Culture Tube Method for the Determination of Total Cholesterol in Egg Yolk Lipid (Culture Tube 방법에 의한 난황중의 Cholesterol 정량)

  • A simple, reproducible , and accurate enzymatic method using a cholesterol assay kit was developed to quantify total cholesterol content in egg yolk. Total egg yolk lipid was extracted with hexane : isopropanol(3 : 2, v/v) mixture. Samples containing various amount of the total lipid(0-3mg) in optically identifical culture tubes were reacted for 10 min in a water bath (37$^{\circ}C$) with the enzyme solution (5ml) from the cholesterol assay kit. Cholesterol content of the reaction mixturesin culture tubes was spectrophotometrically determined by two different ways : (1) using the culture tube as a curvette(designate culture tube method ; CTM) and (2) the quartz cvette containing the reaction mixture transferred from the culture tube (designate standard cvette method, SCM). CTM revealed lower cholesterol content in 0.1-1.0mg lipid sample range that SCM did, but not significant. For more than 2.0mg lipid sample, CTM gave significantly (p<0.01) lower cholesterol content relative to that by SCM, suggesting that SCM give a false positive result from the sample containing more than 2 mg lipid due to the interference of absorbance by lipid dispersed in the reaction solution . Cholesterol content of less than 1.0mg lipid sample by CTM was proportional to the amount of lipid used, but its linear relationship was not seen in more than 2mg lipid sample. Thus, to determine the appropriate lipid amounts (mg) analyzed . A constant level (41$\mu\textrm{g}$/mg) of cholesterol concentration was observed from the sample containing 0.1-1mg lipid. after which the cholesterol level was dropped to less than 41$\mu\textrm{g}$ /mg. Cholesterol concentration in egg yolk samples quantified by CTM was in accordance with that by GC method. These results suggest that CTM is an useful method for the quantification of cholesterol in egg yolk lipid and other lipids as well.

Effect of Fructooligosaccharide on Lipid Metabolism in Hypercholoesterolemic Rat (올리고당 첨가식이가 흰쥐의 지질대사에 미치는 영향)

  • This study was conducted to investigate the effect of fructooligosaccharide on intestinal flora, lipid metabolism and immune response. Thirty two male rats of Sprague-Dawley strain were divided into two groups according to body weight. Each group was fed the diet containing 2% cholesterol or the normal diet, respectively for 4 weeks. Each group was again divided into two sub-groups and they were fed with the diet containing 5% of sucrose and fructooligosaccharide, respectively for 8 weeks. The number of bifidobacteria slight increased, but not significantly, in oligosaccharide groups. Plasma total lipid concentration in cholesterol group was signficantly increased compared to the value in normal group. Dietary oligosaccharide decreased plasma total lipid concentration and triglyceride concentrations in normal group, but not in cholesterol group. Fecal lipid excretion was higher in cholesterol group than in normal group. Fecal cholesterol concentration in cholesterol-oligosaccharide group was significantly increased compared to other groups. Cholesterol had slight effects on TBARS and the immune status, but dietary oligosaccharide had no effects on these variables. The results in this study suggest that dietary oligosaccharide had no effect on serum and liver profiles of rats fed high cholesterol diet but increases the fecal cholesterol excretion.

  • In Med Dept. Daehan Education Insurance, Total 690 cases(male 376 cases, female 314 cases) were inspected to study the change of serum lipid value. The objects of investigation were total cholesterol, HDL cholesterol, triglyceride, phospholipid and total lipid value. The results are as follows: 1) In the normal subjects, with the relation of serum lipid value and the age. triglyceride, phospholipid and total lipid value showed the increasing tendency by 70 age, but total cholesterol value and HDL cholesterol value showed the increasing tendency by 59 age and the decreasing tendency after 59 age. 2) In the normal subject the difference of serum lipid value between sexes did not make the significant difference of level statistically in all items which can be inspected. 3) The weight change and serum lipid value are very significantly related. Total cholesterol, HDL cholesterol and total lipid value are high in female if the weight is light. But they are high in male the weight is heavy phospholipid value is opposite to the above result. Triglyceride value is high in male regardless the weight, but the significant difference happened in the high weight group. 4) As serum lipid value of hypertension patients is compared with that of the normal subjects, HDL cholesterol, triglyceride, phospholipid and total lipid value are significantly high. 5) The doubtful group of the fatty infittration of liver among the hypertension pafients have very significant difference in serum lipid value compared with not only the value of the normal subject but also that of the obesity group. 6) The doubtless group of the fatty infitration of liver among the hypertension patients have the significant difference in all serum lipid value compared with that of the normal subject, but compared with the obesity group males had the significant difference of HDL cholesterol value and both sexes had the significant difference in total lipid value, but had no significant difference in total cholesterol, triglyceride and phosprolipid value. 7) The ratio of total cholesterol per HDL cholesterol had the significant difference in the doubtful hypertension patients of the fatty infittration of liver and the relative weight, but it had no significant difference compared with the normal subject in the doubtless hypertension patients.

