Alfalfa mosaic alfamoviruses(AIMV) were isolated from infected potato (Solanum tuberosum) and azuki bean (Paseolus angularis) in Korea. Two AIMV isolated from potatoes were named as strain KR (AIMV-KR1 and KR2) and AIMV isolated from azuki bean was named as strain Az (AIMV-Az). Each isolated AIMV strain was characterized by using their host ranges, symptom developments, serological relations and nucleotide sequence analysis of coat protein (CP) gene. Strains KR1, KR2, and Az were readily transmitted to 20 of 22 inoculated plant species including bean, cowpea, tomato, tobacco, and potato. AIMV-KR1 and KR2 produced the typical symptoms like chlorotic or necrotic spots in Chenopodium quinoa and Solanum tuberosum cv. Superior. AIMV-Az caused bright yellow mosaic symptom and leaf malformation in Nicotiana glauca, which were different from the common mosaic symptom caused by AIMV-KR1 and KR2. Electron microscope observation of purified virus showed bacilliform virions containing a single-stranded plus-strand RNAs of 3.6, 2.6, 2.0 and 0.9 kbp in length, respectively, similar in size and appearance to those of Alfamovirus. In SDS-PAGE, the coat protein of the two viruses formed a consistent band that estimated to be about 24kDa. The CP genes of the AIMV strains, KR1, KR2, and Az have been amplified by RT-PCR using the specific primers designed to amplify CP gene from viral RNA-3, cloned and sequenced. Computer aided analysis of the amplified cDNA fragment sequence revealed the presence of a single open reading frame capable of encoding 221 amino acids. The nucleotide and peptide sequence of viral CP gene showed that strain KR1, KR2, and Az shared highest nucleotide sequence identities with AIMV strain 425-M at 97.7%, 98.2%, and 97.2%, respectively. CP gene sequences of two strains were almost identical compared with each other. Altogether, physical, serological, biological and molecular properties of the purified virus.
This research was conducted to identify plant regulatory regions by gene tagging method. A promoterless GUS coding sequence was introduced to Populus nigra ${\times}$ P. maximowiczii via Agrobacterium strains(LBA4404/EHA101), and putative transgenic poplars were selected by culturing on medium containing G418($60mg/{\ell}$) and by GUS assay. Among them one positive plant was to amplify the native sequences flanking to the introduced GUS gene in plant genome by inverse PCR method and from this 730 by DNA product was obtained. After subcloning and sequencing, it has 88% homology to the Eucalyptus gunnii CAD(cinnamyl alcohol dehydrogenase) gene. The GUS gene fused with the putative promoter reinserted into poplar leaves by particle bombardment method to test the funtional promoter activity. Upon staining with X-gluc, many blue spots appeared on the leaf segments bombarded by the chimeric gene 2-3 days, thus the isolated DNA fragment contain some possible coding region as well as a putative regulatory sequences of poplar CAD gene.

Tomato spotted wilt virus (TSWV) occurred abruptly with a high incidence rate in 14 vegetable crops in Anyang area, Gyunggido in 2004. TSWV was identified by the symptoms on the indicator plants, immunocaptured reverse transcription-polymerase chain reaction (IC/RT-PCR), virion captured (VC)RT-PCR and RT-PCR using total RNA from the infected plants. Vegetable crops infected with TSWV included table tomato, cherry tomato, red pepper, lettuce, chicory, red leaf chicory, red mustard, dragon tongue, treviso, potato, perilla, sesame, pumpkin, and ssamchoo (hybrid of oriental cabbage and cabbage). The incidence of TSWV in fields ranged from 30 to $100\%$. TSWV usually produced necrosis, wilt and/or severe mosaic with typical single or double ring spots on the leaves. TSWV could be detected in Frankliniella occidentalis collected from the crops in the fields with $90\%$ rate by IC/RT-PCR.

This study was conducted to analyze the influence of simulated acid rain on leaves of Liliodendron tulipifera seedlings. The seedlings were treated with four levels of simulated acid rain with pH of 5.6, 4.9, 3.9 and 2.9, and then chlorophyll contents and the degree of foliar damage were investigated. Differences were statistically significant among treatments in chlorophyll contents in all of the tested soils. The total contents of chlorophyll tended to decrease as the simulated acid rain treatment was maintained. Chlorophyll contents, however, did not decrease according to the decrement of pH levels. Foliar damage increased as the simulated acid rain was maintained in all tested soils. Especially, the damage dramatically increased between May and July as the acidity increased. As the treatment of simulated acid rain continued, deformed new leaves appeared. In all treatments except the control, the leaves turned brown and the damage increased with time. Necrotic spots appeared during the first month of treatment at the pH level of 2.9 in all soil types. The damage by acid rain should be considered when we plant Liliodendron tulipifera.
This experiment was conducted to investigate the influence of amounts of lime dressed on the growth of rice by the treatment of percolation and nonpercolation in the acid sulphate soil. And also analysis of soil chemical components after treatment was carried out. The results obtained were summarized as follows: 1. In the initial stage of growth, number of tillers and plant length showed no distinct differences between the treatments of percolation and nonpercolation, but after August the effect of lime appeared and the percolation treatment was more effective than the nonpercolation. 2. Lime dressing affected good influence on the panicles, grain per panicles and the rate of grain formation, and the treatment of percolation showed better results than nonpercolation. 3. If the yield of rough rice in the control (nonpercolation and lime dressing) was 100, it was 194 in the treatment of nonpercolation 12me/100gr of lime dressed, 268 in the treatment of percolation-4me/100gr of lime and 315 in the 8me/100gr-percolation. 4. Lime dressing affected good influence on the control of Helminthosporium leaf spots. 5. In the case of lime dressing, amounts of available phosphate and soluble silicon dioxide were increased, but ferrous ion ($Fe^{{+}{+}}$) were decreased.

