씀바귀 부위별 용매추출 방법에 따른 항산화 활성 및 항염증 효과 비교 (Antioxidant and Anti-inflammatory Activities of Different Parts of Ixeris dentata According to Extract Methods)

  • 씀바귀(Ixeris dentata)의 잎과 줄기를 열수 및 70% 에탄올 추출하여 추출물의 농도별로 항산화 활성 및 항염증 효과에 미치는 영향을 검토하고자 실시하였다. 추출용매에 따른 총 flavonoid 및 polyphenol 함량은 씀바귀 잎과 줄기 모두 열수 추출물에 비해 70% 에탄올 추출물에서 높게 나타났다. DPPH 및 ABTS radical 소거활성은 씀바귀 잎의 70% 에탄올 추출물에서 열수 추출물에 비해 높게 나타났으나 줄기의 열수 추출물이 70% 에탄올 추출물보다 더 높은 활성을 보였다. 부위별 씀바귀 열수 추출물보다는 70% 에탄올 추출물에서 높은 NO 생성 억제 효과를 나타났다. 본 연구를 통하여 씀바귀의 부위별 추출용매에 띠라 우수한 생리활성에 차이가 있는 것으로 나타났으며 씀바귀를 이용하여 건강 기능성 소재로 활용할 수 있을 것으로 사료된다.

모링가 잎 열수 추출물을 첨가한 설기떡의 항산화, 이화학 및 관능 특성 (Antioxidation, Physicochemical, and Sensory Characteristics of Sulgidduck Fortified with Water Extracts from Moringa oleifera Leaf)

  • The aim of this investigation was to examine the antioxidation, physicochemical, and sensory activity of a Korean steamed-rice cake, Sulgidduk, fortified with water extracts from Moringa oleifera (M. oleifera) leaf. M. oleifera leaf extracts were added to rice powder at rations of 0.1%, 1% and 10%. To examine antioxidation properties, the scavenging activities of DPPH radicals, hydroxyl radicals, ABTS+ radicals, and ferric ion reducing antioxidant power were investigated. M. oleifera extracts significantly increased the antioxidation activities of Sulgidduk in a dose dependent manner (p<0.05). Physicochemical characteristics were measured by proximate composition, color, texture profile analysis, and sensory evaluations. As the concentration of M. oleifera leaf extracts increased, L-values and a-values significantly decreased while b-values increased. Texture profile analysis demonstrated that the control groups showed significantly higher values for hardness, cohesiveness, chewiness, and adhesiveness as compared with groups containing M. oleifera leaf extract (p<0.05). In the sensory evaluation, the sample containing 0.1% of M. oleifera leaf extract obtained the best results in overall preference. Taken together, these results suggest that M. oleifera leaf may have the potential to increase the consumer acceptability and the functionality of Sulgidduk.

인삼엽요병에서 효소활성도의 변화 (Study on the Enzyme Activity in Leaf-Burning Disease of Panax ginseng C.A. Meyer)

  • 인삼엽을 강광(100 KLuw) 및 고온($45^{\circ}C$, 암상태)에 처리하여 효소(glucose-6-phosphate dehydrogenase, acid phosphatase, catalase, peroxidase)의 활성도를 조사한 결과 두 처리구에서 공히 감소하는 경향이었으나, 특기 강광에서 활성도가 현저히 감소하였다. 이와 같은 활성도 감소는 효소의 thermal stabilities나 coagulation 등과 같은 광에 의한 2차적인 엽온상승 효과에 따른 inactivation이 아니며, proteolytic activity 증가로 인한 효소단백질의 함량감소로 확인되었다. 인삼엽에서 proteolytic activity가 강광에 의해 급속히 증가하는 것으로 보아 정상엽(normal leaf)에 inactive 상태로 내재(compartmentation)되어 있는 proteinase가 타 식물에 비해 많은 것으로 사료된다. 또한 chlorphyll bleaching과 효소의 inactivation을 유발시킬 수 있는 superoxide radical(${O_2}^{-}$)의 광화학적 생성율이 비교식물(Solanum nigrum)보다 높게 나타나고 crude saponin이 superoxide의 생성율을 촉진하는 것으로 보아 superoxide에 의한 pigment system의 광산화율이 타 식물에 비해 높을 것으로 사료된다.

Relationship of Nitrate Reductase Activity to Leaf Yield, Protein, Sugar and Physiological Attributes in Mulberry (Morus alba L.)

