한국의 초등 과학 교과서 생명영역에 나오는 탐구 주제 및 생물 재료에 대한 초등 예비교사들의 인식 분석 (The Analysis of Pre-service Elementary Teachers' Perception on Biology Inquiry Subjects, and biology Materials in the Korea Elementary School Science Textbook)

  • The aim of this study was to analyze a group of pre-service elementary teachers experience on the cognitive terms of biology inquiry subjects, and experience on living materials related with the domain of life in the Korea elementary school science textbook. Firstly, elementary school pre-service teachers had the confidence of inquiry subjects related with the domain of life such as 'when we take breath what will happen in human body?' and 'Can we make the model of human body?' But they did have the most diffidence of 'The search for pieris rapae, a cabbage butterfly's life', 'Can we grow the little creature?' The reason why pre-service elementary teachers had diffidence of comprehending experiment procedure and planning ability was that 'they focused on understanding the scientific concepts instead of planning experiments by themselves', and 'they carried out the least amount of experiments' Secondly, elementary school pre-service teachers had never experienced biology materials related with the domain of life in the Korea elementary school biology science textbook such as 'tradescantia reflexa', 'Caddis larva', 'hydrilla verticillata', and 'Plantain lily.' According to the findings obtained from the study, the reason why they had never seen the biology materials was that they had the least opportunity of immediately observing life creatures due to memorization-intensive classes.

2009 개정 교육과정에 따른 5, 6학년 초등과학과 교사용 지도서에 제시된 발문 유형 분석 (The Analysis on Question's Patterns in Elementary School Science Teacher's Guidebooks of 5, 6th Grade under the 2009 Revised Curriculum)

  • The purpose of this study was to analyze question's patterns in elementary school science teacher's guide books of 5, 6th grade under the 2009 revised curriculum. A modified analysis framework based on Blosser's classified system was used to analyze 1,982 questions extracted from elementary science teacher's guide books by grade, by domain, and by teaching and learning stage. The findings of this study were as follows. First, of the 1,982 questions, the most prominent type of question was the propositional question and the following was the reproductive question. And, in comparing the question's patterns between 5, 6th grade, it was found that 6th grade had higher rate of close typed question, while 5th grade had higher rate of open typed question in its curriculum. Secondly, a comparative study about two domains, material and energy science domain and earth and life science domain, showed that the number of questions of each domain was not much different. However, it was found that propositional questions and applicable questions showed a higher rate in material and energy science domain, and anticipated questions and open typed questions including divergent and evaluative question showed higher rate in earth and life science domain. Moreover, although the total number of questions from integration and my fun research domain's contents was small, the rate of open typed questions was higher than any other domains. Finally, as a result of comparing and analyzing question's pattern in teaching and learning stages, the rate of reproductive question and anticipated questions was high at the stage of introduction. At the stage of development, the rate of propositional and reproductive questions was high. At the stage of conclusion, the rate of synthetic and applicable questions was high.

Identification of Critical Residues for Plasminogen Binding by the αX I-domain of the β2 integrin, αXβ2

  • The ${\beta}2$ integrins on leukocytes play important roles in cell adhesion, migration and phagocytosis. One of the ${\beta}2$ integrins, ${\alpha}X{\beta}2$ (CD11c/CD18), is known to bind ligands such as fibrinogen, Thy-1 and iC3b, but its function is not well characterized. To understand its biological roles, we attempted to identify novel ligands. The functional moiety of ${\alpha}X{\beta}2$, the ${\alpha}X$ I-domain, was found to bind plasminogen, the zymogen of plasmin, with moderate affinity ($1.92{\times}10^{-6}M$) in the presence of $Mg^{2+}$ or $Mn^{2+}$. The ${\beta}D-{\alpha}5$ loop of the ${\alpha}X$ I-domain proved to be responsible for binding, and lysine residues ($Lys^{242}$, $Lys^{243}$) in the loop were the most important for recognizing plasminogen. An excess amount of the lysine analog, 6-aminohexanoic acid, inhibited ${\alpha}X$ I-domain binding to plasminogen, indicating that binding is lysine-dependent. The results of this study indicate that leukocytes regulate plasminogen activation, and consequently plasmin activities, through an interaction with ${\alpha}X{\beta}2$ integrin.

