Studies on the Lipid Components of Potato Tubers - I. Lipid Composition in Free and Bound Lipids - (감자의 지방질(脂肪質) 성분(成分)에 관한 연구(硏究) - 제(第) 1보(報) : 유리(遊離) 및 결합(結合) 지질(脂質)중의 지방질(脂肪質) 조성(組成)에 관하여 -)

  • The experimentally cultivated potatoes of 4 varieties, Irish Cobbler, Warba, Shimabara, and Saco were dried in a frozen state, powdered subsequently and subjected to the extraction of free and bound lipids. Constituents of the prepared lipids were fractionated, quantified, and compared by the methods of column and gas-liquid chromatographies. The results were summarized as follows : 1. The total crude lipid content in potato on a dry weight basis was 0.57 % of which 0.2 % was free lipid and 0.37 % was bound lipid. 2. The neutral lipid content in the free lipid was 14.9 %, approximately 3 times as much as the 4.5 % contained in the bound lipid, whereas the glycolipid content in the free lipid was 15.1 %, slightly less than 22.2 % contained in the bound lipid. However, the phospholipid content was 33.9 % in the bound lipid, approximately 4.5 times as much as the 7.2 % contained in the free lipid. This fact revealed that the bound lipid consisted mainly of polar lipid, while the free lipid consisted of neutral lipids, glycolipids and phospholipids in about the same proportion. 3. The main fatty acids constituting more than 90 % in the free and bound lipids were linoleic, palmitic and linolenic acids. The content of the saturated fatty acid was slightly less in the free lipid than in the bound lipid, whereas the unsaturated fatty acids were more abundant in the free lipid.

Lipid Composition of Barley Flour Produced in Korea (한국산(韓國産) 보리가루의 지방질(脂肪質) 조성(組成))

  • Grains of naked barley (Baekdong cultivar) were polished, powdered and subjected to the successive extraction into free and bound liquid fractions. These were further fractionated into lipid classes and quantified by means of thin layer chromatography, column chromatography and gas-liquid chromatography. Contents of free and bound lipids in barley flour were 2.27% and 1.01%, which were decreased to 2.12% and 0.76%, respectively, after purification. Free and bound lipids were consisted of monoglycerides, diglycerides, triglycerides, free sterols, sterol esters, free fatty acids and polar lipids. Major constituents of free lipids were 56.2% triglycerides, 14.9% free fatty acids and 13.4% sterols while those of bound lipids were 73.8% polar lipids, 8.4% free fatty acids and 5.2% triglycerides. The content of non-polar lipids in free lipids was 93.6% as compared with 26.2% in bound lipids. However, phospholipids content in bound lipids was 55.5% as compared with 2.5% in free lipids, and glycolipids content in bound lipids was 19.4% as compared with 3.9% in free lipids. Major fatty acids in the free and bound lipid fractions were linoleic acid 52.1%, 54.8%, palmitic acid 24.8%, 30.0% and oleic acid 15.6%, 8.8%, respectively, showing similar patterns in both fractions. The amount of unsaturated fatty acids in free lipids was 72.8% as compared with 68.0% in bound lipids. In comparing the fatty acid composition of non-polar lipids, glycolipids and phospholipids, no difference was observed between free and bound lipid fractions while a slight difference was found among the lipid constituents.

  • The histological and transmission electron microscopic studies revealed the synthesis activity predominantly in the hypopharyngeal glands of the nurse bees. The biochemical analysis of both, the hypopharyngeal gland extract and royal jelly elucidated unequivocally the proteins and lipids as the major constituents. Further the SDS-PAGE of hypopharyngeal gland extract showed about 17 protein bands, perhaps 14.10, 20.00, 29.00 and 43.00 kDa predominantly while that of royal jelly revealed only two protein bands of 29.00 and 43.00 kDa molecular weight suggesting them as the major royal jelly proteins (MRJP). The lipid profile of royal jelly consists of triglycerides, cholesterol, HDL, LDL and VLDL.

  • The purpose of this study was to investigate of the dietary life and nutritional status of the Buddhist priests. Dietary life, the state of prepare a meal and menu are carried out through the questionnaire and the nutritional status is carried out through the analysis of the constituents of the blood. The results of this study can be summarized as follows. 1. The traditional temple's foods undergo a change with the variety and westernization of dietary life. 2. Buddhist priests take a carbohydrate centered meal and the intake of protein and lipid is tendency to lack, but the intake of vitamin and mineral is sufficient with various vegetables. 3. The content of the lipid compoents and glucose of blood are low and they are free from the danger of geriatric diseases but the content of triglyceride is very higher than the healthy general adults. Therefore it is necessary to improve the nutritional status of the buddhist priests with decrease the intake of rice and increase the various side dish and to develop the standard menu for the Buddhist priests.

