동맥경화증(動脈硬化症)의 지방축적(脂肪蓄積) Mechanism (Mechanism of Lipid Accumulation in Atherosclerosis)

  • Atherosclerosis is associated with the presence of extracellular lipid droplets and large fatty deposits, both of which are to be covered at the surface mainly by zwitterionic phospholipids. The development of atherosclerosis is often associated with the accumulation of calcium. Furthermore, the presence of glycosaminoglycans directly underlying fatty deposits in human aorta has been demonstrated. Also, the possible involvement of the interaction between sulfated polysaccharide and lipoprotein in the development of atherosclerosis has been suggested in view of the presence of both low density lipoproteins and glycosaminoglycans, as well as their complexes, in atherosclerotic aortas. Therefore interactions of sulfated polysaccharides with low density lipoproteins which serve as a vehicle for cholesterol and cholesterol ester and with zwitterionic phospholipids have been studied extensively by a number of workers to provide mechanisms. In this paper, the mechanism of the interaction of sulfated polysaccharides with low density lipoproteins and the mechanism of the interaction between sulfated polysaccharides and zwitterionic phospholipids are reviewed. The possibility of the occurence of these interactions in the body are also considered.

인삼투여가토(人蔘投與家兎) 간지방섭취세포(肝脂肪攝取細胞)의 전자현미경적관찰(電子顯微鏡的觀察) (Electron Microscopic Study of Fat-storing Cells in Liver of Rabbits Given Ginseng)

  • The light and electron microscopic observations were carried out in order to know the morphological changes of the fat-storing cells (FSC) in the liver of rabbits given Ginseng for 30 days. The results obtained were as follows: The FSC of experimental group was shown the appearance of a few large lipid droplets, enlarged rough surfaced endoplasmic reticulum, some residual body in the enlarged rER and the nuclear body in some cases. As these findings, it could be suggested that the Ginseng affect the synthesis and accumulation of lipid on the FSC.

  • The light and electron microscopic observations were carried out in order to know the morphological changes of the testicular Leydig cells in the mouse injected with testosterone propionate for a long period. The results obtained were as follows: With the light microscopic study, atrophy of the Leydig cells and larger sudanophilic lipid granules in the experimental group than normal were observed. 2. By the electron microscopic finding, the small spherical or oval mitochondria, large lipid droplets, a decrease in number of smooth endoplasmic reticutum and distended saccular or vacuolar smooth endoplasmic reticulum were observed in the experimental group. Membranous whorls with droplet increased in number and size in the experimental group.

Endpoint Depth When Removing Xanthelasma Using CO2 Laser Ablation: A Case Report

  • The carbon dioxide (CO2) laser is one of the treatment options used for xanthelasma palpebrarum. However, even if the full dermal layer is removed, the lesion can recur due to the residual lipid deposits. A 44-year-old male patient with xanthelasma on both upper eyelids was treated with a pulsed dye CO2 laser. On the right upper eyelid, we carried out a CO2 laser treatment until the yellowish plaque was almost invisible and the full thickness of the dermal layer was removed. On the left upper eyelid, the dermal layer was partially removed and an additional squeezing out of yellowish particles was done. The lesion treated by the squeezing out of lipid droplets showed better long-term results than the lesion treated up to the deeper dermal layer.

The effects of Brassica juncea L. leaf extract on obesity and lipid profiles of rats fed a high-fat/high-cholesterol diet

