Growth Pattern and Phenology of Mankyua chejuense B.Y. Sun, M.H. Kim & C.H. Kim (제주고사리삼(Mankyua chejuense B.-Y. Sun, M.H. Kim & C.H. Kim)의 생장 형태 및 식물계절학적 특성)

  • This study was conducted to analyze the pattern of growth and phenological characteristics of Mankyua chejuense B.-Y. Sun, M.H. Kim & C.H. Kim, which belong to the Ophioglossaceae family. M. chejuense asexually reproduced using rhizomes, and a clone of the species grew up to 52 cm based on root growth and new leaves was produced from rhizomes. The development of leaves were divided into four stages; leaf emergence-separation and growth of leaf-sporophyll maturation-senescence. Most leaves emerged in July and August with high temperatures and precipitation and most leaves reached full expansion during September-October and died during April-May next year. The life span of leave was 10 months, from July to April, and the start of leaf senescence varied depending on the habitat environment and this might result from micro environmental differences among the habitats.

Psidium guajava L. leaf extract inhibits adipocyte differentiation and improves insulin sensitivity in 3T3-L1 cells

  • BACKGROUND/OBJECTIVES: Psidium guajava L. (guava) leaves have been shown to exhibit hypoglycemic and antidiabetic effects in rodents. This study investigated the effects of guava leaf extract on adipogenesis, glucose uptake, and lipolysis of adipocytes to examine whether the antidiabetic properties are mediated through direct effects on adipocytes. MATERIALS/METHODS: 3T3-L1 cells were treated with 25, 50, 100 ㎍/mL of methanol extract from guava leaf extract (GLE) or 0.1% dimethyl sulfoxide as a control. Lipid accumulation was evaluated with Oil Red O Staining and AdipoRed assay. Immunoblotting was performed to measure the expression of adipogenic transcription factors, fatty acid synthase (FAS), and AMP-activated protein kinase (AMPK). Glucose uptake under basal or insulin-stimulated condition was measured using a glucose analog 2-[N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl) amino]-2-deoxy-D-glucose. Lipolysis from fully differentiated adipocytes was measured by free fatty acids release into the culture medium in the presence or absence of epinephrine. RESULTS: Oil Red O staining and AdipoRed assay have shown that GLE treatment reduced lipid accumulation during adipocyte differentiation. Mitotic clonal expansion, an early essential event for adipocyte differentiation, was inhibited by GLE treatment. GLE inhibited the expression of transcription factors involved in adipocyte differentiation, such as peroxisome proliferator-activated receptor 𝛄 (PPAR𝛄), CCAAT/enhancer-binding protein α (C/EBPα), and sterol regulatory element-binding protein-1c (SREBP-1c). FAS expression was also decreased while the phosphorylation of AMPK was increased by GLE treatment. In addition, GLE increased insulin-induced glucose uptake into adipocytes. In lipid-filled mature adipocytes, GLE enhanced epinephrine-induced lipolysis but reduced basal lipolysis dose-dependently. CONCLUSIONS: The results show that GLE inhibits adipogenesis and improves adipocyte function by reducing basal lipolysis and increasing insulin-stimulated glucose uptake in adipocytes, which can be partly associated with antidiabetic effects of guava leaves.

Quality Characteristics of Domestic Wheat White Bread with Substituted Nelumbo nucifera G. Tea Powder (백련차 분말을 대체한 우리밀 식빵의 품질 특성)

