Stspension Culture of Betalain Producing Cell-Line and Characteristics of Hairy Root of Phytolacca esculenta V. Houtte (장록에서 유도한 Betalain 합성 세포주의 액체 배양과 모상근의 특성)

  • Thc effect of light and phylohormonc on the belalaln synthesis was tested using the suspension culture of a rod(bclalaln produulng) cell line from Phytolacca esculenta V. Houtte. Betalain synthesis of rod-cell 1ine was strongly dependent on the irradiation of blue light, but induced by hormone, IAA and/or kinetin, in dark conditions. In a light condition, however, 2 mg/l of IAA increased the betalain content about 30% (per gram fresh weight), whereas more than 0.5mg/l of kinetin remarkably decreased. The hairy root derived from the same plant was also observed for the blue light-dependent pigmentation in the root-tips. When the hairy root grown in dark was transferred to the light condition, the accumulation of betalain was initiated after 12hours. Such pigmentation was completely inhabited by addition of a protein synthesis inhibitor.

  • In this paper, the emergency light where is being scattered always inspection and will be able to manage from the management center. Is not interfered in data neck of a bottle actual condition and the data communication will be possible and the cluster monitor network will grow and uses establishes the emergency light monitoring system. Will not be interfered in location of emergency light and not to be will be able to establish the system. And the monitoring network there is by a destroyer and the communication relay system is born breakdown but the dead zone without condition of emergency light proposes the emergency light management system where the monitoring and management are possible.

Comparison of the responses of two Dunaliella strains, Dunaliella salina CCAP 19/18 and Dunaliella bardawil to light intensity with special emphasis on carotenogenesis

  • Dunaliella salina and Dunaliella bardawil are well known for carotenogenesis, the overproduction of carotenoids, under stress conditions. The effect of high light (HL) and low light (LL) on the growth, morphology, photosynthetic efficiency, and the ${\beta}$-carotene and zeaxanthin production of D. salina CCAP 19/18 and D. bardawil was investigated and compared. Both strains showed similar growth kinetics under LL growth condition, but D. salina CCAP 19/18 was faster. As the light intensity increased, D. salina CCAP 19/18 cells were elongated and D. bardawil cells became larger. Both strains showed decrease of the maximum quantum yield of PSII ($F_v/F_m$) and election transport rate (ETR) under HL growth condition and D. salina CCAP 19/18 was less liable to the light stress. Both strains had about 1.8 and 5 times difference in the $O_2$ evolution rate at LL and HL conditions, respectively. The ${\beta}$-carotene and zeaxanthin production were increased as the light intensity increased in both strains. D. bardawil was more sensitive to light intensity than D. salina CCAP 19/18. The possible application of D. salina CCAP 19/18 as a carotenogenic strain will be discussed.

Analysis on Suitability Light Intensity and Introduction Plan under the Indoor Lighting for the Native Evergreen Daphniphyllum macropodum (자생 상록 굴거리나무의 실내조명 하에서 적정광도 분석 및 도입방안)

  • This study was performed to determine light-adaptability under indoor lighting for Korean wild Daphniphyllum macropodum, and to provide an introduction plan for indoor spaces of landscape architecture. The experimental plants, Daphniphyllum macropodum were purchased from a farmhouse in Jejudo as anannualplant, and it was acclimatized in the glass-green house of the general farm of Gyeongnam National University of Science and Technology for40 days. The experiment was performed from February 15, 2010 to November 15, 2010. First, in the case of the fluorescent lamp, the growth and development condition was poor because the plants withered, or the leaves fell off under the 100lux to 500lux, but the condition under 1,000lux was good In the aspects of the number of leaves, form of the tree, photosynthesis rate in its body, and the value of sight, the best light intensity for the growth condition was under the 1,000lux. Second, in the case of the LED light, the growth and development condition was poor because the plants withered, or the leaves fell off under 100lux to 1,000lux, but the smooth growth and development was done under a more light intensity. The best intensity for the growth condition was under 2,000lux. Third, in the case of the three-wave light, the withering was serious by 1,000lux, and the growth and development was the worst amongst the four introduced lighting systems, therefore, growth under the three-wave light was incongruous. The best intensity for the growth condition was under 2,000lux. Fourth, in the case of the optical fiver, the withering did not existed under 100lux and growth was possible. The growth and development was the amongst in the four introduced lighting systems. Generally, in the more light intensity, more growth was observed, but the value of sight was higher under 1,000lux than under 2,000lux because the falling rate of leaves and the form of the tree was stable. The most effective light was under 1,000lux due to the high photosynthesis in its body. When Daphniphyllum macropodum is introduced into the indoor landscaping space considering the light, the optical fiber, fluorescent lamp, and LED light are suitable to introduce. The three-wave light is unfit. The most proper light intensity by the light source, in the case of the optical fiber, and fluorescent lamp, is 1,000lux, and, in the case of LED light, is 2,000lux. The wild Daphniphyllum macropodum is the species of tree to substitute the Schefflera actinophylla which is the introduced species, and it is expected to be use as the central tree in indoor spaces.

