Expression level and glycan dynamics determine the net effects of TIMP-1 on cancer progression

  • Tissue inhibitor of metalloproteinases (TIMPs; TIMP-1, -2, -3 and -4) are endogenous inhibitor for matrix metalloproteinases (MMPs) that are responsible for remodeling the extracellular matrix (ECM) and involved in migration, invasion and metastasis of tumor cells. Unlike under normal conditions, the imbalance between MMPs and TIMPs is associated with various diseased states. Among TIMPs, TIMP-1, a 184-residue protein, is the only N-linked glycoprotein with glycosylation sites at N30 and N78. The structural analysis of the catalytic domain of human stromelysin-1 (MMP-3) and human TIMP-1 suggests new possibilities of the role of TIMP-1 glycan moieties as a tuner for the proteolytic activities by MMPs. Because the TIMP-1 glycosylation participate in the interaction, aberrant glycosylation of TIMP-1 presumably affects the interaction, thereby leading to pathogenic dysfunction in cancer cells. TIMP-1 has not only the cell proliferation activities but also anti-oncogenic properties. Cancer cells appear to utilize these bilateral aspects of TIMP-1 for cancer progression; an elevated TIMP-1 level exerts to cancer development via MMP-independent pathway during the early phase of tumor formation, whereas it is the aberrant glycosylation of TIMP-1 that overcome the high anti-proteolytic burden. The aberrant glycosylation of TIMP-1 can thus be used as staging and/or prognostic biomarker in colon cancer.

  • Jacques Derrida, a leading theorist of the post-structuralism, has explicated how the material reality is subordinated to the ideality of form, by noticing the platonic concept, 'chora', which implicates the spatiality as the matrix and also by actualizing its essential meaning as the femininity. On the basis of the idea of the platonic 'chora', the Derridian deconstructive spatiality and its feminist oriented enlargement of Grosz this study is purposed to elucidate the architectural idea of Zaha Hadid that is located in the similar horizon with the deconstructionism. On the one hand it is focused on the explication of the new concept of space, which is dealt with the deconstruction of the traditional concept of space, on the other on the inference of the feminity of the originally understood spatiality. With this inference it is finally intended to reexamine the foundation of the discussion for the architectural space that must be ahead of the discussion for the difference between the masculine and the feminine space. If it is fundamentally and manifoldly executed, not only the architectural paradigm will enter into an renewed phase, but also the various ways toward the new direction of the architecture be explored.

The Effect of a Long-Term Cyclic Strain on Human Dermal Fibroblasts Cultured in a Bioreactor on Chitosan-Based Scaffolds for the Development of Tissue Engineered Artificial Dermis

  • Mechanical stimulation is known to activate several cellular signal transduction pathways, leading to the induction of signaling molecules and extracellular matrix (ECM) proteins, thereby modulating cellular activities, such as proliferation and survival. In this study, primary human dermal fibroblasts (HDFs) were seeded onto chitosan-based scaffolds, and then cultured for 3 weeks in a bioreactor under a cyclic strain of 1 Hz frequency. Compared to control samples cultured under static conditions, the application of a cyclic strain stimulated the proliferation of HDFs in I week, and by week 3 the thickness of the cell/scaffold composites increased 1.56 fold. Moreover, immunohistochemical staining of the culture media obtained from the cell/scaffold samples subjected to the cyclic strain, revealed increases in the expression and secretion of ECM proteins, such as fibronectin and collagen. These results suggest that the preconditioning of cell/scaffold composites with a cyclic strain may enhance the proliferation of HDFs, and even facilitate integration of the engineered artificial dermal tissue into the host graft site.

  • 마이크론 크기의 입자를 분산하기에 적합하나 나노 크기의 입자를 분산하기에는 미흡한 이축압출기를 사용하여 카본나노튜브(CNT) 분산을 시도하였다. 카본나노튜브 분산성 향상을 위해 서로 다른 입자 형상을 가지는 무기 충전제를 부가하였다. 무기 충전제 첨가에 따른 점도 증가는 상대적으로 높은 전단력을 유도하여 응집된 카본나노튜브 입자의 분산을 도모하였다. 또한 충전제 입자들은 전단력이 미치지 않는 구역에서 일어나는 카본나노튜브의 재응집을 억제함으로써 분산 안정화에 기여하였다.

The stability of semi-rigid skeletal structures accounting for shear deformations

  • The analysis and design of skeletal structures is greatly influenced by the behaviour of beam-to-column connections, where patented designs have led to a wide range of types with differing structural quantities. The behaviour of beam-to-column connections plays an important role in the analysis and design of framed structures. This paper presents an overview of the influence of connection behaviour on structural stability, in the in-plane (bending) mode of sway. A computer-based method is presented for geometrically nonlinear plane frames with semi-rigid connections accounting for shear deformations. The analytical procedure employs transcendental modified stability functions to model the effect of axial force on the stiffness of members. The member stiffness matrix were found. The critical load has been searched as a suitable load parameter for the loss of stability of the system. Several examples are presented to demonstrate the validity of the analysis procedure. The method is readily implemented on a computer using matrix structural analysis techniques and is applicable for the efficient nonlinear analysis of frameworks. Combined with a parametric column effective length study, connection and frame stiffness are used to propose a method for the analysis of semi-rigid frames where column effective lengths are greatly reduced and second order (deflection induced) bending moments in the column may be distributed via the connectors to the beams, leading to significant economies.

