Isolation of Alliin in Garlic and Its Quantitative Determination by High Performance Liquid Chromatography and Studies on the Antimicrobial Efforts of Alliin and Ethanol Extracts from Korean Garlic(Alliium sativum L.) (마늘 중 고속 액체 크로마토그래피에 의한 알린의 분리 및 정량과 Alliin과 에탄올 추출물의 항균효과에 관한 연구)

  • First. the purification and analysis of alliin in garlic from different origins by alliin-HPLC determination method were studied. Allinase in garlic was inactivated by heating in boiling water followed by extraction of alliin in garlic with 80% methanol. To remove free amino acids and alliin homologs in garlic, garlic extract was separated by cation exchange column which was packed with amberlite CG-120 resin using 40L d-water as eluent. Alliin in garlic extract was crystallized in a mixture of acetone (50$^{\circ}C$):H$_2$O:acetic acid=70:29:1 and then recrystallized in a mixture of acetone (50$^{\circ}C$):H$_2$O:acetic acid=75:24:1. Obtained alliin was identified by melting point. TLC, microscope observation and mass spectrometry. High performance liquid chromatography (HPLC) following pre-column derivatization of cystein derivatives with o-phthaldialdehyde/2-mercaptoethanol has succeessfully been applied to the analysis of various garlics. Each alliic of standard solution and garlic extract was derivatized to isoindole derivative by o-phthaldialdehyde /2-mercaptoethanol and then analyzed by HPLC. Six point calibration was done by using alliin peak area. Lineality was observed at 0 ∼ 1.0mg/ml of alliin concentration. Weighted regression line function was Y=6254X - 256077. By this function, alliin contents in various garlics were 0.34 ∼ 0.73% fresh weight. Second study was designed to evaluate the effects of garlic extracts of various concentrations on the growth of various pathogenes (Eubacterium limonsum, Bacteroides fragilis, Salmonella typhimurium, Salmonella typhi, Shigella sonnei, Kiebsiella pneumoniae, Enterobacter cloacae, Pserdomonas aeruginosa, Escherichia coli). For antimicrobial effects against microorganism, totally minimal inhibition concentrations (MIC) of alliin were from 5,000 to 20,000ppm. MIC of ethanol extract were 1,250 to 10,000ppm.

  • In diffusion process exhaust line during semiconductor manufacturing process, In order to improve the transportation efficiency in the piping by removing "The reaction by-product, $ZrO_2$ and The unreacted material, TEMAZ, TMA, $O_3$, etc" and "Powder being deposited", the piping temperature was raised to $80^{\circ}C$ or more by using the heater jacket, and the bellows at the rear end of the vacuum pump ruptured. So conducted a case study and try to prevent the similar accidents from occurring through case studies. The causes of the accident were analyzed as follows: the inflow of outside air due to the generation of a gap on the suction side of the vacuum pump and heating the pipe with the heater jacket resulted in the overpressure in the pipe due to the volumetric expansion of the gas generated by decomposition of the unreacted TEMAZ, It can be assumed that the most vulnerable bellows of the piping has been ruptured. In order to prevent such accidents, This study is aimed to identify the cause of pipeline rupture accident and to establish safety measures for the prevention of similar accidents by evaluating physical hazards of TEMAZ, which is assumed to be the cause of pipe rupture accident.