  • Energy is vital to global prosperity, yet dependence on fossil fuels as our primary energy source contributes to global climate change environmental degradation, and health problems. Hydrogen $(H_2)$ offers tremendous potential as a clean renewable energy currency. Hydrogen has the highest gravimetric energy density of any known fuel and is compatible with electrochemical and combustion processes for energy conversion without producing carbon-based emission that contribute to environmental pollution and climate change. Numerous methodologies have been developed for effective hydrogen production. Among them, the biological hydrogen production has gained attention, because hydrogen can be produced by cellular metabolismunder the presence of water and sunlight. The green alga Chlamydomonas reinhardtii is capable of sustained $H_2$ photoproduction when grown under sulfur deprived condition. Under sulfur deprived conditions, PSII and photosynthetic $O_2$ evolution are inactivated, resulting in shift from aerobic to anaerobic condition in the culture. After anaerobiosis, sulfur deprived algal cells induce a reversible hydrogenase and start to evolve $H_2$ gas in the light. According to above principle, we investigated the effect of induction parameters such as cell age, cell density. light intensity, and sulfate concentration under sulfur deprived condition We also developed continuous hydrogen production system by sulfate re-addition under sulfur deprived condition.

Comparison of Gold Biosensor Combined with Light Microscope Imaging System with ELISA for Detecting Salmonella in Chicken after Exposure to Simulated Chilling Condition

  • In this study, the performance of a gold biosensor combined with light microscope imaging system (GB-LMIS) was comparatively evaluated against enzyme-linked immunosorbent assay (ELISA) for detecting Salmonella under simulated chilling condition. The optimum concentration of antiSalmonella polyclonal antibodies (pAbs) was determined to be 12.5 and 100 ㎍/ml for ELISA and GBLMIS, respectively. GB-LMIS exhibited a sufficient and competitive specificity toward three tested Salmonella among only. To mimic a real-world situation, chicken was inoculated with Salmonella cocktail and stored under chilling condition for 48 h. The overall growth of Salmonella under chilling condition was significantly lower than that under non-exposure to the chilling condition (p < 0.05). No significant differences in bacterial growth were observed between brain heart infusion and brilliant green broth during the enrichment period (p > 0.05). Finally, both GB-LMIS and ELISA were employed to detect Salmonella at every 2-h interval. GB-LMIS detected Salmonella with a competitive specificity by the direct observation of bacteria on the sensor using a charge-coupled device camera within a detection time of ~2.5 h. GB-LMIS is a feasible, novel, and rapid method for detecting Salmonella in poultry facilities.

  • The effects of nacl concentration, temperatura, light and growth regulator(GA3, kinetin) on the tuber germination of s. maritimus were investigated. The germination percentage increased with decreasing nacl, showing salt tolerance through time, and with increasing temperature untile 20~30oC light / dark (12/12 hr) of optimal temperature. The multiplication of nacl and temperature on germination percentage and velocity, increased significantly in higher temperature than lower temperature in saline. The germination percentage showed high value in dark condition than in light condition which is the charracteristics of underground organ. and ga3 act as germination stimulator to overcome the inhibitory effect of nacl. The effect of ga3 showed significant differances on tubers of s. maritimus of germination but that of kinetin had a litter sffects on germination. Factors of nacl and temperature interacted significantly and the effects of nacl on germination percentage and velocity depended on temperature condition.

  • The use of daylighting has been increased recently due to energy and visual comfort. The aims of interior daylighting are to adequately illuminate visual tasks, to create an attractive visual environment, and to save electrical energy. Lightpipe can improve the distribution of light to interior spaces. This study aims to evaluate the comparative daylighting performance of lightpipe under different sky conditions with mock-up model, sized $6m{\times}6m{\times}4m$ ($w{\times}d{\times}h$). For the purpose, perpendicular lightpipe system was designed as 650 diameter, with an aspect ratio of 2. Totally 49 measuring points of and two of outdoor illuminance on the horizontal plane were monitored from 09:00 to 18:30 on April 29 and May 15 2008. Agilent data logger and photometric sensor were used. Light factor were used to analyse daylight performance under different sky condition. Under overcast sky condition and clear sky condition, the lightpipe system is suitable for KS recommendation level of illuminance.

  • Natural dyeing is considered to be an alternative to chemical staining in terms of harmful VOC-free and environmentally-friendly ingredient. This study was concerned with an evaluation of color performance of naturally dyed- and painted veneer for cylindrical laminated veneer lumber etc. according to the used condition in terms of existence of light and heat in drying. From the results, the color changes by the color of dyed veneer and the type of paint are likely to be affected by fading condition of light or heat. In the future, long-term outdoor exposure test needs to be additionally followed in order to evaluate durable usability of colored veneers.

Effects of Light Intensity on Photosynthesis and Growth in Seedling of Kalopanax pictus Nakai (광강도에 따른 음나무 유묘의 생장 및 광합성 특성)

  • This study was carried out to determine leaf photosynthetic capacity and growth characteristics of Kalopanax picturs Nakai seedlings under the different light condition. The seedlings grown under low light condition showed larger leaf area and petiole, and higher relative growth rate than those grown under high light condition. Chlorophyll contents as chlorophyll a, chlorophyll b, and chlorophyll a + b were high in the seedlings grown under highlight condition compared to those grown in low light conditions. The mean absorption value of shade leaf within photosynthetically active radiation (400-700 nm) was slightly higher than that of sun leaf, Leaf photosynthetic capacity of seedling was variable under the different light conditions. Seedling grown under high light condition had the higher photosynthetic capacity. Leaf photosynthetic rates under forestry and nursery were 700 and $300\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. However, leaf photosynthetic rates under high and low light conditions were 500 and $300\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively.
