  • Lipid oxidation and browning during fermentation of domestic Meju and Doenjang were examined in order to elucidate the antioxidative effects of browning products and phenol compounds from Meju and Doenjang. Peroxide values of lipids from Meju were detectable and slightly increased until 3 weeks of fermentation, but started to be decreased after 3 weeks of fermentation and notdetectable after 6 weeks. Peroxides were not detected in Doenjang during the whole fermentation, but started to be decreased after 3 weeks of period of 22 weeks fermentation. Carbonyl value were increased during the whole period of Meju fermentation, but started to be decreased at the early stage of Doenjang fermentation. Hydrophilic fraction of browning products from Meju was much higher than lipophilic fraction and the former fraction was dramatically increased at the early stage of the fermentation. But the both fractions maintained high values during Doenjang fermentation. Hydrophilic browning products and phenol and phenol compound in Meju showed strong antioxidative against linoleic acid.

  • Daenjang, a traditional Korean fermented soybean paste, was prepared by the fermentation with Aspergillus oryzae for 3 days, and then ripened with salt addition(13%) for 45 days. The changes of composition and fraction pattern of lipid in Daenjang during ripening period were studied. The total lipid of initial Daenjang(0 day ripening) consisted of 91.8% of neutral lipid, 7.1% of phospholipid and 1.1% of glycolipid, and minor changes of the composition were observed during the ripening. The contents of free fatty acid and esterified sterol in nonpolar lipid fraction were increased, however, no significant changes in polar lipid fraction were observed from the Daenjang in ripening. Linoleic, oleic, palmitic acids were the major fatty acyl moiety in the fractions of total lipid, triglyceride and free fatty acid, respectively. No significant changes in fatty acid compositions of the fractions were noted, although the saturated fatty acyl moiety of the fraction were increased a little during Daenjang ripening.

NON-ENZYMATIC BROWNING REACTIONS IN DRIED ANCHOVY WHEN STORED AT DIFFERENT WATER ACTIVITIES (마른멸치 저장중의 수분활성과 비효소적 갈변반응)

  • In this paper, non-enzymatic browning reactions as a factor of self stability of boiled and dried anchovy were studied to discuss the effect of water activity to the discoloring reaction and the preservative moisture content. The development of rancidity of the fish meat was also mentioned since the fish is fatty and the lipid oxidation is a functional deteriorative reaction. Fresh anchovies were boiled in $10\%$ salt solution immediately after the catch, sun dried, and stored at room temperature ($20^{\circ}C$) for two months in humidistat chambers maintaining different levels of water activity as described in Table 1. The pigments formed by non-enzymatic browning reations were extracted in two fractions, those were chloroform-methanol soluble and water dialyzed fraction, and analyzed spectrophotometrically at the wavelength of 460 nm. These two fractions were considered, respectively to be the brown pigments formed by lipid oxidation reactions for the formler and for the latter, to be the pigments developed by sugar-amino or Maillard reaction. The oxidation of lipid in anchovy meat during the storage was measured as the changes in Peroxide value and the color development of thiobarbituric acid reaction. It is summarized from the results that the rate of both reactions, lipid oxidation and browning, was affected by water activity levels. In regard to the changes in peroxide and TBA value during the storage, the propagation of lipid oxidation was obviously accelerated at lower humidities whereas the development of browning progressed at the higher. These two reactions occurring simultaneously and contrary in activity resulted in that the rate of deterioration occurring oxidatively or by browning, was the minimum at the water activity of 0.32-0.45 which were $7-9\%$ as moisture content and slightly higher value than that of monolayer (Aw=0.21, $5.11\%$ as moisture content). It is also noted that the lipid oxidative browning was presumed to dominate sugar-amino reactions so that the rate of browning of the meat was ultimately depended on the development of rancidity although sugar-amino reactions initiated earlier than the other at the first ten days of storage, particulary at higher humidity. At the lower humidity sugar-amino reactions were occurred gradually but lower levels in color development in contrast to the consistent increase in lipid oxidative browning.