  • Using the Chlorella cells which had been uniformly labeled with $^{32}P$, the distribution of phosphorus in various fractions of cell material was investigated. Uniformly $^{32}P$-labeled Chlorella cells were further grown in a P-free medium, and some protions of the cells were taken out at intervals during the culture, and subjected to analyze the contents of $^{32}P$ in various fractins of the cell constituents. 2. Analysis of the $^{32}P$-labeled Chlorella cells showed that the highest in P-content was the fraction of RNA followed by those of lipid, RNA-polyphosphate complex, acid-insoluble polyphosphate, acid-soluble polyphosphate, DNA and protein. 3. During the culture of $^{32}P$-labeled Chlorella cells in a P-free medium, amounts of phosphate in DNA, protein and lipid fractions increased, while the P-contents in the fraction of RNA-polyphosphate complex decreased as well as those of acid-insoluble polyphosphate and acid-soluble polyphosphate fractions. 4. It was inferred that phosphorus used in the syntheses of DNA and protein was taken from polyphosphates of the cells, and RNA-polyphosphate complex would play an important role as a phosphate pool.

  • Seed lipids and proteins of Crotalaria juncea L were analyzed for fatty acids and amino acids respectively. Gas chromatographic analysis of the oil gave palmitic acid (16.01%), stearic acid (7.29%), oleic acid (14.41%), linoleic acid (54.44%) and linolenic acid (7.86%). The defatted seed cake contained all the essential amino acids except methionine and six non-essential amino acids.

Caponization Effects on Growth Performance and Lipid Metabolism in Taiwan Country Chicken Cockerels

  • This trial was designed to study the caponization effects on the appearance, carcass characteristics, blood constituents and lipid metabolism of Taiwan country chicken cockerels. Cockerels were caponized at 8 weeks of age. Sixteen-week-old chickens, including 10 capons, 5 slips (incomplete caponized male chickens) and 20 normal chickens of equal sexes were selected for a 10 week ad libitum feeding trial. Results showed that the testosterone concentrations in the capons and females were lower (p<0.05) than that of intact males. The comb length, height and weights were also lower (p<0.05). The weight of the slips was between that of the capons and intact males, but was heavier (p<0.05) than that of the capon. The live-weight, carcass weight and shank perimeter in the capons were higher than those of the other groups (p<0.05). Hepatic lipogenic enzyme activity analyses showed that NADP-malic dehydrogenase (MDH) activity in the capons and female chickens was higher than that in intact male chickens (p<0.05). The MDH activity in the slips was between that for the capon and intact male chickens (p>0.05). The abdominal fat weight and relative abdominal fat weight of the capons and females were heavier than that for intact males (p<0.05); the slips were between the capons and intact males. The blood lipid content results showed that the triacylglycerol and cholesterol in the capons were higher than that for intact males (p<0.05). However, the percentage of low-density lipoprotein (LDL) was lower than that in intact male chickens (p<0.05). It appears that the increase in lipid accumulation in caponized male chickens is attributed mainly to an increase in MDH activity and the changes in lipid transportation in the capons.

Beakdugu-tang, Traditional Korean Digestant Medicine, Inhibits Hepatic Steatosis in Insulin Resistance Cell Model with HepG2 and THP-1

  • Objectives: Beakdugu-tang (BDGT) consists of three medicinal herbs, and this prescription has long been used in treatment of various digestant problem in Korea. In this study, we designed to clarify mechanisms by which Korean traditional digestive medicine, BDGT, may exert anti-hepatic steatosis effects via improved insulin resistance cell model in human hepatocellular carcinoma (HepG2) and monocyte (THP-1). Materials and methods: The preparation of BDGT and constituents were extracted with 70% ethanol. HepG2 and THP-1 were treated with different concentrations of BDGT and constituents in the presence and absence of stimulants such as free fatty acids (FFAs) and oxidized low-density lipoprotein (ox-LDL), respectively. Results: The BDGT and its constituents inhibited the FFAs-stimulated lipid accumulation in HepG2 cells. Ethanol extracts of Amomum cardamomum (ACE) improved the ox-LDL induced insulin resistance in THP-1 cells. Also, treatment of monocytic cells with ACE increased anti-hepatic steatosis related gene levels including ABCA, ABCG and SR-B1. Conclusion: The results suggest that the ethanol extract of BDGT and its constituents potently inhibit the FFAs- and ox-LDL induced liver steatosis via improved insulin resistance.

Attenuation of streptozotocin mediated oxidative stress, hyperglycemia and toxicity in rats by treatment with B-20 drpos - a homoeopathic preparation

  • The present study is aimed at finding the effect of B-20 drops, a homoeopathic formulation, in streptozotocin (STZ) induced diabetic rats. B-20 drops comprises of the constituents derived from plants and other natural sources, and are generally prescribed by the homoeopathic physician, in cases of hyperglycemia and diabetes. The elevated levels of fasting blood glucose and pancreatic lipid peroxides observed in rats treated with STZ were significantly reduced by the treatment of B-20 drops. The reduced liver glycogen contents were also brought back to near normal level by B-20 drops treatment in STZ diabetic rats. STZ induced histopathological changes in pancreas and liver was also partially reversed by B-20 drops. The findings indicate that B-20 drops help in improving the glycogen stores in the liver and prevents STZ induced damage through free radicals by decreasing the pancreatic lipid peroxides levels.
