The aim of the present study was to evaluate the effect of commercial monostrain and multistrain probiotics in diets on growth performance, intestinal morphology and mucin gene (MUC2) expression in broiler chicks. Three hundred seventy-eight 1-d-old male Arian broiler chicks were allocated in 3 experimental groups for 6 wk. The birds were fed on a corn-soybean based diet and depending on the addition were labeled as follows: control-unsupplemented (C), birds supplemented with Bacillus subtilis (BS) and lactic acid bacteria (LAB) based probiotics. Each treatment had 6 replicates of 21 broilers each. Treatment effects on body weight, feed intake, feed conversion ratio and biomarkers such as intestinal goblet cell density, villus length, villus width, and mucin gene expression were determined. Total feed intake did not differ significantly between control birds and those fed a diet with probiotics (p>0.05). However, significant differences in growth performance were found. Final body weight at 42 d of age was higher in birds fed a diet with probiotics compared to those fed a diet without probiotic (p<0.05). Inclusion of Bacillus subtilis based probiotic in the diets also significantly affected feed conversion rate (FCR) compared with control birds (p<0.05). No differences in growth performance were observed in birds fed different types of probiotic supplemented diets. Inclusion of lactic acid bacteria based probiotic in the diets significantly increased goblet cell number and villus length (p<0.05). Furthermore, diets with Bacillus subtilis based probiotics significantly increased gene expression (p<0.05), with higher intestinal MUC2 mRNA in birds fed diet with probiotics compared to those fed the control diet. In BS and LAB probiotic fed chicks, higher growth performance may be related to higher expression of the MUC2 gene in goblet cells and/or morphological change of small intestinal tract. The higher synthesis of the mucin gene after probiotic administration may positively affect bacterial interactions in the intestinal digestive tract, intestinal mucosal cell proliferation and consequently efficient nutrient absorption.

We investigated the effects of dietary pearlzyme (mudflat-bacteria origin protease) on growth performance and development of digestive organs in broilers. Two hundred forty, 4 day-old female Ross broiler chicks were divided into 2 groups (control vs. pearlzyme) which were randomly housed in 8 pens with 15 chicks/pen. They were fed one of two diets containing pearlzyme at 0 or 0.1% for 4 weeks. Dietary pearlzyme resulted in significant increase in body weight during the first week of the experiment (p<0.05). With age, weight gain and feed efficiency continued to decrease reaching significant level during the last week. Mortality was 3.3% in control but not in pearlzyme during the entire period of the experiment. Dietary pearlzyme resulted in increased weight (p<0.05) in the ceca and rectum, and increased length in the ceca (p<0.05). However, there were tendencies to increase the weight of the gizzard (p<0.071) but to decrease the length of the small intestine (p<0.068). The results of the current study show that dietary pearlzyme affects weight gain and the development of digestive organs.
To examine functionality of depolymerized alginate obtained by hydrolysis of alginate through a heating process at $121^{\circ}C$ on gastrointestinal physiology, the changes of body weight, organ weight and length, pancreatic and small intestinal composition, and light microscopy (LM) observation of small intestinal microvilli's appearances were checked in the rats. Rats were fed diets containing $1\%, 5\%, and 10\%$ of each depolymerized alginate (HAG-10, HAG-50, HAG-100) and alginate for 35 days, The feeding of 5 and $10\%$ HAG-50 and $10\%$ alginate diets for 35 days significantly depressed the body weight gain, but increased the length and weight of the small intestine and cecum in rats (p<0.01). Pancreatic protease activity was decreased significantly (p<0.01) in all groups except lo/o of HAG-10 diets, but the protein content increased in all groups, However, pancreatic amylase and lipase activities as well as DNA and RNA content were not significantly different. The small intestinal protein and the DNA content were the highest in diets fed $5\%$ HAG-50; RNA content increased significantly (p<0.01) in all groups except in the fiber-free diets. Light microscopy (LM) observation showed growth of small intestinal microvilli with numerous ridges; the multiplication of the convolution goblet cells in rats fed with diets containing $5\%$ of HAG-50 were more than others group.
We observed the morphological characteristics and identified the species of gnathostome larvae obtained from the imported Chinese loaches. The early third-stage larvae ($EL_3$) were collected from viscera of the loaches and a part of them were infected to mice. The advanced third-stage larvae ($AdL_3$) were recovered from the mice at 4 weeks post-infection. both larval worms were fixed loth 10% formalin, cleared in alcohol-glycerin solution, mounted with glycerin-jelly, and observed. A total of 369 $EL_3$ were collected from viscera of 9,493 Chinese loaches. The whole body of $EL_3$ was covered with about 190 transverse rows of minute cuticular spines and $0.624{\;}{\times}{\;}0.101{\;}mm$ in average size. A pair of lips were protruded at the anterior end, and the muscular esophagus and brownish intestine were followed. The characteristic head bulb was provided with 4 rows of hooklets. The average number of hooklets in the respective row was 36.7, 39.5, 41.6 and 44.3 posteriorly $AdL_3$ was $2.660{\;}{\times}{\;}0.346{\;}mm$ in average size, and retained the esophagus (about 0.755 mm length) and cervical sac (about 0.355 mm length). The average number of hooklets in the respective row on the head bulb was 39.0, 41.9, 43.9 and 45.6, posteriorly. On the basis of the morphological characteristics, they were identified as the third-stage larvae of Gnathostomc hispinun.
