The propagation of light radiation within tissues is an important problem that confronts the dosimetry of therapeutic laser delivery and the development of diagnostic spectroscopy. In the clinical application of photodynamic therapy(PDT) and in photobiology, the photon deposition within a tissue determines the spatial distribution of photochemical reactions. Scattered light is measured as a function of the distance (r) between the axis of the incident beam and the detection spot. Consequently, knowledge of the photosensitizer(Chlorophyll-a) function that characterizes a phantom is important. To obtain the results of scattering coefficients(${\mu}s$) of a turbid material from diffusion described by experimental approach. It was measured the energy fluency of photon radiation at the position of penetration depth. From fluorescence experimental method obtained the analytical expression for the scattered light as the values of $(I\;/I_o)_{wavelength}$ vs the distance between the center of the incident beam and optical fiber in terms of the condition of "in situ spectroscopy(optically thick)" and real time by fluorometric measurements.

The distribution of light in a randomly scattering medium can represent problems found in many area. Particularly, in the clinical application of lasers for Photodynamic therapy(PDT) or in the fluorescence spectroscopy for biological tissue, turbidity plays a very important role. The influences of fluorophor, scatterer, and absorber in turbid material by light scattering were interpreted for the scattered fluorescence intensity and wavelength. The molecular properties have been studied by laser induced fluorescence spectroscopy in scattering medium as tissue. It has been found that the effects of optical properties in scattering media could be investigated by the optical $parameters({\mu}_s$, ${\mu}_a$ ,${\mu}t)$. Experimental and Monte Carlo simulation method for modelling light transport in tissue was applied. The experimental results using a randomly distributed scattering medium were discussed and compared with those obtained through Monte Carlo simulation. It'll be also important in designing the best model for oil chemistry, medicine and application of medical engineering.
Photobiomodulation therapy has been proposed as a promising therapeutic approach for retinal degenerative diseases. However, its effect on the regenerative capacity in mammalian retina and its intracellular signalling mechanisms remain unknown. Here, we show that photobiomodulation with 670 nm light stimulates Müller glia cell cycle re-entry and dedifferentiation into a progenitor-like state in both the uninjured and injured retina. We also find that 670 nm light treatment inhibits the Hippo pathway, which is activated in Müller glia following NaIO3-induced retinal injury. YAP, a major downstream effector of the Hippo signalling pathway was translocated into the nucleus of Müller glia along with YAP dephosphorylation in retina treated with 670 nm light. Deficiency of YAP attenuated Müller glia cell cycle re-entry and dedifferentiation. Our data reveal that the Hippo-YAP signalling pathway is associated with the photostimulatory effect on regenerative response in mammalian retina, and suggest a potential therapeutic strategy for retinal degenerative diseases.
The role of the ferric uptake regulator (Fur) of Helicobacter pylori in the oxidative stress was investigated in this study. A fur knockout mutant of H. pylori was constructed by replacing the fur gene with an aphA (kanamycin resistant marker) gene. Photodynamic therapy using methylene blue (MB) and 660 nm light was chosen to induce oxidative stress. The bactericidal effect of photodynamic therapy (PDT) was compared between wild type H. pylori and fur knockout mutant H. pylori. The degree of oxidative damage of DNA was confirmed using alkaline gel electrophoresis and an assay of 8-hydroxy-2-deoxyguanosine (8-OHdG). In control groups, the number of viable cells was maintained constantly during experiment. After PDT, the mutant H. pylori showed 10,000 times decreased viable cell number compared with wild type H. pylori. Depending on the exposure time of 660 nm light, the 3-fold increase in the concentration of 8-OHdG was observed in mutant H. pylori. The results of this study showed that H. pylori's Fur protein may play a role in oxidative stress induced by PDT.
This study was investigated the effects of a direct current and ultrasound on transdermal transport of dexamethasone into the rabbits which had contusion in the thigh. Each group was treated under the tallowing conditions. 1. EXP group I : $10\%$ dexamethasone ointment and ultrasound 2. EXP group II : $1\%$aqueous solution of dexamethasone and iontophoresis 3. EXP group III : the application of $10\%$ dexamethasone ointment 4. Control group : No treatment The degree of anti-inflammation was evaluated by the naked eye, the change in girth of thigh, and a light microscope. The results were as follows. 1.8y the naked eye. an inflammation sign was seen in all groups and especially. symptoms of redness. heat swelling were prominent in EXP group I. 2. In comparision in the change of girth of thigh, only EXP group II showed no significant change. Therefore, it meant that there was effective anti-inflammatory reaction in EXP group II. 3. The infiltration of inflammation cells, the degree of swelling, and the degree of crosslinking of connective tissues were evaluated with a light microscope. As a result, EXP group II showed the most effective anti-inflammatory reaction. And, in order of EXP group III, control group, the effect of anti-inflammation reaction was decreased. 4. EXP group I showed more intensive inflammation than control group.