  • In this study, domestic wheat white breads were prepared with the substitution of 1.5, 3.0, 4.5, and 6.0% Nelumbo nucifera G. tea powder(NNTP). The samples and a control were then compared with regard to quality characteristics, including pH, total titratable acidity, fermentation power of dough expansion, specific volume, baking loss, moisture content, color, textural characteristics, external and internal surface appearances, and sensory qualities in order to determine the optimal ratio of NNTP in the formulation. As the NNTP contents increased, the pH of dough and bread, baking loss, and lightness decreased, whereas the total titratable acidity of dough and bread increased. The fermentation power of dough expansion increased with increasing incubation time. The NNTP samples evidenced significantly higher specific volume, greenness, yellowness, resilience, and crumb consistency than were observed in the control group. However, hardness and fracturability evidenced the opposite effect. The water content and uniformity of the crumb pores were highest at a substitution level of 1.5%, and were lowest at a level of 4.5%. Crumb color, flavor, and delicious taste decreased with increasing NNTP contents, whereas lotus leaf flavor, astringency, bitterness, and off-flavor increased. Density of the crumb pore and crumb springiness were not significantly different among the samples. Softness, chewiness, and overall acceptability were maximal with the 1.5% substitution, and were minimal in the 6.0% group. In conclusion, our results demonstrate that $1.5{\sim}3.0%$ NNTP may prove quite useful as a substitute for domestic wheat flour in the production of white bread, and may evidence favorable nutritional and functional properties.

Quality Characteristics of White Bread with Barley Leaves Tea Powder (보리잎차 분말을 대체한 식빵의 품질 특성)

  • In this study, white bread was prepared containing 1.5, 3.0, 4.5 or 6.0% barley leaves tea powder (BLTP). The samples along with a control were then compared regarding their quality characteristics, including pH, total titratable acidity, fermentation power of dough expansion, specific volume, baking loss, moisture content, color, textural characteristics, internal surface appearances and sensory qualities, all to determine the optimal ratio of BLTP. As the BLTP content increased, the pH of the dough and bread and the lightness decreased, whereas the total titratable acidity increased. The fermentation power of dough expansion increased with a longer incubation time. The control group was evidenced by a significantly higher specific volume and baking loss than that observed in the BLTP samples. However, greenness and yellowness evidenced the opposite effect. Hardness was highest at a substitution level of 1.5% and lowest at a level of 4.5%. Fracturability and resilience were not significantly different among the samples. For the internal surface appearance, darkness and greenness increased both increased at higher BLTP content. In the sensory evaluation, color, flavor and overall acceptability were highest in the control bread samples but minimal at a substitution level of 6.0%. Softness was the highest at the 3.0% substitution level and lowest in the control bread samples. Barley leaf flavor, astringency, bitterness and off-flavor increased as the BLTP content increased. Delicious taste was not significantly different among the samples. In conclusion, the results demonstrated that 4.5% BLTP may prove quite useful as a substitute for wheat flour in the production of white bread and may provide favorable nutritional and functional properties.

Identification of Leaf Characteristics from Various Crosses in Relation with Populus glandulosa U. (Populus glandulosa U.에 유사(類似)한 교잡종(交雜種)의 엽특성(葉特性))

  • This study was conducted to widen the range of characteristics of the hybrids, P.alba ${\times}$ P.glandulosa, i.e. aiming for gene population expansion P.glandulosa seemed to have the similar characteristics as the one segregated from the natural hybrids between P.alba and P.davidiana. Thus the main objectives of this study were to make many crosses among poplars and then to identify leaf characteristics of the crosses similar to P.glandulosa, the results obtained can be summerized as follows; 1. Leaf characteristics such as leaf margin, presence of glands at leaf base and pubescence density, of crosses made from P.alba.davidiana ${\times}$ P.datidiana, and P.davidiana.alba ${\times}$ P.davidina showed 44% and 90%, respectively, of similarity to P.glandulosa. 2. The ratio of leaf size, including leaf length, leaf width, length from leaf base to width line, and petiole length, of the above crosses was similar to P.glandulosa. 3. Pubescence density of the dorsal leaf surface in hybrids between P.alba and P.davidiana showed generally intermediate of the parental appearance. Frequency of pubescence appearance differed from depending upon the use of P.alba, either as a female or a male parent. The use of P.alba as a male parent increased frequency of Pubescence appearance. 4. The presence of glands at the leaf base in P.glandulosa may be inherited from P.davidiana which possesses gland although gland is not present in all P.davidiana rather from P.alba which has no gland.