  • Ten improved mulberry varieties (Vl, C1730, C2016, C2017, Anantha, RFS-175, Thallaghatapura, Vishala, S1 and S1635) were evaluated through enzyme assay and estimation of soluble protein content followed by regression analysis, grown under irrigated conditions in the alluvial soils of Gangetic plains of West Bengal in India for five successive crops in a year, The nitrate reductase (EC No. 1.6.6.1) activity (NRA, $\mu$mol N $O_2$- $h^{-1}$ $g^{-1}$ fr, wt.), total soluble protein (mg $g^{-1}$ fr, wt.) was estimated which showed to vary significantly in the tested varieties. In addition to these, the other parameters like unit leaf fresh and dry weight (g), moisture %, unit leaf area ($\textrm{cm}^2$), specific leaf weight (g c $m^{-2}$ ), total soluble sugar (mg $g^{-1}$ fr, wt.), leaf yield/plant (kg), shoot yield/plant (kg) and net photosynthetic rate (NPR, $\mu$$m^{2}$ $s^{-1}$ ) were also studied which showed to vary significantly in tested varieties. Among them, S1635, haying higher NRA (13.25 $\mu$㏖ N $O_2$- $h^{-l}$ $g^{-1}$ fr, wt.), total soluble protein (39.63mg $g^{-1}$ fr, wt.), NPR(16.66 $\mu$$m^{-2}$ $s^{-1}$ ), total soluble sugar (48.44 mg $g^{-1}$ fr. wt.), leaf yield/plant (0.689 kg) and shoot yield/plant (1.135 kg) showed its superiority over other tested varieties. Regression and correlation coefficients were analysed, and a strong positive correlation was found between NRA & total soluble protein, NRA & NPR, NRA & total soluble sugar, NRA af unit leaf weight, NRA & specific leaf weight, NRA & leaf yield/plant, NRA & shoot yield/plant, NPR & leaf yield and NPR & specific leaf weight.t.

스테비아 잎 분말 첨가 오미자편의 품질 특성 및 항산화 특성 (Quality Characteristics and Antioxidant Properties of Omija-pyun (Schisandra chinensis Jelly) added with Stevia Leaf Powder)

  • This study added stevia leaf powder at ratios of 0.5, 1.0, 1.5, and 2.0 percent to Omija-pyun (Schisandra chinensis Jelly) as a natural low-calorie sweetener instead of sugar which is added to Omija-pyun in considerable amounts to evaluate quality characteristics and antioxidant activities compared to the control group with the addition of sugar. Moisture content of Omija-pyun expanded by increasing the measurement of stevia leaf powder (p<0.001), pH (p<0.01) and sugar content (p<0.001) decreased. L-values and b-values revealed a tendency to increase by adding more stevia leaf powder, but a-value revealed a tendency to decrease (p<0.001). Hardness (p<0.001) and chewiness (p<0.05) decreased by adding more stevia leaf powder. Based on the consumer preference evaluation, Omija-pyun with the addition of 0.5 percent stevia leaf powder was the most preferable in terms of color and flavor (p<0.001). The control group and Omija-pyun with the addition of 0.5 percent stevia leaf powder was the most preferable in terms of taste and texture (p<0.001). There were significant differences in the organoleptic properties except hardness between the samples by quantitative descriptive analysis. The control group revealed the highest preference in terms of redness and transparency, and redness and transparency tended to decrease by adding more stevia leaf powder (p<0.001). Organoleptic properties on bitterness and sourness were enhanced by adding more stevia leaf powder in the principal component analysis (PCA). Regarding antioxidant properties, total phenol compounds and flavonoid contents of Omija-pyun increased by adding more stevia leaf powder, and DPPH radical scavenging capacity also increased (p<0.001). Based on results, it is preferable to serve Omija-pyun with the addition of 1.0 percent stevia leaf powder instead of sugar within the context of quality and antioxidant activity.

In vitro cytotoxic activity of ginseng leaf/stem extracts obtained by subcritical water extraction

  • Ginseng leaf/stem extract produced by subcritical water extraction at high temperature ($190^{\circ}C$) posses higher cytotoxic activity against human cancer cell lines than ethanol extract. Subcritical water extraction can be a great candidate for extraction of functional substance from ginseng leaves/stems.

약용식물 메탄올 추출물의 항산화 및 항당뇨 활성 (Anti-Oxidative and Anti-Diabetic Effects of Methanol Extracts from Medicinal Plants)

  • 약용식물 23종 메탄올 추출물의 폴리페놀 함량을 측정한 후 항산화 활성을 측정하고, 항당뇨 활성으로는 ${\alpha}$-amylase 저해 활성, ${\alpha}$-glucosidase 저해 활성을 검정하여 혈당강하 활성을 나타낼 수 있는 약용식물을 선발하고자 한다. 약용식물의 폴리페놀 함량을 분석한 결과 1.50~15.25 mg GAE/g의 다양한 함량 범위로 나타났으며 가장 높은 함량은 주목열매(15.25 mg GAE/g), 화살나무줄기(15.12 mg GAE/g), 두충잎(14.24 mg GAE/g)으로 3종이 나타났다. 60% 이상 DPPH 라디칼 소거능을 본 약용식물로는 두충 줄기수피(80.10%), 구기자나무 뿌리(64.25%), 화살나무 줄기(73.59%), 비수리(78.20%), 주목열매(70.52%), 뽕잎나무 잎줄기(67.81%) 부위 추출물로 6종이 선발되었다. ${\alpha}$-Glucosidase를 80% 이상 저해한 약용소재로는 두충 줄기심재, 두충 줄기수피, 화살나무 줄기, 마, 율무, 녹두 6종이 선발되었다. ${\alpha}$-Amylase를 80% 이상 저해한 약용식물로는 칡뿌리, 두충 줄기수피, 두충잎, 구기자 열매, 화살나무 잎줄기, 화살나무 줄기, 조릿대 지상부, 마, 뽕잎나무 잎줄기 추출물 등 9종이 선발되었다. 항산화 및 항당뇨 활성이 우수했던 약용식물들은 향후 새로운 의약품 개발 및 성인병 예방의 건강기능식품으로 개발하는 데 있어 좋은 기초자료로 활용될 것으로 기대된다.