  • Radon is one of the substances that pollute the indoor air and is classified as a first-level carcinogen by the International Agency for Research on Cancer(IARC) together with asbestos, and it is reported that it can cause lung cancer. The World Health Organization(WHO) reports that lung cancer is the second leading cause of lung cancer, and 6-15% of lung cancer patients report lung cancer caused by radon. Radon occurs in cracks in concrete and aged buildings, and is detected in soil, rocks, groundwater, and so on. It is a colorless, odorless and tasteless gas which is adsorbed to dust in the air and enters through human respiratory system. This study used vermiculite (expanded vermiculite), which has excellent ion exchange ability and a large number of pores, and industrial by - product red mud which has heavy metal adsorption ability, in order to adsorb radon. A matrix capable of adsorbing radon was prepared, and the characteristics of each material were compared and analyzed.

Novel Suppressive Effects of Ketotifen on Migration and Invasion of MDA-MB-231 and HT-1080 Cancer Cells

  • The high mortality rates associated with cancer reflect the metastatic spread of tumor cells from the site of their origin. Metastasis, in fact, is the cause of 90% of cancer deaths. Therefore, considerable effort is being made to inhibit metastasis. In the present study, we screened ketotifen for anti-migratory and anti-invasive activities against MDA-MB-231 breast cancer and HT-1080 fibrosarcoma cancer cells. Cancer cell migration and invasion were measured using multi-well chambers. Additionally, western blots were used to examine the effects of ketotifen on the expressions of CDC42, Rho, Rac, and matrix metalloproteinase 9 (MMP-9). The results showed that ketotifen dose-dependently suppressed the migration and invasion of MDA-MB-231 and HT-1080 cells. Ketotifen also suppressed the expressions of CDC42, Rac, and Rho, which, significantly, are involved in MDA-MB-231 and HT-1080 cancer cell migration. Moreover, ketotifen suppressed the expression and activity of MMP-9, which is involved in degradation of the extracellular matrix leading to invasion. The overall data suggested that ketotifen suppresses the migration and invasion of MDA-MB-231 and HT-1080 cancer cells via inhibition of CDC42, Rac, Rho, and MMP-9 expression.

Latent Semantic Analysis Approach for Document Summarization Based on Word Embeddings

  • Since the amount of information on the internet is growing rapidly, it is not easy for a user to find relevant information for his/her query. To tackle this issue, the researchers are paying much attention to Document Summarization. The key point in any successful document summarizer is a good document representation. The traditional approaches based on word overlapping mostly fail to produce that kind of representation. Word embedding has shown good performance allowing words to match on a semantic level. Naively concatenating word embeddings makes common words dominant which in turn diminish the representation quality. In this paper, we employ word embeddings to improve the weighting schemes for calculating the Latent Semantic Analysis input matrix. Two embedding-based weighting schemes are proposed and then combined to calculate the values of this matrix. They are modified versions of the augment weight and the entropy frequency that combine the strength of traditional weighting schemes and word embedding. The proposed approach is evaluated on three English datasets, DUC 2002, DUC 2004 and Multilingual 2015 Single-document Summarization. Experimental results on the three datasets show that the proposed model achieved competitive performance compared to the state-of-the-art leading to a conclusion that it provides a better document representation and a better document summary as a result.

초고온 세라믹스의 발전 동향 (Development of Ultra-High Temperature Ceramics)

  • ZrB2, ZrC, HfB2, HfC 및 SiC 등의 초고온 세라믹스(UHTC, ultra-high temperature ceramics)들은 최근 주목 받고 있는 극초음속 체계 및 차세대 우주왕복선 등에 적용하기 위하여 최근 활발한 연구가 진행되고 있다. 본 논문에서는 한국, 중국, 미국, 일본 및 EU 등지에서 최근 5년간 발표된 주요 연구 결과들을 위주로 간략히 소개하였다. 또한 초고온 세라믹스의 단점인 취성 문제를 극복하기 위한 세라믹 섬유강화 복합재료 제조를 목적으로 Horizon 2020 프로그램의 일환으로 범 유럽차원에서 공동연구로 수행된 C3HARME 프로젝트의 내용 및 결과를 간략히 보고하였다.

A spent nuclear fuel source term calculation code BESNA with a new modified predictor-corrector scheme

  • This paper introduces a new point depletion-based source term calculation code named BESNA (Bateman Equation Solver for Nuclear Applications), which is aimed to estimate nuclide inventories and source terms from spent nuclear fuels. The BESNA code employs a new modified CE/CM (Constant Extrapolation - Constant Midpoint) predictor-corrector scheme in depletion calculations for improving computational efficiency. In this modified CE/CM scheme, the decay components leading to the large norm of the depletion matrix are excluded in the corrector, and hence the corrector calculation involves only the reaction components, which can be efficiently solved with the Talyor Expansion Method (TEM). The numerical test shows that the new scheme substantially reduces computing time without loss of accuracy in comparison with the conventional scheme using CRAM (Chebyshev Rational Approximation Method), especially when the substep calculations are applied. The depletion calculation and source term estimation capability of BESNA are verified and validated through several problems, where results from BESNA are compared with those calculated by other codes as well as measured data. The analysis results show the computational efficiency of the new modified scheme and the reliability of BESNA in both isotopic predictions and source term estimations.
