Immobilization of Lipases on Amberlite and Their Interesterification Reaction Characteristics (Amberlite에 고정화된 Lipase 제조 및 효소적 Interesterification을 이용한 반응 특성 연구)

  • Immobilized lipases were prepared by physical adsorption using lipase AK, AY, AH, PS and R on Amberlite$^{(R)}$XAD$^{(R)}$7 HP resin. With the immobilized lipases (10%), structured lipid was synthesized by enzymatic interesterification of canola oil, palmitic ethyl ester, and stearic ethyl ester in order to study the reaction characteristics. Among the lipase, the highest protein content was obtained from lipase AH (11.41%) before immobilization, while the highest levels of bound protein was observed from immobilized lipase AK (63.91%). Immobilized lipase AK had the highest interesterification activity (38.3% of total saturated fatty acid). Lipase AK was also used for a continuous reaction in which the slow flow of reactant resulted in increased reaction rate. Reusability of immobilized AK, AH and PS increased at the second reaction (120-196.5%). However, the activity of immobilized AK, which had the highest bound protein content (63.91%) decreased after the third reaction, while the activity of immobilized AH and PS was maintained until the sixth reaction.

Studies of interesterification properties of irradiated lipases (방사선 조사선량에 따른 lipase의 interesterification 반응특성 변화 연구)

  • This study was conducted to prove the effect of irradiation on lipases (lipase AK, lipase AH, lipase PS-D, Lipozyme TLIM, Lipozyme RMIM and Novozyme SP435) which were used for interesterification reaction using batch type reactor. Through such interesterification, structured lipid (1(3)-palmitoyl-2-oleoyl-3(1)-stearoyl, POS) was synthesized by lipase treated with irradiation at different doses (0, 3, 7, 14, 29 and 59 kGy) using canola oil, palmitic ethyl ester (PEE) and stearic ethyl ester (StEE). After the reaction, fatty acid composition of triacylglycerol (TAG) in structured lipid was analyzed to compare the lipase activity. The results showed that activity of the irradiated lipase AH, PS-D and Novozyme SP435 with certain dose (3 kGy) were slightly improved. Such change of lipase activity suggested that irradiation might affect on the interesterification properties. Especially, Lipase AK, Lipozyme TLIM and Lipozyme RMIM after at 3 kGy irradiation showed that content of stearic acid ($C_{18:0}$) was increased while palmitic acid ($C_{16:0}$) decreased in the interesterified products.

Identification of Alga-lytic Bacterium AK-07 and Its Enzyme Activities Associated with Degradability of Cyanobacterium Anabaena cylindrica (Anabaena cylindrica 분해세균 AK-07의 동정과 분해 관련 효소활성 조사)

  • To investigate bacteria with algal Iytic activities against Anabaena cylindrica when water blooming occurs and to study enzyme profiles of alga-Iytic bacteria, various bacterial strains were isolated from surface waters and sediments in eutrophic lakes or reservoirs in Korea. Abacterial strain AK-07 was characterized and identified as Acinetobacter johnsonii based on its16S rDNA base sequence. When AK-07 was co-cultivated with A. cylindrica, bacterial cells propagated to $8\;{\times}\;10^8$ cfu $ml^{-1}$ and Iyses algal cells. However, culture filtrates of AK-07 did not exhibit algal Iytic activities. That suggesting the enzymes on the surfaces of the bacterium might be effective algal Iytic agents to cause Iyses of cells. Acinetobacter johnsonii AK-07 exhibited high degradation activities against A. cylindrica, and formed alginase, caseinase, lipase, fucodian hydrolase, and laminarinase. Moreover, glycosidases for example ${\beta}$-galatosidase, ${\beta}$-glucosidase, ${\beta}$-glucosaminidase, and ${\beta}$-xylosidase, which hydrolyzed ${\beta}$-0-glycosidic bonds, were found in cell-free extracts of A. johnsonii AK-07. Other glycosidase such as ${\alpha}$-galctosidases, ${\alpha}$-N-Ac-galctosidases, ${\alpha}$-mannosidases, and ${\alpha}$- L-fuco-sidases, which cleavage ${\alpha}$-0-glycosidic bondsare not detected. In the results, enzyme systemsof A. johnsonii AK-07 were very complex to do-grade cell walls of cyanobacteria. The polysaccharides or peptidoglycans of A. cylindrica maybe hydrolyzed and metabolized to a range of easily utilizable monosaccharides or other low molecular weight organic substances by strain AK-07 of A. johnsonii.

