Background: High risk human papillomaviruses (HPVs) are the leading cause of cervical cancer (CC) and Pap smear screening has not been successful in preventing CC in Tunisia. HPV vaccination that targets HPV16 and 18 offers a new efficient prevention tool. Identification of HPV types in CC is thus essential to determine the impact of HPV vaccine implementation. The aim of this study is to provide specific data from Tunisia. Materials and Methods: A total of 89 histological confirmed paraffin embedded samples isolated from patients with CC diagnosed between 2001 and 2011 were collected from five medical centres from Northern and Southern Tunisia. HPV DNA was detected using a nested PCR (MY09/MY11-GP5+/GP6+) and genotyping was assessed using a reverse blot line hybridisation assay that enables the detection of 32 HPV types. Results: HPV DNA was detected in all samples. Twelve high risk types were detected; HPV16 and/or 18 were predominant, accounting together for 92.1% of all the CC cases (HPV16: 83.1%). Single infections accounted for 48.8% of the cases and were mostly linked to HPV 16 (32.6%) and less frequently to HPV 18 (2.4%). The other high risk HPV single infections were linked to HPV 35 (4.6%), 45 (4.6%), 58 (2.3%) and 59 (2.3%). Multiple infections with mixing of 2 to 4 genotypes predominately featrued HPV16 and/or 18 with HPV 35 and 45 (96.6 %) and less frequently with HPV 59, 40, 66, 73 and 58. There was no statistically significant variation in the relative distribution of HPV types with age. Conclusions: These results strongly indicate that prophylactic HPV vaccines can have a major impact in preventing CC in Tunisia.

To develop rice cultivars with increased biomass and grain yield, superior lodging resistance is an essential trait. The new breeding approach can be adopted for the improvement of stem lodging resistance by enhancing culm strength. The resistance to breaking type lodging is attributed to bending moment of basal culm (M), which is composed of the section modulus (SM) and bending stress (BS). The resistance to the bending type lodging is attributed to flexural rigidity (FR) of stem, which is composed of the secondary moment of inertia (SMI) and Young's modulus (YM). Starch and cell wall components such as cellulose, hemicellulose and lignin also play a significant role in physical strength of culm, and thus affect lodging. Leaf Star has a superior lodging resistance due to its thick and stiff culm because of its high M and FR compared with Koshihikari. Furthermore, Leaf Star contains high densities of hemicellulose, cellulose and low lignin density in culm compared with Koshihikari. In this study, we performed QTL analysis for these traits associated with culm strength, using 94 recombinant inbred lines (RILs, $F_8$), derived from a cross between Leaf Star and Koshihikari. The SM in the RILs showed a continuous distribution. QTLs for SM were detected on chrs.2, 3 and 10. Leaf Star alleles increased SM on chrs. 2 and 3, but Koshihikari allele increased on chr.10. These QTLs overlapped with those QTLs identified using backcrossed inbred line derived from a cross between Chugoku 117 and Koshihikari, the parents of Leaf Star. The FR in Leaf Star was higher than that in Koshihikari due to the larger SMI and YM. 3 QTLs for SMI were detected on chrs.2, 3 and 10. Leaf Star alleles increased SMI on chrs.2 and 3, and Koshihikari alleles increased on chr.10. One QTL on chr.3 and two QTLs on chr.5 for hollocelulose content were detected with Leaf Star alleles contribution. Moreover, two QTLs were detected for hemicellulose density on chrs.3 and 5. Leaf Star allele increased hemicellulose density on chr.5, and Koshihikari allele increased on chr.3. Furthermore, two QTLs for cellulose density were detected on chr.5, and one QTL on chr.2. For starch content, one QTL on chr.3 and two QTLs on chr.5 with Leaf Star alleles contribution were detected. TULK-6 carrying a chromosome segment of Leaf Star on chr.5 in the Koshihikari genetic background showed higher densities of starch and hemicellulose than those in Koshihikari. These results suggest that the detected QTLs for culm strength could be utilized for the improvement of lodging resistance in rice by marker-assisted selection.

