양액재배시 배지 및 양액종류의 차이가 백경채, 탑채, 상추, 쑥갓 및 잎들깨의 생육에 미치는 영향을 구명하고자 실시한 실험의 결과는 다음과 같다. 1. 전반적으로 순수담액처리구와 혼합상토의 생육이 크게 차이나지는 않았으나 순수담액처리구의 생육이 가장 양호하였고, 모래배지에서의 생육이 가장 저조하였다. 백경채, 탑채, 상추와 잎들깨의 생육은 순수담액의 산기양액처리구에서 가장 좋았으며 쑥갓의 경우 양액종류에 관계없이 순수담액처리구에서 생육이 양호하였다. 2. 엽록소의 상대적 함량은 처리간에 큰 차이가 없었다. 3. Vitamin C의 함량은 처리구간에 차이가 크지 않았으나 공시작물 공히 전반적으로 양액의 종류에 상관없이 혼합상토처리구가 가장 높았고, 모래배지가 가장 낮았다. 그러나 잎들깨에서는 모래배지와 순수담액배지간에 차이가 없었다.
While many isolates of Alternaria alternata are common saprophytes on trees and shrubs, this study clearly demonstrated that A. alternata is a primary pathogen in lilac (Syringa sp.), causing a leaf-blight that affects different Syringa species. Isolates of Alternaria sp. were collected from leaf blight samples of lilacs in the field. The internal transcribed spacer (ITS) region and morphological characterization were used to identify lilac blight pathogen. Based on 100% ITS nucleotide sequence identities to the Alternaria genus in the GenBank and morphological features, these isolates were identified as A. alternata. Disease symptoms were reproduced in lilac plants inoculated with A. alternata mycelial plugs and sprayed with a fungus-free culture filtrate, indicating that pathogenesis in lilac involves secondary metabolites or toxins. Diagnostic primers were developed to detect Alternaria sp. and A. alternata in lilac leaf blight based on ITS region and four known genes associated with pathogenesis in A. alternata: mixed-linked glucanase precursor, endopolygalacturonase, hsp70, and histone genes. The results from our study indicated A. alternata is a primary pathogen in lilac leaf blight, and these diagnostic primers can be used as a tool for the fast detection of A. alternata associated with lilac leaf blight.
The effect of potassium metabolism on the soybean leaves was studied with comparison of other elements during the successive growing period. The results were as follows; 1. The percentage of potassium content showed remarkable increase not only in the first compound leaf at a stage which was growing vigorously and producing new leaves, but also in the fifth compound leaf at a stage which was taking a active metabolism of nitrogen and carbohydrate but not producing new leaves. However, the percentage of potassium content was decreased in the second compound leaf than in the first one. Such a result could be regarded as a potassium removal from mature leaves into immature and flowing out from stoma through respiration. During the pod-development the percentage of potassium content in the soybean leaf was decreased. 2. If nitrogen, phosphorus and potassium were added excessively in the nutrient solution, the percentage of potassium content in the soybean leaf had increased. The effects of these elements showed a remakable increase in the excessive plot of nitrogen than in that of phosphorus. At early stage the redtarded effect of phosphorus on the growth of soybean could be covered by potassium, however, at late stage it could not. The growth of soybean plant was much more inhibited by potassium, compared with nitrogen and phosphorus. New leaves could not be produced in the potassium deficient soybean plant after the third compound leaf. The normal growth of soybean plant could not be observed if only one element was excessively added to the culture solution, compared with the deficiency of other two elements.
In vitro elimination of Sweet potato leaf curl virus (SPLCV) from infected sweet potato is difficult due to low number of virus-free plants obtained from meristem tip culture and long growth period required for the virus detection. In this study, efficient production of the SPLCV-free sweet potato by in vitro therapy coupled with a PCR assay for virus detection was investigated. Infected shoots cultured on Murashige and Skoog medium were treated at three different temperatures for 7 weeks followed by meristem tip culture on the medium with or without ribavirin at 50 mg/L. The regenerated plantlets were tested for virus infection by a PCR assay. The results showed that the both heat- and cold-treatments, and addition of the ribavirin did not have significant effect on efficiency of the virus elimination. The meristem size, however, greatly affected the survival rate. Meristems sized over 0.4 mm survived better than smaller ones (0.2-0.3 mm). The PCR assay was approved to be a rapid, sensitive and reliable for the SPLCV detection in regenerated plantlets. Therefore, combination of cultivating meristem tips sized 0.4-0.5 mm on the medium at $22^{\circ}C$ without ribavirin and detection of SPLCV in the regenerated plantlets by the PCR assay was an efficient system for the SPLCV elimination from infected sweet potato.
In the present study, a simple one medium formulation protocol for callus culture, somatic embryogenesis and in vitro production of ${\beta}-carboline$ alkaloids and diosgenin in Tribulus terrestris L. was developed. Extensive callus induction and proliferation was obtained in leaf explant on Murashige and Skoog (MS) medium supplemented with $5.0{\mu}M$ 6 benzyl adenine (BA) and $2.5{\mu}M$${\alpha}-naphthaleneacetic$ acid (NAA). The embryogenic callus was maintained on subculture to fresh parental medium at 4-week intervals over a period of 28 months. The frequency of embryo formation was at a maximum ($18.1{\pm}0.9$ per g of callus) on MS medium containing $5.0{\mu}M$ BA and $2.5{\mu}M$ NAA together with $75mg\;1^{-1}$ casein hydrolysate. Globular embryo developed into torpedo stage embryo under the influence of starvation. The accumulation of ${\beta}-carboline$ alkaloids (harmaline and harmine) and steroidal saponin (diosgenin) in non-embryogenic and embryogenic callus culture derived from leaf explant was compared with root, leaf, stem, and fruit of the mother plant. The embryogenic callus accumulated equivalent amounts of harmaline ($66.4{\pm}0.5{\mu}g/g$ dry weight), harmine ($82.7{\pm}0.6{\mu}g/g$ dry weight), and diosgenin ($170.7{\pm}1.0{\mu}g/g$ dry weight) to that of the fruit of T. terrestris. The embryogenic callus culture of this species might offer a potential source for production of important pharmaceuticals.

