$-PRODD complex was able to cross the blood-brain barrier, however the complex showed lower uptake than $^{99m}Tc$-HMPAO in mouse or rat brains. We could suggest possibility that PRODD could be used as another $^{99m}Tc$ chelator.