제6차와 7차 초등학교 과학 교과서에 제시된 비유 비교분석 (A Comparative Analysis of Analogies in Elementary Science Textbook by the 6th and 7th Curriculum)

  • The purpose of this study was to analyze the analogies used in elementary science textbooks. For this, the analogies were classified into the following criteria : analogy-related contents included in elementary science text-books, types of representation style, and the role of analogy. The results of this study were as follows : the total of analogies was 154 in the 6th and 166 in the 7th curriculum. Most of them were expressed in textbooks and experimental texts. More structural/functional, verbal/pictorial, concrete/concrete, concrete/abstract, simple and enriched analogies were included in the science textbooks of the 7th than those of the 6th. Most of them took the role as an aid to understanding. According to the domain of science content, energy, material, life domains were increased in the 7th than the 6th, with the exception of the earth science domain. The ratio of used analogies in the life domain was higher than the others, and in the material domain it was the lowest. From these results, it can be said that, in order to help students' understanding, further research into the application of analogies is needed.

CLIP-domain serine proteases in Drosophila innate immunity

  • Extracellular proteases play an important role in a wide range of host physiological events, such as food digestion, extracellular matrix degradation, coagulation and immunity. Among the large extracellular protease family, serine proteases that contain a "paper clip"-like domain and are therefore referred to as CLIP-domain serine protease (clip-SP), have been found to be involved in unique biological processes, such as immunity and development. Despite the increasing amount of biochemical information available regarding the structure and function of clip-SPs, their in vivo physiological significance is not well known due to a lack of genetic studies. Recently, Drosophila has been shown to be a powerful genetic model system for the dissection of biological functions of the clip-SPs at the organism level. Here, the current knowledge regarding Drosophila clip-SPs has been summarized and future research directions to evaluate the role that clip-SPs play in Drosophila immunity are discussed.

In vitro에서 핵산치환인자 BAP이 단백질-분자 샤페론 복합체 해리에 미치는 영향 (A Nucleotide Exchange Factor, BAP, dissociated Protein-Molecular Chaperone Complex in vitro)

  • 소포체는 세포막의 합성뿐만 아니라 세포막에 존재하거나 세포외로 분비되어져야 할 단백질을 합성하는 세포내 소기관이다. 소포체에서 단백질이 합성되어질 경우 이황화결합이 형성되고 glycosylation 등의 수식이 일어나며, 이와 동시에 folding과 assembly과정을 거쳐 삼차원적 구조로 성숙이 되는데 이 과정은 folding enzyme과 molecular chaperone의 도움을 받아 이루어진다. 소포체 내에 존재하는 molecular chaperone 중 가장 잘 알려진 것으로 BiP이 있다. BiP의 기능은 N-terminus의 ATPase domain에 의해 조절되고 ATPase domain은 이것과 선택적으로 결합하는 조절인자에 의해 ATPase의 활성이 영향을 받는다. BiP의 핵산치환조절인자로서 발견된 BAP은 ATPase domain에 결합된 ADP를 ATP로 치환하는 것으로 기능이 알려져 있다. 이 BAP의 핵산치환기능이 BiP의 샤페론 작용에 어떤 영향을 미치는지를 in vitro에서 항체 heavy chain을 이용하여 알아보았다. BAP은 ATP보다 ADP가 결합되어 있는 BiP과 더 잘 결합을 하며, in vitro에서 BiP과 결합하고 있는 unfolded 단백질을 BAP은 BiP으로부터 해리하였다. 또한 소포체내에 존재하는 Hsp70 homologue chaperone인 BiP과 Grp170에 대한 BAP의 결합특이성을 anti-Grp170과 anti-BAP 항체로 co-immunoprecipitation을 하여 확인한 결과 BAP은 Grp170과 결합을 하지 않았다. 따라서 BAP은 ER내에 존재하는 동일한 family group에 속하는 Grp170과 BiP에 대하여 BiP에만 특이성을 갖는 것으로 나타났다.

The Role of Lozenge in Drosophila Hematopoiesis

  • Drosophila hematopoiesis is comparable to mammalian differentiation of myeloid lineages, and therefore, has been a useful model organism in illustrating the molecular and genetic basis for hematopoiesis. Multiple novel regulators and signals have been uncovered using the tools of Drosophila genetics. A Runt domain protein, lozenge, is one of the first players recognized and closely studied in the hematopoietic lineage specification. Here, we explore the role of lozenge in determination of prohemocytes into a special class of hemocyte, namely the crystal cell, and discuss molecules and signals controlling the lozenge function and its implication in immunity and stress response. Given the highly conserved nature of Runt domain in both invertebrates and vertebrates, studies in Drosophila will enlighten our perspectives on Runx-mediated development and pathologies.