Plantlet regeneration in Asteracantha longifolia(L.) Nees (Acanthaceae), a medicinal herb has been achieved from seedling explants on basal MS medium. Three different seedling explants including node, internode and leaf segments on used. Of these three explant, leaf explants gave better response for both callus mediated organogenesis and direct multiple shoot induction. Number of explants showing differentiation of shout buds was higher on MS media supplemented with BA compared to kinetin. MS medium fortified with BA ($2.0mgl^{-1}$) and NAA ($0.5mgl^{-1}$) was found to be most suitable for both callus mediated organogenesis and elongation of shouts. The elongated shoots were successfully routed on MS medium fortified with NAA or IBA. Among them $0.1mgl^{-1}$ NAA or $0.2mgl^{-1}$ IBA provides better response for rhizogenesis. Regenerated plantlets were successfully established in soil where 85.4% or them developed into morphologically normal and fertile plants. RAPD profiling using four decamer primers confirmed the genetic uniformity of the regenerated plantlets and substantiated the efficacy and suitability of this protocol for in vitro propagation of A. longifolia.
고추의 잎 절편을 BAP(2 mg/l)와 IAA(1 mg/l)가 첨가된 MS 배지에서 분화를 유도하여 40일 후에 완전한 식물체를 얻을 수 있었다. 또한 분화 단계에 따라 peroxidase와 esterase의 효소 활성과 isozyme 양상을 조사하였는데, 조직별 peroxidase 활성은 발아 10일째의 잎에서 가장 높았으며, isozyme 양상은 10일째에 잎에서 pI 7.2와 pI 6.9의 새로운 band가 나타났고, 14일째에는 잎과 뿌리에서 새로운 pI 3.4 band가, 줄기에서는 pI 5.2의 새로운 band가 나타나 peroxidase의 조직 특이적 발현을 알 수 있었다. 또한, 잎의 재분화가 진행됨에 따라 peroxidase 효소활성이 증가하기 시작하여 배양 14일째에는 대조구에 비해 80% 정도 높은 효소 활성을 나타냈으며, 분화가 진행됨에 따라 band의 양이 증가하며 배양 14일째에는 pI 3.2와 pI 6.5의 새로운 isozyme이 유도되어 peroxidase가 분화와 발생에 관여하고 있음을 시사하였다. 한편, esterase 활성은 14일째에 대조구보다 2배 이상 높은 활성을 보였으며 esterase isozyme 양상은 배양 7일째에 3개의 cathodic band와 1개의 anodic band가 나타남으로써, 이들 isozyme이 분화시기에 밀접하게 연관되었음을 알 수 있었다.
Agrobacterium tumefaciens를 매개로 한 형질전환 과정에서 제균제로 사용되고 있는 항생제인 carbenicillin이 담배(Nicotiana tabacum L. cv. BY-4) 잎절편 배양시 기관분화에 미치는 영향을 검토하였다. BAP와 kinetin을 각각 0.5 mg/L 첨가한 MS 배지에 250 mg/L∼2000 mg/L의 carbenicillin을 처리한 배지에서 신초 수는 크게 감소하였다. 또한 carbenicillin을 단독첨가하거나 0.5 mg/L의 2,4-D 또는 NAA와 혼합첨가한 MS 배지에서 250 mg/L∼500mg/L농도의 carbenlcillin은 캘러스 형성을 증가시켰다. 반면 carbenicillin을 0.5 mg/L의 2,4-B, NAA와 각각 혼합첨가한 배지에서 배양 4주째 치상조직의 생중량은 carbenicillin의 농도가 250mg/L에서 2000mg/L로 높아질수록 감소하였다. 이러한 결과는 carbenicillin이 캘러스 형성을 촉진하고 신초형성을 억제하는 auxin과 유사한 효과를 가지고 있다는 것을 시사해 준다. 따라서 담배 잎절편에 Agrobacterium tumefaciens를 매개로 형질전환하고자 할 때 최적 재분화용 식물생장조절제의 농도는 carbenicillin이 제균제뿐만 아니라 auxin 유사효과를 가지는 물질임을 고려하여 결정하여야 한다.
벼의 기공 분포 밀도에 미치는 규소의 영향을 구명코자 동계 온냉조절온실에서 규소 농도를 달리하여 수경재배한 벼 제 7 엽기의 엽신 부위별 기공수 및 기공세포 크기를 관찰한 결과 규소 농도 증가로 밀양2003 엽신중 기공수가 줄어들고 엽의 전 표면적에 대한 기공수$\times$기공세포 크기로 나타낸 기공세포의 분포 밀도가 낮게 되어 양면 표피의 cuticle-silica 2중층의 존재 이외에도 수분경제면에서 유리한 특성을 갖게 될 것으로 간주되었다.

In 2015, a new leaf spot disease was observed on strawberry seedlings in Wanju, Korea. Tanned brown spots appeared on the leaves of the infected plants, and often coalesced to form larger necrotic areas, resulting in the death of foliage. An isolate was obtained in pure culture. On the basis of morphological characteristics and molecular analysis of internal transcribed spacer rDNA sequence, the causal agent was identified as Pilidium concavum. Pathogenicity tests revealed the isolate was pathogenic to the leaves and fruits of strawberry. To the best of our knowledge, this is the first report of P. concavum causing tan-brown leaf spot on strawberry in Korea.