Effect of Cholesterol and Serum Albumin on Sperm Ability and Lipid Peroxidation during the Storage of Miniature Pig Sperm (미니돼지정액의 보존 시 콜레스테롤과 혈청 알부민이 정자 성상과 지질 과산화에 미치는 영향)

  • This study was undertaken to find out the effect of cholesterol and serum albumin on sperm ability and lipid peroxidation levels period to the liquid storage of miniature pig sperm. Ejaculated semen from miniature pigs was collected by gloved-hand method into a pre-warmed ($37^{\circ}C$) thermos bottle, and extended with Modena solution {with and without BSA, methyl-beta-cyclodextrin (-cholesterol) and cholesterol loaded cyclodextrin (+cholesterol)}. Each semen was assessed for viability (SYBR-14/PI staining) and acrosome intactness, intensity and capacitation status by chlorotetracycline (CTC) staining at 1, 3, 5, 7 and 10 days of storage. At for the effects of cholesterol and serum albumin on lipid peroxidation, semen were incubated with $H_2O_2$ ($10\;{\mu}M$), and lipid peroxidation level were measured by flow cytometry using the lipid peroxidation reporter probe $C_{11}-BODIPY^{581/591}$. The result, lipid peroxidation level in sperm added with cholesterol were lower in $10\;{\mu}M$ $H_2O_2$ compared to the added sperm with serum albumin. Also, added cholesterol to sperm had significant (p<0.05) higher viability when storage for 7 and 10 days and lower when 10 days of storage percentage of acrosome-reacted sperm (AR pattern) in acrosome state as say result compared to other treated groups. In conclusion, role of cholesterol during lipid storage in miniature pig spermatozoa was protected boar spermatozoa from lipid peroxidation prior to lipid storage. Addition serum albumin during lipid storage in sperm may be induce sperm membrane damage by lipid peroxidation. Therefore, addition of cholesterol to miniature pig sperm will be lead to extension of liquid storage periods.

Effects of Factors on Serum Lipid Levels in Suwon Adult Males (수원지역 성인 남성들의 혈청지질 수준에 영향을 미치는 요인)

  • This study was performed to find and compare the characteristics of dietary factors and obesity indices on serum lipid profiles using 407 healthy adult males that had physical examinations conducted at the general hospital located in Suwon. The subjects were divided 56.3% for the normal group, 16.7% for TG, 14.5% for TC and 12.5% for TC TG group, respectively. When comparing the blood lipid profiles to normal group, TC group was high in both total cholesterol and LDL-cholesterol, TG group was high in triglyceride and low in HDL-cholesterol, TC TG group was high in triglyceride, total cholesterol, LDL-cholesterol and low in HDL-cholesterol. In comparison of blood lipid profiles with normal group, among anthropometric factors, body weigh, BMI (Body Mass Index) and WHR (Waist Hip Ratio) showed significant differences. TC group was high in both total cholesterol and LDL-cholesterol, and TG group was high in triglyceride and low in HDL-cholesterol. TC TG group was high in triglyceride, total cholesterol and LDL-cholesterol, and was low in HDL-cholesterol. Body weigh of TG group was significantly higher than that of normal group (p<0.05). BMI and WHR of TG group and TC TG group were significantly higher than those of normal group (p<0.05). Total cholesterol was mainly related to the intake of animal lipids and alcoholic beverage. Serum triglyceride levels were closely related with lipid intake and high BMI in this group. In TC TG group, BMI and WHR were high. As well, serum LDL-cholesterol was high and HDL-cholesterol was low which indicates TC TG group had the most undesirable blood lipid profiles. Therefore, it may be necessary to manage BMI and WHR to prevent hyperlipidemia and obesity for adult males in Suwon.

