The methanol extracts of some marine algae were tested for investigating the effects on the formation of lipid peroxide and the activities of free radical generating enzyme in vitro in bromobenzene-treated rat. The extracts of Enteromorpha compressa, Capsosiphon fulvescens, Gelidium amansii, Hizikia fusiformis, Sargassum siliquastrum and Sargassum thunbergii which decreased the formation of lipid peroxide, inhibited the activity of xanthine and aldehyde oxidases by adding of each extracts. Phloroglucinol isolated from Ecklonia stolonifera reduced bromobenzene-induced hepatic lipid peroxidation. This compound administered daily over one week before intoxication with bromobenzene did not affect the activities of aminopyrine N-demethylase, aniline hydroxylase and glu tathione S-transferase. Epoxide hydrolase activity was decreased by bromobenzene, which was restored by pretreatment of phloroglucinol, The results suggest that the bromobenzene-induced hepatic lipid peroxidation by phloroglucinol is reduced by enhancing the activity of epoxide hydrolase.
Inhibitory effects of the methanol extract, hexane extract, methanol soluble fraction (MSF) and juice from 3 weeks fermented Kimchi on the tumor formation in sarcoma-180 cell transplanted mice were studied. Effects of the solvent extracts and juice of the Kimchi on the levels of lipid peroxide, glutathione, and the enzyme activities of the liver were also investigated in normal and sarcoma-180 cell transplanted mice. At 32 days following trans-plantation, MSF reduced the tumor formation by 54% compared with the control group, resulting in the smallest tumor weight. Lipid peroxided content in liver increased by the transplantation of sarcoma-180 cells. However, it decreased when MSF of Kimchi was treated to the mice. MSF also suppressed xanthine oxidase activity in cytosol of the liver cells in mice transplanted by sarcoma-180 cells. Kimchi extracts had no inhibitory effect on hepatic aminopyrine-N-demethylase activity in sarcoma-180 cell transplanted or normal mice. Methanol extract and hexane extract of Kimchi slightly increased hepatic glutathione contents in sarcoma-180 treated mice. The injection of MSF from Kimchi markedly increased glutathione levels in the liver of sarcoma-180 treated mice. The injection of MSF from Kimchi markedly increased glutathione levels in the liver of sarcoma-180 treated mice compared to the controls. The MSF recovered the activities of hepatic glutathione reductase and glutathione S-transferase that decreased by the injection of sarcoma-180 cells. These results showed that MSF of Kimchi could suppress the growth of tumors, inhibiting lipid peroxide production and xanthine oxidase activity, in mice. We also suggested that Kimchi extract might play an important role in the prevention of cancer by enhancement of the glutathione level itself as well as via glutathione reductase and glutathione S-transferase.
This study was performed to investigate the effets of hesperidin extracted from tangerine peel on Cadmium (Cd) and lipid metabolism lipid peroxide formation, and antioxidative enzyme activities in rats. Forty-eight male Sprague-Dawley rats weighing 158.3$\pm$3.5g were blocked into eight groups according to body weight. Rats were raised for three weeks with diets containing 0 or 0.04%(w/w) cadmium chloride and 1%(w/w) extracted hesperidin from tangerine peel, commercial hesperidin or naringin. Food intake, weight gain and food efficiency ratio were significantly lower in the Cd-administered groups. The Cd concentrations in blood and liver and the Cd excretions in urine and feces were significantly higher in the Cd-administered groups. Among the Cd groups, blood Cd concentrations were decreased, fecal Cd excretions were increased, and Cd retenition ratios were decreased by feeding flavonoid diets. Plasma total lipid concentrations were significantly lower in the extracted hesperidin group, plasma triglyceride concentrations were significantly lower in the extracted hesperidin and naringin groups. Plasma HDL-cholesterol concentrations and HDL : total cholesterol ratios were increased by feeding flavonoids. Among the Cd groups, liver total lipid concentratons were decreased by feeding flavonoids. Fecal total lipid, fecal cholesterol, and fecal triglyceride excretions were significantly higher in the naringin group, and they were increased by feeding flavonoids among Cd groups. Thiobarbituric acid reactive substance concentrations in plasma and liver were higher in Cd groups, and were significantly decreased by feeding flavonoids. The activities of erythrocyte catalase, superoxide dismutase and glutathione peroxidase showed a tendency to increase by feeding. The activities of liver catalase, superoxide dismutase and glutathione peroxidase were not significantly affected by administering Cd or flavonoids. In conclusion, all flavonoids that were used in this experiment inhibited lipid peroxide formation in plasma and liver, but this effect was not caused by the increased in the activities of antioxidative enzymes.

The ethanol extract of peony root (Paeonia Lactiflora Pall, Paeoniaceae) and its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. And protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were performed. The ethanol extract of peony root (PRE), gallic acid and methyl gallate were shown to possess the significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and were revealed the inhibitory effect of lipid peroxidation as expressed by malondialdehyde (MDA) formation. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, assessed by single cell gel electrophoresis. Furthermore, oral administration of 50% PRE (50% ethanol extract), gallic acid and methyl gallate potently inhibited micronucleated reticulocyte (MNRET) formation of mouse peripheral blood induced by KBrO3 treatment in vivo. Therefore, PRE containing gallic acid and methyl gallate may be a useful natural antioxidant by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage.

