Residual Characteristics of Bifenthrin and Imidacloprid in Squash (Bifenthrin과 Imidacloprid의 호박 중 관류특성)

  • In order to elucidate the residual characteristics of the insecticides, bifenthrin and imidacloprid, the pesticides were sprayed onto the squash and their residues were analyzed with GC-ECD and HPLC. Detection limits of the pesticides were $0.005\;mg;kg^{-1}$ and recoveries of bifenthrin and imidacloprid in squash were from 100.87 to 104.31 and from 79.71 to 92.54%, respectively. Half-lives of bifenthrin and imidacloprid in squash were from 1.8 to 2.9 and from 1.5 to 2.5 days, respectively. Initial concentration of bifenthrin and imidacloprid sprayed at the recommended rate and double rate of recommendation were less than their MRLs. And also, residueal concentrations of the pesticides were rapidly decreased in squash with time. At harvest, estimated daily intakes (EDIs) of the pesticides were less than 0.6% of their acceptable daily intakes (ADIs).

Efficient Production of Porcine Circovirus Type 2 Capsid Protein using Baculovirus

  • Porcine circovirus type 2 (PCV2) is a single-stranded circular DNA virus associated with Postweaning multisystemic wasting syndrome (PMWS), which is considered to be an important infectious swine viral disease. PCV2 capsid protein encoded by ORF2 is a structural protein and expected as the high immunogenicity protein. In this study, we generated recombinant baculovirus containing ORF2 of PCV2 and analyzed the optimal conditions for the production of capsid protein in insect cell. Production and status of recombinant capsid protein in insect cell were confirmed by SDS-PAGE and Western blot analysis using His tag antibody and anti-PCV2 serum. The yield of recombinant capsid protein was high like as shown visible on SDS-PAGE. Optimal multiplicity of infection (MOI) and infection time of recombinant virus were determined as 5 MOI and 4 days, respectively. ORF2 is known to have N-linked glycosylation site, but we couldn't detect the glycosylation of recombinant protein in insect cells.

A Genetically Engineered Pseudomonas fluorescens Strain Possesses Dual Activity Against Phytopathogenic Fungi and Insects

  • A Pseudomonas fluorescens strain was isolated and found to show antagonistic activity against phytopathogenic fungi and to possess a gene responsible for production of antibiotic 2,4-diacetylphloroglucinol. For the extension of biocontrol range, a gene for an Androetonus australis Hector insect toxin 1 (AaHIT1), one of the most known toxic insect-selective peptides, was designed and synthesized according to the preferred codon usage of Pseudomonas fluorescens, cloned, and transformed into the strain by pSUP106 vector, a broad-host-range plasmid. Bioassays indicated that the engineered strain was able to produce AaHIT1 with insecticidal activity, and at the same time retain the activity against plant pathogen. The experiments for nonplanted soil and rhizosphere colonization showed that, similar to the population of the wild-type strain, that of the engineered strain remained relatively constant in the first 10 days, and the subsequent 50 days, suggesting that AaHIT1 expression in the bacterial cell does not substantially impair its long-term colonization. It is first reported that a Pseudomonas fluorescens strain expressing an active scorpion neurotoxin has dual activity against phytopathogenic fungi and insects, making at attractive for agronomic applications.

Identification and Epigenetic Analysis of a Maternally Imprinted Gene Qpct

  • Most imprinted genes are concerned with embryonic development, especially placental development. Here, we identified a placenta-specific imprinted gene Qpct. Our results show that Qpct is widely expressed during early embryonic development and can be detected in the telecephalon, midbrain, and rhombencephalon at E9.5-E11.5. Moreover, Qpct is strikingly expressed in the brain, lung and liver in E15.5. Expression signals for Qpct achieved a peak at E15.5 during placental development and were only detected in the labyrinth layer in E15.5 placenta. ChIP assay results suggest that the modification of histone H3K4me3 can result in maternal activating of Qpct.

Study on Selective Media for Isolation of Entomopathogenic Fungi

  • To determine the optimal isolation conditions of the entomopathogenic fungi from soil, we compared their growth characteristics with non-entomopathogenic fungi on agar media containing various concentrations of cooper (II) chloride ($CuCl_2$) or dodine. The result showed that dodine medium is more selective, and the optimal concentration of dodine is determined with $50{\mu}g$/ml. We could isolate several putative entomopathogenic fungi from soil using this, and identify them using ITS analysis. As a result, 64% fungi were identified as typical entomopathogenic fungi. This selective medium may be useful to the rapid and simple isolation of entomopathogenic fungi from soil.

  • Life cycle assessment(LCA) is acknowledged as a valuable tool to quantify the environment impact of agricultural practice as well as final product(biodiesel) considering whole life cycle of the target product. As a preliminary research of LCA study for rapeseed(Brassica napus L.) biodiesel, the methodological issues which have to be regarded with high priority were dealt with. No life cycle inventory(LCI) based on local data are currently available for LCA of rapeseed cultivation, crushing, and conversion to rapeseed methyl ester(RME) in Korea. In this paper, the life cycle of rapeseed and methodological factors which have to be measured for building LCI of each process are provided and discussed, which are including seed, fertilizer, energy use in rapeseed cultivation environment; and crushing, RME conversion, and transportation in biodiesel production.