A bacterial disease of small red bean (Phaseolus angularis) was observed on field-grown plants in Suwon in year 2003. Leaf symptoms initially appeared as water-soaked spots that gradually enlarged, became flaccid and necrotic and were often bordered by a small zone of lemon yellow tissue. In the case of severe infection, dead leaves were defoliated. Pod symptoms consisted of the lesions that were generally circular, slightly sunken and dark reddish brown. Isolation made from diseased leaves on yeast extract dextrose calcium carbonate agar yielded nearly pure cultures of a yellow-pigmented bacterium typical of a xanthomonad. Three bacterial strains were purified and used for further tests. Pathogenicity of strains was confirmed on 3-week-old small red bean plants sprayed with bacterial suspensions containing $10^8 cfu/ml$ of phosphate buffered saline. The representative Xanthomonas strains isolated from small red bean were compared with X. axonopodis pv. phaseoli and X. axonopodis pv. phaseoli var. fuscans type strains for fatty acid profiles, biochemical tests and metabolic fingerprints using Biolog GN2 microplate, showing that all outcomes were indistinguishable between our isolates and reference strains. Two of three strains produced a melanin-like brown pigment extracellularly on King's medium B agar. These results suggest that this new small red bean disease observed in Suwon is bacterial fuscous blight caused by X. axonopodis pv. phaseoli and X. axonopodis pv. phaseoli var. fuscans.

An obligate plant pathogenic fungus Puccinia zoysiae Dietel, the causal agent of rust disease, which is implicated in the damage of zoysiagrass (Zoysia spp.) in Korea. The fungus is one of the important pathogens, which attacks and colonizes susceptible zoysiagrass. Zoysia rust disease was observed in Jangseong, Hoengseong and Hapcheon in 2014 and 2015, Korea. The typical symptoms of the disease appeared first as small white spots on the leaf and stem of zoysiagrass plants, and turn brownish, dark brown spores revealed on the lesions. Uredospores were light brown in color, ellipsoid or spherical in shape and $22.0{\sim}25.0{\times}15.0{\sim}17.5{\mu}m$ in size. On the basis of the morphological characteristics, the fungus was identified as P. zoysiae. Pathogenicity of the fungus was proved by artificial inoculation on Z. japonica. The rust disease firstly appeared approx. early or middle June and then increased middle of October in Korea. In field, Z. matrella ecotypes were more resistant than Z. japonica ecotypes against the pathogen. Disease response against the pathogen was different among ecotypes of each species, respectively.
A new peanut cultivar "Jopyeong" (Arachis hypogaea ssp. hypogaea L.) was developed at the Yeongnam Agricultural Research Institute, NICS, in Milyang in 2006. It was developed from the cross between the very short stem cultivar "CUP brittle" and the high-yielding cultivar "Daekwang". "Jopyeong" which is Virginia plant type has 23 branch number per plant with early maturing and ellipse-shaped large kernel. Each pod has two grains with brown testa and 100 seed weight was 87g in the regional yield trials (RYT). This variety also showed more resistant to late leaf spots compared with check one. Especially it has resistance to lodging owing to short stem and erect plant type. In the regional yield trials "Jopyeong" was out-yielded than check variety by 11% with 8.37 ton/ha for fresh pod and by 4% with 3.95 ton/ha for grain.

There is a need to breed and distribute of domestic varieties for Lilium longiflorum because most of the lily bulbs cultivated in Korea were imported from foreign country. The L. longiflorum 'Ace' and 'Nellie White' collected from 1996 had been self-crossed from 1996 to 2010. In 2012, the L. longiflorum 'Bright LoSong' was developed as a F1 hybrid crossed between 'L2-17-4' and 'L2-16-6' that obtained by 7th self-fertilization. The 'Bright LoSong' was tested for homogeneity, regional adaptability test, consumer preference show from 2014 to 2015, and it was registered in 2017. The flower of 'Bright LoSong' is a trumpet shaped single flower with no spots.It has 5.8 flowers per plant, which is a relatively many number compared control 'Mount Carmel', Its flower diamater is 9.8cm, relatively a bit small size. And the flower inflorescence has umbel or raceme while the attitude of longitudinal axis is horizontal. The plant height is 38.4cm with 30.8 leaves per plant and 1.9cm of the leaf width. 'Bright LoSong' has a longer flowering period because of relatively small diameter or many number of flowers than those of the control 'Mount Carmel'. Thus, the 'Bright LoSong' showed a high consumer preference, and the cultivar has the advantage of being able to reproduce since the parents are pure lines.

The collected Lilium Asiatic hybrids 'Butter Pixie' for a maternal line, 'Mona' for a paternal line based on the genetic resources from 2000 had been crossed self-fertilization from 2000 to 2006. In 2008, We developed Lilium Asiatic 'Yellow LiASong' as a F1 hybrid which was crossed 'L2-18-1' and 'L2-21-4' those obtained by 4th self-fertilization. The flower of 'Yellow LiASong' is a bowl shaped single flower with no spots that 5.8 flowers per plant, which is a relatively many number compared other cultivars, and 15.5cm flower diameter with a medium size. And the flower inflorescence has umbellate and racemose while the attitude of longitudinal axis is erect. The plant height is 36.2cm with 35.5 leaves per plant and 10.7cm of the leaf length. Thus, The 'Yellow LiASong' showed a high consumer preference. Which has a longer flowering period due to having a relatively larger diameter and number of flowers than that of 'Tiny Bee', which is a comparable variety.