Information Centric Networking (ICN) has recently attracted great attention. It names the content decoupling from the location and introduces network caching, making the content to be cached anywhere within the network. The benefits of such design are obvious, however, many challenges still need to be solved. Among them, the local caching policy is widely discussed and it can be further divided into two parts, namely the cache permission policy and the cache replacement policy. The former is used to decide whether an incoming content should be cached while the latter is used to evict a cached content if required. The Internet is a user-oriented network and popular contents always have much more requests than unpopular ones. Caching such popular contents closer to the user's location can improve the network performance, and consequently, the local caching policy is required to identify popular contents. However, considering the line speed requirement of ICN routers, the local caching policy whose complexity is larger than O(1) cannot be applied. In terms of the replacement policy, Least Recently Used (LRU) is selected as the default one for ICN because of its low complexity, although its ability to identify the popular content is poor. Hence, the identification of popular contents should be completed by the cache permission policy. In this paper, a cache permission policy called Cache-Filter, whose complexity is O(1), is proposed, aiming to store popular contents closer to users. Cache-Filter takes the content popularity into account and achieves the goal through the collaboration of on-path nodes. Extensive simulations are conducted to evaluate the performance of Cache-Filter. Leave Copy Down (LCD), Move Copy Down (MCD), Betw, ProbCache, ProbCache+, Prob(p) and Probabilistic Caching with Secondary List (PCSL) are also implemented for comparison. The results show that Cache-Filter performs well. For example, in terms of the distance to access to contents, compared with Leave Copy Everywhere (LCE) used by Named Data Networking (NDN) as the permission policy, Cache-Filter saves over 17% number of hops.
The ecotoxicity tests for metal plating wastewater were conducted using Daphnia magna (D. magna) and Euglena agilis (E. agilis). Evaluation for sources of toxicity was performed by 1) Correlation analysis between the concentration of individual metals in the metal plating wastewater and the toxic effects on D. magna, 2) Toxicant identification evaluation methods including graduated pH method, EDTA procedure and sodium thiosulfate procedure, 3) Comparison of toxic effect value ($EC_{50}$ or $LC_{50}$) of individual metal on D. magna and it's concentration in the metal plating wastewater. To evaluate the possibility of E. agilis, a Korean domestic organism, as a test model organism for metal plating waste water, E. agilis toxicity test was also assessed using on-line euglena ecotoxicity system (E-Tox system). Based on toxicant characterization test using D. magna, it was expected that SS, oxidants and heavy metals are responsible for toxicity of metal plating waste water. Especially Cu, Hg, and Ag were the major cationic metals that caused toxicity. E. agilis is less sensitive than D. magna based on the $EC_{50}$ value however it shows prompt response to toxic test substances. E. agilis shows even a significant effect on the cell swimming velocity within 2 min to toxic metal plating wastewater. Our study demonstrates that E. agilis test can be a putative ecotoxicity test for assessing the quality of metal plating waste water.
In a previous paper (Kim et al., 1996a), the immediate 5' -flanking region and coding region of the human UDP-N -acetylglucosamine:-D-mannoside-1,4-Nacetylglucosaminyltransferase III (N-acetylglucosaminyitransferase- III; GnT-III) gene was reported, isolated and analyzed. Herein, we report on amplification of a new 5' -noncoding region of the GnT-III mRNA by single-strand ligation to single-stranded cDNA-PCR (5' -RACE PCR) using poly(A)+ RNA isolated from human fetal liver cells. A cDNA clone was obtained with 5' sequences (96 bp) that diverged seven nucleotides upstream from the ATG (+1) start codon. A concensus splice junction sequence, TCTCCCGCAG, was found immediately 5' to the position where the sequences of the cDNA diverged. The result suggested the presence of an intron in the 5' -noncoding region and that the cDNA was an incompletely reversetranscribed cDNA product derived from an mRNA containing a new noncoding exon. When mRNA expression of GnT-III in various human tissues and cancer cell lines was examined, Northern blot analysis indicated high expression levels of GnT-III in human fetal kidney and brain tissues, as well as for a number of leukemia and lymphoma cancer cell lines. Promoter activities of the 5' -flanking regions of exon 1 and the new noncoding region were measured in a human hepatoma cell line, HepG2, by luciferase assays. The 5'-flanking region of exon 1 was the most active, whilst that of exon 2 was inactive.

