Acrodontium burrowsianum and Pestalotiopsis humicola: Two Previously Unrecorded Fungal Species Isolated from Conifer Leaves in Korea

  • Endophytic fungal strains were isolated from the leaves of two conifer species (Juniperus rigida and Pinus densiflora) in Korea and identified on the basis of their morphological and molecular characteristics. Internal transcribed spacer and large subunit regions of rDNA were used for the phylogenetic analysis, and the translation elongation factor 1-alpha (TEF) and RNA polymerase II second largest subunit (RPB2) genes were analyzed depending on the species. Two fungal species that were previously unrecorded in Korea were identified: Acrodontium burrowsianum and Pestalotiopsis humicola. Their morphological and phylogenetic characteristics are described herein.

Delimitation of Russula Subgenus Amoenula in Korea Using Three Molecular Markers

  • Distinguishing individual Russula species has been difficult due to extensive phenotypic plasticity and obscure morphological and anatomical discontinuities. Due to highly similar macroscopic features, such as the presence of a red-cap, species identification within the Russula subgenus Amoenula is particularly difficult. Three species of the subgenus Amoneula have been reported in Korea. We used a combination of morphology and three molecular markers, the internal transcribed spacer (ITS), 28S nuclear ribosomal large subunit (LSU), and RNA polymerase II gene (RPB2), for identification and study of the genetic diversity of Russula subgenus Amoenula in Korea. We identified only two species in Korea (R. mariae and R. violeipes); these two species were indistinguishable according to morphology and LSU, but were found to be reciprocally monophyletic species using ITS and RPB2. The markers, ITS, LSU, and RPB2, have been tested in the past for use as DNA barcoding markers, and findings of our study suggest that ITS and RPB2 had the best performance for the Russula subgenus Amoneula.

머귀나무에서 녹병균 Coleosporium zanthoxyli의 확인 (Confirmation of Coleosporium zanthoxyli on Zanthoxylum ailanthoides in Korea)

  • 한국 학자들에 의해 머귀나무의 녹병균이 Coleosporium phellodendri로 보고되었으나, 추가적인 연구 없이 외국 학자들에 의해 이 녹병균이 C. zanthoxyli로 기록되었다. 한국에서 머귀나무 녹병균의 정체를 밝히기 위하여, 한국에서 채집되어 고려대학교 진균표본보관소에 보존된 8개 시료를 형태적으로 검경하고 대표적인 3개 시료의 internal transcribed spacer (ITS) 및 28S large subunit (LSU) rDNA 의 염기서열을 분석하였다. 그 결과, 한국의 머귀나무 녹병균은 형태적으로나 분자적으로 C. zanthoxyli로 동정하는 것이 옳다고 결정되었다. 따라서 이 연구를 통하여 머귀나무에 녹병균 C. zanthoxyli가 존재한다는 사실을 처음으로 보고한다.

  • 등수국은 수국과에 속하는 덩굴성 식물로 우리나라의 울릉도, 제주도, 남해안 섬에 분포한다. 2017년 10월 제주도에서 등수국에 발생하는 녹병균을 국내 최초로 발견하였다. 등수국 녹병균을 동정하기 위해, 형태적 특성을 조사하고 internal transcribed spacer (ITS) 및 28S large subunit (LSU) rDNA의 염기서열의 분자계통학적 분석을 수행하였다. 그 결과, 한국의 등수국 녹병균은 일본과 러시아에서 보고된 Pucciniastrum hydrangeae-petiolaris의 형태적 및 분자계통학적 특징과 일치하였으며, 북미에서 보고된 P. hydrangeae와는 상당히 먼 계통학적 관계를 보였다. 본 연구는 한국의 등수국에서 녹병균 P. hydrangeae-petiolaris의 첫 보고이다.

  • 때죽나무는 때죽나무과에 속하는 낙엽 소교목으로 우리나라와 중국, 일본, 대만이 원산이나 캐나다, 미국, 영국 등에도 도입되어 분포하고 있다. 1990년 이후 전국에 분포하는 때죽나무에서 녹병균이 지속적으로 발견되었다. 한국의 때죽나무 녹병균의 형태적 특성은 일본의 쪽동백나무에서 보고된 Pucciniastrum styracinum의 형태적 특성과 가장 유사하였으며 internal transcribed spacer (ITS) 및 28S large subunit (LSU) rDNA의 염기서열의 분자계통학적 분석을 통해 동정을 확인하였다. 본 연구에서는 한국의 때죽나무에서 발견한 녹병균 P. styracinum의 존재를 재확인하였고, 균학적 특성과 형태적 측정치 및 분자적 특성에 대한 정보를 제공하였다.