Fractionation and Quantitative Analysis of Lipid Components in Korean Yam (Dioscorea) Tubers (한국산 마의 지질 성분의 분리 및 분석)

  • Using the lipids extracted from Korean yam(Dioscorea) tubers, D. batatas, D. aimadoimo and D. japonica, fractionation and identification of lipid components and their fatty acid compositions were analysed. Lipid contents determined by Folch's method in D. batatas, D. aimadoimo and D. japonica were 11.0 mg/g, 11.4 mg/g and 6.6 mg/g, respectively. Lipids extracted were fractionated into neutral lipid, glycolipid and phospholipid by silicic acid column chromatography. The content of neutral lipid was over about 60% in lipid. Neutral lipid was composed of sterol ester, triglyceride, 1,3-diglyceride, 1,2-diglyceride and monoglyceride. Main constituents of glycolipid were acylsterylglycoside, monogalactosyldiglyceride, sterylglycoside, digalactosyldiglyceride and sulfolipid, and phospholipid contained phosphatidylethanolamine, phosphatidylcholine and phosphatidylinositol. The fatty acids of the total lipid and its three lipid fractions were analyzed by GC. The major fatty acids were palmitic and linoleic acids. Content of the saturated fatty acids was less than that of the unsaturated fatty acids.

Red pepper seed water extract inhibits preadipocyte differentiation and induces mature adipocyte apoptosis in 3T3-L1 cells

  • BACKGROUND/OBJECTIVES: Reducing the number of adipocytes by inducing apoptosis of mature adipocytes as well as suppressing differentiation of preadipocytes plays an important role in preventing obesity. This study examines the anti-adipogenic and pro-apoptotic effect of red pepper seed water extract (RPS) prepared at $4^{\circ}C$ (RPS4) in 3T3-L1 cells. MATERIALS/METHODS: Effect of RPS4 or its fractions on lipid accumulation was determined in 3T3-L1 cells using oil red O (ORO) staining. The expressions of AMP-activated protein kinase (AMPK) and adipogenic associated proteins [peroxisome proliferator-activated receptor-${\gamma}$ ($PPAR-{\gamma}$), CCAAT/enhancer-binding proteins ${\alpha}$ (C/EBP ${\alpha}$), sterol regulatory element binding protein-1c (SREBP-1c), fatty acid synthase (FAS), and acetyl-CoA carboxylase (ACC)] were measured in 3T3-L1 cells treated with RPS4. Apoptosis and the expression of Akt and Bcl-2 family proteins [B-cell lymphoma 2 (Bcl-2), Bcl-2-associated death promoter (Bad), Bcl-2 like protein 4 (Bax), Bal-2 homologous antagonist/killer (Bak)] were measured in mature 3T3-L1 cells treated with RPS4. RESULTS: Treatment of RPS4 ($0-75{\mu}g/mL$) or its fractions ($0-50{\mu}g/mL$) for 24 h did not have an apparent cytotoxicity on pre and mature 3T3-L1 cells. RPS4 significantly suppressed differentiation and cellular lipid accumulation by increasing the phosphorylation of AMPK and reducing the expression of $PPAR-{\gamma}$, C/EBP ${\alpha}$, SREBP-1c, FAS, and ACC. In addition, all fractions except ethyl acetate fraction significantly suppressed cellular lipid accumulation. RPS4 induced the apoptosis of mature adipocytes by hypophosphorylating Akt, increasing the expression of the pro-apoptotic proteins, Bak, Bax, and Bad, and reducing the expression of the anti-apoptotic proteins, Bcl-2 and p-Bad. CONCLUSIONS: These finding suggest that RPS4 can reduce the numbers as well as the size of adipocytes and might useful for preventing and treating obesity.

Partial Characterization of Physicochemical and Kinetic Properties of $Ca^{++}-ATPase$ System in Luteal Membranes (황체막에서의 $Ca^{++}-ATPase$의 특성)