Phytotoxicity and DPPH Radical Scavenging Activity of Barley Seedling Extracts

  • A series of methanol extracts from leaf and root parts in spring- and winter-barley plants were assayed to determine their allelopathy and antioxidant activity. The methanol extracts applied on filter paper in a Petri-dish significantly inhibited root growth of Chinese milkvetch (Astragalus sinicus L.) seedlings. Leaf extracts at 25 and 50 g $L^{-1}$ inhibited root growth of Chinese milkvetch seedlings more than root extracts. No difference in phytotoxic effects of spring- and winter-barley seedlings extracts on root growth of Chinese milkvetch was observed. Methanol extracts dose-dependently increased DPPH free radical scavenging activity in vitro. DPPH free radical scavenging activity was higher in the methanol extracts from winter-barley seedlings than in those from spring-barley seedlings, and from leaf extracts than from root extracts. The antioxidant potential of the individual fraction from the methanol extracts of spring-barley seedlings was in order of n-butanol>ethyl acetate>water>chloroform>n-hexane fraction. By means of HPLC analysis, spring-barley (200.62 mg $100g^{-1}$) had more amount of total phenol acid than winter-barley (114.08 mg $100g^{-1}$). Especially, ferulic acid was detected in spring-barley extract (183.46 mg $100g^{-1}$) as the greatest amount. These results suggest that early seedlings of barley plants had potent allelopathy and antioxidant activity, and their activities were differently exhibited depending on plant parts and growing condition.

  • Using a flue-cured tobacco variety, BU 109, effect of growth regulators(Fatty Alcohol and C-MH) on the change of invertase activity and sugar metabolites were investigated. Invertase activity in untreated leaf tissue was decreased along with maturation of leaf, However, a momentary increase of the activity was observed in leaves by the dual treatment of fatty alcohol and C-MH regardless of leaf position while sole C-MH treatment resulted in activity increase by 14 days after the treatment. Similar tendency was observed in stalk. Sugar content in leaf was increased immediately after the treatment but no significant increase at large resulted until 14 days after treatment. After harvest, reducing sugar was increased by the growth regulators. Nevertheless, in case of dual treatment, the total sugar content was not different with that of untreated control. R/T ratio was gradually increased after topping stage and reached maximum at 14 days after treatment of growth regulators. It decreased a little after harvest but the RR ratio was relatively higher due to increase of reducing sugar resulted by the treatment of the chemicals. Upon treatment of growth regulators, reducing sugar content was increased in lugs and leaves compared to untreated control and the content of sugar metabolites was increased by the use of the chemicals either at lower or higher than recommended dose. 천연과즙을 이용한 Aloe vera L.의 callus 배양과 이들 추출물의 항산화 및 항균활성 효과 검정 (The Effect of Antioxidant and Antimicrobial Activity on the Extracted Its Material and Aloe vera L. Callus Culture by the Natural Fruit Juice)

  • In order to obtain functional materials from aloe callus, we cultured Aloe vera L. leaf on MS medium added 0.2 mg/L IAA, 0.3 mg/L kinetin and 100 mg/L grape or/and apple juice for 30 days. While a callus differentiation during callus culture did not show, the cultured leaves were uniquely released extracellular material into the agar plate. After cultivation for 18 days, the cultured leaf and agar were harvested for extraction a functional material. The materials extracted were measured on the amount of total phenols, flavonoids and polysaccharides and determined on the antioxidant and antimicrobial activity. In result, callus extracts of additive free (CT) and added apple juice (2T) had more amount of phenol compound ($659{\mu}g/mL$, $533{\mu}g/mL$) and flavonoid ($580{\mu}g/mL$, $501{\mu}g/mL$) than natural leaf (p: $525{\mu}g/mL$, f: $301{\mu}g/mL$). However, the extract of natural leaf had the better effect of lipid peroxidation and polysaccharide content than the culture extract. All samples extracted had same effect on the nitrite scavenging activity. On the other hand, only 2T extract showed excellent 72% nitric oxide scavenging activity. The agar extract was also confirmed to contain polyphenol compound and polysaccharide content that had antioxidant and antimicrobial activity partly.