  • Pseudomonas sp. lipase (lipase AK) catalyzed transesterification reaction between fructose and vinyl laurate in anhydrous pyridine. The product of this process was identified as monoester of fructose and vinyl laurate. The synthetic product has been found to be an excellent emulsifier. The synthetic bioemulsifier showed a good emulsification activity and stability in comparison with other commercial emulsifiers, and good emulsification activity on various emulsifying substrates.

Synthesis of Fructose Ester Compound by Lipase in Organic Solvent (유기용매계에서 Lipase에 의한 Fructose Ester의 합성)

  • Sugar ester compounds were synthesized in organic solvent using lipase. Anhydrous pyridinc was selected as ;I solvent because of reasonable solubility of sugar. The synthesis of sugar ester compound was catalyzed by Pseudomonas sp. lipase in the reaction system containing anhydrous pyridine as .i solvent and vinyl butylate as an acyl donor. The analysis of the reaction product by TLC and GC showed thilt monobutyryl and dibutyryl fructose esters were synthesized by transesterification reaction between fructose and vinyl butyrate. Optimal conditions for the transesterification reaction were as follows: the ratio of fructoselvinyl butylate, I : lO(M : M): reaction temperature, 40^{\circ}C.$, velocity of shaking, 150 rprn: concentration of enzyme, 10 mglml. The longer the reaction period, the higher the conversion rate, and the conversion rate reached up to 90% after about 10 days of reaction. Monobutyryl fructose was mainly synthesized in the early stage of reaction, but the amount of dibutyryl fructose increased gradually as the rcdction progressed. When a small amount of water was added to the reaction mixture (micro-water system), the reaction rate decreased, while that of rnonobutyr~l fructosc increased. Only monobutyryl fructose was obtained when 1% water was added to the reaction mixture.

Enzyme Profiles of Alga-Lytic Bacterial Strain AK-13 Related with Elimination of Cyanobacterium Anabaena cylindrica

  • To investigate bacteria with algalytic activities against Anabaena cylindrica when water blooming occurs and to study enzyme profiles associated with alga-lytic activity, various bacterial strains were isolated from surface waters and sediments in eutrophic lakes or reservoirs in Korea. Among 178 isolates, only nine isolates exhibited lytic abilities against A cylindrica on the agar plates, and then the isolate AK-13 was selected as the strongest in lysing the cyanobacterium A. cytindrica. The strain AK-13 was characterized and identified as Sinorhizobium sp. based on fatty acid methyl ether profiles and 16S rDNA sequence. According to the results of the enzyme assays, in the strain An-13 of Sinorhizobium sp., alginase, amylase, proteinase (caseinase and gelatinase), carboxymethyl-cellulase (CMCase), laminarinase, and lipase was produced, namely CMCase, laminarinase and protease were highly active. None of glycosidase was produced. Therefore, enzyme systems of Sinorhizobium sp. AK-13 were very complex to degrade cell walls of A. cylindrica. The peptidoglycans of A. cylindrica mat be hydrolyzed and metabolized to a range of easily utilizable monosaccharides or other low molecular weight organic substances by Sinorhizobium sp. AK-13.

Synthesis of $\beta$-Sitosterol Esters with Conjugated Linoleic Acid and Medium Chain Fatty Acids by Using Lipase as Catalyst

  • Plant steryl esters have good effects on plasma cholesterol level and are used as functional food ingredient. Conjugated linoleic acid (CLA) presents mainly in animal foods and has a good benefit and medium chain fatty acids (MCFAs) are a rapid energy source for human. In this study, we produced the ${\beta}$-sitosterol esters from CLA and MCFAs using various lipases as catalysts. Among lipases, AYS (from Candida rugosa) was the most effective for synthesis of ${\beta}$-sitosterol esters in the presence of water (24.35% conversion) or hexane (25.33% conversion). The second esterification extent was obtained by lipase AK (from Pseudomonas sp), showing 10.26% conversion in water and 15.94% conversion in hexane, respectively. The reaction condition was 1:3 molar ratio (${\beta}$-sitosterol:fatty acid, 1:3) and stirred (175 rpm) at 55$^{\circ}C$ in water bath shaker for 48h.
