Effects of Drying Conditions on Lipid Oxidation and Patty acid Compositions of Large Anchovy (대멸치의 지질산화 및 지방산 조성의 변화에 미치는 건조조건의 영향)

  • To develop plain dried products of large anchovy, Engraulis japonica, lipid oxidation during drying of large anchovy and optimal drying condition were investigated. The moisture contents of large anchovy on 7 hrs dryness were $ 9.0{\%},\;34.0{\%},\;38.0{\%} and\;38.8{\%}\;in\;60^{\circ}C$ hot-air drying (wind velocity, 1.4m/sec), $40^{\circ}C$ hot-air drying (wind velocity, 1.4 m/sec), sun drying($30{\pm}5^{\circ}C$) and $20^{\circ}C$ cold-air drying (wind velocity, 3.1 m/sec), respectively. The cold-air drying depressed remarkably the lipid oxidation of large anchovy during drying, resulting from AV, POV, COV and the formation of browning pigment. The fatty acid composition of large anchovy was $35.8{\%} in saturates, 20.0{\%} in monoenes and 44.2{\%}$ in polyenes. Saturates and monoenes were increased in proportion to the increase of drying time, while polyenes were decreased. The contents of 20 : 5 and 22 : 6 of polyenes were decreased remarkably in proportion to the progress of lipid oxidation, while 14 : 0, 16 : 0, 16 : 1 and 18 : 1 of saturates and monoenes were increased. The changes in fatty acid compositions by drying conditions were remarkably clarified in sun drying, followed by $60^{\circ}C$ hot-air drying $40^{\circ}C$ hot-air drying and $20^{\circ}C$ cold-air drying in order.

Combination Effect of Modified Atmosphere Packaging and Electron Beam Irradiation on the Oxidative and Microbiological Stability of Ground Pork during Storage (공기 조절 포장과 전자선 조사의 병용이 분쇄돈육의 저장 중 산화와 미생물적 안정성에 미치는 영향)

  • Ground pork was packaged(purged) with modified atmosphere (n$_2$ and CO$_2$) and irradiated with the electron beam in order to find out whether modified atmosphere packaging (MAP) inhibit the microbial growth and lipid oxidation development caused by electron beam irradiation. After packaging and irradiation, ground pork was stored at 4$^{\circ}C$ for 6 days and -15$^{\circ}C$ for 3 months, and periodically the microbial counts and the thiobarbituric acid reactive substances (TBARS) for the determination of lipid oxidation were measured. The inhibition of growth of total aerobic bacteria and mesophiles was confirmed when the ground pork was irradiated with the electron beam dose of 1.5 and 3.0 kGy. The N$_2$ or CO$_2$ purging alone was also effective in reducing the development of lipid oxidation of ground pork during storage at 4 and -15$^{\circ}C$. The combination of electron beam irradiation(1.5 and 3.0 kGy) with MAP (N$_2$ or CO$_2$) was effective to inhibit the growth of total aerobic bacteria and mesophiles, and retard the lipid oxidation of ground pork during storage at 4$^{\circ}C$ for 6 days and -15$^{\circ}C$ for 3 months.

Effects of Natural Antioxidants on Lipid Oxidation of Ground Pork (천연항산화제가 분쇄돈육의 지질산화에 미치는 효과)

  • This study was conducted to investigate the effects of antioxidants on lipid oxidation in uncooked ground pork which was treated with ${\alpha}-tocopherol$, GFSE (grapefruit seed extract), carnosine and rosemary, respectively. The ground pork samples were uncooked and cooked during 10 days of storage at $4{\pm}1^{\circ}C$, respectively. The lipid oxidation and fatty acids composition were analyzed for over a periods of storage day. From the results mentioned above, antioxidative activity on lipid oxidation of uncooked ground pork appeared to be in order of carnosine > rosemary > ${\alpha}-tocopherol$ > GFSE. It would not be problem that addition of carnosine in uncooked ground of resulted in an increase of pH because the high pH could be advantageous on processing of meat. Antioxidative activity on lipid oxidation of cooked ground pork appeared to be in order of carnosine > rosemary > ${\alpha}-tocopherol$ > GFSE. Although the antioxidants were not lost their antioxidative }ctivities after cooking, their antioxidative activities in cooked ground pork were not higher than that of uncooked ground pork. That addition of carnosine increased the pH of cooked ground pork, too.

