Since the discovery of the immunomodulation property of mesenchymal stem cells (MSCs) about a decade ago, it has been extensively investigated whether MSCs can be used for the treatment of immune-related diseases, such as graft versus-host disease (GvHD). However, how to evaluate the efficacy of human MSCs for the clinical trial is still unclear. We used an MHC-mismatched model of GvHD (B6 into BALB/c). Surprisingly, the administration of the human MSCs (hMSCs) could reduce the GvHD-related mortality of the mouse recipients and xenogeneically inhibit mouse T-cell proliferation and $IFN-{\gamma}$ production in vitro. We recently established a new protocol for the isolation of a homogeneous population of MSCs called subfractionation culturing methods (SCM), and established a library of clonal MSC lines. Therefore, we also investigated whether MSCs isolated by the conventional gradient centrifugation method (GCM) and SCM show different efficacy in vivo. Intriguingly, clonal hMSCs (hcMSCs) isolated by SCM showed better efficacy than hMSCs isolated by GCM. Based on these results, the MHC-mismatched model of GvHD may be useful for evaluating the efficacy of human MSCs before the clinical trial. The results of this study suggest that different MSC lines may show different efficacy in vivo and in vitro.
최근 국내 통신 사업자와 가전 사업자 중심으로 자신들의 특정 스마트 기기와 네트워크통신망을 활용한 IoT 홈 서비스가 활발하게 제공되고 있다. 이는 사업자 별 전용 장치를 사용해야 하며 월정액 형태의 사용요금을 지불해야 하는 등 사용자의 요구사항을 충족시키지 못하고 있으며 사용자 주도적으로 쉽게 구성할 수 있는 장치와 서비스 환경에 대한 요구가 증대되고 있다. 따라서 본 논문에서는 IoT 플랫폼과 IoT 홈 게이트웨이로 구성되는 IoT 홈 서비스 환경 구조를 제안한다. 또한 DPWS의 기능을 활용한 IoT 장치의 자동 등록 기능과 MQTT 프로토콜을 통해 게이트웨이와 자동으로 인터페이스를 구축하는 기능을 제공하는 ACOME(Auto-Configuration of MQTT and REST) 메커니즘을 제안한다. 이는 시중에서 쉽게 구할 수 있는 오픈소스 하드웨어인 아두이노에 적용 가능한 라이브러리 형태로 제공하여 사용자의 요구사항을 보다 쉽게 구현할 수 있다. 마지막으로 서비스 및 장치 검색에 관한 성능분석을 위해 ACOME과 DPWS를 비교하였다.

The analysis of expressed sequence tags (ESTs) is an efficient approach for gene discovery, expression profiling, and the development of resources for functional genomics. To analyze the transcriptome of the abalone Haliotis discus hannai, we conducted EST analysis using seven cDNA libraries made from gill, gut, hepatopancreas, skin, muscle, testis, and ovary. Redundant ESTs were assembled into overlapping contiguous sequences using the assembly program ICAtools. We found that the total 1,393 ESTs formed 135 clusters and 951 singletons, indicating that the overall redundancy of the library was 22%. Of the 1,393 clones, BLAST identified 1,278 clones (91.7%) as known genes; 115 clones (8.3%) did not match any previously described gene. Based on the major functions of their encoded proteins, the identified clones were classified into 16 broad categories. Sequence analysis revealed the presence of micro satellite-containing genes that may be valuable for further gene mapping studies. This study contributes to the identification of numerous EST clones that can be applied to further clarifying the genetics and developmental biology of abalone.
Sequences from the clones of full-length enriched cDNA libraries serve as valuable resources for functional genomics related studies, genome annotation and SNP discovery. We analyzed 7,392 high-quality chromatograms (Phred value ${\geq}$30) obtained from sequencing the 5' ends of clones derived from full-length enriched cDNA libraries of Korean native pigs including brainstem, liver, cerebellum, neocortex and spleen libraries. In addition, 50,000 EST sequence trace files obtained from GenBank were combined with our sequences to identify cSNPs in silico. The process generated 11,324 contigs, of which 2,895 contigs contained at least one SNP and among them 610 contigs had a minimum of one sequence from Korean native pigs. Of 610 contigs, we randomly selected 262 contigs and performed in silico analysis for the identification of cSNPs. From the results, we identified 1,531 putative coding single nucleotide polymorphisms (cSNPs) and the SNP detection frequency was one SNP per 465 bp. A large-scale sequencing result of clones from full-length enriched cDNA libraries and identified cSNPs will serve as a useful resource to functional genomics related projects such as a pig HapMap project in the near future.