  • 자연에너지원중 태양에너지를 이용한 자갈축열 태양열 온실의 난방에너지 절감효과를 분석하기 위하여 야간에 내부설정온도를 18℃로 했을 때의 자갈축열 태양열 온실과 동일한 제원의 대조온실의 난방연료소비량을 비교하였다. 자갈축열 태양열 온실의 경우 난방초기에는 연료가 대조온실에 비해 약 10%정도 더 소모되었으나 2일이 경과한 후에는 점차 난방연료소비량이 감소하였다(Fig. 2) 이러한 결과는 자갈축열 태양열 온실의 경우 주간에 온실에 투여되는 일사에 의해 축열이 이루어 질뿐만 아니라 야간에 난방을 할 경우에도 축열층의 온도가 설정온도에 도달할 때까지 축열이 이루지기 때문인 것으로 분석되었다 (Fig. 1). 3월 10일부터 3월 15일까지 6일간의 난방연료소비량이 대조온실은 167℃였으나, 자갈축열 태양열 온실은 109℃로써 대조온실에 비해 약 35%정도 난방연료 절감효과가 있는 것으로 분석되었다(Fig. 2). 6일간의 난방 후 자갈축열 태양열 온실에 난방을 중단한 결과 온실외부의 최저기온이 -2.4℃일 때 자갈축열 태양열 온실의 내부최저온도는 14℃를 유지하였으며, 이후 계속 난방을 수행하지 않은 상태에서도 주간에 축열효과로 인하여 최저외기온이 3℃전후일 때 자갈축열 태양열 온실의 내부온도는 15℃를 유지하였으나 대조온실은 5℃ 전후였다. 그리고, 일기온차가 심한 3월말에는 야간설정온도가 18℃인 경우에 자갈축열 태양열 온실의 난방연료소비량이 대조온실에 비해 월등히 적음을 알 수 있었다(Fig. 1, 2). 3월에 자갈축열 태양열 온실의 난방연료소비량은 대조온실에 비해 약 50%이상의 절감효과가 있는 것으로 나타났다./TEX>3.1cm, 2cm$\times$4.2cm 순으로 나타났다. 5. 저고리의 옆길이 곡선에 대한 평가는 진종의 중심인 겨드랑이가 5cm인 것이 가장 우수한 것으로 나타났으며, 그 다음으로 4cm, 6cm, 3cm 순으로 나타났다. 6. 이에 위의 항목들을 종합하여 제작한 연구저고리는 등길이가 28cm, 어깨선 위치는 1.75cm, 깃크기는 5cm$\times$21.5cm, 소매통 크기는 3cm$\times$4.3cm, 옆길이 곡선은 5cm로써 그 형태에 따른 신체적합성에 대한 외관 관능검사 결과로 3.83의 평균값으로 나타났고, 심미성에 대한 외관 관능검사를 항목별 평균치 값으로 종합하면 4.00의 값으로 나타났다. 또한 각 부위별 동작적합성 관능검사는 7가지 동작을 부위별 항목 평균치 결과 3.95의 유수한 저고리로 평가되었다. 본 연구 실험 결과 앞으로도 전통 저고리를 피복함에 있어 외관과 동작적합성이 좋은 저고리를 만들기 위해 지속적 연구가 요청된다. turned back than the Korean women's. CHEMICAL PROPERTIES OF CORES IN DIFFERENT ENVIRONMENTS; THE ORION A, B AND λ ORIONIS CLOUDS

  • We observed 80 dense cores ($N(H_2)$ > $10^{22}cm^{-2}$) in the Orion molecular cloud complex which contains the Orion A (39 cores), B (26 cores), and ${\lambda}$ Orionis (15 cores) clouds. We investigate the behavior of the different molecular tracers and look for chemical variations of cores in the three clouds in order to systematically investigate the effects of stellar feedback. The most commonly detected molecular lines (with the detection rates higher than 50%) are $N_2H^+$, $HCO^+$, $H^{13}CO^+$, $C_2H$, HCN, and $H_2CO$. The detection rates of dense gas tracers, $N_2H^+$, $HCO^+$, $H^{13}CO^+$, and $C_2H$ show the lowest values in the ${\lambda}$ Orionis cloud. We find differences in the D/H ratio of $H_2CO$ and the $N_2H^+/HCO^+$ abundance ratios among the three clouds. Eight starless cores in the Orion A and B clouds exhibit high deuterium fractionations, larger than 0.10, while in the ${\lambda}$ Orionis cloud, no cores reveal the high ratio. These chemical properties could support that cores in the ${\lambda}$ Orionis cloud are affected by the photo-dissociation and external heating from the nearby H II region. An unexpected trend was found in the $[N_2H^+]/[HCO^+]$ ratio with a higher median value in the ${\lambda}$ Orionis cloud than in the Orion A/B clouds than; typically, the $[N_2H^+]/[HCO^+]$ ratio is lower in higher temperatures and lower column densities. This could be explained by a longer timescale in the prestellar stage in the ${\lambda}$ Orionis cloud, resulting in more abundant nitrogen-bearing molecules. In addition to these chemical differences, the kinematical difference was also found among the three clouds; the blue excess, which is an infall signature found in optically thick line profiles, is 0 in the ${\lambda}$ Orionis cloud while it is 0.11 and 0.16 in the Orion A and B clouds, respectively. This result could be another evidence of the negative feedback of active current star formation to the next generation of star formation.