Effect of Addition of Phosvitin and High Pressure Processing on Microbiological Quality and Lipid and Protein Oxidation of Minced Chicken Leg Meat (닭 다리 분쇄육에 초고압 처리시 Phosvitin의 첨가가 미생물학적 품질과 지방 및 단백질 산화에 미치는 효과)

  • The objective of this study was to investigate the effect of high pressure (HP) processing on shelf life, as well as the addition of phosvitin on lipid and protein oxidation stability of minced chicken leg meat. Minced chicken leg meat was mixed with yolk phosvitin at 500 or 1000 mg/kg meat levels, and divided into raw and cooked groups. Then, the samples were subjected to HP at 0.1, 300, and 600 MPa. The total aerobic bacteria, lipid and protein oxidation, along with instrumental meat color ($L^*$, $a^*$, and $b^*$value) of the samples were measured during storage for 7 d at $4^{\circ}C$. In raw meat, the number of total aerobic bacteria was decreased by HP at 300 MPa (4 Log reductions) and 600 MPa (5 Log reductions) after 7 d of storage (p<0.05). HP at 600 MPa increased lipid oxidation of samples at all storage days and protein oxidation of samples during storage at 3 and 7 d. HP induced the changes of meat color by increase of $L^*$ value and decrease of $a^*$ value (p<0.05). The total aerobic bacteria was not detected in the cooked samples, regardless of HP pressure, and the lipid or protein oxidation of the cooked sample treated by 600 MPa was higher than that of the control (0.1 MPa) on day 7 or control on day 3, respectively (p<0.05). The results suggested that HP can improve the shelf life of minced chicken leg meat. However, phosvitin might be a limited antioxidative agent for the improvement of oxidation stability induced by HP.

Biological Activities of Ethanol Extracts and Fractions of Black Olympia Grape(Vitis Labruscana L.) (거봉 포도종의 에탄올 추출물 및 분획물에 대한 생리활성 효능)

  • This study was conducted to determine biological activities, such as lipid peroxidation inhibition and cytotoxic effect of ethanol extracts of Black Olympia grape seeds and skins, and of organic solvent fractionated ethanol extracts obtained from grape seeds and skins at different temperatures. Among different extraction temperatures, the ethanol extract of grape seed obtained at 30$\^{C}$ had the strongest lipid oxidation inhibition of 60.1%, while the strongest lipid oxidation inhibitory effect of 71.2% was observed in the presence of 20 $\mu\textrm{g}$/㎖ ethylacetate fraction obtained from ethanol extract of grape seeds at 30$\^{C}$. The ethanol extract of grape seeds showed more strong lipid oxidation inhibition than that of skin extracts. Similar results were observed in cytotoxic effects. The ethanol extract of grape seeds at 30$\^{C}$ exhibited more strong cytotoxicity than that of skin extracts on MCF-7, Hep3B, and A549 cell lines. Among organic solvent fractions extracted from the ethanol extracts of gape seeds and skins, the hexane fraction showed the strongest cytotoxic inhibition of 75.15% and 62.50% on MCF-7 and Hep3B cell in the presence of 1.0 $\mu\textrm{g}$/㎖ respectively. On the other hand, the water fraction showed the strongest cytotoxic inhibition of 65.41% on A549 cell in the presence of 1.0 $\mu\textrm{g}$/㎖. Overall, the ethanol extracts and their fractions of Black Olympia grape seeds showed strong lipid oxidation inhibition and cytotoxicity than those of grape skins.