월드 와이드 웹(WWW)과 MBone 응용 등의 출현으로 인터넷 이용자가 급속히 증가되면서 인터넷 주소 공간의 확장과 멀티미디어 응용을 지원할 수 있도록 기존 인터넷 프로토콜의 개선이 요구됨에 따라 IETF에서는 차세대 인터넷 망 계층 프로토콜로써 IPv6를 개발하였다. 이 논문에서는 IPv6 프로토콜을 Windows NT의 커널 내부에 프로토콜 드라이버로 구현한 내용을 기술하고 있다. 본 연구에서는 IPv6 호스트로 동작되기 위해 필수적으로 요구되는 IPv6 헤더 처리기능, IPv6 주소처리기능, 제어메세지 처리기능, 그룹관리 메세지 처리기능, 이웃탐색 기능이 구현 및 시험되었다. 구현된 IPv6 프로토콜 드라이버 모듈은 하부 통신망 접속 카드와 표준 인터페이스인 NDIS를 통해 접속되며, 디스패치 함수화 Lower-Edge 함수를 이용하여 커널 내부에서 동작하는 드라이버 모듈을 상위 사용자 응용 및 하부 NDIS와 접속시키는 형태로 구현하였다. 구현된 IPv6 프로토콜 드라이버는 커널 모드에서 구현됨으로써 향상된 성능을 제공하며, 다이나믹 링크 라이브러리 형태로 사용자 인터페이스를 제공하므로 응용 프로그램 개발자들이 손쉽게 사용할 수 있도록 하였다.
The purpose of this study was to review the herbal medications of asthma through clinical studies in Korea and to be utilized in the treatment of asthma and in other clinical studies. All clinical researches about asthma published up to 13th February 2018 were found in two domestic electric databases, Oriental Medicine Advanced Searching Integrated System(OASIS) and National Discovery for Science Library(NDSL). Twenty-seven articles were selected and of these, there were 14 articles of before and after studies(BAS), 8 of case series(CS), 4 of case reports(CR) and 1 of the randomized controlled trial(RCT). The most frequently used prescription was "Cheongsangboha-tang(淸上補下湯)". There were various TCM patterns, and Tae-Eum-In(太陰人) was the most common Sasang constitution(四象體質). Frequently used evaluations to assess treatment effects were lung function tests such as Forced expiratory volume at one second (FEV1) and Quality of Life Questionnaire for Adult Korean Asthmatics(QLQAKA) questionnaire responses. We have found that herbal medicine treatment can be an effective treatment to improve the symptoms and the quality of life of asthmatic patients. But we consider that large-scale systematically designed clinical researches are needed additionally.
Chitin deacetylase (CDA) inhibitors were developed as novel antifungal agents because CDA participates in critical fungal physiological and metabolic processes and increases virulence in soil-borne fungal pathogens. However, few CDA inhibitors have been reported. In this study, 150 candidate CDA inhibitors were selected from the commercial Chemdiv compound library through structure-based virtual screening. The top-ranked 25 compounds were further evaluated for biological activity. The compound J075-4187 had an IC50 of 4.24 ± 0.16 µM for AnCDA. Molecular docking calculations predicted that compound J075-4187 binds to the amino acid residues, including active sites (H101, D48). Furthermore, compound J075-4187 inhibited food spoilage fungi and plant pathogenic fungi, with minimum inhibitory concentration (MIC) at 260 ㎍/ml and minimum fungicidal concentration (MFC) at 520 ㎍/ml. Therefore, compound J075-4187 is a good candidate for use in developing antifungal agents for fungi control.