  • BACKGROUND/OBJECTIVES: Obesity is a global health problem of significant importance which increases mortality. In place of anti-obesity drugs, natural products are being developed as alternative therapeutic materials. In this study, we investigated the effect of Brassica juncea L. leaf extract (BLE) on fat deposition and lipid profiles in high-fat, high-cholesterol diet (HFC)-induced obese rats. MATERIALS/METHODS: Male Sprague-Dawley rats were divided into four groups (n = 8 per group) according to diet: normal diet group (ND), high-fat/high-cholesterol diet group (HFC), HFC with 3% BLE diet group (HFC-A1), and HFC with 5% BLE diet group (HFC-A2). Each group was fed for 6 weeks. Rat body and adipose tissue weights, serum biochemical parameters, and tissue lipid contents were determined. The expression levels of mRNA and proteins involved in lipid and cholesterol metabolism were determined by reverse transcription polymerase chain reaction and western blot analysis, respectively. RESULTS: The HFC-A2 group showed significantly lower body weight gain and food efficiency ratio than the HFC group. BLE supplementation caused mesenteric, epididymal, and total adipose tissue weights to decrease. The serum levels of triglyceride, total cholesterol, and low-density lipoprotein cholesterol were significantly reduced, and high-density lipoprotein cholesterol was significantly increased in rats fed BLE. These results were related to lower glucose-6-phosphate dehydrogenase, acetyl-coA carboxylase, and fatty acid synthase mRNA expression, and to higher expression of the cholesterol $7{\alpha}$-hydroxylase and low density lipoprotein-receptor, as well as increased protein levels of peroxisome proliferator-activated receptor ${\alpha}$. Histological analysis of the liver revealed decreased lipid droplets in HFC rats treated with BLE. CONCLUSIONS: Supplementation of HFC with 3% or 5% BLE inhibited body fat accumulation, improved lipid profiles, and modulated lipogenesis- and cholesterol metabolism-related gene and protein expression.

녹차추출물이 인체 피지선세포주에서 지질 생성에 미치는 효과 (Effect of Green Tea Extract on Lipid Synthesis in Human Sebocyte Cell Line)

  • The aqueous Green tea comes to be used with the Oriental medicine plant, it has the numerous health benefits, including antioxidant, anti-inflammatory, and anti- carcinogenic properties. Epidermal progenitor cells give rise to multiple skin lineages: hair follicle, sebaceous gland and the overlying interfollicular epidermis. Sebocytes are the cells of the sebaceous gland, which synthesize and accumulate lipid dropolets. In order to determine the effect of Green tea on lipid production, several experiments were performed in SZ95 cells (sebocytes). We found that Green tea increased lipid droplets compared with control in a dose-dependent manner. Human sebaceous glands produce sebum, a lipid mixture of squalene, wax esters, triglycerides, cholesterol esters, and free fatty acids that is secreted onto the skin. Therefore, to investigate the effects of Green tea on intracellular lipid levels, we treated SZ95 cells with Green tea, and then examined cholesterol and triglyceride levels. After treatment of the cells with Green tea, the cholesterol and triglyceride levels of SZ95 cells were increased significantly in a dose-dependent manner.

Glutamic-oxaloacetic transaminase 1 regulates adipocyte differentiation by altering nicotinamide adenine dinucleotide phosphate content

  • Objective: This study was performed to examine whether the porcine glutamic-oxaloacetic transaminase 1 (GOT1) gene has important functions in regulating adipocyte differentiation. Methods: Porcine GOT1 knockout and overexpression vectors were constructed and transfected into the mouse adipogenic 3T3-L1 cells. Lipid droplets levels were measured after 8 days of differentiation. The mechanisms through which GOT1 participated in lipid deposition were examined by measuring the expression of malate dehydrogenase 1 (MDH1) and malic enzyme (ME1) and the cellular nicotinamide adenine dinucleotide phosphate (NADPH) content. Results: GOT1 knockout significantly decreased lipid deposition in the 3T3-L1 cells (p<0.01), whereas GOT1 overexpression significantly increased lipid accumulation (p<0.01). At the same time, GOT1 knockout significantly decreased the NADPH content and the expression of MDH1 and ME1 in the 3T3-L1 cells. Overexpression of GOT1 significantly increased the NADPH content and the expression of MDH1 and ME1, suggesting that GOT1 regulated adipocyte differentiation by altering the NADPH content. Conclusion: The results preliminarily revealed the effector mechanisms of GOT1 in regulating adipose differentiation. Thus, a theoretical basis is provided for improving the quality of pork and studies on diseases associated with lipid metabolism.

