Effects of Peroxisome Proliferator-Activated $Receptor-{\gamma}2$ Pro12Ala Polymorphism on Body Fat Distribution in Female Korean Subjects (Peroxisome Proliferator-Activated $Receptor-{\gamma}$ 2 $(PPAR{\gamma}2)$ Pro12Ala (P12A) 유전자 다형성이 한국여성의 체지방분포에 미치는 영향)

  • Objectives: The effects of peroxisome proliferator-activated receptor ${\gamma}2\;(PPAR{\gamma}2)$ Pro12Ala (P12A) polymorphism on body mass index (BMI) and type 2 diabetes are well documented; however, until now, only a few studies have evaluated the effects of this polymorphism on body fat distribution. This study was conducted to elucidate the effects of this polymorphism on computed tomography (CT)-measured body fat distribution and other obesity-related parameters in Korean female subjects. Methods & Results: The frequencies of $PPAR{\gamma}2$ genotypes were: PP type, 93.0%; PA type, 6.8%; and AA type, 0.2%. The frequency of the A allele was 0.035. Body weight (P .012), BMI (P .012), and waist-to-hip ratio (WHR) (P .001) were significantly higher in subjects with PA/AA compared with subjects with PP. When body composition was analyzed by bioimpedance analysis, lean body mass and body water content were similar between the 2 groups. However, body fat mass (P .003) and body fat percent (P .025) were significantly higher in subjects with PA/AA compared with subjects with PP. Among overweight subjects with BMI of greater than 25, PA/AA was associated with significantly higher abdominal subcutaneous fat (P .000), abdominal visceral fat (P .031), and subcutaneous upper and lower thigh adipose tissue (P .010 and .013). However, among lean subjects with BMI of less than 25, no significant differences associated with $PPAR{\gamma}2$ genotype were found, suggesting that the fat-accumulating effects of the PA/AA genotype were evident only among overweight subjects, but not among lean subjects. When serum lipid profiles, glucose, and liver function indicators were compared among overweight subjects, no significant difference associated with $PPAR{\gamma}2$ genotype was found. Changes in body weight, BMI, WHR, and body fat mass were measured among overweight subjects who finished a 1-month weight lose program of a hypocaloric diet and exercise; no significant differences associated with $PPAR{\gamma}2$ genotype were found. Conclusions: The results of this study suggest that the $PPAR{\gamma}2$ PA/AA genotype is associated with increased subcutaneous and visceral fat areas in overweight Korean female subjects, but does not significantly affect serum biochemical parameters and outcomes of weight loss programs.

Protective effects of dandelion extract against liver damage by repeated administration of thioacetamide in rats (랫드에서 thioacetamide의 반복 투여로 유도된 간 손상에 대한 민들레 추출물의 효과)

  • This study was conducted to investigate of hepatoprotective effect of dandelion water extract (DWE) according to repeated administration of thioacetamide (TAA) induced hepatotoxicity in Spraque-Dawley rats. Thirty rats were randomly assigned to 5 groups; normal control, DWE-control, TAA-control (TAA injection during the feeding of normal diet), TAA&DWE600 (TAA repeated injection during the feeding of DWE 600 mg/kg BW), TAA&DWE1200 (TAA repeated injection during the feeding of DWE 1,200 mg/kg BW). Rats in DWE-control and TAA&DWE groups were treated with DWE (600 or 1,200 mg/kg BW daily) by gavage for 20 days (twice a day). All the rats in the TAA-control and TAA&DWE groups were repeated injection of TAA (100 mg/kg BW) into the abdominal cavity 3 days interval and 12 hrs later, all rats were sacrificed. At the same time, normal control and DWE-control groups were injected normal saline. In TAA&DWE groups, serum alanine and aspartate aminotransferase (ALT, AST) were significantly decreased and triglyceride (TG) synthesis was significantly increased compared to TAA group. As well as total billilubin and GGT were slightly decreased by the treatment of DWE. Lipid peroxidation (MDA) concentration was significantly decreased and hepatic GSH content was slightly or significantly increased in the TAA&DWE groups compared to TAA group. Hepatic anti-oxidative enzyme activities, such as GSH, GST, SOD and catalase were slightly or significantly elevated by the treatment of DWE. According to these results, When dandelion extract was long term supplied, it could be used as a potential protective material for a longer time liver damage by repeated adminstration of the TAA.