Sorghum seed is traditionally used as secondary food sources in addition to rice in Korea. While the hypoglycemia regulating phytochemicals have been found in sorghum seed, peptides related with hypoglycemia never been studied before. To obtain the peptide characteristics and the specifically high-expressed peptides in hypoglycemic sorghum seed, peptide profiles of seven hypoglycemic and five non-hypoglycemic sorghum lines bred in RDA were determined using surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF MS). The twelve sorghum lines exhibited 104 peptides on CM10 protein chip array (weak cation exchange) and 95 peptides on Q10 (weak cation exchange) in the molecular mass range from 2,000 to 20,000 Da. Heat map via supervised hierarchical clustering of the significantly different peptides (p < 0.01) in peak intensity among the 12 lines effectively revealed the specifically upregulated peptides in each line and distinguished between 7 hypoglycemic and 5 non-hypoglycemic lines. Through the comparison with hypoglycemic and non-hypoglycemic lines, 10 peptides including 2231.6, 2845.4, 2907.9, 3063.5, 3132.6, 3520.8, 4078.8, 5066.2, 5296.5, 5375.5 Da were specifically high-expressed in hypoglycemic lines at p < 0.00001. This study characterized seed peptides of 12 sorghums and found ten peptides highly expressed for hypoglycemic sorghum lines, which could be used as peptide biomarkers for identification of hypoglycemic sorghum.
Sclerotium rot of cucumber (Cucumis sativus L.) occurred at the experimental field of Gyeongsangnam-do Agricultural Research and Extension Services in July 2012. The typical symptoms included wilt, rot, and water-soaking on stems and fruits and severely infected plants eventually died. White mycelial mats spread over lesions, and then sclerotia were formed on fruit and near soil line. The sclerotia were globoid in shape, white to brown in color and 1-3 mm in size and the hyphal width was 4-8 ${\mu}m$. The optimum temperature for mycelial growth and sclerotia formation on PDA was $30^{\circ}C$. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. For further identification, the complete internal transcribed spacer (ITS) rDNA region was amplified and sequenced. On the basis of mycological characteristics, ITS rDNA region comparison, and pathogenicity to host plants, this fungus was identified as Sclerotium rolfsii Saccardo. This is the first report of sclerotium rot on cucumber caused by S. rolfsii in Korea.
Development of disease resistant plant is one of the important objectives in rice breeding programs because biotic stresses can adversely affect rice growth and yield losses. This study was conducted to identify lines with multiple-resistance genes to biotic stress among 173 hybrid rice breeding lines and germplasms using DNA-based markers. Our results showed that one hybrid rice line [IR98161-2-1-1-k1-3 (IR86409-3-1-1-1-1-1/IRBB66)] possessed 5 bacterial blight resistance genes (Xa4, xa5, Xa7, Xa13 and Xa21) while two hybrid rice lines [IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66) and 7292s (IR75589-31-27-8-33S(S1)/IR102758B)] possessed 3 bacterial blight resistance genes (Xa4, Xa7 and Xa21, and Xa3, Xa4 and xa5). Molecular survey on rice blast disease revealed that most of these lines had two different resistant genes. Only 11 lines possessed Pib, Pi-5, and Pi-ta. In addition, we further surveyed the distribution of insect resistant genes, such as Bph1, Bph18(t), and Wbph. Three hybrid breeding lines [IR98161-2-1-1-k1-3 (IR86409-3-1-1-1-1-1/IRBB66), IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66), and 7292s (IR75589-31-27-8-33S(S1) /IR102758B)] contained all three resistance genes. Finally, we obtained four hybrid rice breeding lines and germplasms [IR98161-2-1-1-k1-2 (IR86409-3-1-1-1-1-1/IRBB66), Damm-Noeub Khmau, 7290s, and 7292s (IR75589-31-27-8-33S(S1)/IR102758B)] possessing six-gene combination. They are expected to provide higher level of multiple resistance to biotic stress. This study is important for genotyping hybrid rice with resistance to diverse diseases and pests. Results obtained in this study suggest that identification of pyramided resistance genes is very important for screening hybrid rice breeding lines and germplasms accurately for disease and pest resistance. We will expand their cultivation safely through bioassays against diseases, pests, and disaster in its main export countries.
The root of Panax ginseng C.A. Meyer were extracted with 70% aqueous EtOH and the concentrates were partitioned into MeOH and H2O fractions using Diaion HP-20. The repeated SiO2 or octadecyl SiO2 column, and MPLC for the MeOH fraction led to isolation of four malonyl ginsenosides. The chemical structures of these compounds were determined as malonyl ginsenoside Rd (1) malonyl ginsenoside Rc (2) malonyl ginsenoside Rb2 (3) malonyl ginsenoside Rb1 (4) based on spectroscopic analyses including Nuclear magnetic resonance and HR-TOF/MS. The contents of malonyl ginsenoside Rb1 was highist as 5.44 mg/g of five years of ginseng. And malonyl ginsenoside Rd was lowest as 0.11 mg/g of six years of ginseng. Additionally, the malonyl ginsenoside Rd exhibited hepatoprotective effect against ethanol-induced hepatotoxicity in HepG2 cell line.