Effects of Testosterone, 17β-estradiol, and Progesterone on the Differentiation of Bovine Intramuscular Adipocytes

  • The aim of this study was to investigate the effects of testosterone, 17$\beta$-estradiol, and progesterone on the differentiation of bovine intramuscular adipocytes (BIA). Stromal-vascular (SV) cells were obtained from M. longissimus dorsi of 20 months old Korean (Hanwoo) steers, and were cultured in DMEM containing 5% FBS. The proliferated BIA were induced to differentiate with 0.25 $\mu$M dexamethasone, 0.5 mM 1-methyl-3-isobutyl-xanthine and 10 $\mu$g/ml insulin. During differentiation, the cells were treated with testosterone, 17$\beta$-estradiol, and progesterone at concentrations of $10^{-10}$, $10^{-9}$, and $10^{-8}$ M, respectively, for 12 days. Regardless of its concentration, testosterone remarkably reduced lipid droplets in the cytosol of BIA. On the other hand, 17$\beta$-estradiol and progesterone increased the accumulation of lipid droplets in BIA. Testosterone significantly (p<0.05) decreased GPDH activities with a dose-dependent pattern. 17$\beta$-Estradiol treatment onto BIA during differentiation, however, increased GPDH activity showing the highest activity (11.3 nmol/mg protein/min) at $10^{-10}$ M. Treatment of BIA with progesterone also increased (p<0.05) GPDH activity with the highest activity (13.8 nmol/mg protein/min) at $10^{-9}$ M. In conclusion, the results in the current study suggest that testosterone inhibits differentiation of BIA by suppressing GPDH activity while 17$\beta$-estradiol and progesterone have adverse effects.

난소를 절제한 흰쥐 자궁상피의 호르몬투여에 대한 전자현미경적 연구 (Ultrastructural Study on the Luminal Epithelium of the Ovariectomized Rat Uterus after Hormonal Treatment)

  • 난소절제한 흰쥐에서 $17{\beta}$-estradiol과 progesterone을 장기간 주사햐여 자궁상피의 형태적 변화를 전자현미경으로 관찰하였다. 난소절제 후 자궁상피는 막성구조물과 지방구 기타의 구조물을 포함하는 vacuole의 출현이 특이적으로 나타났다. 상피는 낮은 입방형이며 세포 유리면에 소수의 짧은 microvilli를 볼 수 있었다. Estradiol투여시 상피는 높은 윈주형을 나타내며, 분비계는 비교적 잘 발달되고, 세포 유리면에 소수의 긴 microvilli도 관찰되었다. Progesterone 처리시의 상피는 세포 첨단부에 다수의 vacuole과 핵하부에 다수의 지방구의 집적이 관찰되었다. 이상의 결과로 보아 자궁상피는 형태학적 및 기능적인 상태에서 estrogen과 Progesterone 양자에 의해 변화를 받으며 이들 호르몬은 상피세포에 대해 독특한 영향을 나타낸다고 생각된다.

팜유발효에 있어서 리파제의 특성과 팜유자화와의 관계 (Properties of Lipases and Palm Oil Assimilating Patterns in Palm Oil Fermentation)

  • 미생물에 의한 천연유지의 분해자화과정을 극명함으로써 값싼 유지를 발효원으로서 착용하기 위하여 팜유자화성 유용균주인 Torulopsis candida Y-128과 Acinetobacter calcoaceticus KB-2가 생산하는 리파제의 특성과 이들 균주의 생리적인 특성을 검토하였다. T. candida Y-128은 팜유입자에 부착·자화하며 리파제의 작용에 의해 유리되는 분포화지방산을 포화지방산에 비해 쉽게 자화이용 함으로써 균체증식이 이루어지고 있었다. T. candida Y-128의 리파제는 대부분 균체내에 존재하는데 비해, A calcoaceticus KB-2는 배양시에 균체증식 대수기부터 균체외로 리파제가 생성됨을 알수 있었으며, 리파제에 의해 유리된 포화 지방산도 다른 균주에 비해 자화이용이 용이함을 알 수 있었다. 두 균주는 배양액중에 리파제를 축적하지 않고 균체생육에 필요한 정도를 생산하며 천연중지중 1(3-)-위치의 지방산에 작용하는 위치특이성을 나타내었다. 따라서 두 균주는 천연유지입자에 부착하여 1(3-)- 위치의 지방산을 분해하고, 분해생성물은 지방산대사경로를 거쳐 자화이용되는 것으로 보여진다.