Effect of Chitosan and N. O-Carboxymethyl Chitosan of Different Sources and Molecular Weights on Lipid Metabolism (급원과 분자량이 다른 Chitosan 과 N, O-Carboxymethyl Chitosan이 지방대사에 미치는 영향)

  • This study was performed to investigate the effect of chitosan and NOCC from different sources and of different molecular weights on lipid metabolism. Sprague-Dawley rats were blocked into 26 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400 ppm in diet. Various chitosan and NOCC sources were given at the level of 0%(w/w) of diet. Total serum cholesterol and serum triglyceride concentrations were little affected by chitosan and NOCC supplements. However, HDL-cholesterol concentration and HDL : total cholesterol ratio were increased , and liver lipid, cholesterol , and triglyceride concentration were decreased by chitosan and NOCC supplements. The cholesterol and lipid lowering activity depends on fiber sources(crab and shrimp); type (chitosan and NOCC); and molecular weight(low , medium , and high). Among cadmium-free groups, chitosan-fed groups showed greater activity than NOCC -fed groups in lowering cholesterol and lipid levels, and greater fecal excretion of lipids and bile acids. Crab chitosans were more effective in cholesterol and lipid lowering activity than shrimp chitosans. The group fed high molecular weight crab chitosan showed the highest fecal excretion of lipids and bile acids. This indicated that high molecular weight crab chitosan was most effective in interfering with cholesterol and lipid absorption.

A Retrospective Study on the Effect of Daeshiho-tang on the Lipid Profile in Patients with Uncontrolled Dyslipidemia by Statins (스타틴으로 조절되지 않는 이상지질혈증 환자에게 대시호탕 복용이 미치는 영향)

  • Objective: This study was conducted to investigate the lipid-lowering effect and safety of Daeshiho-tang in patients with uncontrolled lipid levels by statins. Methods: We investigated patients who had an abnormal lipid profile even when taking statins and who were administered Daeshiho-tang at Kyung-Hee University Korean Medical Hospital for at least one day between January 2008 and December 2018. Their basal characteristics and examinations were reviewed retrospectively with respect to lipid profile, AST, ALT, GGT, BUN, and creatinine. The lipid profile was composed of total cholesterol, triglyceride, LDL-cholesterol, and HDL-cholesterol. Subgroup analysis was performed on each component of dyslipidemia. Results: Among 20 participants, there were 10 males and 10 females. The mean BMI was 23.52. Eighty five percent of the participants were diagnosed as having cerebral infarction. After the administration of Daeshiho-tang, total cholesterol and LDL cholesterol were significantly reduced, to 41.3 mg/dl and 33.95 mg/dl, respectively. In subgroup analyses, total cholesterol and LDL cholesterol were significantly decreased, to 63 mg/dl and 54.6 mg/dl, respectively. Liver and kidney function showed no significant difference after taking Daeshiho-tang. Conclusions: Daeshiho-tang as a decoction or powder had significant lipid-lowering effects on total cholesterol and LDL cholesterol in patients with dyslipidemia. The lipid-lowering effect on total and LDL cholesterol increased in patients with hypercholesterolemia and hyper-LDL-cholesterolemia, respectively. Based on the minimal changes in the liver and kidney function test, Daeshiho-tang would be safe enough to be used in clinics.

Effects of Dietary Lobster Shrimp Chitosan on Lipid Metabolism in Diet-Induced Hyperlipidemic Rats (가시발새우 키토산 식이가 식이성 고지혈증 흰쥐의 지질대사에 미치는 영향)