Previous studies have investigated stepping over obstacles in treadmill walking training (TWT-OS) and treadmill walking training (TWT) alone for walking capacity not considering real physical activity. As such, we investigated the effects of TWT-OS on physical activity and changes in different levels of physical activity based on community ambulation in stroke patients. Thirty subjects were randomly assigned to either the experimental group or the control group, with 15 and 15 subjects, respectively. However, one subject from the control group was excluded because of inadequate treatment sessions. All subjects underwent routine physical therapy in the form of treadmill walking. The subjects in the experimental group underwent simultaneous training in obstacle-stepping while walking on the treadmill for 30 min/day, five times/week, for four weeks. Subjects were given a three-axis accelerometer to wear at the hip on a belt for one-week pre- and post-training physical activity. Step counts for seven days, average daily step counts, and the average of minutes spent in sedentary, light, and above moderate activity were chosen as outcome measures of physical activity. No significant differences between the groups were found in terms of step counts for seven days, average daily activity, or daily activity spent at sedentary levels after four-week interventions. However, the average daily activity spent at light levels (-42.60 min vs. -6.71 min) was significantly lower in the experimental group than in the controls. Conversely, average daily activity spent at above moderate levels was higher (19.86 min vs. 11.07 min) (p<.05) after adjusting for each baseline value. Significant pre- and post-training differences were found in all variables of the experimental group (p<.05). Thus, TWT-OS could improve physical levels above moderate activity as a community-oriented task more than simple repetitive waking on a treadmill, and it could provide an opportunity for patients ambulatory after stroke to participate in the community again.
Background: Monoclonal antibodies (mAb) of rodent origin are produced with ease by hybridoma fusion technique, and have been successfully used as therapeutic reagents for humans after humanization by genetic engineering. However, utilization of these antibodies for therapeutic purpose has been limited by the fact that they act as immunogens in human body causing undesired side effects. So far, there have been several attempts to produce human mAbs for effective in vivo diagnostic or therapeutic reagents including the use of humanized xenomouse that is generated by mating knockout mice which lost Ig heavy and light chain genes by homologous recombination and transgenic mice having both human Ig heavy and light gene loci in their genome. Methods: Genomic DNA fragments of mouse Ig heavy and light chain were obtained from a mouse brain ${\lambda}$ genomic library by PCR screening and cloned into a targeting vector with ultimate goal of generating Ig knockout mouse. Results: Through PCR screening of the genomic library, three heavy chain and three light chain Ig gene fragments were identified, and restriction map of one of the heavy chain gene fragments was determined. Then heavy chain Ig gene fragments were subcloned into a targeting vector. The resulting construct was introduced into embryonic stem cells. Antibiotic selection of transfected cells is under the progress. Conclusion: Generation of xenomouse is particularly important in medical biotechnology. However, this goal is not easily achieved due to the technical difficulties as well as huge financial expenses. Although we are in the early stage of a long-term project, our results, at least, partially contribute the successful generation of humanized xenomouse in Korea.
Background: Light-emitting diodes curing unit (LCU), which emit blue light, is used for polymerization of composite resins in many dentistry. Although the use of LCU for light-cured composite resin polymerization is considered safe, it is still controversial whether it can directly or indirectly have harmful biological influences on oral tissues. The aim of this study was to elucidate the biological effects of LCU in wavelengths ranging from 440 to 490 nm, on the cell viability and secretion of inflammatory cytokines in MDPC-23 odontoblastic cells and inflammatory-induced MDPC-23 cells by lipopolysaccharide (LPS). Methods: The MTT assay and observation using microscope were performed on MDPC-23 cells to investigate the cell viability and cytotoxic effects on LCU irradiation. Results: MDPC-23 cells and LPS stimulated MDPC-23 cells were found to have no effects on cell viability and cell morphology in the LCU irradiation. Nitric oxide (NO) and prostaglandin $E_2$ which are the pro-inflammatory mediators, and interleukin-$1{\beta}$ and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) which are the proinflammatory cytokines were significantly increased in MCPD-23 cells after LCU irradiation as time increased in comparison with the control. LCU irradiation has the potential to induce inflammation or biological damages in normal dental tissues, including MDPC-23 cells. Conclusion: Therefore, it is necessary to limit the use of LCU except for the appropriate dose and irradiation time. In addition, LCU irradiation of inflammatory-induced MDPC-23 cells by LPS was reduced the secretion of NO compared to the LPS alone treatment group and was significantly reduced the secretion of TNF-${\alpha}$ in all the time groups. Therefore, LCU application in LPS stimulated MDPC-23 odontoblastic cells has a photodynamic therapy like effect as well as inflammation relief.