This paper deals with the morphology and taxonomy of the first reported species of echinostome belonging to the genus Himasthla Dietz, 1909, found from dunlins (Calidris alpina sakhalina) in Korea. Birds were captured with mist-nets at the tidal flat of Yeochari, Kanghwa-gun, Kyunggi-do, Korea. The worms were removed from avian intestines in physiological saline, fixed, acetocarmine stained and observed. Himasthla kusasigi Yamaguti, which was found in 1939, was characterized morphologically by a long and slender body (5.27 mm in length), besetting with spines and reniform head collar (0.27 mm) in a form of a single and uninterrupted row of 31 spines. The oral sucker (0.057$\times$0.0684 mm) is smaller than the ventral sucker (0.35$\times$0.29 mm). The ventral sucker was close to the anterior extremity of the body. The worm had two elliptical testes (anterior 0.47$\times$0.30 mm, posterior 0.59$\times$0.27 mm). The testes were close to the posterior end of the body. The uterus was very long, winding and extending through at least two thirds of the body length. The ovary (0.13$\times$0.14mm) was round and small. Below the ovary, a round and small seminal receptacle was found.
This study was conducted to investigate the effects of dietary supplementation of the mixture of probiotics (MP) on growth performance, size of small intestine, cecal microflora and ammonia concentrations in broiler chicks. A total of 700, one-day-old male broiler chicks were randomly allotted to four treatments with seven replications having 25 birds per pen. The birds were fed one of the four experimental diets; containing no antibiotics nor MP (negative control, NC), containing antibiotics without MP(positive control, PC), negative control with MP 0.1% and negative control with MP 0.2% for 5 weeks. During overall experiment, birds in PC and MP treatments had higher final BW and daily BW gains than birds in NC treatments; however, the significance was not identified. The feed conversion ratio of the chicks fed the diet containing MP was significantly improved as compared to those fed the NC diets. The weights of jejunum were increased by the MP (p<0.05), but weights of duodenum, ileum and length of small intestine were similar among the groups. Birds in PC treatment showed lower populations of total microbes and lactic acid bacteria than other groups (p<0.05), cecal ammonia concentrations of the chicks fed the diet containing MP were significantly decreased as compared to those of NC and PC (p<0.01). In conclusion, MP added to the broiler diets improved the feed conversion rate and reduced cecal ammonia concentration.
This study was conducted to investigate the feeding effect of $bio-silverlite^{(R)}$ on growth performance, organ phenomenon and cecum microflora in broiler chicks. The $bio-silverlite^{(R)}$ was made by an ion exchange between illite and $silver(Ag^+)$. There were four treatment groups: negative control group(non-treatment), antibiotic supplement group (positive control), 0.5% $bio-silverlite^{(R)}$ supplment group and 1.5% $bio-silverlite^{(R)}$ supplement group. Total 200 birds was assigned for this five replication tests, allocating 10 birds into each treatment. Experimental diets were formulated on isocalories and isonitrogen for the whole experimental period. Body weight gain was higher in antibiotic supplementation (+C) and $bio-silverlite^{(R)}$ supplement groups(S 0.5% and 51.5%) than the negative control group(-C), and feed efficiency was significantly enhanced with increase of the level of $bio-silverlite^{(R)}$ supplement. The length of small intestine was longer in +C than in -C and $bio-silverlite^{(R)}$ supplement groups (P<0.05), and the weight of small intestine was proportional to the level of $bio-silverlite^{(R)}$ supplement. Crop weight was lower in $bio-silverlite^{(R)}$ supplement group than in -C and +C groups (P<0.05), and the cecum weight was heavier in $bio-silverlite^{(R)}$ supplementation group. Intestinal villi height was longer in 51.5% group at 3 weeks and 6 weeks of age than in -C and +C groups. With the respect of the formation of intestinal microflora, TBC and CBC was not affected by age and feed additive. However, the number of LAB was slightly higher in $bio-silverlite^{(R)}$ supplement group than in -C and +C groups.