  • This study was designed to investigate effects of lobster shrimp chitosan on serum and liver lipid levels in rats fed high fat and cholesterol diets. Male Sprague-Dawley rats of 4-weeks old, weighing 70$\pm$5g, were fed experimental diets for 3 weeks: basal diet+5% cellulose(group 1), high lipid diet+5% cellulose(group 2, control group), high lipid diet+5% balloon flower root(group 3), high lipid diet+5% cellulose(group 4, high lipid diet+5% lobster shrimp chitosan(group 5). Concentrations of total cholesterol, HDL-cholesterol, LDL-cholesterol, free cholesterol, cholesteryl ester, triglyceride, phospholipid in serum and liver were determined. The results obtained were as followes: Serum total cholesterol concentration were significantly reduced in all supplemented group compared with control. HDL-cholesterol concentrations were higher in groups 1, 4, and 5 and the ratios of HDL-cholesterol to total cholesterol concentration were higher in groups 1, 4, while the atherogenic-indices were lower in groups 1, and 4 than control. Concentrations of LDL, LDL-cholesterol and free cholesterol, cholesteryl ester were significantly lower in all supplemented groups than the control group. Concentrations of phospholipid and triglyceride were lower in groups 3, 4, 5, while activities of GPT and GOT were significantly lower in groups 4, 5 than the other group. Concentrations of calcium, magnesium and phosphorous were high in all treatment groups compared to the control group. Liver contents of total cholesterol, free cholesterol and cholesteryl ester were lower in the groups 1, 3, 4, and 5 than in the control group, especially lower in groups 1, and 5. Liver triglyceride and phospholipid concentrations were significantly lower in the groups 1, and 5 than in the other groups. The results from the data suggested that the experimental diet containing 5% lobster shrimp chitosan was effective to reduce the serum and liver lipids.

Fruiting Body Extracts of Paecilomyces tenuipes Ameliorate Lipid and Antioxidant Metabolism in Rats Fed a High Fat-Cholesterol Diet

  • The present study examined the lipid lowering and antioxidant activity of Paecilomyces tenuipes. Eight week-old male Sprague-Dawley rats were fed one of the three diets, a reference diet without cholesterol addition (NC), a high fat (17 g/ 100 g)-high cholesterol (1 g/100 g) diet (HC) and a HC diet supplemented with 3% P. tenuipes (PT) for 30 days. Total lipid and total cholesterol were reduced significantly by 33 and 37%, respectively, in the PT diet group compared with controls. A similar reduction was found for low density lipoprotein (LDL) cholesterol levels, while plasma high density lipoprotein (HDL) cholesterol and triglyceride (TG) concentrations were not significantly different among groups. Hepatic total lipid and total cholesterol levels, but not hepatic TG levels, were significantly decreased in the PT group compared to the HC group. The administration of P. tenuipes increased the plasma total antioxidant potential and decreased the levels of lipid peroxidation. These results suggest that P. tenuipes exerts significant health benefits through the modulation of physiological functions including a variety of atherogenic lipid profiles and antioxidants in hypercholesterolemia.

Preventive Effect of $\beta$-Glucan on the Experimental Atherosclerosis in Rats (랫트의 실험적 동맥경화증에 대한 $\beta$-Glucan의 예방 효과)

  • The present studies were undertaken in attempt to investigate the preventive effect of $\beta$-glucan from barley ad diltiazem on cholesterol and vitamin D2 induced-atherosclerotic rat. The results obtained were summerized as follows. 1.The group, fed only the mixture of cholesterol and vitamin D2, showed significant increase of calcium, inorganic phosphorus, total cholesterol, lipid LDL-cholesterol and phospholipid in serum, and total lipid in the liver (p<0.05) as comparing with normal group. The aorta showed severe damage of disorganization, necrosis and lipid deposition in the elastic membrane. 2. The group fed mixture of cholesterol and vitamin D2 plus diltiazem simultaneously, showed significant increase of total cholesterol, total lipid and phospholipid in serum, and total lipid and triglyceride n the liver (p<0.05) as comparing with normal group, but the significant decrease of calcium and inorganic phosphorus in serum(p<0.05) as comparing with the atherogenic control group. The aorta showed slight damage of elastic membrane and lipid deposition as comparing with the atherogenic control group. 3.The group, fed mixture of cholesterol and vitamin D2 puls $\beta$-glucan simultaneously, showed significant decrease of total cholesterol, LDL and VLDL-cholesterol, total lipid, phospholipid and triglyceride in serum, and total lipid in the liver as comparing with the atherogenic control group(p<0.05), but the significant increase of calcium in serum as comparing with the normal group(p<0.05). The aorta showed no changes in elastic fiber and no lipid deposition in comparing with the atherogenic control group.
