오미자 수집종별 리그난 함량의 연차간 차이 (Varietal and Yearly Differences of Lignan Contents in Fruits of Collected Lines of Schizandra chinensis Baillon)

  • 수집종의 리그난 분석결과 schizandrin, gomisin A, gomisin N의 평균함량은 1996년산 실생 6년생인 24개 계통에서 각각 평균 0.78%, 0.25%, 0.63%를 나타냈고, 1997년산 실생 7년생인 59개 계통에서는 각각 0.72%, 0.22%, 0.63%를 나타냈다. 리그난 함량은 수집종 및 연차간에 통계적으로 유의하고 범위가 큰 변이를 보였으며, 조사된 23개 계통 중 충주 143, 충주 453, 충주 532 계통은 비교적 높은 0.9% 이상의 3개년$(1995{\sim}1997)$ 평균 schizandrin 함량을 보였다. 연차별 schizandrin 함량의 변이계수가 낮으면서 0.8% 이상의 높은 함량을 나타낸 것은 충주 542, 충주 547, 충주 580 계통들이었다. 상관 분석에서 오미자 과실중과 리그난 함량과는 고도의 부의 상관을 보이고 리그난 성분들 간에는 고도의 정의 상관을 보였다.

팽화처리에 의한 오미자의 품질 특성 (A Quality Characteristic in Schisandra chinensis by Puff processing)

  • 본 연구에서는 팽화처리 된 오미자의 lignan 성분 함량과 항산화 활성 평가를 통해 가공에 따른 품질 특성을 비교하고자 하였다. 팽화오미자의 lignan 성분 함량은 대조구에 비해 모두 증가하였고, $5kgf/cm^2$의 압력에서는 다소 감소하는 경향을 보였다. 항산화 활성 또한 오미자를 팽화처리 하였을 때 총 폴리페놀 함량과 플라보노이드 함량은 대조구에 비해 모두 증가하였고, 압력이 높아질수록 함량이 증가하다가 $5kgf/cm^2$의 압력에서는 다시 감소하였으며 DPPH radical, ABTS radical 소거능에서도 유사한 경향을 보였다. 이는 팽화 처리에 의한 추출수율의 증가 및 약리성분이 변화됨으로써 생리활성 변화에 기인한 것으로 판단되어지며 어느 일정 압력의 이상으로 처리할 경우 오히려 lignan 성분 함량과 항산화 활성이 감소하는 것을 확인할 수 있었다. 따라서, 오미자를 팽화처리 하였을 때 lignan 성분 함량이 증가하였으며 항산화 활성 또한 대조구에 비해 증가하였고 $3kgf/cm^2$의 압력으로 처리하였을 때 가장 효과적이었다.

Changes of Sesamin and Sesamolin Contents of Sesame Seeds during Grain Filling in Different Sowing Dates

  • To obtain some information on the change of antioxidant components of seeds during grain filling stage as affected by the sowing dates, lignan compounds were investigated according to days of flowering under different sowing dates. Sesamin and sesamolin contents showed significantly different by days of flowering and varieties. Both of sesamin and sesamolin content increased after flowering and reached highest at 40 days of flowering, but they started to decrease thereafter, Sesamin and sesamolin contents of sesame seeds changed with sowing dates. Generally, late sowing date of May 30 showed relatively higher accumulation rate of sesamin and sesamolin contents rather than other sowing dates, but overall patterns were a little different by varieties and lignan compounds. In Yangbaekkae, sesamin and sesamolin contents showed relatively higher at sowing date of May 30, but Yanghuckkae showed higher sesamin and sesamolin contents till 20 days of flowering when sowing date of May 30, but it showed to change that both lignan contents were relatively higher under sowing date of May 10.

Antioxidant Activity of Lignan Compounds Extracted from Roasted Sesame Oil on the Oxidation of Sunflower Oil

  • Effects of lignan compounds (sesamol, sesamin, and sesamolin) extracted from roasted sesame oil on the autoxidation at $60^{\circ}C$ for 7 days and thermal oxidation at $180^{\circ}C$ for 10 hr of sunflower oil were studied by determining conjugated dienoic acid (CDA) contents, p-anisidine values (PAV), and fatty acid composition. Contents of lignan compounds during the oxidations were also monitored. ${\alpha}$-Tocopherol was used as a reference antioxidant. Addition of lignan compounds decreased CDA contents and PAY of the oils during oxidation at $60^{\circ}C$ or heating at $180^{\circ}C$, which indicated that sesame oil lignans lowered the autoxidation and thermal oxidation of sunflower oil. Sesamol was the most effective in decreasing CDA formation and hydroperoxide decomposition in the auto- and thermo-oxidation of oil, and its antioxidant activity was significantly higher than that of ${\alpha}$-tocopherol. Sesamol, sesamin, and sesamolin added to sunflower oil were degraded during the oxidations of oils, with the fastest degradation of sesamol. Degradation of sesamin and sesamolin during the oxidations of the oil were lower than that of ${\alpha}$-tocopherol. The results strongly indicate that the oxidative stability of sunflower oil can be improved by the addition of sesamol, sesamin, or sesamolin extracted from roasted sesame oil.