A Study on Lipids Oxidation Boiled Whale Meat’s in Process of Circulation Market (유통되고 있는 삶은 고래고기의 안정성 연구)

  • The purpose of the study was to analyze in process of circulation market boiled whale meat's rancidity. oxidative rancidity is oil or fat food depend on oxygen in air oxidative change in quality. boiled whale meat faty come to oxidative rancidity food stability and hygiene reasons for people health poisonous point out, however it is not indication study of support. Accordingly confirm lead into circulation process boiled whale meat's rancidity examination and microorganism examination boiled whale meat's stability whether or not. The result obtained were summarized as follows: 1. Proximate percentage of boiled whale meat(pectoral, pelvic, fin, flank) of moisture and crude lipid and crude protein from samples shown to be : moisture was pectoral 16.4%, pelvic 36.2%, fin 46.2%, flank 19.2%, crude lipid was pectoral 54.1%, pelvic 42.8%, fin 15.8%, flank 40.6%,crude protein was pectoral 29.4%, pelvic 20.5%, fin 29.5%, flank 28.6%. 2. The fatty acid composition of total lipid were composed of pectoral 27.2%, pelvic 28.9%, fin 33.3%, flank 23.4% of oleic acid and pectoral 12.7%, pelvic 11.1%, fin 11.3%, flank 14.0% of palmitic acid pectoral 10.8%, pelvic 7.9%, fin 7.6%, flank 2.1% of docosahexaenoic acid, pectoral 14.2%, pelvic 7.5%, fin 1.9%, flank 7.2% of eicosenoic acid, pectoral 5.1%, pelvic 5.7%, fin 4.4%, flank 5.7% of myristic acid, 16: 0 11∼14 % of high saturated fatty acid. generally most of 18: 1ω9 of boiled whale meat's each portion, 22:6 7∼12%, 20:5 1∼14% of polyenoic fatty acid. 18:3 showen to be 1% make an expection of pectoral and fin portion the total lipid were most of netural lipid's about 90%, monoenic fatty acid were most of 19∼22% of saturated fatty acid, 77∼80% of monoenic fatty acid level of 47∼56% of 18:1 16:1 was markelly high to those of total lipid. 3. The storage number days variation of oxidation were shown to be by stages process favorably the past days of boiled whale meat's acid value for 5days. pectoral the day 0.1, five days 1.3, pelvic the day 0.1, five days 1.6, fin the day 0.3, five days 0.7, flank the day 0.2, five days 0.4. 4.   • Fatty acid oxidation (FAO) defects cause abnormal lipid accumulation in various tissues, which provides an opportunity to uncover novel genes that are involved in lipid metabolism. During a gene expression study in the riboflavin deficient induced FAO disorder in the chicken, we discovered the dramatic increase in mRNA levels of an uncharacterized gene, ANKRD9. No functions have been ascribed to ANKRD9 and its orthologs, although their sequences are well conserved among vertebrates. To provide insight into the function of ANKRD9, the expression of ANKRD9 mRNA in lipidperturbed paradigms was examined. The hepatic mRNA level of ANKRD9 was repressed by thyroid hormone ($T_3$) and fasting, elevated by re-feeding upon fasting. However, ANKRD9 mRNA level is reduced in response to apoptosis. Transient transfection assay with green fluorescent protein tagged- ANKRD9 showed that this protein is localized within the cytoplasm. These findings point to the possibility that ANKRD9 is involved in intracellular lipid accumulation.