Effects of Methanol Extract from Terminalia chebulae on Renal and Pulmonary Toxicities Induced by Paraquat in Rats (가자(Terminalia chebulae) 메탄올 추출물이 paraquat 독성에 의한 흰 쥐의 신장 및 폐조직에 미치는 영향)

  • Paraquat (1,1-dimethyl 4,4' -dipyridium dichloride; PQ) is a kind of herbicide. Terminalia chebulae (TC) has been used as a medicine in China and in Korea for treating illnesses such as diarrhea, collapsed anus, spasmodic, diphtheria, asthma etc.. This study was to examin new physiological activities of methanol extract of TC (TCM) on the toxicity of PQ. It was observed biochemical effects on the toxicity of PQ in kidney and lung tissues after treatment orally administered 100, 200, 300 mg/kg of TCM daily for two weeks. In the experiment related to the toxicity of PQ, we got following results: renal and pulmonary lipid peroxide contents, activities of aminopyrine N-demethylase, aldehyde oxidase and xanthine oxidase were significantly increased in control group as compared with normal group, in the treatment of TCM the values were decreased as compared with control group. Activities of superoxide dismutase, catalase and glutathione peroxidase which are free radical scavenging enzymes were also increased in control group as compared with normal goup, but were decreased in TCM group as compared with control group. Collagen content and glucose-6-phosphatase activity in lung tissue were increased in control group as compared with normal, but was decreased in TCM group as compared with control group. From these results, we concluded that TCM can playa role as an effective agent to decrease toxicity of PQ.

Peptides-derived from Scales of Branchiostegus japonicus Inhibit Ultraviolet B-induced Oxidative Damage and Photo-aging in Skin Cells (피부세포에서 옥돔 비늘로부터 추출한 펩타이드의 UVB에 대한 산화적 손상 및 광 노화 억제)

  • Collagen peptides, which are found at high concentrations in the human body, are present in animal bones and the skin of marine organisms, namely, fish scales. Collagen is the most abundant structural protein of various connective tissues in animals. Furthermore, it is widely used in biomedical material, pharmaceutical, cosmetic, food, and leather industries. Peptides extracted from scales of various fish protect against ultraviolet B (UVB)-induced skin damage and photo-aging. However, the protective effects of collagen peptides derived from the scales of Branchiostegus japonicus against UVB exposure are unclear. This study investigated the effects of peptides larger than 1 kDa (high-molecular weight peptides [HMP]) and smaller than 1 kDa (low-molecular weight peptides [LMP]), derived from extracts of B. japonicus scales, against UVB-induced skin damage and photo-aging. These peptides scavenged 1,1-diphenyl-2-picrylhydrazyl radicals in a dose-dependent manner. In UVB-exposed HaCaT human keratinocytes, LMP inhibited 8-isoprostane generation, a marker of cellular lipid peroxidation. The peptides also suppressed the UVB-induced increase in tyrosinase activity and melanin content in B16F10 mouse melanoma cells. In addition, the LMP and HMP treatment suppressed UVB-induced elastase and matrix metalloproteinase-1 activities in the HaCaT cells. These results indicate that peptides derived from B. japonicus scales have antioxidant, antiphoto-aging, and skin-whitening effects.

The Involvement of p38 MAPK and JNK Activation in Palmitic Acid-Induced Apoptosis in Rat Hepatocytes (Palmitic acid에 의한 간세포 사멸효과에 대한 p38 MAPK 및 JNK 관련성)

  • Hyperlipidemia has been reported to be associated with the development of fatty liver. Palmitic acid, a major saturated fatty acid, is involved in the development of diverse diseases. The activation of mitogen activated protein kinases (MAPKs), such as Jun N-terminal kinase (INKs) and p38 MAPK is implicated in the apoptosis in diverse cells. Thus, this study was conducted to investigate the effects of palmitic acid on apoptosis and its relationship between JNK and p38 MAPK in cultured rat hepatocytes. In the present study, palmitic acid (>50 uM) decreased cell proliferation and increased lactate dehydrogenase activity in hepatocytes, which was blocked by the treatment of SP600125 (a JNK inhibitor) and SB203580 (a p38 MAPK inhibitor). Indeed, palmitic acid decreased Bcl-2 expression but increased Bax expression in rat hepatocytes, which was blocked by the treatment of SP600125 and SB203580. In addition, palmitic acid decreased glutathione (GSH) content and increased lipid peroxide formation, which was blocked by the treatment of SP600125 and SB203580. Western immunoblotting analysis also revealed that palmitic acid increased JNK and p38 MAPK. In conclusion, palmitic acid induced apoptosis through oxidative stress via JNK and p38 MAPK activation in rat hepatocytes.

Purification and Characterization of Fish Oil Obtained from Hydrolysates of Mackerel (Scomber japonicus) By-products (고등어(Scomber japonicus) 부산물 가수 분해물로부터 분리된 유지의 정제 및 특성)

  • This study aimed to investigate an effective method of purifying oil from hydrolysates of mackerel by-products. A hydrolyzed sample was separated into lipids and other material. After degumming of crude lipids with 6% citric acid solution, the phosphorous content (45 mg/kg) decreased to 5.8 mg/kg, and the acid value (18.03 mg /g) decreased to 1.19 mg KOH/g. Following the addition of 8% sodium hydroxide solution, the peroxide value (30.25 meq/kg) dropped to 3.18 meq/kg, and the chromaticity decreased from 0.488 nm to 0.057 nm. Bleaching was performed by adding 5% activated charcoal for 60℃ for 20 min under vacuum conditions. After bleaching, the acid value was 0.17 mg KOH/g, and the peroxide value was 1.21 meq/kg, but the chromaticity was not changed remarkably (0.062 nm). Crystallization was conducted to increase the amount of unsaturated fatty acids. After crystallization, the total amount of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) was 228.83 mg/g. The results suggest that the product can be used as an omega-3 fatty acid resource for functional food.