Morphological Variation and Characteristics of Native Medium-Leaf Type Zoysiagrasses (Zoysia spp.) by Site Environment (입지환경에 따른 자생 중엽형 한국잔디의 형태적 변이 및 특성)

  • It is important for genetic resources to collect and identify in native medium-leaf type zoysiagrasses species distributed in Korea. This study was conducted to investigate morphological variation and characteristics of native medium-leaf type zoysiagrasses from coastal, island and inland regions in Korea. Among them, 75 collected lines was confirmed to have various morphological variations, accessions were classified into 2 main based group coastal and inland regions by morphological characteristics and site environment. Group I included Z. sinica type, this group showed 3.7 mm in leaf width, 29 in number of seed per spikelet and 5.0 mm in seed length. Group II included Z. japonica type, this group showed 4.4 mm in leaf width, 42 in number of seed per spikelet and 3.5 mm in seed length. There is a need for additional research on growth characteristics and the molecular level for the introgressive hybridization between species which confirmed that cross-pollination is possible due to protogyny. The individuals showing variations should be preserved as valuable genetic resources for the expansion of variations in zoysiagrasses, and the results of this investigation on the genetic resources collected will be highly valuable in breeding high quality turfgrass.

Physical Properties of Pan Bread Made with Various Amounts of Squeezed Danggui Leaf (Angelica acutiloba Kitagawa) Juice (당귀잎 착즙액을 이용하여 제조한 식빵의 물리적 특성)

  • Danggui leaf (Angelica acutiloba Kitagawa) has numerous dietary fiber and healthy functional properties such as anti-inflammatory activity. However, it is usually discarded after its roots are harvested. For wide application of danggui leaf, squeezed danggui leaf juice was used for making pan bread. Treatments included imported wheat flour (IMWF) and domestic wheat flour (DOWF) as a control, 1% soluble solid contents of squeezed danggui leaf juice instead of wheat flour (1% SDLJ), 2% SDLJ, 3% SDLJ, and 4% SDLJ. In the fermentation expansion, dough volume was not significantly different between DOWF and 1% SDLJ, whereas it decreased according to increased amount of squeezed danggui leaf juice. Regarding physical properties, springiness and cohesiveness decreased according to increased amount of squeezed danggui leaf juice and longer storage period. Gumminess, brittleness, and hardness increased according to increased amount of squeezed danggui leaf juice, although these were not statistically different among IMWF, DOWF, and 1% SDLJ after making pan bread. Principal component analysis (PCA) was performed to assess the correlation between storage period and physical properties. In the PCA, variance proportion of principal component (PC) 1 was 0.87 while that of PC 2 was 0.10. Further, 1% SDLJ showed similar results as DOWF and INWF after making pan bread and after 3 days of storage. In conclusion, use of 1% SDLJ is desirable for making squeezed danggui leaf juice.

  • This study was conducted to investigate the effects of temperature and light conditions on the vigor and growth responses of red pepper(Capsicum annuum L.) seedlings in the nonheated plastic houses with triple and quadruple coverings during winter. The results obtained were as follows ; 1. The growth in terms of plant height, number of leaves, leaf area and dry weight of each organ was reduced up to 50% in the triple coverings compared to quadruple ones. The greatest difference between the triple coverings and quadruple ones was the leaf area, showing two to five times larger in the quadruple ones. Therefore, the differences of the early environmental conditions during raising seedling stage was well reflected. 2. The leaf expansion in the early stage of seedling was delayed in the triple coverings and subsequently crop growth rate(CGR) significantly reduced. The specific leaf area (SLA) in the triple coverings was less than quadruple ones. The net assimilation rate (NAR) was increased in the early stage of seedlings and then reduced in the quadruple coverings. In triple ones, however, the great reduction showed at six weeks after sowing and then increased sharply. 3. The linear relationship among leaf area, total dry weight and leaf dry weight was observed in the all experimental areas, while between leaf area and crop growth rate only in the quadruple coverings, and between leaf area and net assimilation rate in both triple and quadruple ones. 4. The dry matter partitioned to the leaf increased up to six weeks after sowing in the triple coverings but reduced in the stem and root. However, the adverse results were obtained in the quadruple ones. 5. In the triple coverings, the number of branches showed the exponential increment while fruit weight showed linear increment in both triple and quadruple ones. Higher yield was obtained in the quadruple ones. 6. The shoot dry weight among the characteristics of seedlings was greatly contributed to the yield of red pepper after transplanting. Accordingly, one of the decisive criterion for good quality of red pepper seedlings was determined by the highest correlation between shoot dry weight of growing plant and fruit yield of pepper plant.