Purpose: This study evaluated the antibacterial effects of curcuma, ginger, and finger root extracts in water-soluble powder on planktonic Streptococcus mutans(S.mutans), which is known to cause dental caries, in order to confirm whether these extracts could perform as photosensitizers for the effects of photodynamic therapy (PDT). Methods: This study used the strain of streptococcus mutans ATCC 25175 distributed by the Korean Collection for Type Cultures of the Korea Research Institute of Bioscience & Biotechnology. Commercial edible curcuma, ginger and finger root were used as the natural extracts for the use of photosensitizer. To extract organic solvent, 3 g of each powder was mixed in $30m{\ell}$ of dimethyl sulfoxide (DMSO, VWR, Germany) before extraction. $1.8m{\ell}$ of the photosensitizer solution, manufactured in the concentrations of 5, 0.5, and $0.05mg/m{\ell}$, was mixed with $0.2m{\ell}$ of the S. mutans culture medium that had been cultured for 2 days. To induce the photodynamic reaction, Qraycam (AIOBIO, Seoul, Korea) equipped with 405 nm LED was used to expose light for 5 minutes to irradiate 59 nW energy for 300 seconds. Results: Compared with the case with no light, a higher photodynamic therapeutic effect was confirmed with $0.05mg/m{\ell}$ curcuma powder extract, the concentration of $0.5mg/m{\ell}$ and LED light of 405 nm wavelength (p=0.000, p=0.003). $0.05mg/m{\ell}$ of curcuma powder extract and the concentration of $0.5mg/m{\ell}$ showed 100% antibacterial effect when exposed to light, whereas the concentration of $5mg/m{\ell}$ showed 11.95% antibacterial effect. When exposed to light, $0.05mg/m{\ell}$ of ginger powder extract showed an antibacterial effect which didn't statistically decrease. The concentrations of $0.5mg/m{\ell}$ and $5mg/m{\ell}$ did not show any antibacterial effects. As a result of examining any photodynamic therapeutic effects of finger root powder extract on S. mutans, no statistically significant effect was found. Conclusion: The curcuma powder extract is expected to perform as a photosensitizer. Even though belonging to the same ginger family, ginger powder and finger root powder seem difficult to perform as photosensitizer.
The purpose of study was to compare body image between diabetes mellitus patients who used insulin pump therapy and didn't use insulin pump therapy. The study design was comparative survey study the subjects were 60 diabetes mellitus patients who used insulin pump therapy and 60 diabetes mellitus patients who didn't use insulin pump therapy at B hospital in Busan. The data were collected from 15th April to 20th August, 1998. The instrument used for this study were Osgood's body image scale. The collected data were analyzed frequency, percentage, $X^2$-test, mean, standard deviation, t-test, ANOVA, Scheffe test. The results were as follows 1. Demographical characteristics between diabetes mellitus patients who used insulin pump therapy and didn't use insulin pump therapy were no significant difference. 2. Characteristics related disease between diabetes mellitus patients who used insulin pump therapy and didn't use insulin pump therapy were significant difference in paticipation of D.M. meeting, no of paticipation of D.M. meeting. 3. Body inmage score of diabetes mellitus patients was $69.08{\pm}18.13$. In body image, diabetes mellitus patients who used insulin pump therapy were higher than that didn't use insulin pump therapy(t=1.964, P<.05) 4. In body image's each item, common-strange item, noble-humble item, competent-incompetent item, light-heavy item, diabetes mellitus patients who used insulin pump therapy were higher than diabetes mellitus patients who didn't use insulin pump therapy(P<.05). 5. In body image according to economic status, marital status, occupational status were significantly difference. 6. In body image according to causes of regular hospital visiting, paticipation of diabetes mellitus class were significantly difference. In conclusion, diabetes mellitus patients who used insulin pump therapy were more positive than diabetes mellitus patients who didn't use insulin pump therapy.