Characteristics and Application of Soybean Hull Fractions Obtained by Microparticulation/Air-Classification (초미세분쇄/공기분급을 이용한 대두피 분획물의 특성과 응용)

  • Soybean hull was microparticulated at cut-off wheel speed of 9,000 rpm and air-classified into fine and coarse fraction at air classifying wheel speeds (ACWS) of 10.000, 15,000 and 20,000 rpm. respectively. Proximate analysis of the coarse and fine fractions of the microparticulated/air-classified soybean hull showed that ash, protein and lipid were enriched in the coarse fractions and carbohydrates in the fine fraction. Dietary fiber were enriched in coarse fractions and dietary fiber and dietary fiber content increased with decreasing ACWS. Mean particle size of fine fractions ranged from $3.1{\mu}m\;to\;5.5{\mu}m$ that of coarse fractions ranged from $9.9{\mu}m\;to\;20.0{\mu}m$ and both increased with decreasing ACWS. The particles were mostly oval-shaped regardless of the fractions. Generally the fraction of low ACWS showed higher viscosity. In water holding capacity, oil absorption and emulsion capacity, there was significant difference between coarse and fine fractions while there was no significant difference as a function of different ACWS. Oil absorption of flying batter was decreased significantly in the fraction of ACWS 15,000. when cakes and cooking were prepared partially substituted with soybean hull fraction, there was no significant changes on their physical and sensory properties up to 10% substitution.

Clinical Study on Food Safety evaluation of Panax ginseng (고려인삼의 식품 안전성에 관한 임상 연구)

  • In this study, the clinical safety and toxicology of oral ingestion of supplement capsules containing ginseng radix was investigated in healthy young volunteers. This study was a pilot randomized, double blinded, placebo controlled trial. The healthy volunteers were divided into 6 groups of 20 each (10 males and 10 females). They took the ginseng powder for 35 days (3g/day) for safety evaluation. There were measured general healthy levels such as hematological, biochemical and electrocardiographic parameters. After the first week, besides Korean white ginseng the other treatments led to an significant increase of white blood cells. Korean red ginseng increased UREA (blood urea nitrogen) in healthy volunteers, but it didn't exceed the range of normal values, and in the subsequent process of treatment there is no effect of elevating UREA. After the three weeks, Korean white ginseng showed relatively low the content of blood glucose and low-density lipoprotein cholesterol. After the five weeks, compared with the other treatments, Korean red ginseng increased white blood cells, platelet distribution width and average volume of platelet. Korean white ginseng decreased low-density lipoprotein cholesterol. American ginseng decreased blood creatinine in healthy volunteers. In conclusion, through test the blood routine, urine routine, liver function, renal function, blood glucose, blood lipid and electrocardiogram, the healthy volunteers continuous taking ginseng for 35 days (3 g/day) is safe and reliable, and have no obvious adverse reactions and side effects.

  • Rainbow trout, Oncorhynchus mykiss, were cultured with different levels of carotenoids in odor to investigate the feeding effect of ascidian tunic extracts on the liver fatty acid compositions of them. Dominant monoenoic fatty acids of the ascidian tunic extracts were 16:1n-7($5.9\%$), 18:1n-9($21.9\%$), and 18:1n-7($3.5\%$) and polyenoic fatty acids of them were linoleic(18:2n-6, $14.2\%$), eicosapentaenoic(20:5n-3, $3.5\%$), and docosahexaenoic acid(22:6n-3, $8.3\%$). The compositions of fatty acid in the liver lipids were affected by the tunic extract levels during the feeding. The percentage of monoenoic acids in extract diets was decreased, and that of n-3PUFA was increased during feeding 2 weeks. But the n-3PUFA contents were decreased in 4 weeks. The 20:4n-6 content in rainbow trout fed extract diet was higher than that in the control and pink diet groups. The rainbow trout fed with ascidian tunic extracts showed an increase of essential fatty acids in the fish tissue, compared to the control or pink diet feeding groups.

Comparison of Quality among Boiled-Dried Anchovies Caught from Different Sea (어획지 차이에 따른 마른멸치의 품질 비교)

  • This study was conducted to evaluate the quality of the commercial boiled-dried anchovies (Engraulis japonicus) caught from different sea (southern, eastern and western sea) by determining chemical components and sensory evaluation. Product S was from the southern sea. product W was from the western sea and product E was from the eastern. The moisture content of product S was $22.9\%$ which was low compared with those of the product I ($27.4\%$%) and the product W ($27.8\%$). There are no difference in the acid-insoluble ash total amino acid (dry basis), mineral (dry basis) contents and fatty acid composition among products caught from different sea. On the other hand, The product S was superior in the lipid properties (peroxide value and (20:5n-3+22:6n-3)/16:0) and sensory Properties (appearance and color) to those of product I and product W. Judging from the above results of chemical analyses and sensory evaluation, the product S was the best quality among commercial boiled-dried anchovy, followed by product W, and then product E.