국내 참깨 품종의 리그난 함량 및 지방산 조성 (Varietal Difference of Lignan Contents and Fatty Acids Composition in Korean Sesame Cultivars)

  • Although lignans of sesame seed, sesamolin and sesamin have been known as possessing an antioxidant activity, it is less known about their contents of the sesame cultivated in Korea. Collections of sesame cultivated in Korea were used for studies on their lignans content of the seed and fatty acids composition of the oil. The sesamin content of sesame seed with white-coat were 370.29 mg/100g seed, while that of sesame seed with black-coat were 246.58mg/100g seed. Also, the sesamolin contents of sesame seed were 202.22 mg/100g seed in white-coat cultivars and 132.68 mg/100g seed in black-coat sesames. Hence, the lignan content of white-coat sesame cultivars was significantly hi임or than that of black-coat ones. Korean sesame cultivars also showed considerably higher sesamin content than sesamolin content in seeds. The correlation between sesamin and sesamolin contents was not recognized in Korean sesame cultivars. The stearic acid of white-coat sesame was significantly higher than that of black-coat one (p<0.05).

아시아 및 아프리카 원산 참깨(Sesame indicum L.) 유전자원의 농업형질과 리그난 함량 평가 (Characterization of Agronomic Traits and Evaluation of Lignan Contents in Asian and African Sesame (Sesamum indicum L.) Germplasms)

  • 본 연구에서는 다수성 및 고리그난 참깨 품종 육성에 활용될 수 있는 자원을 발굴하고자 아시아 및 아프리카 원산 참깨 165자원을 대상으로 농업형질 조사와 리그난 함량 분석을 수행하였다. 그 결과, 참깨 유전자원은 다양한 농업형질을 보였으며, 세사민, 세사몰린, 리그난 함량은 각각 0.5-12.6 mg/g, 0.1-3.5 mg/g, 1.1-16.1 mg/g의 높은 변이를 보였다. 대륙 원산에 따른 농업형질을 분석한 결과, 대부분의 농업형질이 아시아 원산자원과 아프리카 원산자원 간에 차이를 나타냈다. 아시아 원산자원이 아프리카 원산자원에 비해 개화기와 성숙기가 짧고, 초삭고가 낮고, 착삭부위장이 길며, 천립중이 가벼운 특성을 보였다. 그러나 대륙 원산에 따른 리그난 함량의 차이는 보이지 않았다. 농업형질과 리그난 함량 간의 상관관계를 분석한 결과, 리그난 함량은 개화기, 초삭고 형질과 양의 상관관계를, 착삭부위장, 천립중 형질과 음의 상관관계를 보여, 수량성 관련 농업형질과 리그난 함량은 음의 상관관계를 가짐을 알 수 있었다. 농업형질과 리그난 함량에 대한 주성분 분석결과, 세 개의 주성분이 전체 변이의 73.6%를 설명하였다. 대부분의 농업형질과 리그난 함량이 세 개의 주성분과 높은 상관관계를 보여, 본 연구에서 평가한 농업형질 및 리그난 함량은 참깨 165자원의 다양성을 설명하는 주요인으로 판단되었다. 또한, 농업형질과 리그난 함량에 대한 군집 분석 결과, 수량성이 낮고 리그난이 높은 군집 1 (67자원), 수량성이 높고 리그난 함량이 높은 군집 2 (34자원), 수량성이 군집 1과 3의 중간 정도이며 리그난 함량이 낮은 군집 3 (64자원)으로 나눌 수 있었다. 최종적으로, 참깨 165자원에 대한 농업형질 및 리그난 함량 분석 결과를 기반으로 다수성(IT29416, IT167042, K276848, K276849)및 고리그난(IT169254, IT170031, IT169250, IT154876, IT170034) 우수자원을 각각 발굴하였다. 선발된 우수자원은 다수성 및 고리그난 참깨 품종 육성 시 유용한 육종소재로 활용될 것으로 기대된다.

수확시기에 따른 오미자의 항산화 활성과 리그난 성분 함량 변화 (Changes of Antioxidant Activity and Lignan Contents in Schisandra chinensis by Harvesting Times)

  • We investigated antioxidant activity and lignan contents by harvesting times to expand use of Schisandra chinensis (Turcz.) Baillon. Total polyphenol and flavonoid contents of seed was higher than those of flesh but there is not much difference in harvesting times. As $RC_{50}$ value, that was, the concentration of sample required for 50% reduction of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical, was 13.7~24.2 ${\mu}g/m{\ell}$ in seed thus it showed a high antioxidant activity. Among lignan components, schizandrin content was the highest and followed by gomisin N and gomisin A in all of flesh and seed. Also these components in seed were 4~9 times more contained than those of flesh. All of them were decreased by harvesting times in flesh. But the contents of schizandrin and gomisin N were high in August 3rd and September 15th in seed, respectively. As the results, the seed of S. chinensis had high antioxidant activity and lignan contents so it could be potentially developed as a resource.