  • It has been reported that the luteal function may be regulated by the intracellular calcium in luteal cells (Higuchi et al, 1976; Dorflinger et at, 1984; Gore and Behrman, 1984) which is adjusted partially by $Ca^{++}-ATPase$ activities in luteal cell membranes (Verma and Pennistion, 1981). However, the physicochemical and kinetic properties of $Ca^{++}-ATPase$ in luteal membranes were not fully characterized. This study was, therefore, undertaken to partially characterize the physicochemical and kinetic properties of $Ca^{++}-ATPase$ system in luteal membranes and microsomal fractions, known as an one of the major $Ca^{++}$ storge sites (Moore and Pastan, 1978), from the highly luteinized ovary Highly luteinized ovaries were obtained from PMSG-hCG injected immautre female rats. Light membrane and heavy membrane fractions and microsomal fractions were prepared by the differential and discontinuous sucrose density gradient centrifugation method desribed by Bramley and Ryan (1980). Light membrane and heavy membrane fractions and microsomal fractions from highly luteinized ovaries are composed of the two different kinds of $Ca^{++}-ATPase$ system. One is the high affinity $Ca^{++}-ATPase$ which is activated in low $Ca^{++}$ concentration (Km, 10-30 nM), the other is low affinity $Ca^{++}-ATPase$ activated in higher $Ca^{++}$ concentration $(K_{1/2},\;40\;{\mu}M)$. At certain $Ca^{++}$ concentrations, activities of high and low affinity $Ca^{++}-ATPase$ are the highest in light membrane fractions and are the lowest in microsomal fractions. It appeares that high affinity $Ca^{++}-ATPase$ system have 2 binding sites for ATP (Hill's coefficient; around 2 in all membrane fractions measured) and the positive cooperativity of ATP bindings obviously existed in each membrane fractions. The optimum pH for high affinity $Ca^{++}-ATPase$ activation is around S in all membrane fractions measured. The lipid phase transition temperature measured by Arrhenius plots of high affinity $Ca^{++}-ATPase$ activity is around $25^{\circ}C$. The activation energies of high affinity $Ca^{++}-ATPase$ below the transition temperature are similar in each membrane fractions, but at the above transition temperature, it is the hightest in heavy membrane fractions and the lowest in microsomal fractions. According to the above results, it is suggested that intracellular $Ca^{++}$ level, which may regulate the luteal function, may be adjusted primarily by the high affinity $Ca^{++}-ATPase$ system activated in intracellular $Ca^{++}$ concentration range $(below\;0.1\;{\mu}M)$.

The Effect of Each Fraction of Methanol Extract of Alisma canaliculatum on Blood Glucose Levels and Lipid Metabolism in Streptozotocin Induced Diabetic Rats (택사 분획물의 투여가 Streptozotocin 유발 당뇨 흰쥐의 혈당수준과 지질대사에 미치는 영향)

  • Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-245g by injection of streptozotocin(STZ) into the tail vein at a dose of 45mg/kg and were divided into seven groups ; normal, diabetic control, and five experimental groups(fractions of hexane, chloroform(CHCl$_3$), ethylacetate(EtOAc), butanol(BuOH) and water($H_2O$)). The rats of all groups were fed on AIN-93 diet and the five experimental groups were orally administered each fraction for 14 days. The body weight and diet intake were monitored daily. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol, free fatty acid and aspartate and alanine aminotransferase activities were analyzed. Diabetic rats showed the lower weight gain compared to the normal rats. The plasma glucose levels of the CHCl$_3$, and $H_2O$ fraction groups were significantly lower than the other experimental groups. The plasma insulin level of the CHCl$_3$ fraction group was much higher than that of diabetic control group. The plasma cholesterol levels were increased in all the experimental groups. The groups of hexane, BuOH and H2() fractions showed the lower plasma triglyceride levels compared to diabetic control group. The plasma free fatty acid levels were not significantly different in all groups. HDL-cholesterol levels were definitely higher in hexane, CHCl$_3$ and EtOAc fraction groups than that of diabetic control group. In conclusion, administration of CHCl$_3$ and $H_2O$ fractions of methanol extract of Alisma canaliculatum exhibited hypoglycemic effects in STZ induced diabetic rats, showing the possibility of therapeutic use of Alisma canaliculatum to the diabetes mellitus.

