Biosynthesis of L-Azetidine-2-carboxylic acid in actinoplanes ferrugineus

  • L-Azetidine-2-carboxylic acid(A-2-C) is a four-membered cyclic imino acid which was first discovered from Convalaria majalis and Polygonatum officinalis in 1955(1,2). The imino acid A-2-C has been identified in at least 16 species of plants (3) (mostly the families Liliaceae, Agavaceae and Amaryllidaceae); in two marine sponges (Haliclona sp. and Chalinospilla sp.) (4); in the red algae (Lophocladia lamenandi) (5); in the sugar beet Beta vulgaris (6) ; and the microorganism Actinoplanes ferrugineus (7).

  • A hitherto unknown substance was isolated from the commercial sample of Aloe arborescens, which was shown to be the 4',6'-O-ethylidene-aloenin, most likely an artifact caused by hot-air drying procedure for the product preparation.

  • Five known diosgenin glycosides have been isolated from the MeOH extract of Smilax china rhizomes and characterized as prosapogenin A of dioscin, dioscin, gracillin, methyl protogracillin, methyl protodioscin and its corresponding 22-hydroxy analog. This is the first isolation of the former four compounds from this plant. ${\beta}-Sitosterol\;glucoside$ was also isolated and identified.

  • The plants medicinal resources of Mt. Wol-chul were investigated 8 times from July, 1988 to July 1990. In orther to analyze the vegetation of Wol-chulmountain area, medical wild plants structure and distribution. Medical wildplants of Wol-chul mourltain consisted of 11'0 familis, 338 species in all. Theresources of important herb drugs were Polypodiaceae, Graminea, Liliaceae ,Polygonaceae, Ranunculaceae, Brassicaceae, Rosaceae, Apiaceae, Labiatae, Com-positae .

  • In the course of continuous work on the leaves of Allium tuberosum (Liliaceae), compounds 1 and 2 were isolated from the ethylacetate and butanol-soluble fraction, and identified as N-p-coumaroyl tyramine and bis(p-hydroxyphenyl) ether, respectively, on the basis of spectral data and physicochemical results.

  • A furostanol glycoside, $mp\;190{\sim}198^{\circ}$, was isolated from the MeOH extract of Smilax china rhizomes. The structure was established as a mixture of $26-O-{\beta}-_D-glucopyranosyl-(25R)-22-methoxy-furost-5-en-3\beta,26-diol\;3-O-\alpha-_L-rhamnopyranosyl-(1\rightarrow2)-\beta-_D-glucopyranoside$ and its 22-hydroxy derivative on the basis of spectral data and chemical correlations.

  • Rhizomes of Polygonatum species belong to Liliaceae are important herbal drugs in the traditional medical practice of Asian region. Two representative Chinese drugs derived from this genus are Hwangjeong and Okjuk. Though botanical origins of these drugs are officially listed as P. falcatum, P. sibiricum and P. kingianum for Hwangjeong and P. odoratum var. pluriflorum for Okjuk in the Korean Pharmacopoeia and Korean Herbal Pharmacopoeia, respectively, they are often sold as a mixture of several different species in the market.   • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var.platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of IS. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks (p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract of Mt. Odae eco-type showed more potent activity than that of Is. Ullung one. These results suggest that use of the fresh leaves may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.