  • The important results which have been obtained in the investigation can be recapitulated as follows. 1. As demonstrated by the experimental results and analyses concerning their effects in the on-ground type mushroom house, the constructions in relation to the side wall and ceiling of the experimental house showed a sufficient heat insulation on effect to protect insides of the house from outside climatic conditions. 2. As the effect on the solar type experimental mushroom house which was constructed in a half basement has been shown by the experimental results and analyses, it has been proved to be effective for making use of solar heat. However there were found two problems to be improved for putting solar house to practical use in the farm mushroom growing: (1) the construction of the roof and ceiling should be the same as for the on ground type house, and (2) the solar heat generating system should be reconstructed properly. 3. Among several ventilation systems which have been studied in the experiments, the underground earthen pipe and ceiling ventilation, and vertical side wall and ceiling ventilation systems have been proved to be most effective for natural ventilation. 4. The experimental results have shown that ventilation systems such as the vertical side wall and underground ventilation systems are suitable to put to practical use as natural ventilation systems for farm mushroom house. These ventilation systems can remarkably improve the temperature of fresh air which is introduced into the house by heat transfers within the ventilation passages, so as to approach to the desired temperature of the house without any cooling or heating operation. For example, if it is assuming that X is the outside temperature and Y is the amount of temperature adjustment made by the influence of the ventilation system, the relationships that exist between X and Y can be expressed by the following regression lines. Underground iron pipe ventilation system. Y=0.9X-12.8 Underground earthen pipe ventilation system. Y=0.96X-15.11 Vertical side wall ventilation system. Y=0.94X-17.57 5. Optimal Operation of Gas Engine for Biogas Plant in Sewage Treatment Plant (하수처리장 바이오가스 플랜트의 가스엔진 최적 운영 방안)

  • The Korea District Heating Corporation operates a gas engine generator with a capacity of $4500m^3 /day$ of biogas generated from the sewage treatment plant of the Nanji Water Recycling Center and 1,500 kW. However, the actual operation experience of the biogas power plant is insufficient, and due to lack of accumulated technology and know-how, frequent breakdown and stoppage of the gas engine causes a lot of economic loss. Therefore, it is necessary to prepare technical fundamental measures for stable operation of the power plant In this study, a series of process problems of the gas engine plant using the biogas generated in the sewage treatment plant of the Nanji Water Recovery Center were identified and the optimization of the actual operation was made by minimizing the problems in each step. In order to purify the gas, which is the main cause of the failure stop, the conditions for establishing the quality standard of the adsorption capacity of the activated carbon were established through the analysis of the components and the adsorption test for the active carbon being used at present. In addition, the system was applied to actual operation by applying standards for replacement cycle of activated carbon to minimize impurities, strengthening measurement period of hydrogen sulfide, localization of activated carbon, and strengthening and improving the operation standards of the plant. As a result, the operating performance of gas engine # 1 was increased by 530% and the operation of the second engine was increased by 250%. In addition, improvement of vent line equipment has reduced work process and increased normal operation time and operation rate. In terms of economic efficiency, it also showed a sales increase of KRW 77,000 / year. By applying the strengthening and improvement measures of operating standards, it is possible to reduce the stoppage of the biogas plant, increase the utilization rate, It is judged to be an operational plan.

Anti-Allergic Inflammatory Effect of Bacteria Isolated from Fermented Soybean and Jeotgal on Human Mast Cell Line (HMC-1) (장류 및 젓갈 분리 균주 추출물의 비만세포 매개 항염증효과)

  • The mast cell is one of the major effector cells in inflammatory reactions and can be found in most tissues throughout the body. Activated mast cells can produce histamine, as well as a wide variety of other inflammatory mediators such as eicosanoids, proteoglycans, proteases, and several pro-inflammatory cytokines, such as tumor necrosis factor (TNF)-${\alpha}$, and interleukins (IL-6), IL-8, IL-4, IL-13. In the present study, we isolated two bacterial strains (J80 and G147) from fermented soybean and Jeotgal, and investigated the inhibitory effects of their extracts which were prepared by several pretreatment methods (sonication for 20 min, heating at $100^{\circ}C$ for 30 min, autoclaving at $121^{\circ}C$ for 15 min) on the mast cell-mediated inflammatory response. The pretreated bacterial extracts had no cytotoxicity against Human Mast Cell (HMC-1). Among various pretreatments, the extracts treated at $100^{\circ}C$ showed highest inhibition of histamine release (J80, 28.46%; G147, 41.14%). The J80 and G147 extracts treated at $100^{\circ}C$ resulted in the inhibition of IL-6 secretion by 38.46% and 56.45%, respectively. The J80 extract treated at $100^{\circ}C$ resulted in the inhibition of TNF-${\alpha}$ secretion by 66.67%, but G147 extract showed the highest inhibition effect by 41.1% when treated with sonication. These results suggest that bacterial extracts treated at $100^{\circ}C$ have a higher level of anti-inflammatory effects than other treatments such as sonication or autoclaving.