Potential of Benthic Diatoms Achnanthes longipes, Amphora coffeaeformisand Navicula sp. (Bacillariophyceae) as Antioxidant Sources

  • Recently, interest in plant-derived food additives has developed natural antioxidants, in order to alternate syn-thetic antioxidants with several disadvantages. In the present study, different organic fractions from solvent parti-tions of 80% methanol extract from Jeju benthic diatoms, Achnanthes longipes, Navicula sp. and Amphora coffeaeformis was assessed for their potential antioxidant effects. Among the solvent fractions tested, n-hexane (80.4%) and 80% methanol extract (76.6%) from A. longipes, chloroform (63.2%) from Navicula sp. and n-hexane (67.4%) from A. cof-feaeformis were effective in DPPH free radical scavenging. Fractions of chloroform (53.4%) and n-hexane (53.1%) from A. longipes exhibited higher activities on $H_2O_2$ scavengin. Fraction of n-Hexane from A. longipes exhibited the highest hydroxyl radical scavenging activity and NO. scavenging activity (56.5% and 75.6%, respectively). Aqueous residue from A. coffeaeformis (75.6%) showed the highest metal chelating effect. chloroform and ethyl acetate frac-tion of all the diatoms exhibited significant antioxidant activities in lipid peroxidation inhibitory activity. In particu-lar, both chloroform and the ethyl ecetate fraction from A. longipes and A. coffeaeformis exhibited lipid peroxidation inhibitory activity significantly higher than that of $\alpha$- tocopherol. These data suggest that the Jeju benthic diatoms tested are rich in hydrophilic and hydrophobic antioxidative compounds with different antioxidative properties that can be applied in food industry.

Evaluation of Cytotoxicity, Carbohydrate, and Lipid Inhibitory Activity of Codonopsis lanceolata using Different Solvent Fractions

  • This study was conducted to evaluate the cytotoxicity and ${\alpha}-Amylase$, ${\alpha}-Glucosidase$, pancreatic lipase inhibition in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The values of $IC_{50}$ against Calu-6 cell showed a high effect in n-hexane fraction ($10.13{\mu}g/mL$) whereas DW fraction exhibited the weakest inhibition on cell viability, having an $IC_{50}$ value of over $1,000{\mu}g/mL$. The values of $IC_{50}$ against HCT-116 cell showed the highest activity in n-BuOH fraction ($102.01{\mu}g/mL$), followed by n-hexane fraction ($145.85{\mu}g/mL$), methylene chloride fraction ($332.02{\mu}g/mL$), ethyl acetate fraction ($462.93{\mu}g/mL$) and DW fracion ($>1,000{\mu}g/mL$). ${\alpha}-Amylase$ inhibitory activity in methylene chloride fraction and ethyl acetate fraction was found to have a higher inhibitory effect with 24.5% and 25.6% than the other fractions. The highest ${\alpha}-Glucosidase$ inhibitory activity was observed from the ethyl acetate fraction extract, while the extract of DW fraction showed the lowest level of inhibitory activity at given experiment concentration. The pancreatic lipase inhibitory activity of C. lanceolata was found to have a higher the effect in ethyl acetate fraction. Inhibition of lipase activity of the ethyl acetate fraction and n-hexane fraction showed a relatively high, while the extract of DW fraction showed the lowest level at given experiment concentration. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on carbohydrate, lipid Inhibitory activity and anticancer activity.

Effects of Human Plasma Lipid Transfer Protein on the Distribution of Lipids Between Lipoprotein Fractions of Rat Plasma (인체의 혈장에서 분리한 지질전이단백질이 흰쥐의 혈장 Lipoprotein 의 지질분포에 미치는 영향)

  • Rat lipoprotein-deficient plasma possessed a lipid tramsfer inhibitory activity when it was added to purified human plasma lipid transfer protien, while it lacked a lipid transfer activity. Incubation of whole rat plasma with partially purified human lipid transfer protein resulted in big changes in lipid distribution of rat plasma lipoproteins. There w was a 4-fold increase in cholesteryl ester(CE) and 4 47 % reduction in triglyceride(TG) in very low density lipoproteins after 2싹lour incubation. In high density lipoprotein $2(HDL_2)$ there was a 9­fold increase in TG and 33 % reduction in CEo HDL3 had 82 % reduction in CE. The result indi­c cates that the absence of the lipid transfer activity in rat plasma can be ascribed not to the inability of rat lipoproteins to serve as substrates but to the lack of 야Ie lipid transfer protein in rat plasma. Th­e erefore, species differences in lipid transfer betwe­e en lipoproteins should be taken into consideration to interpret results of studies on lipoprotein m.eta­b bolism using rats.