Nucleoside deoxyribosyltransferase (NDT) is an enzyme that replaces the purine or pyrimidine base of 2'-deoxyribonucleoside. This enzyme is generally used in the nucleotide salvage pathway in vivo and synthesizes many nucleoside analogs in vitro for various biotechnological purposes. Since NDT is known to exhibit relatively low reactivity toward nucleoside analogs such as 2'-fluoro-2'-deoxynucleoside, it is necessary to develop an enhanced NDT mutant enzyme suitable for nucleoside analogs. In this study, molecular evolution strategy via error-prone PCR was performed with ndt gene derived from Lactobacillus leichmannii as a template to obtain an engineered NDT with higher substrate specificity to 2FDU (2'-fluoro-2'-deoxyuridine). A mutant library of 214 ndt genes with different sequences was obtained and performed for the conversion of 2FDU to 2FDA (2'-fluoro-2'-deoxyadenosine). The E. coli containing a mutant NDT, named NDTL59Q, showed 1.7-fold (at 40℃) and 4.4-fold (at 50℃) higher 2FDU-to-2FDA conversions compared to the NDTWT, respectively. Subsequently, both NDTWT and NDTL59Q enzymes were over-expressed and purified using a His-tag system in E. coli. Characterization and enzyme kinetics revealed that the NDTL59Q mutant enzyme containing a single point mutation of leucine to glutamine at the 59th position exhibited superior thermal stability with enhanced substrate specificity to 2FDU.
현재 우리는 소셜 네트워크 서비스(Social Network Service, 이하 SNS) 상에서 수많은 데이터를 만들어 내고 있다. 특히, 모바일 기기와 SNS의 결합은 과거와는 비교할 수 없는 대량의 데이터를 생성하면서 사회적으로도 큰 영향을 미치고 있다. 이렇게 방대한 SNS 데이터 안에서 사람들이 많이 이야기하는 이슈를 찾아낼 수 있다면 이 정보는 사회 전반에 걸쳐 새로운 가치 창출을 위한 중요한 원천으로 활용될 수 있다. 본 연구는 이러한 SNS 빅데이터 분석에 대한 요구에 부응하기 위해, 트위터 데이터를 활용하여 트위터 상에서 어떤 이슈가 있었는지 추출하고 이를 웹 상에서 시각화 하는 트위터이슈 트래킹 시스템 TITS(Twitter Issue Tracking System)를 설계하고 구축 하였다. TITS는 1) 일별 순위에 따른 토픽 키워드 집합 제공 2) 토픽의 한달 간 일별 시계열 그래프 시각화 3) 토픽으로서의 중요도를 점수와 빈도수에 따라 Treemap으로 제공 4) 키워드 검색을 통한 키워드의 한달 간 일별 시계열 그래프 시각화의 기능을 갖는다. 본 연구는 SNS 상에서 실시간으로 발생하는 빅데이터를 Open Source인 Hadoop과 MongoDB를 활용하여 분석하였고, 이는 빅데이터의 실시간 처리가 점점 중요해지고 있는 현재 매우 주요한 방법론을 제시한다. 둘째, 문헌정보학 분야뿐만 아니라 다양한 연구 영역에서 사용하고 있는 토픽 모델링 기법을 실제 트위터 데이터에 적용하여 스토리텔링과 시계열 분석 측면에서 유용성을 확인할 수 있었다. 셋째, 연구 실험을 바탕으로 시각화와 웹 시스템 구축을 통해 실제 사용 가능한 시스템으로 구현하였다. 이를 통해 소셜미디어에서 생성되는 사회적 트렌드를 마이닝하여 데이터 분석을 통한 의미 있는 정보를 제공하는 실제적인 방법을 제시할 수 있었다는 점에서 주요한 의의를 갖는다. 본 연구는 JSON(JavaScript Object Notation) 파일 포맷의 1억 5천만개 가량의 2013년 3월 한국어 트위터 데이터를 실험 대상으로 한다.
목적 : 본 연구는 감각통합치료효과에 대한 단일대상연구의 특성을 파악하고 질적 수준을 알아보고자 하였다. 연구방법 : 대한작업치료학회, 국가과학기술정보센터(NDSL), 누리미디어(DBpia), 학술연구정보서비스(RISS), 한국학술정보(KISS), 국회도서관의 원문제공 서비스를 이용하여 2002년부터 2013년 까지 발간된 논문 중 '감각통합', '단일대상연구', '응용행동분석'을 주요검색용어를 사용하여 검색하였다. 17편의 단일대상연구 논문을 선별하여 연구방법과 연구 설계의 질적 수준을 분석하였다. 결과 : 연구 설계 방법으로는 반전설계가 가장 많았으며, 총 회기와 연구기간, 중재시간은 다양하였고 중재 장소는 작업치료실과 감각통합실에서 이루어졌다. 대상자는 지적장애가 가장 많았다. 질적 수준은 모든 연구들이 중간수준이상으로 나타났다. 결론 : 단일대상연구의 질적 수준은 어느 정도 보장되고 있었으나 연구방법에 대한 논의와 보완이 필요한 것으로 보인다.