  • Skeletonema pseudocostatum의 세포는 규산질 성분의 돌기에 의한 사슬 형태로, 길이는 6~17.3 ㎛였고, 엽록체는 세포 당 1~2개를 포함하고 있었다. 주사전자현미경 관찰결과 Skeletonema 종을 구분할 수 있는 가장자리 받침돌기끝(terminal fultoportula process)은 끝이 갈라지거나, 갈고리 모양이었고, 길이가 1.67±0.5 ㎛이고, 개각의 가장자리에 위치해 있으며, 개수는 8.10±1.1개로 개각의 크기에 따라 다양하게 나타났다. 말단세포 입술돌기(terminal rimoportula process)는 두꺼운 원통형의 나팔관 모양으로, 개각의 중앙 근처에 위치하였고, 길이는 1.1±0.6 ㎛였으며, 개수는 1개였다. 연결세포 받침돌기(intercalary fultoportula process)는 대부분 1 : 1 결합으로 서로 맞물려 있는 형태였고, 1 : 2 결합도 종종 발견되었다. 계통분석 결과는 형태적 특징이 유사한 종 간의 유전학적 거리가 가깝다는 것을 나타냈고, 지리적 기원이 다른 동일 종의 경우, 유전적 차이를 보이지 않았다. 이는 S. pseudocostatum은 지리적 위치와 상관없이 유전적 차이가 나타나지 않는 단일계통(monophyly)이라는 것을 의미한다.

Fungal Diversity in Composting Process of Pig Manure and Mushroom Cultural Waste Based on Partial Sequence of Large Subunit rRNA

  • Fungal diversity during composting was investigated by culture-independent rDNA sequence analysis. Composting was carried out with pig manure and mushroom cultural waste using a field-scale composter (Hazaka system), and samples were collected at various stages. Based on partial sequence analysis of large subunit (LSU) ribosomal RNA (rRNA) and sequence identity values, a total of 12 different fungal species were found at six sampling sites; Geotrichum sp., Debaryomyces hansenii, Monographella nivalis, Acremonium strictum, Acremonium alternatum, Cladosporium sphaerospermum, Myriangium durosai, Pleurotus eryngii, Malassezia globosa, Malassezia restricta, Rhodotorula glutinis, and Fusarium sporotrichioides. Geotrichum sp. of the class Saccharomycetes was the most predominant fungal species throughout the composting process (185 out of a total of 236 identified clones, or 78.4%), followed by Acremonium strictum (7.6%), Monographella nivalis (5.1%), and Pleurotus eryngii (3.8%). The prevalence of Geotrichum sp. was the lowest (61.1%) at the beginning of composting, and then gradually increased to 92.5% after 10 days of composting.

Unreported Endophytic Fungi Isolated from the Leaves of Evergreen Woody Plants in Korea: Muyocopron lithocarpi and Didymocyrtis cladoniicola

  • In this study, endophytic fungi were isolated from the leaves of evergreen woody plants in Hansando Island in South Korea. The fungal isolates were identified based on morphological characteristics and phylogenetic analysis of the DNA sequences of the internal transcribed spacer and the large subunit regions of rDNA. It was confirmed that two endophytic fungal species, Muyocopron lithocarpi and Didymocyrtis cladoniicola, isolated in this study have not previously been reported in South Korea.

Report on Tuber huidongense, a Truffle Species Previously Unrecorded in Korea

  • The fruiting bodies and roots of a Tuber species were collected from the rhizosphere of Quercus dentata in Pohang, Korea. The morphological characteristics of the ascoma and ectomycorrhizal roots were studied, and phylogenetic analyses were performed using the sequences of the internal transcribed spacer, large subunit rDNA, and β-tubulin DNA. Based on the features of the fruiting bodies, the species was identified to be Tuber huidongense, which has not been reported earlier in Korea. The morphological characteristics and phylogenetic analysis of T. huidongense are described in the present study.

Two anthozoans, Entacmaea quadricolor (order Actiniaria) and Alveopora japonica (order Scleractinia), host consistent genotypes of Symbiodinium spp. across geographic ranges in the northwestern Pacific Ocean

  • The actiniarian sea anemone, Entacmaea quadricolor, and the scleractinian coral, Alveopora japonica, host symbiotic dinoflagellates belonging to the genus Symbiodinium (Freudenthal). We studied the host-symbiont specificity of these two anthozoan hosts in the northwestern Pacific Ocean. Symbionts within the two hosts were identified using partial large subunit (LSU) ribosomal DNA (rDNA) and complete internal transcribed spacers (ITS) 1 rDNA regions. The host, E. quadricolor, was identified using the partial LSU rDNA molecular marker. Genetic analysis showed that E. quadricolor only harbors dinoflagellates belonging to subclade C1/3 of the genus Symbiodinium. Moreover, no genetic variation was detected among the symbionts of E. quadricolor within the study region (Korea and Japan), even though the two distant sites were separated by more than 1000 km, at collection depths of 1 m in shallow and 13-16 m in deep water. Whilst scleractinian corals host multiple Symbiodinium clades in tropical waters, A. japonica, sampled over a wide geographical range (800 km) within the study region, only hosts Symbiodinium sp. clade F3. The high specificity of endosymbionts in E. quadricolor and A. japonica within the northwestern Pacific Ocean could be accounted for because symbiotic dinoflagellates within the host anemones appear to be acquired maternally, and the Kuroshio Current might affect the marine biota of the northwestern Pacific. However, the consistency of the symbiotic relationships between these two anthozoan hosts and their endosymbionts could change after climate change, so this symbiotic specificity should be monitored.