Physicochemical and Sensory Properties of Pan Bread Made with Various Amounts of Squeezed Perilla Leaf Juice (깻잎착즙액을 이용하여 제조한 식빵의 이화학적 및 관능적 특성)

  • For wide application of perilla leaf, which has various healthy functions and can be easily cultured across Korea, the physicochemical and sensory properties of pan bread made with various amounts of squeezed perilla leaf juice (SPLJ) were analyzed. When dough characteristics were analyzed by using farinograph, consistency and dough development time were not significantly different between the control and bread dough made with various amounts of SPLJ, whereas dough stability time increased with increasing SPLJ amount. Expansion rate of dough decreased with increasing SPLJ amount. The volume, specific volume, and baking loss rate of pan bread made with various SPLJ amounts decreased with increasing SPLJ amount. Pan bread crumb colors became thickened and greenish with increasing SPLJ amount. For physical properties of pan bread made with various SPLJ amounts, springiness and cohesiveness decreased with increasing SPLJ amount, whereas brittleness, chewiness, and hardness increased with increasing SPLJ amount. In the sensory strength analysis, pore uniformity and soft texture decreased with increasing SPLJ amount, whereas crumb color (dark greenish), perilla leaf odor, perilla leaf taste, and chewing texture increased with increasing SPLJ amount. In the overall acceptance analysis, 1.5% SPLJ was the most preferred with a score of 7.10. However, statistical differences between 1.5% and 1.0% SPLJ were not detected at P<0.05. In the partial least squares analysis, consumers liked bread with a green crumb color, perilla leaf odor, perilla leaf taste, and soft and chewing texture. In conclusion, physicochemical properties of pan bread made with SPLJ were less desirable than those of the control; however, consumer acceptance of pan bread made with 1.5% SPLJ was shown the highest. Therefore, methods for increasing physicochemical properties of pan bread made with SPLJ need to be developed for wide application of perilla leaf.

Physico-chemical Properties of leaf tobacco by expansion processes. (잎담배의 팽화방법에 따른 팽화율 및 내용성분 특성)

  • Flue-cured tobaccos(cv. NC82, grade AB3O-1) expanded by super steam, volatile organic solvents(tricuorofluoromethane, propane), carbon dioxide, nitrogen, and by freeze-drying were examined for their physico-chemical properties. The expanding rate by using the organic solvents or carbon dioxide at the impregnated pressure was 70% or more, whereas the rate by super steam and freeze-drying was 49.4 and 42.9%, respectively. The contents of total sugar, alkaloid and ether extracts of the expanded tobacco by organic solvents or carbondioxide were decreased to 27.6, 20% or more, and 13%, respectively, when compared to non-expanded one. The contents of citric acid of the tobacco expanded by using trichlorofluoromethane and carbon dioxide were increased to 19.1, 14.0%, respectively. Linoleic acid in the tobacco expanded by super steam or trichlorofluoromethane was also increased to 17.3%, but malonic acid in the tobacco expanded by super steam was 18.5% less than that in the non-expanded one. Nineteen essential oil among 22 assayed in this experiment were decreased in the expanded tobaccos. Key words : expanding method, tobacco, physico-chemical properties.
