The use of microalgal biomass as feedstock for biofuels has been discussed for decades as it provides a sustainable approach to producing fuels for the future. Nonetheless, its feasibility has not been established yet and various aspects of biomass applications such as CO2 biofixation should also be explored. Therefore, in this study, the CO2 biofixation and lipid/carbohydrate production potential of Chlorella sp. ABC-001 were examined under various nitrogen concentrations. The highest biomass productivity and CO2 biofixation rate of 0.422 g/l/d and 0.683 g/l/d, respectively, were achieved under a nitrogen-rich condition (15 mM nitrate). Carbohydrate content was generally proportional to initial nitrate concentration and showed the highest value of 41.5% with 15 mM. However, lipid content showed an inverse relationship with nitrogen supplementation and showed the highest value of 47.4% with 2.5 mM. In consideration as feedstock for biofuels (bioethanol, biodiesel, and biogas), the sum of carbohydrate and lipid contents were examined and the highest value of 79.6% was achieved under low nitrogen condition (2.5 mM). For lipid-based biofuel production, low nitrogen supplementation should be pursued. However, considering the lower feasibility of biodiesel, pursuing CO2 biofixation and the production of carbohydrate-based fuels under nitrogen-rich condition might be more rational. Thus, nitrogen status as a cultivation strategy must be optimized according to the objective, and this was confirmed with the promising alga Chlorella sp. ABC-001.
Although most algae cultivation systems are operated in suspended culture, an attached growth system can offer several advantages over suspended systems. Algal cultivation becomes light-limited as the microalgal concentration increases in the suspended system; on the other hand, sunlight penetrates deeper and stronger in attached systems owing to the more transparent water. Such higher availability of sunlight makes it possible to operate a raceway pond deeper than usual, resulting in a higher areal productivity. The attached system achieved 2.8-times higher biomass productivity and total lipid productivity of $9.1g\;m^{-2}day^{-1}$ and $1.9g\;m^{-2}day^{-1}$, respectively, than the suspended system. Biomass productivity can be further increased by optimization of the culture conditions. Moreover, algal biomass harvesting and dewatering were made simpler and cheaper in attached systems, because mesh-type substrates with attached microalgae were easily removed from the culture and the remaining treated wastewater could be discharged directly. When the algal biomass was dewatered using natural sunlight, the palmitic acid (C16:0) content increased by 16% compared with the freeze-drying method. There was no great difference in other fatty acid composition. Therefore, the attached system for algal cultivation is a promising cultivation system for mass biodiesel production.
해양 미세조류 Dunaliella tertiolecta에서 바이오디젤 원료인 지질생산을 위하여 철 함량 변화 및 빛 공급과 $CO_2$ 공급에 의한 다중스트레스 인자의 조합이 세포성장 및 지질함량의 변화에 미치는 영향을 조사하였다. 1차 스트레스 인자로 정상보다 높거나 부족한 철 함량 조건이 지질 합성을 유도할 수 있음을 확인하였다. 2차 스트레스 인자로 빛 또는$CO_2$ 공급이 제한될 때 지질함량이 증가하였지만 오랜 시간 배양할 때 세포성장이 감소하는 단점이 있었다. 이와 같이 스트레스 조건에서 세포의 성장과 지질생산이 서로 다른 경향을 보이면 단일 배양기에서 지질생산성을 높이기 어려우므로, 세포성장과 지질생산을 분리한 2단계 배양 전략을 적용하였다. 1단계 배양에서는 성장 위주의 조건으로 고농도배양을 얻은 후, 2단계에서 지질생산을 유도하는 스트레스 조건을 부여하는 것이다. 암소 조건이 다른 조건에 비해 세포농도 감소폭이 작고 지질함량이 높아졌기 때문에, 세포 2 g/L의 고농도로 접종한 2단계에서 5X 철 농도(3.25 mg/L as Fe) 및 암소 조건을 사용하여 12 h의 짧은 배양을 통하여 1.44 g/L/d의 높은 지질생산성을 얻을 수 있었다.
단세포유지 생산에 이용되는 미생물은 효모, 곰팡이, 미세조류 등이다. 단세포유지의 생산 효율은 배지의 조성을 조절하여 영양공급을 최적화하거나, 배양 조건을 조절하거나, 최신의 생명공학기술을 활용하여 균주를 개량함으로써 증가시킬 수 있다. 단세포유지를 상업적으로 대량 생산하기 위해서는 값이 싼 탄소원을 확보하는 것이 무엇보다 중요하므로 지구 상에 풍부한 섬유질 자원을 활용하는 방법이 주목을 끌고 있다. 미세조류는 대기 중의 탄산가스를 탄소원으로 이용하므로 탄소원의 비용이 들지 않는 는 장점이 있다. 단세포유지 생산 미생물의 유전자를 조작하거나 대사공학기술을 이용하는 방법은 균주의 생산성을 획기적으로 높이기도 한다. 단세포유지는 그 동안 식품, 사료, 의약품 등으로 사용되어 왔으나 최근에는 바이오디젤(biodiesel)의 원료로도 사용될 수 있어 또 다른 관심을 끌고 있다.