Use of Near-Infrared Spectroscopy for Estimating Lignan Glucosides Contents in Intact Sesame Seeds

  • Near-infrared spectroscopy(NIRS) was used to develop a rapid and efficient method to determine lignan glucosides in intact seeds of sesame(Sesamum indicum L.) germplasm accessions in Korea. A total of 93 samples(about 2 g of intact seeds) were scanned in the reflectance mode of a scanning monochromator, and the reference values for lignan glucosides contents were measured by high performance liquid chromatography. Calibration equations for sesaminol triglucoside, sesaminol($1{\rightarrow}2$) diglucoside, sesamolinol diglucoside, sesaminol($1{\rightarrow}6$) diglucoside, and total amount of lignan glucosides were developed using modified partial least square regression with internal cross validation(n=63), which exhibited lower SECV(standard errors of cross-validation), higher $R^2$(coefficient of determination in calibration), and higher 1-VR(ratio of unexplained variance divided by variance) values. Prediction of an external validation set(n=30) showed a significant correlation between reference values and NIRS estimated values based on the SEP(standard error of prediction), $r^2$(coefficient of determination in prediction), and the ratio of standard deviation(SD) of reference data to SEP, as factors used to evaluate the accuracy of equations. The models for each glucoside content had relatively higher values of SD/SEP(C) and $r^2$(more than 2.0 and 0.80, respectively), thereby characterizing those equations as having good quantitative information, while those of sesaminol($1{\rightarrow}2$) diglucoside showing a minor quantity had the lowest SD/SEP(C) and $r^2$ values(1.7 and 0.74, respectively), indicating a poor correlation between reference values and NIRS estimated values. The results indicated that NIRS could be used to rapidly determine lignan glucosides content in sesame seeds in the breeding programs for high quality sesame varieties.

종자류 식품에 함유된 5종 리그난의 동시 분석법 개발 (Development of Simultaneous Analytical Method for Five Lignans in Edible Seeds)

  • There has been increased interest in lignans due to their potential effect in reducing the risk of developing several diseases. To evaluate lignan contents, sensitive and accurate methods should be developed for their quantification in food. The present study aimed to validate a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the quantification of 5 lignans: lariciresinol (Lar), matairesinol (Mat), pinoresinol (Pin), secoisolariciresinol (Seco), and syringaresinol (Syr). The validation included selectivity, linearity, recovery, accuracy, and precision. The method was proved to be specific, with a linear response (R2≥0.99). The limits of detection were 0.040~0.765 ㎍/100 g and the limits of quantification were 0.114~1.532 ㎍/100 g. Recoveries were 90.588~109.053% for black sesame powder. Relative standard deviations of repeatability and reproducibility were below 5%. Total lignan contents of roasted coffee bean, oat, and blacksoy bean were 105.702 ㎍/100 g, 78.965 ㎍/100 g, and 165.521 ㎍/100 g, respectively. These results showed that LC-MS/MS analysis would be effective in producing acceptable sensitivity, accuracy, and precision in five lignan analyses.

Determination of Sesamin and Sesamolin in Sesame (Sesamum indicum L.) Seeds Using UV Spectrophotometer and HPLC

  • Sesamin and sesamolin, antioxidant lipidsoluble lignan compounds, are abundant in sesame (Sesamum indicum L.) seed oil and provide oxidative stability of oil related to sesame quality. The sesamin and sesamolin contents of 403 sesame land races of Korea were determined by HPLC analysis of methanol extract (HPLC value), and their total lignan content was compared with those by using UV-Vis spectrophotometric analysis (UV method) of methanol (UV-MeOH value) and hexane (UV-Hexane value) extracts. HPLC values of total lignan content were strongly associated with UV-Hexane (r=0.705**) and UV-MeOH (r=0.811**) values. The UV values from both the extracts were 3.8-4.7 times higher than those of HPLC values. Lignan content was overestimated by UV method because total compounds in the mixture solution were quantified by absorbing at the same ultraviolet wavelength as in HPLC method. UV method could more rapidly analyze small amount of sample with higher sensitivity of detection than HPLC method. Average contents of lignans in sesame germplasm evaluated in this study were $2.09{\pm}1.02mg/g$ of sesamin, and $1.65{\pm}0.61mg/g$ of sesamolin, respectively, showing significant variation for lignan components. The results showed that UV method for the determination of sesamin and sesamolin could be practically used as a faster and easier method than HPLC by using the regression equations developed in this study.