벼메뚜기 건제품의 가공저장중 품질의 열화에 가장 큰 영향을 미친 것으로 예상되는 지질의 조성을 분석하고 저장중 생성된 갈변반응물질이 지질산화에 미치는 영향을 검토하였다. 성숙한 벼메뚜기 암컷의 지질함량은 5.12%로서 인지질함량(27.35%)이 높았고 불포화지방산 함량이 68.18%나 되었다. 벼메뚜기 천일 및 열풍건조 과정중 COV는 크게 증가하였으나 POV는 거의 측정되지 않았다. 동결건조 과정중에는 과산화물과 카아보닐 화합물이 거의 생성되지 않았다. 저장중에 POV는 초기부터 급속히 증가하였으나 COV는 저장 98일까지 지속적인 증가를 보였고 저장 초기에 당-아미노반응에 의한 수용성갈변반응물질이 많이 생성됨과 동시에 환원당과 아미노질소 함량은 크게 감소하였다. 벼메뚜기 저장중 생성된 갈변반응물질이 지질화에 미치는 영향을 조사한 결과 수용성 구분에서 지질산화의 억제효과가 다소 있는 것처럼 보였으나 지용성 구분에서는 산화촉진 효과를 나타내었다.
Objectives : The purpose of this study is to examine the antioxidant effects on male mouse reproductive cells of the extract of Platycladi Semen. Methods : The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, cell viability by a modified MTT assay, the effects on $H_2O_2$-induced cytotoxicity by MTT assay, lipid peroxidation by malondialdehyde (MDA) formation and super oxide dismutase (SOD), respectively. Results : The results showed that the extract scavenged DPPH radical in a dose-dependent manner by up to 74.87%. The cell viability of the extract was within 72~96% on Leydig cells and GC-2 cells at concentrations of 1, 5, 10, 50 and 100 ug/ml. The hydrogen peroxide-induced cytotoxicity of Leydig cells was protected to 72.09% by the extract at concentration of 100 ${\mu}g/ml$. The hydrogen peroxide-induced lipid peroxidation of MDA formation was decreased to 1.80 and 1.65 nmoles/mg protein by the extract at concentrations of 50 and 100 ${\mu}g/ml$. The extract at all concentrations, SOD activity was not significantly changed. Conclusions : In conclusion, the extract of Platycladi Semen has antioxidant effects on Leydig cells and protect male reproductive system against oxidative stress.
Objectives : Previously we reported that the roots of Panax ginseng C.A. Meyer (Araliaceae) increased sperm count and motility. also induced spermatogenesis via cAMP-responsive element modulator(CREM) activation in rat testes. In this study, for the first step of spermatogenesis in germ cell lines, the antioxidant activity of Panax ginseng were examined in mouse GC-1 spermatogonia cells. Methods : The extract was studied on diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, GC-1 cell viability by a modified MIT assay. H202-induced cytotoxicity by MIT assay and lipid peroxidation by malondialdehyde (MDA) formation. respectively. Results: The results showed that the extract scavenged DPPH radical with the IC50 being 0.631 mg/mi. The extract at concentrations of 5, and 10, 50, 100, 250 ${\mu}$g/mi increased GC-1 cell viability significantly(p < 0.05, and p < O.O1). Hydrogen peroxide-induced cytotoxicity (73.8%, p < O.O1) was blocked by the extract at concentrations of 50, and 100, 250, 500 ${\mu}$g/ml significantly (p < 0.05, and p < O.O1). The extract at concentrations of 10. and 50 ${\mu}$g/ml decreased the MDA formation on hydrogen peroxide-induced lipid peroxidation. Conclusions : In conclusion, the extract of Panax ginseng has potent antioxidant activity and increases the survival rate of GC-1 spg cells against $H_20_2$-induced cytotoxicity.
The intradermal injection of Propionibacterium acnes (ATCC 6919) into the ears of female Sprague-Dawley rats produced a chronic inflammation with the formation of acneiform lesions. Inflammation was characterized by more than four times of ear thickness and 2.8 times of ear weight at day 21. Histologically, massive infiltration of neutrophils, macrophage and lymphocytes, hyperplastic epidermis, comedones containing keratin mass and inflammatory materials were observed. Both ginseng saponin and extract from Korean red ginseng significantly redliced the ear thickness and their effects were similar to that of benzoyl peroxide. Ginseng samples and beneoyl peroxide modified lipid constituents of P. acnesinjected rat ear tissues. Even though no marked histological changes in inflammatory lesions were observed in ginseng-treated ear tissues, Korean red ginseng showed a possibility of reduce in the risk of acne development.
This study was conducted to investigate the antioxidant activity of paprika in the lard-pork model system adding ground fresh paprika (3%) and paprika powders (5%). Paprika powders were obtained through 4 drying methods (freeze, vacuum, far infrared-ray, and hot-air). In the lard and meat-fat mixture (containing lard 30%) containing paprika powders, the rate of increase in the peroxide value (POV) and thiobarbituric acid (TBA) value decreased notably during the refrigerated storage ($4^{\circ}C$) compared to the control without paprika. Therefore, paprika powders showed potent antioxidant activity and especially the freeze dried paprika powder revealed the most effective activity among them. However, its antioxidant activity was still lower than that of the fresh paprika because the addition of fresh paprika in the lard and meat-fat mixture merely increased the POV and TBA value. In linoleic acid oxidation, the addition of capsanthin 500 ppm to mixed linoleic acid and 10 ppm of $FeCl_3$ (LF) inhibited the formation of peroxides by 15.2% compared to LF, showing its iron scavenging ability. When mixed antioxidants (${\beta}$-carotene 200 ppm + ascorbic acid 100 ppm, capsanthin 200 ppm + ascorbic acid 100 ppm) were added in LF, synergistic effects were obtained with 57.7 and 60.4% of inhibition of peroxide formation, respectively.