Objective: This experiment investigated the effects of dietary supplementation with conjugated linoleic acid (CLA) on the serum components, laying hen productivity, lipid composition of egg yolk, egg flavor and egg quality. Methods: Healthy 28-week-old Hy-Line white laying hens (n = 480) were divided randomly into 4 groups, 6 replicates/group, 20 birds/replicate. The 30-day experimental diets included 0% (control), 0.4%, 0.8%, and 1.6% CLA. Some serum indices of the birds, and egg production, quality, fatty acid composition, egg quality were measured. Results: The dietary supplementation with 0.4%, 0.8%, and 1.6% CLA did not significantly affect the laying rate and feed intake, as well as calcium ion and phosphorus ion concentration in serum (p>0.05). However, the CLA had significantly increased the strength of eggshell, decreased the odor, flavor, and taste of egg yolk, deepened the color of egg yolk, increased saturated fatty acids and polyunsaturated fatty acids, and reduced the monounsaturated fatty acids (p<0.05). On the other hand, the dietary supplementation with 1.6% CLA had significant effects on feed/gain, and improved serum hormones. Dietary supplementation with 0.4% and 0.8% CLA can significantly enhance the activity of alkaline phosphates. Conclusion: CLA has no effect on production performance, but does enhance the lipid content of the egg yolk and the strength of the eggshell.
미세조류는 통상적인 에너지 작물에 비하여 빠른 성장속도와 높은 오일함량으로 바이오디젤 생산의 원료로 관심을 받고 있다. 미세조류의 지질은 주로 트리글리세라이드인 중성지방으로 에스테르교환반응을 통하여 바이오디젤인 지방산 메틸에스테르로 전환할 수 있다. 본 논문에서는 영양분의 제한, 염도 및 금속 성분의 변화와 같이 미세조류의 지질 생산을 촉진할 수 있는 배양환경 스트레스 조건의 영향들을 비교 고찰하였다. 사용하는 미세조류 종에 따라 스트레스에 대응하여 지질의 양이 변하거나 구성하는 지방산의 조성이 변화될 수 있다. 비록 질소원 제한 조건이 가장 많이 사용되는 지질생산 촉진조건이긴 하지만, 미세조류로부터 바이오디젤 생산성의 향상을 위해서는 그 외에도 영양분 과잉 조건, 염도의 변화, pH, 온도, 금속 성분 농도 변화 등의 다른 조건들도 고려되어야 한다.

Chlorella spp. are green algae that are found across wide-ranging habitats from deserts to arctic regions, with various strains having adapted to survive under diverse environmental conditions. In this study, two novel Chlorella strains (ABC-002, ABC-008) were isolated from a freshwater lake in South Korea during the winter season and examined for possible use in the biofuel production process. The comparison of ABC-002 and ABC-008 strains with Chlorella vulgaris UTEX265 under two different temperatures ($10^{\circ}C$, $25^{\circ}C$) revealed their cold-tolerant phenotypes as well as high biomass yields. The maximum quantum yields of UTEX25, ABC-002, and ABC-008 at $10^{\circ}C$ were 0.5594, 0.6747, and 0.7150, respectively, providing evidence of the relatively higher cold-resistance capabilities of these two strains. Furthermore, both the biomass yields and lipid content of the two novel strains were found to be higher than those of UTEX265; the overall lipid productivities of ABC-002 and ABC-008 were 1.7 ~ 2.8 fold and 1.6 ~ 4.2 fold higher compared to that of UTEX265, respectively. Thus, the high biomass and lipid productivity over a wide range of temperatures indicate that C. vulgaris ABC-002 and ABC-008 are promising candidates for applications in biofuel productions via outdoor biomass cultivation.
Mortierella isabellina IFO 8183을 이용하여 ${\gamma}-linolenic$ acid를 생산하기 위한 최적 배양조건을 검토한 결과, 최적 탄소원과 최적 질소원은 각각 glucose와 $NH_4NO_3$이었으며, 지질 생산배지내의 최적 C/N율은 40이었다. 또한, 최적 초기 pH와 배양온도는 각각 6.0과 $30^{\circ}C$이었으며, 균체내에 축적된 지질의 지방산 조성은 pH와 온도에 의해 변화되었다. Sodium acetate를 배양 초기에 0.5%(w/v) 첨가하였을 때 대조구에 비해 ${\gamma}-linolenic$ acid 함량과 ${\gamma}-linolenic$ acid 생성량이 각각 15.4%와 37.8% 증가되었다. 최적 조건하에서 8일간 진탕배양하였을 때 균체수율, 지질수율, 지질함량, ${\gamma}-linolenic$ 함량 및 ${\gamma}-linolenic$생성량은 각각 0.347g/g, 0.18g/g, 0.52g/g, 60mg/g 및 347mg/l이었다.
바이오디젤은 신재생연료이면서 환경 친화적인 수송용 액상 바이오연료이다. 곤충은 새로운 바이오디젤 원료로 여겨지고 있다. 특히, 동애등에는 높은 지질을 함유하고 있어 재생가능한 바이오디젤 원료이다. 동애등에 유래 바이오디젤은 포화지방산 함량이 높고 다불포화지방산 함량이 낮아 품질이 좋은 바이오디젤을 만들 수 있다. 동애등에 유래 바이오디젤은 EN 14214의 대부분 품질기준을 만족한다. 동애등에는 기존의 식물성 원료, 미세조류에 비해 지질 수율이 높아 바이오디젤 생산성이 높다. 본 논문에서는 곤충 유래 바이오디젤의 전반적인 생산 방법과 품질 특성에 대해 서술하였다.