Effect of Eicosapentaenoic Acid on Cellular Lipid Accumulation and Transcription Factors Involving Glucose Utilization (에이코사펜타에노익산에 의한 세포 내에서의 지방 축적 억제 효과 및 포도당 대사에 관여하는 전사인자의 변화)

  • Previous studies suggest that polyunsaturated fatty acids with long carbon chains such as eicosapentaenoic acid(EPA) and docosahexaenoic acid(DHA) have several health benefits. However metabolic consequences of these fatty acids themselves and their regulation of transcriptional activity involving glucose utilization are not well established. Thus, the purpose of this study was to investigate how EPA influx affects cellular lipid accumulation and gene expressions involving $de$ $novo$ lipogenesis in hepatocyte cultures. Compared to oleic acid treatment, EPA treatment showed remarkably decreased cellular TG conversion and accumulation, along with phospholipids at a lower extent. As expected, EPA increased mRNA expression involving fatty acid influx and lipid droplet formation, but did not affect mRNA expression involving glucose utilization. EPA increased transcriptional activity of PPAR-${\alpha}$ and glucose responsive transcription factor when transcription factor binding protein was activated. Taken together, these data suggest that EPA decreases lipid accumulation through increases of the ${\beta}$-oxidation pathway without interruption of glucose utilization.

Effects of Prostaglandin E2 Analogue, Enprostil, on Lipid Metabolism in Mice

  • This study was conducted to investigate the effects of effects of enprostil, a prostaglandin $E_2$, analogue, on liver triacylglycerol content and factors that regulate liver lipid metabolism in mice. Mice received vehicle or $10{\mu}g$ enprostil/kg body weight intraperitoneally every 6 h, and were killed at 0, 6, 12, 18 and 24 h after the first injection. Enprostil significantly lowered liver triacylglycerol content after 12 h of the first injection. However, the peroxisomal ${\beta}$-oxidation activity was inconsistent with the result of liver triacylglycerol content, because its activity was lovered by enprosil. In another experiment, the effect of enprostil on lipid metabolism in mice was investigated in a short period. Mice received $10{\mu}g$ enprostil/kg body weight intraperitoneally, and were killed after 0, 5, 10, 30 and 60 min. After 30 min, malic enzyme activity was significantly increased by the administration of enprostil compared with the activity at 5 min after. No significant changes in liver carnitine palmitoyltransferase and peroxisomal ${\beta}$-oxidation activities were observed. Plasma free fatty acid concentrations were markedly reduced from 5 through 60 min after the administration of enprostil. Consequently, enprostil suppressive effect on liver triacylglycerol concentration might result from the decreased entry of free fatty acid into liver.

Effect of Different Sources of Carbohydrate and N-3 Fatty Acid on Lipid Metabolism in Hypertriglyceridemic Rats (탄수화물과 N-3 지방산급원의 차이가 혈중중성지방함량이 높은 흰쥐의 지질대사에 미치는 영향)

  • This study intended to compare the hypolipidemic effects among six experimental groups fed by three different dietary carbohydrates on hyperlipodemic rats. Sixty experimental animals were divided into 6 groups, SB, ST, SP and CB, CT, CP after production of hyperlipidemia fed by SB diet on Sprague-Dawley rats for 4 weeks. Rats were fed by six experimental diets for eight weeks. Hyjperlipidemic rats showed three times higher in plasma TG level not in cholesterol content compare to control group fed by stock diet. Two different dietary carbohydrates seem to be effective in body weight gain and fat cumulation as weight of epididymal fat pad. In comparison of S and C groups, C fed groups showed lowering effect in plasma TG and total lipid contents, but among S fed groups, ST and SP group showed lower than SB in this respects. Dietary carbohydrates seem to be more effective than fat in plasma lipid contents. When we compare among three different fat groups, only T groups with different carbohydrates increased in peroxisomal lipid oxidation and decreased in lipogenic enzyme activites. As same token, sucrose fed group with three fat sources seem to increase activiteies enzyme activities. In epididymal fat pad and Heart, SP and CP effect more in LPL activites than other groups. In conclusion, we can recommed to consume polysaccharides rather than disaccharide and n-3 fatty acids such as perilla and tuna oils to alleviate hypertringlycemic condition.