Purpose: Anastomotic leakage (AL) is associated with high morbidity and mortality, high reoperation rates, and increased hospital length of stay. Here we investigated the risk factors for AL after anterior resection for rectal cancer with a double stapling technique. Patients and Methods: Data for 460 patients who underwent primary anterior resection with a double stapling technique for rectal carcinoma at a single institution from 2003 to 2007 were prospectively collected. All patients experienced a total mesorectal excision (TME) operation. Clinical AL was defined as the presence of leakage signs and confirmed by diagnostic work-up according to ICD-9 codes 997.4, 567.22 (abdominopelvic abscess), and 569.81 (fistula of the intestine). Univariate and logistic regression analyses of 20 variables were undertaken to determine risk factors for AL. Survival was analysed using the Cox regression method. Results: AL was noted in 35 (7.6%) of 460 patients with rectal cancer. :Median age of the patients was 65 (50-74) and 161 (35%) were male. The diagnosis of AL was made between the 6th and 12th postoperative day (POD; mean 8th POD). After univariate and multivariate analysis, age (p=0.004), gender (p=0.007), tumor site (p<0.001), preoperative body mass index (EMI) (p<0.001), the reduction of TSGF on 5th POD less than 10U/ml (p=0.044) and the pH value of pelvic dranage less than or equal to 6.978 on 3rd POD (p<0.001) were selected as 6 independent risk factors for AL. Background: Damage to the healthy intestinal epithelial layer and regulation of the intestinal immune system, closely interrelated, are considered pivotal parts of the curative treatment for inflammatory bowel disease (IBD). Plant-based diets and phytochemicals can support the immune microenvironment in the intestinal epithelial barrier for a balanced immune system by improving the intestinal microecological balance and may have therapeutic potential in colitis. However, there have been only a few reports on the therapeutic potential of plant-derived exosome-like nanoparticles (PENs) and the underlying mechanism in colitis. This study aimed to assess the therapeutic effect of PENs from Panax ginseng, ginseng-derived exosome-like nanoparticles (GENs), in a mouse model of IBD, with a focus on the intestinal immune microenvironment. Method: To evaluate the anti-inflammatory effect of GENs on acute colitis, we treated GENs in Caco2 and lipopolysaccharide (LPS) -induced RAW 264.7 macrophages and analyzed the gene expression of proinflammatory cytokines and anti-inflammatory cytokines such as TNF-α, IL-6, and IL-10 by real-time PCR (RT-PCR). Furthermore, we further examined bacterial DNA from feces and determined the alteration of gut microbiota composition in DSS-induced colitis mice after administration of GENs through 16S rRNA gene sequencing analysis. Result: GENs with low toxicity showed a long-lasting intestinal retention effect for 48 h, which could lead to effective suppression of pro-inflammatory cytokines such as TNF-α and IL-6 production through inhibition of NF-κB in DSS-induced colitis. As a result, it showed longer colon length and suppressed thickening of the colon wall in the mice treated with GENs. Due to the amelioration of the progression of DSS-induced colitis with GENs treatment, the prolonged survival rate was observed for 17 days compared to 9 days in the PBS-treated group. In the gut microbiota analysis, the ratio of Firmicutes/Bacteroidota was decreased, which means GENs have therapeutic effectiveness against IBD. Ingesting GENs would be expected to slow colitis progression, strengthen the gut microbiota, and maintain gut homeostasis by preventing bacterial dysbiosis. Conclusion: GENs have a therapeutic effect on colitis through modulation of the intestinal microbiota and immune microenvironment. GENs not only ameliorate the inflammation in the damaged intestine by downregulating pro-inflammatory cytokines but also help balance the microbiota on the intestinal barrier and thereby improve the digestive system.
Scutellariae Radix (Scu.), one of the immune-regulatory substances, is recognized to play the role in the metabolic process of inflammation, allergy and immunity. It has been traditionally used in the Oriental medicine to treat inflammatory bowel diseases (IBD). The purpose of this study was to evaluate the effects of water extracts of Scutellariae Radix on the spleen lymphocyte immune function in the Balb/c female mice treated with dextran sodium sulfate (DSS) to induce colitis. Water extract of Scutellariae Radix (100 mg/kg) and sulfasalazine (50 mg/kg) were administrated orally for 2 weeks of experimental period. Mice were divided into three experimental groups randomly: DSS group (5% DSS was ad libitum for 5 days) as control group, DSS + Scu. (water extracts of Scutellariae Radix for 2 weeks after 5% DSS was ad libitum for 5 days) as experimental group, and DSS + Sulfasalazine group (Sulfasalazine for 2 weeks after 5% DSS was ad libitum for 5 days) as positive control group. Levels of Ig A, Ig E, CD4$^{+}$, CD8$^{+}$, TNF-$\alpha$ and other cytokines were measured. Treatment of DSS for 5 days induced bowel inflammation and the treatment with Scu. water exteract and sulfasalazine significantly recovered the damage. The length of intestine of DSS group was significantly shorter than that of other groups. The serum and fecal concentration of Ig A of SS + Scu group was higher than those of DSS group. The contents of CD4$^{+}$ T cells was higher in the DSS + Scu. group than the other groups and CD8$^{+}$ T cells was the lowest in DSS + Sulfasalazine group. The Ig A level of cultured supernatant of spleen lymphocyte was the highest, while the Ig E level was the lowest in SS + Scu group. The concentration of TNF-$\alpha$, cytokine secreted from the Th1 cell in the supernatant spleen lymphocyte, was the highest in the DSS group and the lowest in the DSS + Scu. group. The concentration of IFN-${\gamma}$ and ll...-12 was lower in the DSS + Scu. group than those of the other groups. The concentration of IL-4 in the supernatant of spleen lymphocyte was the lowest in the DSS + Scu. group but IL-10 was not significantly different. Based on these findings, water extract of Scutellariae Radix exhibited the inhibitory effect via IL-4 production thereby inhibited the production of Ig E and strengthened immune system, and alleviated injury in DSS- induced colitis mice model.
